Poly(HEMA‐MAA) hydrogel particles were synthesized by redox free‐radical polymerization using 2‐hydroxyethylmethacrylate, different concentration of methacrylic acid as monomer, ethyleneglycol dimethacrylate as crosslinking agent, and APS/TEMED as free‐radical initiator. Fourier transform infrared spectrum of poly(HEMA‐MAA) hydrogels showed intense absorption peak of carbonyl group at ~ 1700 cm?1 due to carboxylic acid groups of MAA, peak at ~ 2960 cm?1 due to CH stretching and vinylic peak at 1700 cm?1 independent of MAA concentration. Highest swelling percentage 587% was observed in case of poly(HEMA‐MAA) hydrogel synthesized using 30% of MAA while lowest swelling percentage 413% was observed in hydrogel synthesized 10% of MAA at basic pH (8.0). Scanning electron micrograph of copolymeric particles showed the irregular shape of poly(HEMA‐MAA) particles with conglomeration with each due to ionization of carboxylic groups. Insulin was radiolabeled using technetium‐99m radionuclide and the radiolabeling efficiency was found to be 99%. Poly(HEMA‐MAA) hydrogel having 60% of MAA showed the highest insulin loading efficiency of 68% while lowest 37% was observed in case of 10% MAA hydrogel. Insulin release studies showed only 35–65% of insulin was released into the medium from particles at pH 2.5 in 60 min, while insulin release was significantly higher at pH 7.4. Hypoglycemic effect of the 60 and 80 I.U./kg insulin dose loaded in poly(HEMA‐MAA) copolymeric particles were carried out in fasted diabetic rats and highest decrease in blood glucose level from 506 mg/dL to 170 mg/dL was observed within first 3 h. © 2011 Wiley Periodicals, Inc. Chitosan (CS) grafted poly[(acrylic acid)‐co‐(2‐hydroxyethyl methacrylate)] (CS‐g‐poly(AA‐co‐HEMA)) at different molar ratios of AA and HEMA, and the associated nanocomposite hydrogels of CS‐g‐poly(AA‐co‐HEMA)/mica were synthesized by radical copolymerization. The grafting positions at the amino or hydroxyl groups in the CS were identified by Fourier transform infrared spectroscopy. CS‐g‐poly(AA‐co‐HEMA) hydrogels were intercalated in the mica and the amount of hydrogel insertion did not affect the spacing of the silicate layers in mica. The higher mica loadings produced a rougher surface of the nanocomposite hydrogel. The water absorbency of the CS‐g‐poly(AA‐co‐HEMA)/mica nanocomposite hydrogels decreased with increasing levels of mica loading to a lower level than those of the CS‐g‐poly(AA‐co‐HEMA) hydrogels. Both CS‐g‐poly(AA) and CS‐g‐poly(AA‐co‐HEMA)/mica nanocomposite hydrogels exhibited a higher antiproliferative activity against Staphylococcus aureus than did the neat CS hydrogel with CS‐g‐poly(AA) revealing a very pronounced minimum inhibition concentration (MIC) of 1.56 mg mL?1. The extent of mica loading in the CS‐g‐poly(AA‐co‐HEMA) nanocomposite hydrogels did not affect the MIC (12.5 mg mL?1). © 2012 Wiley Periodicals, Inc. J. Appl. Polym. Sci., 2013  相似文献

In this work, biocompatible hydrogel matrices for wound‐dressing materials and controlled drug‐release systems were prepared from poly[hydroxyethyl methacrylate‐co‐poly(ethylene glycol)–methacrylate] [p(HEMA‐co‐PEG–MA] films via UV‐initiated photopolymerization. The characterization of the hydrogels was conducted with swelling experiments, Fourier transform infrared spectroscopy, scanning electron microscopy, thermogravimetric analysis (differential scanning calorimetry), and contact‐angle studies. The water absorbency of the hydrogel films significantly changed with the change of the medium pH from 4.0 to 7.4. The thermal stability of the copolymer was lowered by an increase in the ratio of poly(ethylene glycol) (PEG) to methacrylate (MA) in the film structure. Contact‐angle measurements on the surface of the p(HEMA‐co‐PEG–MA) films demonstrated that the copolymer gave rise to a significant hydrophilic surface