In view of the transition to hydrogen as a major energy carrier in the future, new routes for bringing down the cost of biological hydrogen production need to be explored. The current study was devoted to optimizing the dark fermentation by Escherichia coli HD701 for hydrogen production from an acid-hydrolyzed potato starch residue stream without nitrogen sparging to reduce the cost. To further increase the economic feasibility of hydrogen production by E. coli, this study explores the use of the waste culture after hydrogen production in mass scale one-pot green synthesis of silver nanoparticles.  相似文献

Defined co-cultures of hydrogen (H2) producers belonging to Citrobacter, Enterobacter, Klebsiella and Bacillus were used for enhancing the efficiency of biological H2 production. Out of 11 co-cultures consisting of 2–4 strains, two co-cultures composed of Bacillus cereus EGU43, Enterobacter cloacae HPC123, and Klebsiella sp. HPC793 resulted in H2 yield up to 3.0 mol mol−1 of glucose. Up-scaling of the reactor by 16-fold resulted in a corresponding increase in H2 production with an actual evolution of 7.44 L of H2. It constituted 58.2% of the total biogas. Continuous culture evolution of H2 by co-cultures (B. cereus EGU43 and E. cloacae HPC123) immobilized on ligno-cellulosic materials resulted in 6.4-fold improvement in H2 yield compared to free floating bacteria. This synergistic influence of B. cereus and E. cloacae can offer a better strategy for H2 production than undefined or mixed cultures.  相似文献

The redox balance and bacteriochlorophyll (Bchl) synthesis are both significant to hydrogen generation in photosynthetic bacteria. In this study, spbA and hupSL genes were knocked out from the genome of Rhodobacter sphaeroides HY01. The UV–vis spectra showed that the Bchl contents of spbA mutants were enhanced under photosynthetic conditions. The hydrogen yields of WH04 (hupSL) and WSH10 (spbA, hupSL) mutants increased by 19.4%, 21.8%, and the maximum hydrogen evolution rates increased by 29.9% and 55.0% respectively using glutamate as sole nitrogen source. The maximum hydrogen production rate of WSH10 was up to 141.9 mL/(L·h). The nifH expression levels of the mutants and the wild type supported the correlation between hydrogen production and nitrogenase activity. The results demonstrate that disruption of spbA in R. sphaeroides can partially derepress the ammonium inhibition in nitrogenase activity, and indicate that spbA is a negative regulator in nitrogenase synthesis in the presence of ammonium.  相似文献

Nanosized nickel particles have been synthesized by three different routes: polyol, microemulsion and precipitation/reduction methods. Nickel nanoparticles have been evaluated as electrocatalysts for the hydrogen evolution reaction (HER). The electrocatalysts have been characterized by using X-ray diffraction (XRD), thermogravimetric analysis (TGA) and X-ray photoelectron spectroscopy (XPS). Their electrocatalytic performance in the hydrogen evolution reaction has been evaluated by means of the Tafel curves recorded in alkaline medium. The activity for the hydrogen evolution reaction increases with the increasing amount of reduced Ni in the electrocatalysts. Remarkably, the formation of a nickel-boride alloyed phase (Ni3B) is responsible for the higher activity of the sample prepared by the precipitation/reduction method for the HER. The crystalline phase Ni3B appears to be responsible for the very high activity in hydrogen production.  相似文献

Escherichia coli produces molecular hydrogen (H2) during glucose fermentation. This production of H2 occurs via multiple and reversible membrane-associated hydrogenases (Hyd). Dependence of H2 producing rate (VH2)(VH2) by Hyd-4 (hyf) on glucose concentration was studied at different pHs. During growth on 0.2% glucose at pH 7.5 in JRG3615 (hyfA-B) and JRG3621 (hyfB-R  ) mutants (VH2)(VH2) was decreased ∼6.7 and ∼5 fold, respectively, compared to wild type. Only in JRG3621 mutant at pH 6.5 and 5.5 (VH2)(VH2) was severely decreased ∼7.8 and ∼3.8 fold, respectively. But when cells were grown on 0.8% glucose no difference between wild type and mutants was detected at any of the tested pHs. The results indicate Hyd-4 H2 producing activity inhibition by high concentration of glucose mainly at pH 7.5. This is of significance to regulate Hyd activity and H2 production by E. coli during fermentation.  相似文献

