Hydrogen productivities of different photosynthetic bacteria have been searched on real thermophilic dark fermentation effluents (DFE). The results obtained with potato steam peels hydrolysate (PSP) DFE were compared to glucose DFE. Photobiological hydrogen production has been carried out in indoor, batch photobioreactors using several strains of purple non-sulfur (PNS) bacteria such as Rhodobacter capsulatus (DSM1710), Rhodobacter capsulatus hup- (YO3), Rhodobacter sphaeroides O.U.001 (DSM5864), Rb. sphaeroides O.U.001 hup- and Rhodopseudomonas palustris.The efficiency of photofermentation depends highly on the composition of the effluent and the PNS bacterial strain used. Rb. sphaeroides produced the highest amount of hydrogen on glucose DFE. Rb. capsulatus gave better results on PSP DFE. This study demonstrates that photobiological hydrogen production with high efficiency and productivity is possible on thermophilic dark fermentation effluents. Consequently, a sequential operation of dark fermentation and photofermentation is a promising route to produce hydrogen, and it provides a higher hydrogen yield compared to single step processes.  相似文献

Mixed bacteria were used to improve hydrogen yield from cassava starch in combination of dark and photo fermentation. In dark fermentation, mixed anaerobic bacteria (mainly Clostridium species) were used to produce hydrogen from cassava starch. Substrate concentration, fermentation temperature and pH were optimized as 10.4 g/l, 31 °C and 6.3 by response surface methodology (RSM). The maximum hydrogen yield and production rate in dark fermentation were 351 ml H2/g starch (2.53 mol H2/mol hexose) and 334.8 ml H2/l/h, respectively. In photo fermentation, immobilized mixed photosynthetic bacteria (PSB, mainly Rhodopseudomonaspalustris species) were used to produce hydrogen from soluble metabolite products (SMP, mainly acetate and butyrate) of dark fermentation. The maximum hydrogen yield in photo fermentation was 489 ml H2/g starch (3.54 mol H2/mol hexose). The total hydrogen yield was significantly increased from 402 to 840 ml H2/g starch (from 2.91 to 6.07 mol H2/mol hexose) by mixed bacteria and cell immobilization in combination of dark and photo fermentation.  相似文献

In this study we described the isolation of eight new strains of purple non-sulfur bacteria resistant to salinity ≥30 g L−1 and high concentration of VFAs (200 mM). These strains were characterized by their general physiological properties and the occurrence of hupSL genes. Some correlation was observed between the rate of H2 photoproduction, the absence of hupSL genes and hydrogenase activity. Two fast-growing strains without hupSL genes showed high nitrogenase activity and hydrogen accumulation during growth on Ormerod medium. These strains were capable of H2 photoproduction using non-treated dark culture (75% in water) after dark fermentation of starch at 30 g L−1, unlike control strains, Rhodobacter capsulatus B10 and Rb. sphaeroides GL. New N7 and 13 strains identified as Rb. sphaeroides can be recommended for application in a two-stage H2 production system.  相似文献

Hydrogen gas production from sugar solution derived from acid hydrolysis of ground wheat starch by photo-fermentation was investigated. Three different pure strains of Rhodobacter sphaeroides (RV, NRLL and DSZM) were used in batch experiments to select the most suitable strain. The ground wheat was hydrolyzed in acid solution at pH = 3 and 90 °C in an autoclave for 15 min. The resulting sugar solution was used for hydrogen production by photo-fermentation after neutralization and nutrient addition. R. sphaeroides RV resulted in the highest cumulative hydrogen gas formation (178 ml), hydrogen yield (1.23 mol H2 mol−1 glucose) and specific hydrogen production rate (46 ml H2 g−1 biomass h−1) at 5 g l−1 initial total sugar concentration among the other pure cultures. Effects of initial sugar concentration on photo-fermentation performance were investigated by varying sugar concentration between 2.2 and 13 g l−1 using the pure culture of R. sphaeroides RV. Cumulative hydrogen volume increased from 30 to 232 ml when total sugar concentration was increased from 2.2 to 8.5 g l−1. Further increases in initial sugar concentration resulted in decreases in cumulative hydrogen formation. The highest hydrogen formation rate (3.69 ml h−1) and yield (1.23 mol H2 mol−1 glucose) were obtained at a sugar concentration of 5 g l−1.  相似文献

