The current study was conducted to characterize objective meat quality, fiber type and their relations to postmortem proteolysis in longissimus muscle of Landrace and Korean native black (KNP) pigs. Longissimus muscles from each 10 market-weighted male pigs were removed after conventional slaughtering and chilling procedures, and aged for 1 or 7 days at 4 °C to determine WB-shear force, objective meat color, proportion of myosin heavy chain I (MyHC I), intramuscular fat content and rate of proteolysis by a proteomics approach. KNP had a significantly (p < 0.05) higher content of MyHC I, and that concurred with greatly (p < 0.05) higher intramuscular fat content and Hunter a* value, and significantly (p < 0.05) lower drip loss than those seen in Landrace. One-dimension SDS-PAGE indicated that GAPDH, troponin I and creatine kinase were prominent proteolytic products during chiller ageing. By applying a gel-based proteome analysis, 26 proteins were identified, which showed different degradation properties during ageing between the breeds. Biopsied sample revealed that myosin regulatory light chain 2, myosin light chain isotype v/sb, fatty acid-binding protein and albumin were expressed at a greatly higher level for KNP, but their relation to fiber type (or genetic background) is unclear. It was particular noticeable that different actin isoforms showed various degradation behavior during ageing time.  相似文献

Drip loss increased with ageing while cooking loss was unchanged in both breeds. Shear force varied between animals and decreased with ageing. Analysis of the proteomes showed four types of temporal expression profiles. One factor affecting meat quality is pre-slaughter stress. We investigated the effects of exercise stress on drip loss and toughness in relation to resting times of 0, 1 or 3 h following exercise on a treadmill. This exercise stress was regarded as combined physical and physiological stress. Exercise stress increased the muscle temperature, reduced the creatine phosphate, ATP and glycogen content of pigs slaughtered immediately after stress exposure. These conditions lead to a reduced pH early post mortem and an increased drip loss, while only 1 h of rest after exercise stress normalised these effects. However, an overshooting effect was noted when pigs were rested for 1–3 h before slaughter, emphasising the importance of critical control of the resting period when studying exercise stress-induced effects on meat quality. Furthermore, meat from exercise stressed pigs, irrespective of resting, had increased toughness compared to controls, indicating that the toughness was not related to drip loss in meat from exercise stressed pigs.  相似文献

In this study, migration rate and spatial distribution of colloidal particles with and without proteolytic enzymes were studied after injection in pork loin muscle cubes for 5 days at 25 °C to simulate behavior of non-motile bacteria. Samples were monitored daily by light microscopy and confocal laser scanning microscopy and particle distribution and movement analyzed by image analysis (MATLAB®). The extent of proteolytic activity was assessed by determining the tyrosine content in muscle tissue. Results showed that particles diffused from the injection hot spot into the inner structure of the meat (1.35 ± 0.99 × 10 12 to 2.39 ± 2.02 × 10 12 m2/s). Upon addition of protease, the migration rate increased, i.e. the particle distribution became more widespread due to proteolysis of meat proteins reducing resistance to mass transfer. Results indicate that particles are able to diffuse into the densely packed fiber structure of meat muscles, which is contrary to the long held belief that such penetration may not occur in the absence of extensive proteolysis or mechanical damage of tissue.  相似文献

Meat high-hydrostatic pressure treatment causes severe decolouration, preventing its commercialisation due to consumer rejection. Novel procedures involving product freezing plus low-temperature pressure processing are here investigated. Room temperature (20 °C) pressurisation (650 MPa/10 min) and air blast freezing (−30 °C) are compared to air blast freezing plus high pressure at subzero temperature (−35 °C) in terms of drip loss, expressible moisture, shear force, colour, microbial quality and storage stability of fresh and salt-added beef samples (Longissimus dorsi muscle). The latter treatment induced solid water transitions among ice phases. Fresh beef high pressure treatment (650 MPa/20 °C/10 min) increased significantly expressible moisture while it decreased in pressurised (650 MPa/−35 °C/10 min) frozen beef. Salt addition reduced high pressure-induced water loss. Treatments studied did not change fresh or salt-added samples shear force. Frozen beef pressurised at low temperature showed L, a and b values after thawing close to fresh samples. However, these samples in frozen state, presented chromatic parameters similar to unfrozen beef pressurised at room temperature. Apparently, freezing protects meat against pressure colour deterioration, fresh colour being recovered after thawing. High pressure processing (20 °C or −35 °C) was very effective reducing aerobic total (2-log10 cycles) and lactic acid bacteria counts (2.4-log10 cycles), in fresh and salt-added samples. Frozen + pressurised beef stored at −18 °C during 45 days recovered its original colour after thawing, similarly to just-treated samples while their counts remain below detection limits during storage.  相似文献