To explore the process kinetics of hydrogen evolution reaction (HER) on carbon-based Ni/NiO nanocomposite in the microbial electrolysis cells (MECs), the performance was systematically studied by different time-course sampling of five parallel single-chamber MECs operated under identical operating conditions, which included the electrochemical performance of anodes and cathodes, and the mechanism and kinetics of HER. It was hypothesized that the decreased performance of the nickel cathodes was due to corrosion and Ni dissolution. These results provide valuable insights into the effects of long-term operation on MEC performance.  相似文献

In this study, recombinant plasmid was constructed to analyze the effect of hydrogen production on the expression HupSL hydrogenase isolated from Rhodobacter sphaeroides in Escherichia coli. Although most of recombinant HupSL hydrogenase was produced as inclusion bodies the solubility of the protein increased significantly when the expression temperature shifted from 37 °C to 30 °C. Hydrogen production by expression of HupSL hydrogenase from recombinant E. coli increased 20.9-fold compared to control E. coli and 218-fold compared to wild type R. sphaeroides under anaerobic dark condition. The results demonstrate that HupSL hydrogenase, consisting of small and large subunits of hydrogenase isolated from R. sphaeroides, increases hydrogen production in recombinant E. coli. In addition conditions for enhancing the activity of HupSL hydrogenase in E. coli were suggested and were used to increase bacterial hydrogen production.  相似文献

Biological mycelia pellets, which are formed spontaneously in the process of Aspergillus niger Y3 fermentation, were explored as carrier for immobilization of Clostridium sp. T2 to improve hydrogen production. Batch fermentation tests showed that optimal dosage and size of mycelia pellets for hydrogen production were 0.350 g 150 ml−1 medium and 1.5 mm. Under these conditions, hydrogen production with immobilized cells on mycelia pellets was further investigated in continuous stirred-tank reactor (CSTR) with hydraulic retention time (HRT) ranging from 12 to 8 h. It obtained that the maximum hydrogen production rate reached 2.76 mmol H2 L−1 h−1 at 10 h HRT, which was 40.8% higher than the carrier-free process, but slightly lower than the counterpart immobilized in sodium alginate with the value of 3.15 mmol H2 L−1 h−1. SEM observation showed that abundant cells were closely adhered to mycelia pellets. The present results indicate the potential of using mycelia pellets as biological carrier for enhancing hydrogen production.  相似文献

The efficiency of hydrogen production by different cyanobacterial species depends on several external factors. We report here the factors enhancing hydrogen production by filamentous non-heterocystous cyanobacterium Arthrospira sp. PCC 8005. Cells adapted to dark-anaerobic conditions produced hydrogen consistent with increased hydrogenase activity when supplemented with Fe2+. Stimulation of hydrogen production could be achieved by addition of reductants, either dithiothreitol or β-mercaptoethanol with higher production observed with the latter. Additionally, Fe2+ and β-mercaptoethanol added to nitrogen- and sulphur-deprived cells significantly stimulated H2 production with maximal value of 5.91 ± 0.14 μmol H2 mg Chla−1 h−1. Glucose and a small increase of osmolality imposed by either NaCl or sorbitol enhanced hydrogen production. High rates of hydrogen production were obtained in cells adapted in nitrogen-deprived medium with neutral and alkaline external pH, significant decrease of hydrogen production occurred under acidic external pH.  相似文献

