The purpose of this study was to investigate whether smokers outside buildings with work-place smoking bans smoke "harder" than those smoking in social settings. An unobtrusive random observational study of smokers followed by structured interview was used, with 143 smokers taking smoking breaks outside their office buildings and 113 smokers in social settings. The main outcome measurements were number of puffs per cigarette and cigarette smoking duration. The mean number of puffs per cigarette for the office building group was 18.7% greater than that for the social settings group (10.7 +/- 3.2 vs. 8.7 +/- 2.7, t = 5.58, df = 253, p < 0.001); 74.8% of smokers outside offices took more than the mean number of puffs for the group compared to 42.5% of smokers in social settings (chi 2 df 1 = 26.31, p < 0.0001). Mean cigarette smoking duration was 30.4% shorter for the work-place group than the social settings group (3.9 +/- 1.2 minutes vs. 5.6 +/- 2.6 minutes). Of smokers outside offices, 55.2% had a cigarette smoking duration between 3 and 4.59 minutes, while 53.1% of smokers in social settings took > or = 5 minutes to smoke the observed cigarette (chi 2 df 2 = 31.55, p < 0.0001). Smokers who scored at the 75th percentile on the Fagerstrom Tolerance Scale took a mean 9.5 +/- 2.6 puffs per cigarette compared to 9.3 +/- 2.7 puffs by those who scored in the 25th percentile on the scale (t = 0.34, df = 145, p = 0.73). Regardless of degree of nicotine dependency, smokers leaving work-stations to smoke outside buildings smoked their cigarettes nearly 19% "harder" than cigarettes smoked in social settings. The individual and public health benefits of reduced smoking frequency engendered by work-place smoking bans may be lessened by policies which allow smokers to take smoking breaks. 相似文献

Ozone exposure results in an acute decrease in the serum levels of thyroid hormones; the physiologic sequelae of this are unclear. Whereas thyroid hormone supplementation appears to benefit pulmonary function in septic, oxyradical models of injury, thyroid hormone increases ozone toxicity. We demonstrated an increase in metabolic rate and pulmonary injury in lungs from ozone exposed, T3 treated animals. This was evidenced by an increase in pulmonary weight gain, vascular perfusion pressure, and decrease in compliance in the supplemented animals. However, an increase in alkane generation, as an index of lipid peroxidation, was not seen in the ozone exposed, hormonally treated animals. This suggests that although thyroid hormone supplementation increases metabolic rate and ozone toxicity, an increased rate of lipid peroxidation plays a minimal role. 相似文献

Recent evidence indicates that neocortical electrocorticographic (ECoG) activation depends critically on cholinergic (ACh) inputs from the basal forebrain and serotonergic (5-hydroxytryptamine, 5-HT) inputs from the midbrain raphe to the neocortex. It appears that ACh and 5-HT block synchronized ECoG activity and induce cortical activation by a direct, local action in the neocortex that is not mediated by secondary systems. Concurrent blockade of cholinergic and serotonergic inputs to the cortex abolishes all ECoG activation, suggesting that: (a) together, ACh and 5-HT are essential for cortical activation to occur; and (b) other systems cannot maintain cortical activation in the absence of the cholinergic and serotonergic inputs. Nevertheless, additional neural systems (amygdala, locus coeruleus-noradrenaline (-NA), superior colliculus, orbitofrontal cortex, dopamine, histamine, glutamate) also modulate the ECoG and can produce cortical activation. In contrast to ACh and 5-HT, however, none of these systems are essential for the maintenance of cortical activation. Further, rather than producing activation via direct inputs to the neocortex, these systems appear to produce cortical activation by stimulating the cholinergic or serotonergic inputs to cortex. Thus, neocortical activation is maintained by multiple, parallel systems; cholinergic and serotonergic pathways are essential for activation and produce activation directly and locally in the neocortex, whereas additional pathways are not essential and contribute to activation indirectly by acting through these two direct activating inputs to the cortex. 相似文献