Hydrogen production by suspended culture, entrapped culture of Rhodobacter sphaeroides and Clostridium butyricum, and entrapped co-culture of these two bacteria in the presence of NH_4+ was investigated and compared. Hydrogen production by suspended culture of Rhodobacter sphaeroides was most affected by NH_4+ among the four cultures studied. Nevertheless, hydrogen production rate was the highest after the NH_4+ was removed metabolically. Hydrogen production by entrapped culture of Rb. sphaeroides was not readily inhibited by NH_4+ compared with that by suspended culture, and the hydrogen production rate was also kept high. Hydrogen production by entrapped culture of C. butyricum was not inhibited by NH_4+ in the initial period of time, but NH_4+ had some influence on hydrogen production by this culture in the later period. The hydrogen production rate from this culture was low compared with that from the culture of Rb. sphaeroides as expected theoretically. The former was only 1 of 5.6 of the latter, on average. The pattern of NH_4+ effect on the hydrogen production by co-culture of these two bacteria was similar to that by the single culture of C. butyricum. Nevertheless, hydrogen production by co-culture was enhanced by 160% compared with that by the single culture of anaerobic bacterium.  相似文献

Continuous combined fermentation of ground wheat starch was realized in an annular-hybrid bioreactor (AHB) for hydrogen gas production. A mixture of pure cultures of Clostridium beijerinkii (DSMZ-791) and Rhodobacter sphaeroides-RV were used as seed cultures in combined fermentation. The feed contained 5 g L−1 ground wheat with some nutrient supplementation. Effects of hydraulic residence time (HRT) on the rate and yield of hydrogen gas formation were investigated. Steady-state daily hydrogen production decreased but, hydrogen yield increased with increasing HRT. The highest hydrogen yield was 90 ml g−1 starch at HRT of 6 days. Hydrolysis of starch and fermentation of glucose to volatile fatty acids (VFA) were readily realized at all HRTs. However, slow conversion of VFAs to H2 and CO2 by photo-fermentation caused accumulation of VFAs in the medium. Specific and volumetric rates of hydrogen formation also decreased with increasing HRT. High hydrogen yields obtained at high HRTs are due to partial fermentation of VFAs by Rhodobacter sp. The system should be operated at HRTs longer than 5 days for effective hydrogen gas formation by the dark and photo-fermentation bacteria.  相似文献

Ground wheat powder solution (10 g L−1) was subjected to combined dark and light fermentations for bio-hydrogen production by fed-batch operation. A mixture of heat treated anaerobic sludge (AN) and Rhodobacter sphaeroides-NRRL (RS-NRRL) were used as the mixed culture of dark and light fermentation bacteria with an initial dark/light biomass ratio of 1/2. Effects of wheat starch loading rate on the rate and yield of bio-hydrogen formation were investigated. The highest cumulative hydrogen formation (CHF = 3460 ml), hydrogen yield (201 ml H2 g−1 starch) and formation rate (18.1 ml h−1) were obtained with a starch loading rate of 80.4 mg S h−1. Complete starch hydrolysis and glucose fermentation were achieved within 96 h of fed-batch operation producing volatile fatty acids (VFA) and H2. Fermentation of VFAs by photo-fermentation for bio-hydrogen production was most effective at starch loading rate of 80.4 mg S h−1. Hydrogen formation by combined fermentation took place by a fast dark fermentation followed by a rather slow light fermentation after a lag period.  相似文献

The microbial community structure of thermophilic mixed culture sludge used for biohydrogen production from palm oil mill effluent was analyzed by fluorescence in situ hybridization (FISH) and 16S rRNA gene clone library techniques. The hydrogen-producing bacteria were isolated and their ability to produce hydrogen was confirmed. The microbial community was dominated by Thermoanaerobacterium species (∼66%). The remaining microorganisms belonged to Clostridium and Desulfotomaculum spp. (∼28% and ∼6%, respectively). Three hydrogen-producing strains, namely HPB-1, HPB-2, and HPB-3, were isolated. 16S rRNA gene sequence analysis of HPB-1 and HPB-2 revealed a high similarity to Thermoanaerobacterium thermosaccharolyticum (98.6% and 99.0%, respectively). The Thermoanaerobacterium HPB-2 strain was a promising candidate for thermophilic fermentative hydrogen production with a hydrogen yield of 2.53 mol H2 mol−1hexose from organic waste and wastewater containing a mixture of hexose and pentose sugars. Thermoanaerobacterium species play a major role in thermophilic hydrogen production as confirmed both by molecular and cultivation-based analyses.  相似文献