Poly(N-vinyl-2-pyrrolidone)(PVP)-protected Au/Ni bimetallic nanoparticles (BNPs) were prepared in one-vessel via chemical reduction of the corresponding ions with dropwise addition of NaBH4, and their catalytic activity in the hydrogen generation from hydrolysis of a basic NaBH4 solution was examined. The structure, particle size, and chemical composition of the resultant BNPs were characterized by Ultraviolet–visible spectrophotometry (UV–Vis), X-ray photoelectron spectroscopy (XPS), Transmission electron microscopy (TEM) and High-resolution transmission electron microscopy (HR-TEM). The effects of processing parameters such as metal composition, metal ion concentration, and mole ratio of PVP to metal ion on the hydrolysis of a basic NaBH4 solution were studied in detail. The results indicated that as-prepared Au/Ni BNPs showed a higher catalytic activity than corresponding monometallic NPs (MNPs) in the hydrogen generation from the hydrolysis reaction of a basic NaBH4 solution. Among all the MNPs and BNPs, Au/Ni BNPs with the atomic ratio of 50/50 exhibited the highest catalytic activity, showing a hydrogen generation rate as high as 2597 mL-H2 min−1 g-catalyst−1 at 30 °C, which can be ascribed to the presence of negatively charged Au atoms and positively charged Ni atoms. Based on the kinetic study of the hydrogen generation from the hydrolysis reaction of a basic NaBH4 solution over the PVP-protected Au/Ni BNPs, the corresponding apparent activation energy was determined as 30.3 kJ/mol for the BNPs with the atomic ratio of 50/50.  相似文献

The present work evaluated the influence of magnetite nanoparticles in chicken fat methyl-ester blend (CFME20) and hydrogen induction in CFME20 nano-fuels combustion performance and exhaust emissions using a 1-cylinder dual DICI engine. Results revealed that CFME20 blend exhibits a significant reduction in smoke, hydrocarbon, and carbon-monoxide emissions; however, increment in oxides of nitrogen and insignificant reduction in BTE also noted. Magnetite nanoparticles mixed with CFME20 through ultrasonication to make nano-additive fuel which noticeably decreased the exhaust emissions including oxides of nitrogen and slightly enhanced the BTE as well. Performance parameters gradually enhanced with escalating nanoparticles dosage up to 100 ppm. Hydrogen induction during nano-additive pilot fuel combustion further decreased the smoke, carbon-monoxide, and hydrocarbon emissions and enhanced the BTE to an appreciable level. However, oxides of nitrogen also enhanced with increasing hydrogen flow rate, but for 10 lpm trivial increased. Hence, the results confirmed that nano-fuel (100 ppm) with hydrogen (10 lpm) addition provides optimum outcomes.  相似文献

PtRuMoOx and PtRuWOx catalysts supported on multi-wall carbon nanotubes (MWCNTs) are prepared by ultrasonic-assisted chemical reduction method. XRD measurements indicate that Pt exists as face-centered cubic structure, Ru is alloyed with platinum, and the metal oxides exist as an amorphous structure. TEM pictures show that PtRuMoOx and PtRuWOx catalysts are well dispersed on the surface of MWCNTs with the particle size of about 3 nm and a narrow particle size distribution. The electrochemical properties of the catalysts for methanol electrooxidation are studied by cyclic voltammetry (CV), chronoamperometry (CA) and chronopotentiometry (CP). The onset potentials for methanol oxidation on PtRuMoOx and PtRuWOx are more negative than that of pure Pt catalyst, shifting negatively by about 0.20 V and have better electrocatalytic activities than PtRu/MWCNTs.  相似文献

A preliminary study on photoproduction of hydrogen by Rhodobacter capsulatus KU002 isolated from leather industry effluents under different cultural conditions with various carbon and nitrogen sources was investigated. Hydrogen production was measured using a Gas chromatograph. Lactate promoted more amounts of hydrogen production under anaerobic light conditions and aerobic light conditions. Cumulative hydrogen production by the organism was recorded at various time intervals. Incubation period of 120 h was optimum for production of hydrogen. pH 7.0 ± 0.2 was optimum for production of hydrogen by growing cells, while pH 7.5 ± 0.26 for resting cells. l-cystine was a good nitrogen source for production of hydrogen. Growing cells produced more amount of hydrogen than resting cells. Glutamine was a poor nitrogen source for hydrogen production by Rb. capsulatus. Significance of the above results in the presence of existing literature is discussed.  相似文献

The batch fermentations of two hyperthermophilic eubacteria Thermotoga maritima strain DSM 3109 and Thermotoga neapolitana strain DSM 4359 were carried out to optimize the hydrogen production. The simple and economical culture medium using cheap salts with strong buffering capacity was designed based on T. maritima basal medium (TMB). Both strains cultivated under strictly anaerobic conditions showed the best growth at temperature of 75–80 °C and pH of 6.5–7.0. The maximum cell growth of 3.14 g DCW/L and hydrogen production of 342 mL H2 gas/L were obtained, respectively, in the modified TB medium containing 7.5 g/L of glucose and 4 g/L of yeast extract. Hydrogen accumulation in the headspace was more than 30% of the gaseous phase. Cells were also cultivated in cellulose-containing medium to test the feasibility of hydrogen production.  相似文献