In fresh meat production fast and non-destructive quality monitoring along the distribution chain is a key aspect to guaranteeing high quality and safe products for consumption. The applicability of fluorescence spectroscopy using protoporphyrins as indicators for meat ageing was investigated. Porcine musculus longissimus dorsi (MLD) was stored in slices over 20 days at 5 and 12 °C and measured every day with an excitation of 420 nm and an emission range of 550–750 nm. Additionally, pH, drip loss and colour were examined to assess possible correlations.The obtained spectra of the MLD showed an increase in three peaks at 592, 638 and 705 nm which could be reconstructed using the spectra of standard solutions of protoporphyrin IX (PP) and zinc protoporphyrin IX (ZnPP) or magnesium protoporphyrin (MgPP), respectively.Using principal component analysis (PCA) on the fluorescence spectral data, the meat slices stored at 5 °C showed differences in the fluorescence signal after the 10th day and 5th day when stored at 12 °C. An interrelationship between the additional analyses and the fluorescence intensities on these relevant days could not be established.In conclusion, the increase of ZnPP fluorescence due to temperature related changes of physiological meat properties is capable of serving as a quality indicator with regards to inadequate conditioning (e.g. during transportation and/or storage) of pork meat.  相似文献

The use of dielectric spectra to detect quality defects in porcine muscle during postmortem period was evaluated. The changes in dielectric spectra during meat ageing were also studied. For these purposes, dielectric spectra were measured by using an Agilent 85070E Open-ended Coaxial Probe connected to an Agilent E8362B Vector Network Analyzer from 500 MHz to 20 GHz parallel and perpendicular to muscle fiber direction. This study reveals that dielectric properties at two punctual frequencies (0.5 GHz and 10 GHz) are a useful tool to determine meat quality classes (RFN, PSE, and DFD) soon (6 h) after slaughter. Based on these results, it is possible to develop control algorithms in order to discriminate low quality pork meats and to decide their best uses. Moreover, the evolution of dielectric spectra during meat ageing showed important variations only in DFD meats, while in PSE and RFN meats the variations were observed only at some punctual frequencies of the spectrum during the 24 h after slaughter.  相似文献

The effect of post mortem temperature treatment on suckling lamb carcass and meat quality was study. Conventional (2 °C for 24 h), ultra-rapid (− 20 °C for 3.5 h, 2 °C until 24 h) and slow chillings (12 °C for 7 h, 2 °C until 24 h) were compared. Total viable counts (TVC), weight losses, and pH and temperature falls were recorded on carcasses. Meat colour, water holding capacity (WHC), Warner-Bratzler shear force, sarcomere length and sensory analysis were evaluated in M. longissimus. Ultra-rapid treatment reduced TVC and weight losses. The pH decline was faster in slow chilled carcasses than in faster chilled carcasses. No significant differences were found for colour and WHC. Slow treatment carcasses showed significantly lower shear force and higher sarcomere length. In the sensory analysis, tasters also rated the early post mortem slow-treated meat as more tender, less fibrous and chewy. Therefore, delay chilling in suckling lamb carcasses made it possible to obtain meat with better organoleptic characteristics, without affecting weight loss or hygienic quality.  相似文献