Eight natural microbial consortia collected from different sites were tested for dark, hydrogen production during starch degradation. The most active consortium was from silo pit liquid under mesophilic (37 °C) conditions. The fermentation medium for this consortium was optimized (Fe, NH4+, phosphates, peptone, and starch content) for both dark fermentation and for subsequent purple photosynthetic bacterial H2 photoproduction [Laurinavichene TV, Tekucheva DN, Laurinavichius KS, Ghirardi ML, Seibert M, Tsygankov AA. Towards the integration of dark and photo fermentative waste treatment. 1. Hydrogen photoproduction by purple bacterium Rhodobacter capsulatus using potential products of starch fermentation. Int J Hydrogen Energy 2008;33(23):7020–26], in the presence of the spent dark, fermentation effluent. The addition of Zn (10 mg L−1), as a methanogenesis inhibitor that does not inhibit purple bacteria at this concentration, also did not inhibit dark, fermentative H2 production. The influence of various fermentation end products at different concentrations (up to 30 g L−1) on dark, H2 production was also examined. Added lactate stimulated, but added isobutyrate and butanol strongly inhibited gas production. Under optimal conditions the fermentation of starch (30 g L−1) resulted in 5.7 L H2 L−1 of culture (1.6 mol H2 per mole of hexose) with the co-production mainly of butyrate and acetate.  相似文献

Biohydrogen produced from cellulosic feedstock is a promising candidate as a clean energy carrier to meet future energy needs. This work identified a functional consortium that can produce biohydrogen from three cellulosic feedstocks, the Whatman No. 2 filter, xylan, and Chlorella cells, using a novel dilution-to-stimulation approach. By serially diluting an anaerobic inoculum, biohydrogen production increased, peaked at a specific dilution ratio, and then declined. With filter paper tests at 10−9 dilution, the functional consortium was mainly composed of Clostridium sp. clone UC61. At dilutions greater than 10−9, the presence of other heterotrophic strains diminished the ability of consortium to produce hydrogen. In the xylan tests, Clostridium sp. clone UC61, Clostridium sp. BN2, and Bacteroides species comprised the functional consortium. Klebsiella sp. competed with Clostridium sp. for carbon and decreased hydrogen production. In the Chlorella cells tests, Bacillus, Acinetobacter, Elusimicrobium, and Clostridium comprised the functional consortium. The loss of Bacillus and Acinetobacter species under high dilution deteriorated the synergistic relationships among different strains. The proposed dilution-to-stimulation approach is effective for isolating functional strains for simultaneous hydrolysis and hydrogen production by cellulosic materials.  相似文献

Purple non-sulfur (PNS) bacteria can convert volatile fatty acids into hydrogen with a high substrate conversion efficiency. However, when PNS bacteria utilize sugars as a carbon source, such as glucose and sucrose, the substrate conversion efficiency is relatively low. In order to investigate the contributions of the glucose catabolic pathways in Rhodobacter sphaeroides 6016 to its hydrogen production, the cfxA gene from the Embden–Meyerhof–Parnas (EMP) pathway, edd from the Entner–Doudoroff (ED) pathway, and kdg from the semi-phosphorylative ED bypass were knocked out to construct the mutant strains edd, cfxA, and kdg, respectively. Additionally, two of these three genes were knocked out to construct the mutant strains kdgedd, kdgcfxA, and cfxAedd. Hydrogen productions by these mutant strains were compared to that of the wild type strain 6016 using 25 mM glucose as a carbon source. Compared to 6016, variations in hydrogen production and growth were detected in the edd mutant strains (kdgedd, cfxAedd, and edd), while no obvious changes were detected in the others. Notably, the kdgedd mutant did not produce hydrogen, and its maximum growth was 70% less than that of R. sphaeroides 6016. These results indicate that the ED pathway and semi-phosphorylative ED bypass have a governing impact on cell growth and hydrogen production from glucose in R. sphaeroides 6016. The potential synergistic function of the ED pathway and semi-phosphorylative ED bypass and the reasons for the low hydrogen yield from sugar carbon sources in R. sphaeroides 6016 are discussed.  相似文献