in comparison with the homopolymer of 2‐hydroxyethyl methacrylate (HEMA). The blood protein adsorption was significantly reduced on the surface of the copolymer hydrogels in comparison with the control homopolymer of HEMA. Model antibiotic (i.e., minocycline) release experiments were performed in physiological buffer saline solutions with a continuous flow release system. The amount of minocycline release was shown to be dependent on the HEMA/PEG–MA ratio. The hydrogels have good antifouling properties and therefore are suitable candidates for wound dressing and other tissue engineering applications. © 2009 Wiley Periodicals, Inc. In this study, N‐allylsuccinamic acid (NASA) was synthesized in a single step with a yield of 85%. Carboxylic acid containing NASA was characterized through Fourier transform infrared (FTIR) radiation and 1H‐NMR and 13C‐NMR analysis, and then it was used for synthesis of poly(2‐hydroxylethyl methacrylate‐co‐N‐allylsuccinamic acid) [p(HEMA‐co‐NASA)] hydrogels. The structure of the obtained pH‐responsive p(HEMA‐co‐NASA) hydrogels were characterized with FTIR spectroscopy and scanning electron microscopy analysis, and their swelling characterization was carried out under different drug‐release conditions. In the application step of the study, the hydrogels were used for the in vitro release of vitamin B12 and Rhodamine 6G, which were selected as model drugs. We determined that the hydrogels used as a drug‐delivery matrix could release the drug they had absorbed under different release conditions (phosphate‐buffered saline, 0.9% NaCl, and pH 1.2) at high rates for time periods of up to 24 h. © 2013 Wiley Periodicals, Inc. This works aims at (i) studying the antiadhesive properties and the hemocompatibility of poly[2‐(dimethylamino)ethyl methacrylate]‐co‐poly[(ethylene glycol)methacrylate] [poly(DMAEMA‐co‐PEGMA)] copolymers and (ii) investigating the insulin delivery kinetics through hydrogels at physiological pH. A series of poly(DMAEMA‐co‐PEGMA) hydrogels have been synthesized, and their controlled composition was confirmed by X‐ray photoelectron spectroscopy. Then, antibiofouling properties of hydrogels—fibrinogen, erythrocytes, and thrombocytes adhesion—are correlated to their molecular compositions through their hydrophilic properties. As DMAEMA/PEGMA ratio of 70/30 (D70) offers the best compromise between pH sensitivity and hemocompatibility, it is selected for investigating the kinetic rate of insulin release at physiological pH, and the diffusion coefficient of insulin in gel is found to be 0.64 × 10?7 cm2 s?1. Overall, this study unveils that poly(DMAEMA‐co‐PEGMA) copolymers are promising hemocompatible materials for drug delivery systems. © 2015 Wiley Periodicals, Inc. J. Appl. Polym. Radiation‐crosslinked 2‐hydroxyethylmethacrylate/citric acid (HEMA/CAc), 2‐hydroxyethylmethacrylate/tartaric acid (HEMA/TA), and 2‐hydroxyethylmethacrylate/succinic acid (HEMA/Sc) copolymers were prepared by using 60Co γ‐rays. The gel fraction yield and the swelling behavior of the prepared hydrogels were studied. It was shown that increasing irradiation doses was accompanied by an increase in yield of gel fraction and a decrease in swelling degree. The parameters of equilibrium swelling, maximum swelling, initial swelling rate, swelling exponent, and diffusion coefficient of the hydrogels were determined by studying the swelling behavior of the hydrogels prepared. It was seen that the equilibrium swelling degree increases as the content of acid increases, as a result of introducing more hydrophilic groups. When the hydrophilic polymer (acids) varies in the content range of 40–80 mg, swelling exponents (n) decreases, thereby indicating a shift in the water‐transport mechanism from the anomalous (non‐Fickian)‐type to the Fickian‐type. Characterization and some selected properties of the prepared hydrogels were studied, and accordingly the possibility of its practical use in the treatment of industrial wastes such as dyes and heavy metals (Fe, Ni, Co, and Cu) were also studied. The effect of treatment time, pH of feed solution, initial feed concentration, and temperature on the dye and heavy metals uptake was determined. The uptake order for a given metal was HEMA/TA hydrogel > HEMA/CAc > HEMA/Sc hydrogel. © 2007 Wiley Periodicals, Inc. J Appl Polym Sci, 2008  相似文献

