Increased liver blood flow (LBF) resulting from elevated feed intake in lactating dairy cows may increase steroid metabolism. Continuous infusion of bromosulphthalein (BSP; specifically metabolized in liver) was used to measure LBF. Similarly, progesterone (P4) and estradiol-17beta (E2) were administered by continuous infusion. Circulating concentrations at steady state were used to calculate the metabolic clearance rate (MCR) of BSP, P4, and E2. Experiment 1: Variation in LBF was determined in thee nonlactating and four lactating cows over 3 d at 3 to 5 h after feeding. Coefficients of variation ranged from 14 to 31% among cows within day and from 4 to 8% within cows across days. Experiment 2: Six nonlactating cows were used in a 3 x 3 Latin-square design with three feed regimens: no feed, 0.5 maintenance diet (M), and 1.5 M. Experiment 3: Eight lactating cows were used in a 4 x 4 Latin-square design with four feed regimens: no feed, 0.5 M, 1.5 M, and 2.2 M. In experiments 2 and 3, LBF and MCR of P4 increased immediately after feed consumption and increases persisted longer at higher intakes. The LBF reached a maximum at 2 h after feeding and MCR of P4 reached maximum at 3 h after feeding with a positive correlation (r = 0.92) between LBF and MCR for P4. Experiment 4: A crossover design was used to determine MCR of E2 in unfed or full-fed lactating dairy cows. The MCR of E2 increased immediately after feeding and stayed elevated throughout the 4.5-h infusion period. Thus, LBF and steroid metabolism were acutely elevated by feed consumption in lactating and nonlactating cows. Higher rates of LBF and steroid metabolism in lactating than in nonlactating cows may indicate chronic effects of higher feed intakes as well.  相似文献

This study was conducted to investigate hormonal imbalances preceding stillbirths and dystocia in primiparous heifers. The study was conducted between 2003 and 2004 on a German dairy farm, including 433 heifers. Starting 3 wk before calving, a weekly blood sample was collected. At calving, another blood sample was obtained, and the calving ease (grade 0 = unassisted to grade 2 = heavy pull with mechanical calf puller), sex, birth weight, as well as vitality status (stillborn, alive) of the calf were recorded. The blood serum was analyzed for estradiol-17β and progesterone concentration. At parturition, the measured estradiol-17β concentration was greater in heifers delivering bulls than in those with female calves and was increasing with greater birth weight of the calf and increasing calving difficulty score. Already 2 wk before calving, the serum estradiol-17β concentration was significantly smaller in heifers with stillborn than live calves. On the other hand, the progesterone concentration was greater 2 wk before calving in heifers with stillborn calves, but it was unaffected by the birth weight or sex of the calf or the calving difficulty score. Stillborn and live calves did not differ in birth weight or pregnancy duration. The smaller estradiol-17β concentrations of the heifers with stillborn calves could indicate an abnormality of the placenta or an abnormality of hormonal signals from the calf to the placenta in the weeks before the calving.  相似文献

Natural sex hormones, estradiol-17 beta (EST), progesterone (PRO) and testosterone (TES), in sixty domestic bovine muscles and forty bovine muscles imported from USA and Australia were determined by radioimmunoassay. The EST, PRO and TES levels (mean +/- standard deviation) in domestic samples (n = 60) was 1.15 +/- 1.87 ppt, 3.19 +/- 5.80 ppb and 30.9 +/- 122.1 ppt on a whole basis, respectively. The hormone levels in muscles was presumed to reflect sex, estrous cycle, and so on. The hormone levels in imported bovine muscles (n = 40) were 3.33 +/- 2.83 ppt (EST), 0.52 +/- 0.50 ppb (PRO) and 8.8 +/- 13.0 ppt (TES), respectively.  相似文献

