Isolation of a potentially functional HPRT processed pseudogene from the hill kangaroo Macropus robustus

A highly conserved hypoxanthine phosphoribosyltransferase processed pseudogene (KPH) has been isolated from a female kangaroo (Macropus robustus) lambda EMBL3 genomic library. The pseudogene contains only transcribed material with all of the introns precisely removed and has possible direct repeats at either end of the message. It has a 654-nucleotide open reading frame (ORF) from the Met start codon to the stop codon that contains no additions, deletions or premature stops relative to expressed HPRT genes and, therefore, the possibility exists that it is expressed in vivo. Possible CAAT and GC boxes are present in the region 5' to the ORF and a polyadenylation signal is present in the region 3' to the ORF. If not expressed, the age of the pseudogene is estimated to be 10.7 million years. We propose that integration into the genome occurred specifically in a homocopolymeric region within a highly repeated region unique to the kangaroo genome.     

A Civil Engineer's View from Space

From the perspective of a 250‐nautical‐mile orbit aboard the Space Shuttle, the author has had the opportunity to observe the effects of man on the earth, to reflect on his future in space, and to examine the role civil engineers may have in building our future. In the decades to come, civil engineers will require skills that are not currently provided by universities and which are not adequately represented in professional societies. All disciplines of the civil engineering profession will need to examine their strategies to enable them to establish a significant place on the team. From launch pads to remote sensing satellites, from space stations to lunar bases, civil engineers can and should play a significant role in design requirements, engineering, testing, assembly, and operation. The Aerospace Division of the ASCE should take the lead to insure that civil engineers are prepared to meet the challenge.     

Structural Testing of Prestressed Concrete Girders from the Lake View Drive Bridge

On the evening of December 27, 2005, the fascia beam supporting the east-side parapet wall of the third span of the Lake View Drive Bridge failed under the action of dead load. This paper describes the structural testing and posttest forensic examination of two girders recovered from the partially collapsed Lake View Drive Bridge. The objective of this paper is to describe the tests conducted and report the observations. An interior and an exterior prestressed concrete adjacent box girder were tested to failure in flexure. Prior to testing, an extensive visual inspection was conducted to assess the extant condition of the 42-year-old girders. Following testing, the girders were sawcut near their failure regions to permit an extensive forensic investigation. Conclusions based on the pre- and posttest inspections and test results are presented. Recommendations intended to reinforce issues that need to be considered in the bridge inspection and rating process of similar structures are presented.     

Plant Sucrose Transporters from a Biophysical Point of View

T The majority of higher plants use sucrose as their main mobile carbohydrate. Proton-driven sucrose transporters play a crucial role in cell-to-cell and long-distance distribution of sucrose throughout the plant. A very negative plant membrane potential and the ability of sucrose transporters to accumulate sucrose concentrations of more than 1 M indicate that plants evolved transporters with unique structural and functional features. The knowledge about the transport mechanism and structural/functional domains of these nano-machines is, however, still fragmentary. In this review,the current knowledge about the biophysical properties of plant sucrose transporters is summarized and discussed.     

Analysis of Martensite Pole Figure from Crystallographic View Point

Martensite pole figures from three different steels have been studied using electron backscatter diffraction (EBSD) and mathematical models to show that the two stage transformation theory is not necessary for correct prediction of pole figure and popular orientation relationships, like Kurdjumov–Sachs. These theories can give misleading prediction. It has been proved that the use of correct crystallographic data can lead to a better texture prediction. The typical features of a pole figure in a {2 5 2} _γ habit system have been studied in detail.

View from the bridge: reflections of a recovering staff model HMO psychiatrist

The phenomenon of managed care is misperceived by its critics as a movement. Atheoretical in nature, and driven by spiralling health care costs, it represents the introduction of an executive function into the free-for-all (a supremely ironic term) of American medicine. Part one of this article traces the origin and development of the mental health carve-out, relating it to its antecedents, and describing its 3 overlapping phases: utilization review, discounted fees, and network development and management. Part two describes the key concept of the continuum of care and the role of the case manager in monitoring a care episode. The article concludes by anticipating seven future trends and calling for mental health leadership to recognize and ally with the need to manage resources in a more rational and efficient manner.

Processing of Agglomerated Red Filter Dust in the Converter Operation from Metallurgical Point of View

Red filter dust (RFD) from steel works contains up to 50 mass% iron, which therefore can serve as raw material for steel production. It should be possible to recycle a fraction of the RFD in the converter process of a steel works wherein also scrap for recycling is used. The aim was the investigation of the reduction behavior of the iron oxide in the RFD. This was accomplished by contact of the dust with pig iron containing up to 3.9 mass% carbon and also by addition of bio‐char to the dust, creating self‐reducing briquettes. The experimental results were compared to the theoretical achievable iron oxide reduction. The reaction time of selected briquettes was calculated by a kinetic approach. Additional the behavior of lead and zinc in the dust was investigated. The mass balance of the converter process indicated the influence of the dust recycling especially regarding the zinc mass flow.     

Durability of Brick Lined Steel Ladles from a Mechanical Point of View

The refractory lining in steel ladles is exposed to chemical and mechanical loads during the heats. Mechanical loads develop from the thermal expansion of the refractories which is confined either by the steel construction or by regions of different temperature within the refractory material. The aim of this work was the investigation of factors influencing the mechanical durability of refractory steel ladle linings and the clarification of failure mechanisms. Especially irreversible strains at the hot face of the working lining caused by compressive stresses induce an opening of joints at the hot face. The irreversible strains reduce the compressive stresses in circumferential direction and increase the probability of tensile failure. A further effect of the irreversible strains is a reduced stability of the working lining. Of special interest in this context is a possible controlled expansion of bricks to counterbalance the irreversible compressive strains.     

从汽车衡检定看《数字指示秤检定规程》修订的必要性

对汽车衡在执行标准JJG 539—97《数字指示秤检定规程》进行检定过程中遇到的问题进行探讨,指出该规程因制定时间比较早,目前已经不太适应汽车衡的检定,期望相关管理部门考虑对此进行修订,以更好地适应汽车衡的检定。

中国古典文学名著<西游记>中懦弱短视的"御弟"唐僧在当今一些日本影视动漫版<西游记>中被大胆变异成女性,并成为爱情的主角,引发了中国观众的强烈反感.但其在日本高居不下的收视率不禁引发人们深层次的思考.分析唐僧形象女性化变异的原因可以看出,是日本神道教与佛教介入其中,将唐僧形象加以分解后进行集体再创造和重新诠释,从而形成了女性唐僧这一形象变异体.