In this study, a new cationic monomer 2‐(3‐indol‐yl)ethylmethacrylamide (IEMA) derived from tryptamine was synthesized in a single step and characterized by Fourier transform infrared (FTIR), 1H‐NMR, and 13C‐NMR. Then, one‐step preparation of novel poly[2‐hydroxyethylmethacrylate‐c‐2‐(3‐indol‐yl)ethylmethacrylamide], or p(HEMA‐c‐IEMA), copolymeric hydrogels has been performed successfully with IEMA and 2‐hydroxyethylmethacrylate (HEMA) as monomers using free radical aqueous polymerization. The hydrogels were characterized with scanning electron microscopy, FTIR, elemental analysis, thermogravimetric analysis, and texture profile analysis instruments. p(HEMA‐c‐IEMA) hydrogels were used for swelling, diffusion, drug release, and antibacterial activity studies. The drug‐release behavior of the hydrogels was determined as a function of time at 37 °C in pH 1.2 and 7.2. The swelling and drug‐release studies showed that an increased IEMA amount caused a higher increase in swelling and drug‐release values. Additionally, zero‐order, first‐order, and Higuchi equation kinetic models were applied to the drug‐release data, and the data fit well in the Higuchi model, and the Peppas power‐law model was applied to the release mechanism. Finally, the antibacterial activities of the hydrogels were screened against Gram‐positive bacteria (Bacillus cereus and Staphylococcus aureus) and Gram‐negative bacteria (Escherichia coli and Salmonella typhimurium). © 2017 Wiley Periodicals, Inc. 2‐Hydroxyethyl methacrylate (HEMA) based polymeric hydrogels were synthesized by free‐radical redox bulk polymerization technique using 1% ethyleneglycol dimethacrylate (EGDMA) as crosslinking monomer and ammonium persulfate (APS) and N,N,N′,N′‐tetramethyl ethylenediamine (TEMED) as redox initiator. Polymeric hydrogel samples were loaded with solid elemental iodine. Thermal and physical characteristics of polymer before loading and after 3 months release of iodine were evaluated by differential scanning calorimetry (DSC), Fourier transform infrared spectroscopy (FTIR), and scanning electron microscopy (SEM). On immersing in water, different forms of iodine were released from the hydrogel device. The amount and rate of release of I? and I were measured by analytical techniques. Released iodine species showed broad spectrum antimicrobial properties and release was sustained for about 120 days. Polymeric hydrogel iodine‐based system developed can be used as a device for controlled release of iodine species at concentration levels sufficient for disinfection to get potable water. © 2006 Wiley Periodicals, Inc. Composite hydrogels of poly(acrylamide‐co‐acrylate) with rice husk ash (RHA) were synthesized and studies of the swelling variables were accomplished comparatively with commercial polyacrylamide gel and PAMACRYL, a poly(acrylamide‐co‐acrylate) hydrogel without RHA. FT‐IR and WAXS were the techniques employed for characterizing a series of hydrogel obtained by varying the percentage of RHA (1, 2, 5, 10, and 20 wt %) and the amount of crosslinking agent (0.05, 0.1, and 0.2 mol %) relative to sum of AAm and AAc. Superabsorbent hydrogel with Weq > 800 g H2O/g gel was obtained with percentage of 10 wt % of RHA and 0.1 of crosslinking agent mol %. The hydrogel showed to be sensitive to the pH variation and to the presence of salts. The hydrogels, even though submitted through