This paper reports investigations carried out to determine the optimum culture conditions for the production of hydrogen with a recently isolated strain Clostridium butyricum CWBI1009. The production rates and yields were investigated at 30 °C in a 2.3 L bioreactor operated in batch and sequenced-batch mode using glucose and starch as substrates. In order to study the precise effect of a stable pH on hydrogen production, and the metabolite pathway involved, cultures were conducted with pH controlled at different levels ranging from 4.7 to 7.3 (maximum range of 0.15 pH unit around the pH level). For glucose the maximum yield (1.7 mol H2 mol−1 glucose) was measured when the pH was maintained at 5.2. The acetate and butyrate yields were 0.35 mol acetate mol−1 glucose and 0.6 mol butyrate mol−1 glucose. For starch a maximum yield of 2.0 mol H2 mol−1 hexose, and a maximum production rate of 15 mol H2 mol−1 hexose h−1 were obtained at pH 5.6 when the acetate and butyrate yields were 0.47 mol acetate mol−1 hexose and 0.67 mol butyrate mol−1 hexose.  相似文献

Combined dark and photo-fermentation of ground wheat starch was carried out by using different light sources, intensities and lighting regime. A mixture of heat treated anaerobic sludge and Rhodobacter sphaeroides-RV with a certain light/dark bacteria ratio was used in batch experiments. Tungsten, fluorescent, infrared (IR), tungsten + infrared, halogen lamps were used as light sources with a light intensity of 270 Wm−2 along with sunlight. Halogen lamp was found to be the most suitable light source yielding the highest cumulative hydrogen formation (178 ml) and yield (218 ml g−1 starch). Combined fermentations were performed at different light intensities (1–10 klux) using the halogen lamp in the second set of experiments. The optimum light intensity was found to be 10 klux (approx. 352 Wm−2) resulting in the highest cumulative hydrogen (111 ml) and hydrogen yield (139 ml H2 g−1 starch). Hydrogen formation was limited by the availability of light at low light intensities below 10 klux. Durations of dark/light cycles were changed to determine the most suitable lighting regime. Hydrogen gas formation increased with increasing cycle time and continuous lighting resulted in the highest cumulative hydrogen formation and hydrogen yield.  相似文献

In this study, an active photoheterotrophic growth of purple non-sulfur bacteria (PNSB) on Tris Acetate Phosphate Yeast extract (TAPY) medium is reported. TAPY medium is a modified TAP medium supplemented with filter sterilized yeast extract (0.3 g L−1) and vitamin B12. Heterotrophic growth of PNSB on nitrogen replete TAPY medium in dark could be obtained but much slower than that in light where cells could grow but without formation of pigments. The medium showed high potency for hydrogen production under nitrogen deprivation in light by Rhodobacter sphaeroides DSM 5864. Through using TAPY medium, 1 mole of acetate provided as glacial acetic acid produced 0.819 mole of hydrogen gas by R. sphaeroides on nitrogen deprived TAPY medium (initial pH 7 adjusted by HCl) with a maximum H2 production rate of (0.669 mmol H2 h−1 L−1) obtained at 42 h after start of fermentation. Repeated-batch hydrogen production could be achieved with high efficiency for three cycles by supplementing the nitrogen deprived culture with filter sterilized sodium acetate at the end of the log hydrogen production phase of each cycle. The medium was also applicable for photoheterotrophic growth in light and heterotrophic growth in dark of other PNSB namely Rhodobacter capsulatus JCM-21090 and Rhodospirillum rubrum DSM 467. Although R. rubrum could actively grow on TAPY medium under nitrogen deprivation in light, it was the lowest in hydrogen production compared to R. sphaeroides and R. capsulatus. The active growth of R. rubrum on nitrogen deprived TAPY medium suggests its possible use for producing biodegradable plastic polymers better than hydrogen. The results suggest that it's possible to use TAPY medium for growth and producing variable bioproducts by PNSB in future studies and applications. The repeated-batch hydrogen production by R. sphaeroides on nitrogen deprived TAPY medium is promising for large-scale applications of hydrogen production industry by only sequential supply with sodium acetate to the culture for three rounds of batch fermentation. This is the first report in using TAPY medium for growth and hydrogen production by PNSB.  相似文献