The effect of, and associations between, loin muscle morphology and pork quality indicator traits were assessed at three cooked temperatures in loin chops from 38 purebred Berkshire and 52 purebred Landrace swine. Three loin chops from each pig were randomly assigned to cooked temperature treatments of 62, 71, or 79 °C and loin tenderness was assessed as Warner–Bratzler shear force (WBSF). Cooked temperature (P < 0.001), breed (P < 0.001) and breed × cooked temperature (P < 0.001) effects influenced loin chop WBSF, whereby WBSF increased as cooked temperature increased. Chops from Landrace pigs had greater WBSF at each cooked temperature compared with chops from Berkshire pigs. Chops from Landrace pigs became less tender with increasing cooked temperature, whereas chops from Berkshire pigs became less tender only when cooked to 79 °C. In loins from Landrace pigs, Minolta a1 at 62 °C (R2 = 0.07), and average muscle fiber diameter at 71 °C and 79 °C (R2 = 0.07 and 0.24, respectively), contributed to WBSF variation. In contrast, for loins from Berkshire pigs, loin ultimate pH and intramuscular fat percentage accounted for 27% and 30% of the variation in WBSF at 62 °C and 71 °C, respectively, and loin ultimate pH accounted for 7% of variation in WBSF at 79 °C. Results suggest that loins from Berkshire pigs have properties that resist toughening at greater cooked temperatures and that associations between quality measures and loin tenderness differ between Landrace and Berkshire pigs.  相似文献

This study aimed to determine how small heat shock proteins (sHSPs) protect myofibrillar proteins from μ-calpain degradation during ageing. Immunoprecipitation experiments with M. longissimus dorsi (LD) from Angus heifers (n = 14) examined the interaction between αβ-crystallin, desmin, titin, HSP20, HSP27 and μ-calpain. Results showed that αβ-crystallin associated with desmin, titin, HSP20, HSP27 and μ-calpain. Exogenous αβ-crystallin reduced desmin and titin degradations in myofibrillar extracts and attenuated μ-calpain activity. In a second experiment, bull LD (n = 94) were aged at − 1.5 °C for up to 28 days post mortem. μ-Calpain autolysed faster in high ultimate pH (pHu) meat (pHu  6.2) and this was concomitant with the more rapid degradation of titin and filamin in this pHu group. Desmin stability in intermediate pHu meat (pHu 5.8 to 6.19) may be due to the protection of myofibril-bound sHSPs combined with the competitive inhibition of μ-calpain by sHSPs.  相似文献

The objective of the present study was to elucidate the physicochemical changes and protein oxidation of porcine longissimus muscle as influenced by different thawing methods. Five kinds of thawing methods, comprising of refrigerator thawing (RT, 4 °C), ambient temperature thawing (AT, 20 °C), water immersion thawing (WT, 14 °C), lotic water thawing (LT, 9 °C), and microwave thawing (MT), were used. There were significant effects on the porcine meat quality due to different thawing methods. RT had the least quality loss and the physicochemical characteristics of pork were closer to fresh muscle than the other thawing methods. MT significantly increased thawing loss, cooking loss, cutting force, carbonyl content, and TBARS (thiobarbituric acid-reactive substances) value, but decreased a* value and Ca-, K-ATPase activities (P < 0.05). Microstructural changes in experimental muscle showed that MT induced visibly larger gap between muscle fibers and tore more muscle fiber bundles compared to the other thawing methods. The reduction of Ca- and K-ATPase activities (P < 0.05) of myofibrillar protein was consistent with the increases in carbonyl content and TBARS value (P < 0.05). The results demonstrated all the thawing methods could cause porcine lipid and protein oxidation. Gel electrophoresis patterns of porcine muscle displayed that different thawing methods did not induce obvious protein aggregates and fragments.  相似文献

Chilled meat exportation comprises chilling within 48 h post-mortem to temperatures < 0 °C without freezing and holding under these conditions for several weeks. The effects of this ageing on sensory quality of pork are unknown and hence the objective of this study was to compare the sensory quality of Canadian pork as found in an export (Japan) market and locally. Regardless that the Japanese market's quality criteria were met, pork sorted on-line differed (P < 0.05) from that for the domestic market only for lightness, exudate and cooking loss; no differences in intramuscular fat content were observed. Overall, a trained panel scored weaker pork and meat flavours and odours in the export than the domestic pork as a result of either the quality by selection if roasted or the ageing (− 1.7 °C, 43 days exported chilled or 3.1 °C, 5 days domestic) if grilled or shabu shabu. Grilled pork was also more tender, sweeter and had stronger caramel flavour with the chilled ageing.  相似文献