cycles of drying and swelling, preserved their superabsorbent characteristics and demonstrated better water absorbance properties when compared with commercial polyacrylamide gel. The composite hydrogels of poly(acrylamide‐co‐acrylate) with RHA presented good characteristics to be applied as soil conditioner for using in agriculture. © 2011 Wiley Periodicals, Inc. A cholic acid (CA)‐containing biodegradable hydrogel (PLA‐PEG‐PLA‐co‐MACAH) was synthesized from the photoinduced copolymerization of a CA‐modified methacrylate monomer (MACAH), bearing a spacer of hexane‐1,6‐diol spacer between the methacryloyl and the cholanoate moieties, and a macromonomer (PLA‐PEG‐PLA‐DA), bearing two acryloyl end groups derived from a poly(lactic acid)‐b‐poly(ethylene glycol)‐b‐poly(lactic acid) triblock copolymer. The structure of MACAH was confirmed by FTIR, 1H‐NMR, and MS. The hydrogel PLA‐PEG‐PLA‐co‐MACAH was characterized by scanning electron microscopy and X‐ray diffraction. The experiment results showed that the swelling ratios of the hydrogels decreased with the increase of the CA fraction. The investigation on the in vitro degradation of the hydrogel showed that the CA‐containing hydrogels degraded much slower than the hydrogels without CA component. The bioactivity of the synthesized hydrogels was assessed by the simulated body fluid method. The observed formation of hydroxyapatite on the scaffold of the hydrogels indicated that the hydrogels possess good bioactivity. © 2009 Wiley Periodicals, Inc. J Appl Polym Sci, 2009  相似文献

Semi‐interpenetrating polymer network hydrogels with different compositions of chitosan (Cs), acrylic acid, and citraconic acid were synthesized via free‐radical polymerization with ethylene glycol dimethacrylate as a crosslinker. The variations of the swelling percentages of the hydrogels with time, temperature, and pH were determined, and Cs–poly(acrylic acid) (PAA) hydrogels were found to be most swollen at pH 7.4 and 37°C. Scanning electron micrographs of Cs–PAA and Cs–P(AA‐co‐CA)‐1 (Cs‐poly(acrylicacid‐co‐citraconir acid)?1) were taken to observe the morphological differences in the hydrogels. Although the less swollen hydrogel, Cs–P(AA‐co‐CA)‐1, had a sponge‐type structure, the most swollen hydrogel, Cs–PAA, displayed a uniform porous appearance. Fluconazole was entrapped in Cs–P(AA‐co‐CA)‐1 and Cs–PAA hydrogels, and the release was investigated at pH 4.0 and 37°C. The kinetic release parameters of the hydrogels (the gel characteristic constant and the swelling exponent) were calculated, and non‐Fickian diffusion was established for Cs–PAA, which released fluconazole much more slowly than the Cs–P(AA‐co‐CA)‐1 hydrogel. A therapeutic range was reached at close to 1 h for both hydrogels. © 2009 Wiley Periodicals, Inc. J Appl Polym Sci, 2009  相似文献

New hybrid poly(hydroxyethyl methacrylate‐co‐methyl methacrylate)‐g‐polyhedral oligosilsesquioxane [poly(HEMA‐co‐MMA)‐g‐POSS] nanocomposites were synthesized by the combination of reversible addition fragmentation chain transfer (RAFT) polymerization and click chemistry using a grafting to protocol. Initially, the random copolymer poly(HEMA‐co‐MMA) was prepared by RAFT polymerization of HEMA and MMA. Alkynyl side groups were introduced onto the polymeric backbones by esterification reaction between 4‐pentynoic acid and the hydroxyl groups on poly(HEMA‐co‐MMA). Azide‐substituted POSS (POSS? N3) was prepared by the reaction of chloropropyl‐heptaisobutyl‐substituted POSS with NaN3. The click reaction of poly(HEMA‐co‐MMA)‐alkyne and POSS? N3 using CuBr/PMDEATA as a catalyst afforded poly(HEMA‐co‐MMA)‐g‐POSS. The structure of the organic/inorganic hybrid material was investigated by Fourier transformed infrared, 1H‐NMR, and 29Si‐NMR. The elemental mapping analysis of the hybrid using X‐ray photoelectron spectroscopy and EDX also suggest the formation