The effects of dietary gentian violet upon certain physiological parameters of broiler chickens were studied. Gentian violet exhibited no effect upon growth rates or feed conversion ratios at dietary levels of 16, 32, or 64 micrograms/g. In vitro intestinal absorption of methionine and glucose was also unaltered. Dietary gentian violet significantly increased hemoglobin concentration without an effect on packed cell volume. Furthermore, commercial gentian violet containing preparations, when incorporated into the diet, resulted in increased intestinal absorption of Fe59 but this increase was dependent upon type of inert carrier used. Dietary gentian violet alleviated some of the growth suppression caused by dietary aflatoxin; however, no effect was observed on plasma pigmentation. These data suggest that dietary gentian violet possesses effects other than those for which it has been traditionally employed. 相似文献

PURPOSE: We conducted this study to examine differences in characteristics of immunoreactivity for free PSA and alpha(1)-antichymotrypsin complex PSA (ACT-PSA) as well as in compositions and concentrations of PSA reference materials among commercially available PSA kits. METHODS: Fractionated serum samples using a Sephacryl S-200 column were measured by Tandem-R, Delfia-PSA, Ab bead PSA, ACS-PSA, Markit-M and gamma-seminoprotein (gamma-Sm) kits. The calibrators of Tandem-R, Delfia-PSA, Ab bead PSA and Markit-M were fractionated by the same method and measured by Tandem-R. The calibrators of Delfia-PSA, Ab bead PSA and Markit-M and control serums of ACS-PSA were measured by Tandem-R. RESULTS: Although the characteristic of immunoreactivity of Tandem-R, Delfia-PSA, and Ab bead PSA were found to be similar, they were not shown identical. ACS-PSA was proved to recognize free PSA greater than above three PSA kits, while Markit-M could scarcely detect free PSA. gamma-Sm recognized only free PSA. The calibrators of Tandem-R, Delfia-PSA, Ab bead PSA and Markit-M were proved to be only free PSA. The linear correlation was obtained between Tandem-R and Delfia-PSA or Ab bead PSA or Markit-M. The ratio of Delfia-PSA to Tandem-R, Ab bead PSA to Tandem-R and Markit-M to Tandem-R was 0.66, 0.93 and 2.2, respectively. With regard to relation of ACS-PSA and Tandem-R, two ratios of 0.22 and 0.25 were obtained between the two kits according to the different concentrations of control sera. CONCLUSION: The present studies suggest that the difference in PSA values among the commercial PSA kits results from (1) different characteristics of immunoreactivity for ACT-PSA and free PSA among PSA kits, (2) compositions of PSA calibrators among the kits, and (3) different concentrations of PSA calibrators among the kits. 相似文献