The objective of this study was to determine effects of pre rigor stretching on beef tenderness development. Beef loins (M. longissimus dorsi; n = 24) were assigned to either stretching or non-stretching treatments and aged for 14 days at − 1.5 °C. Sarcomere length, shear force, water-holding capacity and proteolysis were determined for the loins at 1, 7 and 14 days of ageing. Stretching increased the length of the loins by 33% resulting in a trend of increasing sarcomere length (P = 0.19). However, no significant differences for shear force values, purge, drip and cooking losses between treatments were found. Shear force values decreased with ageing times (P < 0.05). Western blot assay found that stretching did not affect the extent of desmin degradation. The findings from the present study suggest that pre rigor stretching with the current increase in length will not contribute to tenderness improvement of the beef loins.  相似文献

Restructured meat is made by binding individual pieces of meat together. To study the effect of mechanical work on the meat binding process, a standard cooking protocol must be established. This paper details the establishment of a standard cooking protocol for the cooking process using untreated beef semitendinosus muscle. The effect of different cooking temperatures and applied loads during cooking were investigated. Meat samples were cut in 20 mm cubes size from beef semitendinosus muscle and two pieces were held together with the muscle fibre parallel to each other by wrapping them with a plastic food wrap. Then the samples were placed inside square steel tubes, that act as a mold for cooking, and different weights (0, 250, 500, 750 and 1000 g) were placed on top of the meat cubes during cooking. The temperatures used for cooking were 60, 70 and 80 °C. There was a significant temperature effect, with increases noted between 60 °C and 70 °C and between 70 °C and 80 °C. At 60 °C neither myosin nor collagen has gelatinized, leading to low binding strengths. At 70 °C the myosin component will have gelatinized. At 80 °C the collagen component will be contributing to the bond. To keep the collagen effect to a minimum the meat should be cooked at 70 °C. The effect of applied cooking load was significant at all cooking temperatures once sufficient load had been applied against no load to ensure good contact at the joint. There was a significant effect of applied load noted at 80 °C with increases up to 750 g and a drop occurring between the 750 and 1000 g loadings. The drop has been attributed to collagen being squeezed out of the joint as a bead of white material was noted around the joint.  相似文献

The aim of the study was to quantify the effect of meat quality characteristics and some processing conditions on the softness of dry-cured biceps femoris (BF) muscles. The BF muscles were dissected from forty hams and classified according to their pHBF into three groups: LpH (pH < 5.66), MpH (5.66  pH  6.00) and HpH (pH > 6.00). BF muscles within each pHBF group were distributed into three different Salting levels (1%, 2% or 4% of added NaCl). Muscles were salted, vacuum-packed and stored at 3 °C for 30 days. The post-salting BF muscles were classified into two intramuscular fat (IMF) levels: Low (IMF < 4%) and High (IMF  4%). Thereafter, the muscles were divided into two pieces and dried at two of the three different Drying levels (1.5, 2 and 2.5 g H2O/g desalted dry matter). Then, each piece was divided into two samples that were packed in N2 and stored at 5 °C or 30 °C for 1 month. Stress Relaxation was used to evaluate texture. Dry-cured BF muscles with initial pH > 6.0, with IMF > 4% or with added NaCl levels less than 2% were more prone to show soft texture. Softness in dry-cured muscles can be reduced by applying an ageing temperature of 30 °C for 30 days, despite increasing proteolysis. The softness reduction by ageing at 30 °C compared with 5 °C is expected to be higher when applied to drier samples, which show a smaller increase in proteolysis.  相似文献