An intravaginal sponge impregnated with 500 mg estradiol-17 beta and 1000 mg progesterone to induce lactation in nonpregnant dairy cattle was investigated. Sponge retention (greater than 95%) was achieved by its attachment to a nylon collar. Treatment with the sponge for 10 days resulted in lactogenesis in 25% of treated cows compared with an 89% success rate when an adjunct injection of dexamethasone esters (20 mg intramuscular) was given on day 6 or a 96% success rate when reserpine injections (2.5 mg intramuscular) were given on days 6, 8, and 10 10 after sponge insertion. Milking was begun 12 days after sponge insertion (2 days after removal). Peak milk yield was unaffected by adjunct treatments but was greater in spring (mean 11.5 kg/day) than in autumn treated cows (mean 3 and 6 kg/day) in 2 yr of trials. Concentration of estradiol-17 beta in plasma rose rapidly after sponge insertion to a peak similar to that in cows in late pregnancy. Measurement of estradiol-17 beta concentration in milk indicated less, even at first milking than in commercially distributed pasteurized milk. Milk fat and protein content were higher in milk from induced lactations compared with that from normally calving cows on the same day.  相似文献

Data from artificial insemination, rectal palpation, and hormone assays were used to characterize postpartum reproductive activity in 54 dairy cows. Progesterone and estradiol-17 beta were measured in milk samples collected for 120 d (Trial 1) or 65 d (Trial 2). Progesterone was higher and estradiol was lower in milk than in serum. Values for both hormones in milk were highly correlated with those in serum. Most cows (64%) had short first luteal phases (less than or equal to 12 d). First rise (28 d) in progesterone was later (33.4 vs. 24.9 d) for cows having short rather than normal (greater than 12 d) luteal phases. Cows were classified as having a short luteal phase followed by a normal luteal phase or as having normal luteal phases for the first two estrous cycles. Estradiol for the 6 d prior to each luteal phase was higher preceding the second phase than the short phase or those preceding both phases of cows with normal phases. Follicular function prior to ovulation, as measured by estradiol, was not responsible for short-lived corpora lutea. Concentrations of progesterone in milk in the late luteal phase prior to insemination were related to fertility.  相似文献

The aim of this study was to determine whether an increase in circulating estrogen concentrations would increase percentage pregnant per artificial insemination (PP/AI) in a timed AI protocol in high-producing lactating dairy cows. We analyzed only cows having a synchronized ovulation to the last GnRH of the Ovsynch protocol (867/1,084). The control group (n = 420) received Ovsynch (GnRH - 7 d - PGF - 56 h - GnRH - 16 h - timed AI). The treatment group (n = 447) had the same timed AI protocol with the addition of 1 mg of estradiol-17β (E2) at 8 h before the second GnRH injection. Ovarian ultrasound and blood samples were taken just before E2 treatment of both groups. In a subset of cows (n = 563), pressure-activated estrus detection devices were used to assess expression of estrus at 48 to 72 h after PGF2α treatment. Ovulation was confirmed by ultrasound 7 d after timed AI. Treatment with E2 increased expression of estrus but overall PP/AI did not differ between E2 and control cows. There was an interaction between treatment and expression of estrus such that PP/AI was greater in E2-treated cows that showed estrus than in E2-treated or control cows that did not show estrus and tended to be greater than control cows that showed estrus. There was evidence for a treatment by ovulatory follicle size interaction on PP/AI. Supplementation with E2 improved PP/AI in cows ovulating medium (15 to 19 mm) but not smaller or larger follicles. The E2 treatment also tended to improve PP/AI in primiparous cows with low (≤2.5) body condition score, and in cows at first postpartum service compared with Ovsynch alone. In conclusion, any improvements in PP/AI because of E2 treatment during a timed AI protocol appear to depend on expression of estrus, parity, body condition score, and size of ovulatory follicle.  相似文献