Four experiments were conducted to determine the effects and interactions of feeding different levels of vitamins A, cholecalciferol (vitamin D3), and E on broiler chicks. In Experiment 1, chicks were fed marginal vitamin D3 (500 IU/kg) and increasing dietary levels of vitamin A (5,000, 10,000, 20,000, 40,000, 80,000, and 160,000 IU/kg). Bone ash was reduced by 10,000 IU/kg of vitamin A in the diet and at vitamin A levels above 20,000 IU/kg of diet body weight was reduced. In Experiment 2, two levels of vitamin A (1,500 and 15,000 IU/kg) and six levels of vitamin E (10, 500, 1,000, 2,500, 5,000, and 10,000 IU/kg) were added to the basal diet. High levels of vitamins A and E significantly (P < 0.001) reduced bone ash. The vitamin A x E interaction was significant (P < or = 0.05) for rickets. In Experiment 3, the same two levels of vitamin A as Experiment 2 and six levels of vitamin D3 (500, 1,000, 1,500, 2,000, 2,500, and 3,000 IU/kg) were added to the basal diet that contained 10,000 IU/kg of vitamin E. Body weight and bone ash were increased by increasing vitamin D3 with a corresponding reduction (P < or = 0.05) in rickets. In Experiment 4, three levels of vitamin A (1,500, 15,000, and 45,000 IU/kg), three levels of vitamin D3 (500, 1,500, and 2,500 IU/kg), and three levels of vitamin E (10, 5,000, and 10,000 IU/kg) were added to the basal diet. Significant negative responses (P < or = 0.05) to increasing dietary vitamin A were observed for bone ash, rickets, and plasma and liver vitamin E. A significant (P < 0.001) increase in bone ash and plasma calcium with a corresponding reduction in rickets was observed by increasing vitamin D3. Increasing dietary vitamin E adversely affected (P < or = 0.01) bone ash, plasma calcium, and plasma and liver vitamin A concentrations. Six isolates of Mycoplasma synoviae, identified as WVU 1853, K1968, K1858, 92D8034, F10-2AS, and FMT, were compared for pathogenicity in broiler chickens. Specific-pathogen-free chickens were inoculated, in two groups of 20, with each isolate by footpad or eyedrop inoculation at 1 day of age and were examined at necropsy 7, 14, 28, and 42 days postinoculation. Specimens were taken for histopathology, culture, polymerase chain reaction assay, and hemagglutination-inhibition serology. Isolates were grouped according to pathogenicity on the basis of differences in lesion development and tissue distribution in the respiratory system, other viscera, and the skeletal system. K1968 (pathogenic) induced lesions in all sites examined in both the footpad and eyedrop inoculation groups. It was detected in all sites following footpad inoculation and in all sites except viscera following eyedrop inoculation. WVU 1853, K1858, and 92D8034 (moderately pathogenic) induced lesions and were detected in all sites following footpad inoculation. With eyedrop inoculation, lesions were identified only in upper and lower respiratory sites, and organisms were detected only in upper respiratory sites. F10-2AS (moderately pathogenic) was similar; however, footpad inoculation failed to induce visceral lesions or permit organism detection in any site. F10-2AS was detected in upper and lower respiratory tissues following eyedrop inoculation. FMT (mildly pathogenic) induced only upper respiratory lesions when either footpad or eyedrop inoculation was used, and detection was restricted to upper respiratory sites following eyedrop inoculation. These results are useful in comparative evaluations of the virulence of other M. synoviae isolates and form a basis for characterization of virulence factors of M. synoviae. 相似文献

The present experiments were undertaken to investigate the effects of environmental temperatures on growth, abdominal fat content, rate of muscle protein turnover, and heat production in tube-fed intact male broiler chickens. Plasma concentrations of thyroxine (T4), triiodothyronine (T3), and corticosterone (CTC) were also examined. Chicks (15 d old) were kept at different environmental temperatures (16, 19, 22, 25, 28, 31, and 34 degrees) and given the experimental diet (200 g crude protein/ kg, 13.57 MJ/kg metabolizable energy) by tube three times daily throughout the 12 d experimental period. In the hot conditions, except for 34 degrees, body-weight gain was significantly higher than in the cold conditions. Thus, food conversion ratios (food:gain ratios) were lower when the birds were exposed to the hot conditions other than 34 degrees. Likewise, abdominal fat content was significantly increased, and heat production was lower in the groups kept under the hot conditions other than 34 degrees. The rate of skeletal muscle protein turnover and plasma concentration of CTC were decreased when the birds were exposed to hot conditions other than 34 degrees, suggesting a role of CTC in the regulation of muscle protein turnover. Plasma concentrations of T4 and T3 were significantly decreased as environmental temperature increased. These results clearly show that plasma concentrations of thyroid hormones and CTC are associated with accelerated muscle protein turnover and heat production. 相似文献

Experiments were conducted to determine the ability of a hydrated sodium calcium aluminosilicate (T-Bind) sorbent to reduce the toxicity of aflatoxins (AF) or T-2 toxin in male broiler chickens from day of hatch to 21 d of age. In Experiment 1, the sorbent was added at 0.250 or 0.375% to diets containing AF at 5 or T-2 toxin at 8 mg/kg of diet. When compared with controls, AF reduced BW gain by 27% and T-2 toxin reduced BW gain by 17%. The addition of the sorbent at 0.250 or 0.375%, in the absence of added mycotoxins, did not alter the performance of the chicks. The sorbent reduced the toxic effects of 5 mg AF/kg of diet on BW gain by 43% but did not significantly diminish the toxic effects of 8 mg T-2 toxin/kg of diet. The decreased efficiency of feed utilization and the increased relative organ weights caused by AF were significantly diminished to differing degrees by the sorbent. Oral lesions caused by T-2 toxin were not affected by the sorbent. In Experiment 2, the sorbent was added at 0.80% to a diet containing 8 mg T-2 toxin/kg of diet. The sorbent did not diminish the toxic effects of T-2 toxin when added at 0.80% of the diet. These data demonstrate that this specific sorbent can provide protection against the toxicity of AF, but not T-2 toxin, in young broiler chicks. 相似文献

