Ochratoxin A and Brewing Technology: A Review

Ochratoxins are a hazardous group of mycotoxins produced as secondary metabolites by several fungi of the Aspergillus and Penicillium families. Ochratoxins have been detected and determined in foods and beverages, including barley, malt and beer, at ppb levels. Varied analytical methods have been developed for the detection of ochratoxin A [OTA] in cereals and beer. Ochratoxin A has hazardous effects on health and has been classified within group 2B, as a possible human carcinogen by IARC. Scientists have expressed great concern about the presence of ochratoxin A in foods. In this article, chemical structure, biosynthesis by microorganisms, analytical methods for testing, regulations, changes during brewing, and detoxification of OTA, are reviewed. Beer poses very little risk as a source of ochratoxin in the diet.     

Biochanin A: A phytoestrogen with therapeutic potential

BackgroundBiochanin A, a bioactive constituent obtained from Trifolium pratense (red clover) and many other legumes, has drawn considerable attention of researchers in recent years owing to its wide spectrum of pharmacological activities. It exhibits multifaceted biological activities viz. neuroprotective, anticancer, anti-oxidant, anti-inflammatory, osteogenic, anti-hyperglycemic and protective effect on endothelial cell integrity and function. The therapeutic potential of this isoflavone has been explored in various in vitro, in vivo and ex vivo models. However, in vivo evidences are limited due to its low bioavailability.Scope and approachThis review highlights the therapeutic potential of biochanin A along with the methods of its extraction, isolation and identification, and supplemented further with approaches to enhance its bioavailability. The various molecular targets through which it displays different biological roles have also been spotlighted.Key findings and conclusionBiochanin A, a potent molecule exhibits its molecular actions by targeting different phases of cell cycle; various signal transduction pathways involved in cell proliferation, cell differentiation, apoptosis, nuclear factor kappa B (NF-κB), and peroxisome proliferator-activated receptor gamma (PPAR γ). In cancer cells, biochanin A suppresses the activation of Akt and mitogen activated protein kinases (MAPKs) and also increases the expression of estrogen receptors (ERs) in other cellular models. Despite its huge potential, the clinical use of this isoflavone is limited due to its low bioavailability. Various strategies developed to enhance the bioavailability of biochanin A have led to foresee the potential of this promising molecule to increase the health benefits in future.

Carbohydrate-Concanavalin A Interactions A Note on Recent Studies

A large number of biologically important systems can be studied by use of the carbohydrate complexing protein, concanavalin A. Carbohydrate-concanavalin A interactions have proven useful in examining the structure and functions of isolated carbohydrate-containing compounds and to study the surface properties of cell membranes. Present knowledge concerning the carbohydrate binding site of concanavalin A is reviewed. Based on current data, it was suggested that an accurate model to account for concanavalin A-carbohydrate complex formation could not be constructed.     

A VISCOSITY MODEL FOR A COOKING DOUGH1

A five parameter semi-empirical model is developed describing the apparent viscosity of a cooking dough as a function of applied shear rate, temperature, and time-temperature history. Such a model will be useful in the mathematical simulation of the extrusion cooking process. Other factors which can affect the apparent viscosity of a cooking dough are also discussed. The viscosity model is verified and the constants evaluated for a typical dough by combining the results of Amylograph and capillary viscometer experiments.

A superantigen bioassay to detect staphylococcal enterotoxin A

Current detection methods for enterotoxins of Staphylococcus aureus are labor intensive and limited in sensitivity. Furthermore, these immunochemical protocols fail to adequately detect heat-treated enterotoxins. Staphylococcal enterotoxins cause severe gastrointestinal illness at relatively low concentrations and retain toxigenicity even after heat treatment. Presented here is a novel method to detect staphylococcal enterotoxin A (SEA). This method is a bioassay that exploits SEA's activity as a superantigen in that it induces in cytotoxic T lymphocytes a cytotoxic response against SEA-bound Raji cells. Target cell death is assayed colorimetrically with the CytoTox 96 cell lysis detection kit. In the experiments presented here, this bioassay was also able to detect heat-treated SEA, albeit with a slight compromise in sensitivity. This system detected SEA at picomolar concentrations. Because of the sensitivity of this assay, it is conceivable that it could be incorporated into current detection methods as a confirmatory test.     

PERFORMANCE OF A STEAM EDUCTOR IN A PILOT BREWERY

Steam eductors have the potential for reducing the demand for steam during the evaporation of wort by up to 50% with low capital expenditure and without affecting the process of wort boiling or the flavour of beer. The theory describing the operation of eductors is discussed together with sample calculations for the prediction of performance. A small unit was installed on a kettle in the Holyrood pilot brewery to demonstrate the practicality of the principle and to obtain experience in operation. The unit performed in a manner in keeping with theory and had an efficiency of 90%. There is a great likelihood that the economic advantage of steam eductor systems will be very close to theoretical prediction.