of poly(HEMA‐co‐MMA)‐anchored POSS nanocomposites. The XRD spectrum of the nanocomposites gives evidence that the incorporation of POSS moiety leads to a hybrid physical structure. The morphological feature of the hybrid nanocomposites as captured by field emission scanning electron microscopy and transmission electron microscopic analyses indicate that a thick layer of polymer brushes was immobilized on the POSS cubic nanostructures. The gel permeation chromatography analysis of poly(HEMA‐co‐MMA) and poly(HEMA‐co‐MMA)‐g‐POSS further suggests the preparation of nanocomposites by the combination of RAFT and click chemistry. The thermogravimetric analysis revealed that the thermal property of the poly(HEMA‐co‐MMA) copolymer was significantly improved by the inclusion of POSS in the copolymer matrix. © 2012 Wiley Periodicals, Inc. Hydrophobically modified poly[2‐(diethylamino)ethylmethacrylate‐co‐N‐vinyl‐2‐pyrrolidone/octadecyl acrylate) [P(DEAEMA‐co‐NVP/OA)] hydrogels were synthesized by free‐radical crosslinking copolymerization of 2‐(diethylamino)ethylmethacrylate (DEAEMA), N‐vinyl‐2‐pyrrolidone (NVP) with different amounts of hydrophobic comonomer octadecyl acrylate (OA) in tert‐butanol with ethylene glycole dimethacrylate (EGDMA) as a crosslinker. The swelling equilibrium of the hydrogels was investigated as a function of temperature and hydrophobic comonomer content in aqueous solutions of the anionic surfactant sodium dodecyl sulfate (SDS) and the cationic surfactant dodecyltrimethylammonium bromide (DTAB). The results indicated that the swelling behavior and temperature sensitivity of the hydrogels were affected by the type and concentration of surfactant solutions. Additionally, the amount of the adsorbed SDS and DTAB molecules onto the hydrogels was determined by fluorescence measurements. An increase of OA content in the hydrogel caused an increase in the amount of adsorbed surfactant molecules in both media. © 2006 Wiley Periodicals, Inc. Macroporous beads, poly(ethylene glycol dimethacrylate‐co‐acrylic acid) [poly(EGDMA‐co‐AAc)], and poly(ethylene glycol dimethacrylate‐co‐hydroxyethyl methacrylate) [poly(EGDMA‐co‐HEMA)] were prepared by the suspension polymerization technique in the presence of a porogen agent. Different experimental conditions such as amount of initiator, porogen type, and temperature were studied to optimize the polymerization systems. These hydrophilic copolymers were characterized by IR spectroscopy, scanning electron microscopy, specific surface area, and swelling in water. A new parameter, H, defined as the ratio between the equilibrium weight swelling ratio (qw) and equilibrium volume swelling ratio (qv), allowed to select the reaction conditions from which matrices with high capacity of water sorption and low stretching degree were reached. © 2000 John Wiley & Sons, Inc. In this study, a series of poly(acrylamide‐co‐acrylic acid‐co‐2‐hydroxy ethyl actylate) [AM‐co‐AA‐co‐HEA] hydrogels have been synthesized by varying the acrylic acid (AA)content over eightfold in feed in the range of 33.34–93.76% by keeping other monomer constant. These hydrogels were characterized by FTIR, SEM analysis, elemental analysis, residual acrylic acid analysis, network parameters, and dynamic swelling behavior. The swelling study showed that equilibrium swelling ratio was nonlinearly increased with increasing AA content. Interestingly, the equilibrium swelling ratio decreased from 53.42 to 48.52 for 75–80% AA content hydrogel. The swelling data were found to satisfactorily fit Fick's second law, demonstrating that diffusion rate of water uptake was primarily Fickian. From model fitting, it was observed that early model was applicable for first 30% water absorption, and late model was applicable for latter 70% water absorption for increasing AA content from 33.34–90.90%. For 93.76% AA, early‐time model was extended up to first 50% of water absorption and late model was contracted for latter 50% water absorption, indicating that excessive AA content affects the applicability range of early‐time and late‐time diffusion models for water absorption. Etters model was best applicable to all type of hydrogels and followed over all swelling range. © 2011 Wiley Periodicals, Inc. J Appl Polym Sci, 2012  相似文献