Chicken carcasses from a supermarket and from a poultry abattoir were examined using methods designed to isolate as many strains of campylobacters and related organisms as possible. Strains of arcobacter, but no campylobacters, were isolated from every carcass after enrichment. Campylobacter jejuni subsp. jejuni was isolated from all carcasses examined by direct plating and other Campylobacter-like strains were isolated from nine out of 15 abattoir carcasses by direct plating but not after enrichment. Only the Camp. jejuni subsp. jejuni strains could be identified to species level using a readily available identification scheme and/or a commercial identification kit. Examination of caecal contents from the 15 abattoir poultry yielded Camp. jejuni subsp. jejuni and Campylobacter-like strains from 15 and eight by direct plating, and from six and nine after enrichment, respectively. Four sites in the intestine of the abattoir birds (60 samples) were examined for arcobacters and only one strain was isolated. This indicates that arcobacters are probably not normal inhabitants of the poultry intestine. Poultry is a rich source of other campylobacteria besides the thermophilic Campylobacter spp. 相似文献

OBJECTIVE: To compare the Bard BTA (bladder tumour antigen) test with voided urine cytology (VUC) in the diagnosis of recurrent bladder cancer (BC). METHODS: Urine specimens for the BTA test and VUC were collected on the same day as before cystoscopy from patients undergoing routine surveillance cystoscopy for recurrent BC. The pathologists performing VUC were blinded to the results of the BTA test. RESULTS: BC was identified by cystoscopy and biopsy in 39 of 164 study participants. The overall sensitivities of the BTA test and VUC were 54 and 28%, respectively (p < 0.05). The BTA test was more sensitive than VUC for all tumour stages and grades. For > or = T2 tumours and grade 3 tumours, respectively, the difference was statistically significant (p < 0.05). The specificities of the BTA test and VUC were 92 and 97%, respectively. Both a false-positive BTA test and VUC were found to predict recurrence. CONCLUSION: The BTA test is equal or superior to VUC in the detection of BC in patients undergoing routine surveillance for recurrent BC. 相似文献

Effect of T3 therapy on tanned red cell agglutinating thyroglobulin (TRC-TG) antibodies in 10 obese subjects without apparent thyroid disease was investigated. Six other obese subjects without thyroid dysfunction and of approximately the same mean age who also had circulating TRC-TG antibodies served as control subjects and were untreated. In vitro thyroid tests (TSH, total and free T4) performed before T3 therapy, as well as clinical examination, showed thyroid function to be normal in all subjects, and there was no evidence of thyroiditis. TRC-TG antibodies were present in low to moderate titers of 40-1280 in control subjects as well as in subjects selected for T3 treatment. Therapy with T3 was started at 50 mug/day and gradually increased to a maximum of 250 mug/day, depending on clinical needs. T3-treated as well as untreated obese control subjects were all maintained on a high protein, low fat, low carbohydrate diet. Duration of T3 therapy varied from 2-8 mo, and in all but one T3-treated subject, TRC-TG antibodies completely disappeared. In the one exceptional case, TRC-TG antibody titer decreased from 1280 to 80 after 7 mo of therapy. In non-T3-treated obese control subjects, antibody titers remained at the same levels throughout the observation period, thereby indicating a lack of spontaneous regression of circulating immune response. Therapy with T3, by inhibiting TSH, may have caused regression of inapparent immunologic thyroid lesion, thus leading to the disappearance of circulating TRC antibodies; alternatively, T3 specifically may have accelerated catabolism of thyroid antibodies. The latter possibility is favored in the absence of clinical and laboratory evidence of thyroiditis in T3-treated subjects. 相似文献