Concentrations of estradiol-17 beta in follicular fluid were correlated to follicular size, stage of estrous cycle, location of corpus luteum, and presence of large follicles. Paired ovaries were obtained from 481 nonpregnant cows at slaughter and follicles were classified as ipsilateral or contralateral to the corpus luteum. Follicular fluid estradiol-17 beta concentrations from 2494 small, 1485 medium, and 396 large follicles were quantified by radioimmunoassay. Stage of estrous cycle was estimated by visual examination of the corpus luteum. Follicles in stage 1 of the estrous cycle (d 1 to 4) had the highest estradiol-17 beta concentration and the smallest mean follicular diameter. Location of follicles relative to the corpus luteum had no influence on estradiol-17 beta concentrations. As follicular size increased, concentration of estradiol-17 beta also increased. The presence of a single large follicle did not affect the concentration of estradiol-17 beta in medium or small follicles. In contrast, if multiple large follicles occurred in the same cow, concentrations of estradiol-17 beta were significantly lower in medium but not small follicles.  相似文献

Lactations were induced in nonpregnant, nonlactating dairy cows by subcutaneous injections of estradiol-17 beta and progesterone for 21 d (.10 and .25 mg/kg body weight/d) and dexamethasone (.028 mg/kg body weight/d) on d 31 to 34. Milking was initiated on d 35. Each cow was biopsied two or three times during the experiment with five to eight mammary tissue biopsies on d 0, 7, 14, 21, 28, 35, 49, and 130. Mammary tissue preinjection had abundant connective and adipose tissues with limited lobuloalveolar structures. Beginning on d 7, there was decreased stroma, increased epithelial cell area, increased lobuloalveolar architecture, plus the accumulation of intracellular and intraluminal secretions which were high in lipid droplets. From d 7 through 35, these changes were progressive although variable among cows. Changes in activities of enzymes and concentrations of ribonucleic acid and deoxyribonucleic acid were gradual during this time but essentially paralleled histological development. Tissue samples during lactation (d 49 and 130) showed increased histological and biochemical development; development was maximal for d 130 samples. Fourteen of 15 cows that lactated had mean daily yields of milk more than 5 kg and yields of milk of 12 cows with projected or actual 305-d lactations were 63.0% of that during their previous natural lactations. Reasons for less yields of milk and for varied patterns of tissue development were not identified nor explained by concentrations of several selected hormones in plasma.  相似文献

Blood is the standard source for DNA analysis, but requires venipuncture of cows by veterinarian and tedious and costly DNA extraction. A procedure was developed for sampling of vaginal cells from cows, establishing a cell lysis protocol using robotics, and applying fluorescent analysis of genetic markers. Two insemination technicians collected vaginal cells from 254 elite Israeli Holstein cows located in 152 herds using commercial Catch-All sample collection brushes. Cells were lysed in a 400-microliters solution, and 5 microliters was used as template for polymerase chain reaction (PCR). Sensitivity of the PCR was enhanced using only 1 microliter of lysed cells. Eight markers of the International Society of Animal Genetics paternity panel were amplified in four separate PCR. ILSTS039, a marker for a quantitative trait loci on BTA14, was amplified in a separate reaction. Genotypes from one to nine genetic markers were obtained for 253 out of 254 samples, and 244 cows had genotypes for at least three markers (96%). Cows that did not inherit either paternal allele for at least two loci were considered not to be daughters of the sire listed. Fifteen cows met this criterion, for a paternity misidentification rate of 6.25%. The frequency of allele 225 of ILSTS039, which was associated with increased milk fat content, was 11.1% in the bull-dam population, similar to the 12% found in the cow population in Israel. The use of vaginal cells for genetic analysis is accurate, as demonstrated by replicated analysis and by comparison to individual and population analysis based on DNA derived from blood.  相似文献