Immobilization glucoamylase onto plain and a six‐carbon spacer arm (i.e., hexamethylene diamine, HMDA) attached poly(2‐hydroxyethylmethacrylate‐ethyleneglycol dimethacrylate) [poly(HEMA‐EGDMA] microspheres was studied. The microspheres were prepared by suspension polymerization and the spacer arm was attached covalently by the reaction of carbonyl groups of poly(HEMA‐EGDMA). Glucoamylase was then covalently immobilized either on the plain of microspheres via CNBr activation or on the spacer arm‐attached microspheres via CNBr activation and/or using carbodiimide (CDI) as a coupling agent. Incorporation of the spacer arm resulted an increase in the apparent activity of the immobilized enzyme with respect to enzyme immobilized on the plain of the microspheres. The activity yield of the immobilized glucoamylase on the spacer arm‐attached poly(HEMA‐EGDMA) microspheres was 63% for CDI coupling and 82% for CNBr coupling. This was 44% for the enzyme, which was immobilized on the plain of the unmodified poly(HEMA‐EGDMA) microspheres via CNBr coupling. The Km values for the immobilized glucoamylase preparations (on the spacer arm‐attached microspheres) via CDI coupling 0.9% dextrin (w/v) and CNBr coupling 0.6% dextrin (w/v) were higher than that of the free enzyme 0.2% dextrin (w/v).The temperature profiles were broader for both immobilized preparations than that of the free enzyme. The operational inactivation rate constants (kiop) of immobilized enzymes were found to be 1.42 × 10?5 min?1 for CNBr coupled and 3.23 × 10?5 min?1 for CDI coupled glucoamylase. © 2001 John Wiley & Sons, Inc. A series of amphoteric, superabsorbent polymers [poly(acrylic acid‐co‐diallyldimethylammonium chloride)] with different molar ratios of anionic groups to cationic groups were prepared by solution polymerization to investigate their swelling behaviors and the controlled release of agrochemicals. Various factors, including the solution pH, the concentrations of different salt solutions, and the temperature, were studied. The dynamic parameters of hydrogels at different temperatures suggested that diffusion was Fickian at lower temperatures, whereas non‐Fickian diffusion prevailed at higher temperatures. A copolymer hydrogel with a low anionic‐group/cationic‐group ratio showed a higher swelling capacity in water and higher salt tolerance. Also, the anionic‐group/cationic‐group ratio was not the dominant factor in determining the water retention. A poly(acrylic acid‐co‐diallyldimethylammonium chloride) hydrogel could control the release of agrochemicals effectively. © 2006 Wiley Periodicals, Inc. J Appl Polym Sci 102: 986–991, 2006  相似文献

N‐Methacryloyl‐(L )‐histidine methyl ester (MAH) was synthesized from metharyloyl chloride and histidine. Spherical beads with an average size of 150–250 μm were obtained by the suspension polymerization of ethylene glycol dimethacrylate and MAH in an aqueous dispersion medium. Magnetic poly(ethylene glycol dimethacrylate‐coN‐Methacryloyl‐(L )‐histidine methyl ester) [m‐p(EGDMA‐co‐MAH)] microbeads were characterized with swelling tests, electron spin resonance, elemental analysis, and scanning electron microscopy. The specific surface area of the beads was 80.1 m2/g. m‐p(EGDMA‐co‐MAH) microbeads with a swelling ratio of 40.2% and 43.9 μmol of MAH/g were used for the adsorption of bovine serum albumin (BSA) in a batch system. The Cu(II) concentration was 4.1 μmol/g. The adsorption capacity of