A thermotolerant Klebsiella sp. TR17 for production of hydrogen from crude glycerol was investigated. Results from Plackett–Burman design indicated that the significant variables, which influenced hydrogen production, were KH2PO4 and NH4Cl (for buffer capacity and nitrogen source). Subsequently, the two selected variables and crude glycerol were optimized by the Central Composite design for achieving maximum hydrogen and ethanol yield. The concentration of crude glycerol, KH2PO4, and NH4Cl had an individual effect on both hydrogen and ethanol yield (p < 0.05), while KH2PO4 and NH4Cl had an interactive effect on ethanol yield (p < 0.05). The optimum medium components were 11.14 g/L crude glycerol, 2.47 g/L KH2PO4, and 6.03 g/L NH4Cl. The predicted maximum simultaneous hydrogen and ethanol yield were 0.27 mol H2/mol glycerol and 0.63 mol EtOH/mol glycerol, respectively. Validation of the predicted optimal conditions exhibited similar hydrogen and ethanol yield of 0.26 mol H2/mol glycerol and 0.58 mol EtOH/mol glycerol, respectively.  相似文献

Magnetite nanoparticles (MNP)-caused enhancements for H2 yield (HY) are usually justified based on oxidation-reduction potential (ORP), iron ions (Fe+2/+3) concentration, and enzymatic activity, that is pH-dependent. However, the questions “If pH, ORP, and Fe+2/+3 impacts are excluded, will MNP-caused HY enhancement be still present? and how electrical energy input (EEI) affects HY?” are still unanswered. Herein, control, MNP-supplemented, EEI-applied, FeCl3-, and Na2S-supplemented batches, referred as G1, G2, G3, G5, and G6, respectively were conducted, under fixed pH (6.0 ± 0.1). G5 and G6 targeted quantifying sole impact of Fe+2/+3, and ORP, respectively on HY. G1, G2, G3, G5, and G6 achieved HY values of 1.10 ± 0.05, 1.66 ± 0.07, 1.38 ± 0.06, 1.18 ± 0.04, and 1.16 ± 0.05 mol H2/molhexose, respectively. Neither Fe+2/+3 release nor ORP reduction significantly affected HY. Further, Clostridium amylolyticum dominance was almost similar among G2, and G3. Metabolites flux analysis and functional genes’ prediction highlighted that G2 achieved highest hydrogenase expression and lowest homoacetogenic H2 consumption.  相似文献

Chlamydomonas reinhardtii UTEX 90 accumulated 1.45 g dry cell weight and 0.77 g starch/L during photosynthetic growth using TAP media at 25 °C°C in presence of 2% CO2CO2 for 3 days. C. reinhardtii biomass was concentrated and then converted into hydrogen and organic acids by anaerobic fermentation with Clostridium butyricum. Organic acids in the fermentate of algal biomass were consecutively photo-dissimilated to hydrogen by Rhodobacter sphaeroides KD131. In the concentrated algal biomass 52% of the starch was hydrolyzed to 37.1 mmol H2H2/L-concentrated algal biomass and 13.6, 25.5, 7.4 and 493 mM of formate, acetate, propionate, and butyrate, respectively by C. butyricum. R. sphaeroides KD131 evolved 5.72 mmol H2H2 per ml-fermentate of algal biomass under illumination of 8 klux at 30 °C°C. Only 80% of the organic acids, mainly butyrate, were hydrolyzed during photo-incubation. During anaerobic conversion, 2.58 mol H2/molH2/mol starch–glucose was evolved using C. butyricum and then 5.72 mol H2/LH2/L-anaerobic fermentate was produced by R. sphaeroides KD131. Thus, the two-step conversion process produced 8.30 mol H2H2 from 1 mol starch–glucose equivalent algal biomass via organic acids.  相似文献