1. The effects of varying the dietary arginine:lysine (Arg:Lys) ratio for broiler chickens at thermoneutral and high temperatures was studied in a series of 5 experiments which measured intestinal epithelial transport or evaluated growth and food efficiency with practical diets or diets supplemented with L-arginine free base. 2. The growth studies showed that increasing the Arg:Lys ratio at high temperatures produced consistent improvements in food conversion without any loss in growth. 3. Increasing dietary sodium chloride concentration reduced the Arg:Lys ratio necessary for optimum food conversion. 4. Food conversion responses were improved whether L-arginine free base was used as a dietary supplement in place of an inert filler or practical diets with differing ingredients were used to vary the Arg:Lys ratio. 5. In the presence of an equimolar concentration of lysine the uptake of arginine by the intestinal epithelium of heat-stressed broilers was reduced significantly compared with that of broilers at thermoneutral temperatures. 6. The results indicate that the ideal amino acid balance for broilers varies with ambient temperature. 相似文献

We investigated the effects of thyroid hormone treatment on morphology, growth behaviour, and cytoskeletal structures of long-term cultured MC3T3-E1 cells. Morphological investigations were carried out on native cells by phase contrast microscopy and on epon-embedded semithin sections. The area covered by the cell and matrix layers (tissue-like area), percent extracellular matrix, average height of tissue-like area, and length and height of single cells were measured histomorphometrically on the cross sections. F-actin was analyzed histochemically and quantitated after fluorochrome-labeled phalloidin staining using confocal microscopy and fluorometry. Significant differences between control and T3-treated cells were found after confluency, but not in subconfluent cultures. Control cells continued to proliferate forming multilayers, and produced increasing amounts of extracellular matrix. In contrast, T3-treated cells stopped to proliferate forming two cell layers at the maximum. These cells were flattened, distinctly enlarged, and polygonal in shape. Histochemical staining for F-actin revealed three different staining patterns, depending on the position of the cell within the multilayer of control cultures. Basal cells contained a large number of thick stress fibers in parallel arrangement. Intermediate cells exhibited only a few thick actin filament bundles located at the outermost periphery. The superficial cells were characterized by a large number of thin, parallel-oriented microfilament bundles extending across the entire cytoplasm. The actin pattern of T3-treated cells resembled that of the basal cell layer of the control cells. The amount of F-actin increased with the prolonged T3 treatment. We conclude from these data that the known specific cellular responses to T3 treatment are accompanied by significant morphological alterations indicating pivotal effects of thyroid hormones on osteoblastic differentiation. 相似文献

Ninety-nine strains of Arcobacter spp., isolated from 10 chicken carcasses purchased from a supermarket and 15 chicken carcasses collected from a poultry abattoir, were speciated using a variety of phenotypic identification methods. All were tested using API Campy test strips and the 16-test (Preston) identification scheme developed for campylobacters. Fifty strains were selected for examination using a more comprehensive probabilistic identification scheme, and the identity of representative strains confirmed by protein profiling using SDS-PAGE. All 25 carcasses yielded Arcobacter butzleri. Three supermarket and 10 abattoir carcasses also carried A. cryaerophilus, and two abattoir carcasses carried A. skirrowii. The API Campy scheme proved unsatisfactory for identifying these strains: only 20 of 99 strains were accurately identified, all of which were A. cryaerophilus, the only Arcobacter sp. included in the database. Moreover, 76 of 99 strains were misidentified. The 16-test scheme identified all the arcobacter strains as A. cryaerophilus, since neither A. butzleri nor A. skirrowii had been described when the scheme was developed. The computer-assisted probabilistic scheme succeeded in identifying all but one strain, the identity of which was clarified by the use of SDS-PAGE. To our knowledge this is the first time that arcobacters other than A. butzleri have been reported in poultry meat or any other food of animal origin. Their high prevalence in poultry products may be of significance to public health. 相似文献