BSA on the Cu(II)‐incorporated beads was 19.2 mg of BSA/g. The BSA adsorption first increased with the BSA concentration and then reached a plateau, which was about 19.2 mg of BSA/g. The maximum adsorption was observed at pH 5.0, which was the isoelectric point of BSA. The BSA adsorption increased with decreasing temperature, and the maximum adsorption was achieved at 4°C. High desorption ratios (>90% of the adsorbed BSA) were achieved with 1.0M NaSCN (pH 8.0) in 30 min. The nonspecific adsorption of BSA onto the m‐p(EGDMA‐co‐MAH) beads was negligible. © 2004 Wiley Periodicals, Inc. J Appl Polym Sci 93: 2669–2677, 2004  相似文献

To achieve the injectable hydrogel system in order to improve bone regeneration by locally delivering a protein drug including bone morphogenetic proteins, thermo‐responsive injectable hydrogels composed of N‐isopropylacrylamide (NIPAAm) and vinyl phosphonic acid (VPAc) were prepared. The P(NIPAAm‐co‐VPAc) hydrogels were also biomineralized by urea‐mediation method to create functional polymer hydrogels that deliver the protein drug and mimic the bone extracellular matrix. The loosely cross‐linked P(NIPAAm‐co‐VPAc) hydrogels were pliable and fluid‐like at room temperature and could be injected through a small‐diameter aperture. The lower critical solution temperature (LCST) of P(NIPAAm‐co‐VPAc) hydrogel was influenced by the monomer ratio of NIPAAm/VPAc and the hydrogel with a 96/4 molar ratio of NIPAAm/VPAc exhibited an LCST of ~34.5°C. Water content was influenced by temperature, NIPAAm/VPAc monomer ratio, and biomineralization; however, all hydrogels maintained more than about 77% of the water content even at 37°C. In a cytotoxicity study, the P(NIPAAm‐co‐VPAc) and biomineralized P(NIPAAm‐co‐VPAc) hydrogels did not significantly affect cell viability. The loading content of bovine serum albumin in hydrogel, which was used as a model drug, gradually increased with the amount of VPAc in the hydrogel owing to the ionic interaction between VPAc groups and BSA molecules. In addition, the release behavior of BSA from the P(NIPAAm‐co‐VPAc) hydrogels was mainly influenced by the drug loading content, water content, and biomineralization of the hydrogels. © 2009 Wiley Periodicals, Inc. To reach sustained drug release, a new composite drug‐delivery system consisting of poly(d,l ‐lactide‐co‐glycolide) (PLGA) nanoparticles (NPs) embedded in thermosensitive poly(N‐isopropyl acrylamide) (PNIPAAm) hydrogels was developed. The PNIPAAm hydrogels were synthesized by free‐radical polymerization and were crosslinked with poly(ethylene glycol) diacrylate, and the PLGA NPs were prepared by a water‐in‐oil‐in‐water double‐emulsion solvent‐evaporation method. The release behavior of the composite hydrogels loaded with albumin–fluorescein isothiocyanate conjugate was studied and compared with that of the drug‐loaded neat hydrogel and PLGA NPs. The results indicate that we could best control the release rate of the drug by loading it to the PLGA NPs and then embedding the whole system in the PNIPAAm hydrogels. The developed composite hydrogel systems showed near zero‐order drug‐release kinetics along with a reduction or omission of initial burst release. The differential scanning calorimetry results reveal that the lower critical solution temperature of the developed composite systems remained almost unchanged (<1°C increase only). Such a characteristic indicated that the thermosensitivity of the PNIPAAm hydrogel was not distinctively affected by the addition of PLGA NPs. In conclusion, an approach was demonstrated for the successful preparation of a new hybrid hydrogel system having improved drug‐release behavior with retained thermosensitivity. The developed systems have enormous potential for many biotechnological applications. © 2014 Wiley Periodicals, Inc. J. Appl. Polym. Sci. 2014 , 131, 40625.  相似文献