A mesophilic alkaline tolerant fermentative microbe was isolated from estuarine sediment samples and designated as Clostridium butyricum TM-9A, based on 16S rRNA gene sequence. Batch experiments were conducted for investigation of TM-9A strain for its growth and hydrogen productivity from glucose, in an iron containing basal solution supplemented with yeast extract as organic nitrogen source. Hydrogen production started to evolve when cell growth entered exponential phase and reached maximum production rate at late exponential phase. Maximum hydrogen production was observed at 37 °C, initial pH of 8.0 in the presence of 1% glucose. Optimization of process parameters resulted in increase in hydrogen yield from 1.64 to 2.67 mol of H2/mol glucose. Molar yield of H2 increased further from 2.67 to 3.1 mol of H2/mol of glucose with the decrease in hydrogen partial pressure, obtained by lowering the total pressure in the head space of the batch reactor. Acetate and butyrate were the measure volatile fatty acids generated during hydrogen fermentation. TM-9A strain produced hydrogen efficiently from a range of pentose and hexose sugars including di-, tri and poly-saccharides like; xylose, ribose, glucose, rhamnose, galactose, fructose, mannose, sucrose, arabinose, raffinose, cellulose, cellobiose and starch.  相似文献

The anaerobic photosynthetic bacterium Rhodopseudomonas sphaeroides was used to produce hydrogen gas from glucose. Light served as an energy source for the bacterium to synthesize ATP through photosynthetic phosphorylation. A new strain was developed R. sphaeroides VM81 through successive culturing and adaptation to glucose growth, which gave high yields and rates of hydrogen production. This paper reports optimization experiments on the growth and hydrogen production of R. sphaeroides VM81 at different initial glucose concentrations and pH. The optimum conditions for cell growth and hydrogen gas production were found to be at 7 g glu/l and pH = 7.2.  相似文献

In order to understand some limiting factors in microbial hydrogen fermentation we have examined hydrogen production by different strains of Escherichia coli grown in batch cultures under different limiting nutrient regimes. The effect of mutations in uptake hydrogenases, in lactate dehydrogenase (ldhA), and fhlA, coding for the regulator of formate hydrogen lyase (fhl) component synthesis, were studied. Each mutation contributed to a modest increase in hydrogen evolution and the effects were synergistic. Various elements were used as limiting nutrient. In batch experiments, limitation for sulfate was without great effect. There was some affect of limiting phosphate with yields approaching 1 mol per mol of glucose. However, strains showed the highest yield of hydrogen per glucose (∼22) when cultured at limiting concentrations of either ammonia or glucose.  相似文献

Rhodobacter sphaeroides O.U.001 is one of the candidates for photobiological hydrogen production among purple non-sulfur bacteria. Hydrogen is produced by Mo-nitrogenase from organic acids such as malate or lactate. A hupSL in frame deletion mutant strain was constructed without using any antibiotic resistance gene. The hydrogen production potential of the R. sphaeroides O.U.001 and its newly constructed hupSL deleted mutant strain in acetate media was evaluated and compared with malate containing media. The hupSLR. sphaeroides produced 2.42 l H2/l culture and 0.25 l H2/l culture in 15 mM malate and 30 mM acetate containing media, respectively, as compared to the wild type cells which evolved 1.97 l H2/l culture and 0.21 l H2/l culture in malate and acetate containing media, correspondingly. According to the results, hupSLR. sphaeroides is a better hydrogen producer but acetate alone does not seem to be an efficient carbon source for photoheterotrophic H2 production by R. sphaeroides.  相似文献

A novel hydrogen-producing strain was isolated from gamma irradiated digested sludge and identified as Clostridium butyricum INET1. The fermentative hydrogen production performance of the newly isolated C. butyricum INET1 was characterized. Various carbon sources, including glucose, xylose, sucrose, lactose, starch and glycerol were used as substrate for hydrogen production. The operational conditions, including temperature, initial pH, substrate concentration and inoculation proportion were evaluated for their effects on hydrogen production, and the optimal condition was determined to be 35 °C, initial pH 7.0, 10 g/L glucose and 10% inoculation ratio. Cumulative hydrogen production of 218 mL/100 mL and hydrogen yield of 2.07 mol H2/mol hexose was obtained. The results showed that C. butyricum INET1 is capable of utilizing different substrates (glucose, xylose, sucrose, lactose, starch and glycerol) for efficient hydrogen production, which is a potential candidate for fermentative hydrogen production.  相似文献

Hydrogen production with glucose by using co-immobilized cultures of a lactic acid bacterium, Lactobacillus delbrueckii NBRC13953, and a photosynthetic bacterium, Rhodobacter sphaeroides RV, in agar gels was studied.