The effect of early (5 month) or late castration (13 month) on meat quality of hypertrophied Piemontese cattle was investigated. Twenty four animals, equally divided into three groups (early castrated, EC; late castrated, LC; intact males, IM) were reared under the same experimental conditions and slaughtered at the same age. Twenty four hours after slaughter the pH was measured on the longissimus thoracis of the right side. After 11 days of ageing the following analyses were performed on the longissimus thoracis et lumborum: water, protein and ether extract contents, hydroxyproline content and collagen solubility, colour (L, a(L), b(L), hue and chroma), drip and cooking losses, Warner-Bratzler shear values and sensory analysis (appearance of the raw meat and eating qualities of the cooked meat). Compared with intact males, the castrates had lower water and hydroxyproline contents and higher contents of protein and ether extract. No significant differences were observed between early and late castration, except for cooking losses.  相似文献

This study was designed to evaluate whether decreasing circulating progesterone (P4) or increasing circulating estradiol-17β (E2) near the time of artificial insemination (AI) in an Ovsynch protocol would increase pregnancies per AI (P/AI) in lactating dairy cows. Six hundred nineteen lactating Holstein cows (n = 772 inseminations) received Ovsynch (GnRH-7 d-PGF-56 h-GnRH-16 h-timed AI). Cows were randomized in a 2 × 2 factorial experiment of 4 treatments to receive or not receive 25 mg of PGF 24 h after the standard PGF of Ovsynch, or 0.5 mg of E2 at the time of the final GnRH of Ovsynch, or both. Blood samples were collected 24 h after normal PGF and at final GnRH to evaluate circulating P4. Ovarian ultrasound was done at final GnRH to determine preovulatory follicle size. Ovulation was confirmed by ultrasound 5 d after AI. Treatment with additional PGF increased the percentage of cows that had complete luteal regression (95.6%) compared with control cows (84.6%). In contrast, additional PGF had no detectable effect on P/AI (control = 41.5% vs. + PGF = 44.7%). Supplementation with E2 increased expression of estrus (84.4 vs. 37.2%), but had no effect on overall fertility and even tended to have a negative effect on fertility in cows that ovulated to the second GnRH (control = 51.5% vs. +E2 = 44.0%). Thus, additional treatments with PGF or E2 during Ovsynch can be used to increase synchronization and expression of estrus during Ovsynch, although the lack of improvement in fertility makes these treatments unwarranted.  相似文献

为研究不同坡度的装卸台装卸牛对牛肉品质的影响,选取西门塔尔肉牛20头,随机分为4组,各组分别通过(30,40,50°)坡度装卸台装卸牛后,然后对牛进行屠宰,分析宰后牛肉pH值、肉色、失水率、蒸煮损失率和剪切力。结果表明:随着装卸台坡度的增大,宰后牛肉pH值、牛肉L*值、a*值呈下降趋势(P0.05),牛肉b*值、剪切力、肉失水率、蒸煮损失率呈增加趋势(P0.05)。通过坡度为30°的装卸台装卸牛,宰后牛肉品质较好,在实际生产中应降低装卸台坡度,以提高牛肉品质。  相似文献

The health, longevity, and performance of dairy cattle can be adversely affected by heat stress. This study evaluated the in-barn condition [i.e., temperature, relative humidity, and resulting temperature-humidity index (THI)] at 9 dairy barns with various climates and farm design-management combinations. Hourly and daily indoor and outdoor conditions were compared at each farm, including both mechanically and naturally ventilated barns. On-site conditions were compared with on-farm outdoor conditions, meteorological stations up to 125 km away, and NASA Power data. Canadian dairy cattle face periods of extreme cold and periods of high THI, dependent on the regional climate and season. The northernmost location (53°N) experienced about 75% fewer hours of THI >68 compared with the southernmost location (42°N). Milking parlors had higher THI than the rest of the barn during milking times. The THI conditions inside dairy barns were well correlated with THI conditions measured outside the barns. Naturally ventilated barns with metal roofs and without sprinklers fit a linear relationship (hourly and daily means) with a slope <1, indicating that in-barn THI exceeded outdoor THI more at lower THI and reached equality at higher THI. Mechanically ventilated barns fit nonlinear relationships, which showed the in-barn THI exceeded outdoor THI more at lower THI (e.g., 55–65) and approached equality at higher THI. In-barn THI exceedance was greater in the evening and overnight due to factors such as decreased wind speed and latent heat retention. Eight regression equations were developed (4 hourly, 4 daily) to predict in-barn conditions based on outdoor conditions, considering different barn designs and management systems. Correlations between in-barn and outdoor THI were best when using the on-site weather data from the study, but publicly available weather data from stations within 50 km provided reasonable estimates. Climate stations 75 to 125 km away and NASA Power ensemble data gave poorer fit statistics. For studies involving many dairy barns, the use of NASA Power data with equations for estimating average in-barn conditions in a population is likely appropriate especially when public stations have incomplete data. Results from this study show the importance of adapting recommendation on heat stress to the barn design and guide the selection of appropriate weather data depending on the aim of the study.  相似文献