Impact of fresh ham quality on finished ham product characteristics was evaluated. Bone-in hams destined for spiral-sliced ham manufacturing were sorted into two pH groups before processing: pH  5.5 and pH  5.6. For boneless hams, raw materials were sorted into groups with different levels of pale, soft, and exudative (PSE) product before manufacturing into sliced vacuum packaged hams: “Low PSE” (⩽5% PSE muscle), “Intermediate PSE” (20–30% PSE muscle) or “High PSE” (40–60% PSE muscle). Few differences were observed between the pH  5.5 and pH  5.6 groups in objective color measures and drip loss in bone-in spiral-sliced hams stored under refrigeration, however, after frozen storage, hams from the pH  5.5 group had lower L*- and a*-values and had much higher drip loss than those from the pH  5.6 group. Processing yields for bone-in spiral-sliced hams were similar through cooking and chilling, however, the pH  5.6 group had higher yields after slicing. For boneless hams, defects occurred at a greater frequency in hams formulated with a greater percentage of PSE raw materials than those with lower amounts of PSE. Differences in objective color measures and purge were minimal over the duration of storage time, but hams formulated with greater percentages of PSE raw materials were lighter in appearance and had less redness. Consumers gave lower color responses for hams formulated with “High PSE” amounts, but did not differentiate between hams manufactured with lower quantities of PSE muscle. However, when consumers directly compared packages of ham, there was distinct discrimination against hams manufactured with greater amounts of PSE. Purchase intent showed that consumers favored ham manufactured from fresh ham muscles containing low quantities of PSE tissue. Further research is needed to determine the optimal ratio of allowable PSE product in formulation that enables processors to maximize consumer appeal with the economic realities of sorting out PSE pork.  相似文献

The influence of different cooking treatments on tenderness and cooking loss, as main quality characteristics of chicken breast meat, was investigated. Industrial skinless chicken breast meat samples were designated as raw and marinated and cooked in the oven by hot air and hot air-steam mixture at 130, 150 and 170 °C, for 4, 8 and 12 min. Cooking losses were evaluated by weight changes before and after cooking, and tenderness changes were determined on cooked samples by measuring shear force using instrumental texture analysis. Results showed that marination, followed by air-steam cooking is the best combination to obtain the most tender chicken breast slices. The time and temperature of cooking showed similar effects on cooking loss and tenderness: short cooking time (4 min) and temperatures of 130–150 °C resulted in lower cooking losses and best meat tenderness, in both not marinated and marinated meat. Statistically significant correlations between tenderness and cooking loss indicated that the cooking loss correlated better with cooking time than with cooking temperature. An opposite phenomenon was observed for meat tenderness.  相似文献

The objective of this study was to investigate beef quality of longissimus muscle after ageing in dry ageing bags, traditional dry ageing or vacuum for 8 or 19 days. Lower ageing weight loss, odour score and microbial growth were found in meat aged in dry ageing bags than after traditional dry ageing. The sensory panel detected no differences for most of the sensory attributes between samples using the two dry ageing methods, except for the odour of the cutting surface. The dry-aged steaks had more umami and butter fried meat taste compared with vacuum-aged steaks. Ageing time affected most of the sensory traits in this study, which improved as ageing time increased from 8 to 19 days. In a consumer test, meat aged for 21 days in dry ageing bags was preferred than the samples aged in vacuum. This may be due to the higher tenderness and juiciness obtained during storage in dry ageing bags than meat aged in vacuum.  相似文献

The amount of nitrite in sliced-pastirma made, from fresh or frozen (which was stored at −18 °C for 240 days and then thawed at 10 °C for 24 h) M. Longissimus dorsi muscle was determined. Sliced-pastirma samples were stored in modified atmosphere packages (50% N2 + 50% CO2) at 4 and 10 °C for 150 days, and the amount of residual nitrite was measured after 0, 30, 60, 90, and 150 days of storage. The residual nitrite of pastirma samples made with frozen/thawed meat was higher than that of the pastirma made from fresh meat at both 0 day and at the end of the storage (150 days). The storage temperature (p < 0.01), storage period (p < 0.01) and the storage period × the storage temperature interaction (p < 0.01) had significant effects on the amount of the residual nitrite.  相似文献