The efficacy of exogenous IGFs to stimulate growth and modulate protein and fat deposition was examined in a number of broiler chicken lines. From around 600 g body weight the chickens received a continuous infusion of vehicle (0.1 M acetic acid), human recombinant IGF-I or [Gly1]IGF-II at 300 microg/kg body weight per day, or a combined infusion of 150 microg/kg/day of each IGF for 2 weeks. Experiment 1 used commercial broiler female chickens and included measurements of nitrogen balance, Ntau-methylhistidine excretion and muscle protein synthesis rates. In Experiment 2 the same treatments were applied to three experimental lines of chickens selected for high food consumption (relatively fat), high food utilisation efficiency (relatively lean), or at random (control). IGF-I, but not IGF-II, significantly increased growth rate and food utilisation efficiency by around 10-15% in each experiment, an effect which was consistent across all genotypes. Nitrogen balance was significantly increased by IGF-I in Experiment 1 as was carcass nitrogen content in Experiment 2, indicating that the increased growth was in lean tissue. Carcass fat was consistently reduced in chickens receiving IGF-I and was related to the levels of circulating IGF-I (r2 = 0.30, P < 0.01) but not triiodothyronine. Protein synthesis rates were unaffected by treatment and could not account for increased growth rate. However, there was a significant reduction in Ntau-methylhistidine excretion indicating a reduced rate of muscle protein breakdown in IGF-I-treated chickens (1. 56%/day vs 2.05%/day for IGF-I-treated vs controls, P < 0.05). The efficiency of feed utilisation was inversely related to the rate of protein breakdown (r2 = 0.25, P < 0.01). In conclusion, these experiments are the first to report an enhancement of growth and food utilisation efficiency by broiler chickens receiving exogenous IGF-I. Our results show that IGF-I may be important in controlling the growth and efficiency of food utilisation of young chickens at least in part by modulating the rates of protein breakdown. 相似文献

Previous research has identified cecal and intestinal contents as sources for Campylobacter contamination of broiler carcasses in the processing plant. During the present study, we evaluated the crop contents of preharvest market-age broilers as a potential reservoir of field-derived Campylobacter in the processing plant. Crops were collected aseptically from 40 randomly selected market-age broilers in each of nine commercial broiler flocks. Ceca were collected from broilers in six of the same flocks for comparison with the crop samples. The presence of Campylobacter in the crops and ceca was determined by enrichment culture in Bolton broth followed by culture on Campy-Ceflex plates. Campylobacter was isolated from the crop contents of broilers in seven of the nine flocks and from the cecal contents in three of six flocks. The incidence of Campylobacter-positive crop samples among all birds evaluated (224/359; 62%) was significantly higher (P < 0.001) than the number of positive cecal samples (9/240; 4%). The results indicate that the incidence of Campylobacter contamination of crop contents may exceed that of the cecal contents by as much as 37-fold in some broiler flocks, and may represent a critical preprocessing control point in reducing Campylobacter entry into the processing plant. 相似文献

Studies were conducted to evaluate spent hen meal (SHM) produced in commercial rendering plants as a nutrient source in diets for broiler chickens. Utilizing previously determined nutrient composition values, including digestible amino acid and TMEn content, diets were formulated to include 0, 5, 10, and 15% of SHM from three different locations. In the first experiment, conducted in battery pens from 1 to 21 d posthatch, diets were formulated with digestible amino acid requirements set at 90, 95, or 100% of NRC (1994) total amino acid requirements. In the second experiment, conducted in floor pens from 1 to 49 d posthatch, diets were formulated with digestible amino acid requirements set at 95% of NRC (1994) total amino acid requirements. Samples of birds from the second experiment were processed to determine the possible influence of SHM inclusion on carcass yield. Results of the present studies indicate that SHM from commercial rendering facilities can be utilized in diets for growing broiler chickens provided adjustments are made in nutrient content and digestibility. When formulated on the basis of digestible amino acid content, levels of SHM up to 10% appear acceptable based upon body weight, feed conversion, bone ash, and carcass yield. Higher inclusion rates generally reduced performance. It is apparent that differences in nutritional quality may exist among products produced by different rendering facilities; however, evaluation of products to assess nutrient quality may be difficult under commercial conditions.