Transrectal palpation of the reproductive tract is the most common method for pregnancy determination in cattle and is considered a veterinary skill that new veterinary medicine (DVM) graduates should perform proficiently. However, using privately owned cattle to train students can be difficult because producers may believe that transrectal palpation by inexperienced students increases the risk of pregnancy wastage compared with examination by an experienced clinician. We used a randomized field trial of 1,216 healthy Holstein and Jersey cattle in 2 commercial dairy herds to estimate the effect of veterinary student transrectal palpation on early pregnancy loss. All cattle were determined to be pregnant using transrectal ultrasonography at approximately 37 d after artificial insemination. Cattle were then allocated into 2 groups based upon their ear tag number (study group = 598; control group = 618). Cattle in the study group were immediately palpated after ultrasonography by a fourth-year veterinary student, whereas control cattle were not subject to any additional pregnancy assessment. For analysis, the student palpators were divided into 2 groups: students who had previously had formal palpation training via an elective bovine palpation class (n = 30) and students who had not had palpation training (n = 134). All cattle were reevaluated using transrectal ultrasonography approximately 70 d after artificial insemination. A total of 53 (4.36%) animals lost their pregnancy between the first and second pregnancy assessments. Of the animals that lost their pregnancy, 26 (4.35%) were study group cows and 27 (4.37%) were control cows. Of the 26 cows documented to have had pregnancy loss within the study group, 20 out of 378 (5.3%) had been palpated by students who had not taken the palpation elective and 6 out of 220 (2.7%) had been palpated by students who had completed the elective. We found no difference in pregnancy loss between student-palpated and clinician-ultrasounded cattle, supporting the safety of using privately owned animals for student bovine palpation and pregnancy diagnosis training without affecting early pregnancy loss.  相似文献

Among the costs attributed to Mycobacterium avium ssp. paratuberculosis (MAP) infection in dairy cattle, the effects on reproduction and culling are the least documented. To estimate the cost of MAP infections and Johne's disease in a dairy herd, the rates of calving and culling were calculated for cows in each stage of MAP infection relative to uninfected cows. Data from 6 commercial dairy herds, consisting of 2,818 cows with 2,754 calvings and 1,483 cullings, were used for analysis. Every cow in each study herd was tested regularly for MAP, and herds were followed for between 4 and 7 yr. An ordinal categorical variable for Johne's disease status [test-negative, low-positive (low-shedding or ELISA-positive only), or high-shedding] was defined as a time-dependent variable for all cows with at least 1 positive test result or 2 negative test results. A Cox regression model, stratified on herd and controlling for the time-dependent infection variable, was used to analyze time to culling. Nonshedding animals were significantly less likely to be culled in comparison with animals in the low-shedding or ELISA-positive category, and high-shedding animals had nonsignificantly higher culling rates than low-shedding or ELISA-positive animals. Time to calving was analyzed using a proportional rates model, an analog to the Andersen-Gill regression model suitable for recurrent event data, stratifying on herd and weighted to adjust for the dependent censoring caused by the culling effects described above. High-shedding animals had lower calving rates in comparison with low-shedding or ELISA-positive animals, which tended to have higher calving rates than test-negative animals.  相似文献

Previous studies have indicated that lame cows have reduced milk yield both before and after they are treated for lameness. One explanation for the reduction in yield before treatment is delay to treatment; that is, cows have impaired mobility for some time before they are treated. The aim of this study was to test this hypothesis by investigating temporal associations between change in milk yield and change in mobility score. Mobility score (MS, on a scale from 0 to 3), milk yield, treatments for lameness, and cow activity were recorded on 312 cows in a dairy herd in Somerset, UK, for 1 yr. The MS was scored every 2 wk and compared with daily yield and activity (mean log steps/h) averaged over the previous 16 d. Approximately 52% of MS changed within 14 d, usually by 1 unit. Overall, milk yields of cows with MS 1 were greater than those of cows with other scores. Cows with MS 2 and 3 produced 0.7 (95% confidence interval: 0.35–0.97) and 1.6 (0.98–2.23) kg less milk/d, respectively, compared with cows with MS 1. In addition, cows with MS 1 were significantly more active than cows with MS 0, 2, or 3. Cows with MS 2 and 3 were 0.02 (0.01–0.03) and 0.03 (0.01–0.05) mean log steps less active than cows with MS 1. Six to 8 wk before nonlame cows became MS 2 or 3, their daily milk yield decreased by a mean (95% CI) of 0.5 kg (0.12–0.47) and 0.9 kg (0.16–1.65) respectively. Daily yield remained lower by 0.42 kg (0.09–0.75) for 4 wk after cows with MS 2 had recovered. The activity of cows was significantly less (0.01 mean log steps) with increasing MS; the associations between activity and parity (means 0.03–0.11) and month of lactation (means 0.03–0.36) were quantitatively larger. Results from a multistate model indicated that once cows were lame they remained lame or became lame again despite treatment. In conclusion, cows’ milk production started to decline before their mobility was visibly impaired.  相似文献

Embryonic losses contribute considerably to low pregnancy rates. Between d 8 and 17 after breeding, the conceptus secretes interferon-τ as a mechanism for maternal recognition of pregnancy and maintenance of the corpus luteum. Nonsteroidal antiinflammatory drugs inhibit the synthesis of prostaglandin F by suppressing the enzyme cyclooxygenase. Flunixin meglumine (FM) has been demonstrated to delay luteolysis and to support embryonic survival. The objective of this study was to evaluate the effect of FM and carprofen on conception rates in dairy heifers and cows, respectively. In experiment 1, the effect of FM on pregnancy rates and progesterone concentrations in dairy heifers was tested. A total of 391 heifers were randomly assigned to 1 of 2 groups. Heifers in the treatment group (n = 197) received 2.2 mg of FM i.m./kg of body weight twice on d 14/15 and 15/16 after insemination, whereas heifers in the control group (n = 194) remained untreated. Blood samples from 388 heifers were taken on d 14/15 and 21/22 after artificial insemination and analyzed for progesterone. Pregnancy rates were 58.2 and 54.8% for the control and treatment groups, respectively. Mean progesterone concentrations were not affected by treatment and number of artificial insemination service (first or second artificial insemination service), but were affected by time and time × pregnancy status. In experiment 2, the objective was to verify the effects of carprofen, a longer acting nonsteroidal antiinflammatory drug and to evaluate its effect on conception rate to first service in dairy cows. A total of 380 cows were randomly assigned to 1 of 2 treatment groups. Cows in the treatment group (n = 194) received 1.4 mg of carprofen s.c./kg of body weight on d 15 after insemination, whereas cows in the control group (n = 186) remained untreated. Pregnancy was diagnosed between d 40 and 47 after insemination. Conception rates to first service were 35.5 and 33.0% in the control and treatment groups, respectively. Neither flunixin meglumine nor carprofen improved conception rates to first service in dairy cattle in the dosage and administration schedule tested.  相似文献
