Absorption of adequate IgG from colostrum is critical to provide the newborn calf with adequate immunological protection and resistance to disease. Excessive iodine supplementation of the prepartum ewe reduces IgG absorption of her offspring; it is possible that excessive iodine supplementation of the prepartum dairy cow may similarly impair the ability of the calf to acquire immunological protection. The objectives of this study were to determine whether the iodine status, health status, and ability of calves to absorb IgG from colostrum were affected by prepartum iodine supplementation strategies of their dams. Dairy cows (n = 127) received one of the following levels of iodine supplementation precalving: 15 mg of iodine/kg of dietary dry matter (DM) (HI); no additional iodine supplementation (MI); 5 mg/kg of dietary DM (SI); and 15 mg of iodine/kg of DM for the first 3.5 wk of the precalving period and no additional supplementation for the second 3.5 wk (HMI). Calves were assigned to 1 of 6 experimental treatments, based on the prepartum iodine supplementation treatment of their dam and the precalving treatment group of the cows from which the colostrum fed was obtained: (1) HI_HI: born to HI dams, fed HI colostrum (i.e., colostrum produced by cows in the HI group); (2) MI_MI: born to MI dams, fed MI colostrum; (3) SI_SI: born to SI dams, fed SI colostrum; (4) HI_MI: born to HI dams, fed MI colostrum; (5) MI_HI: born to MI dams, fed HI colostrum; and (6) HMI_HMI: born to HMI dams, fed HMI colostrum. Concentration of calf serum IgG and plasma inorganic iodine (PII) was measured at 0 and 24 h of age. Apparent efficiency of absorption for IgG was determined. Health scores were assigned to calves twice weekly and all episodes of disease were recorded. Cow experimental treatment group affected calf PII at 0 h of age; the PII of calves born to HI dams (987.2 µg/L) was greater than that of calves born to MI dams (510.1 µg/L), SI (585.2 µg/L), and HMI dams (692.9 µg/L). Calf experimental treatment group affected calf PII at 24 h of age; the PII of HI_HI (1,259.2 µg/L) and HI_MI (1,177.8 µg/L) calves was greater than MI_MI (240.7 µg/L), SI_SI (302.2 µg/L), HMI_HMI (320.7 µg/L), and MI_HI (216.3 µg/L) calves. No effect of experimental treatment was observed on the concentration of IgG measured in calf serum at 24 h of age, or on apparent efficiency of absorption. Experimental treatment had no effect on the likelihood of a calf being assigned a worse nasal, eye and ear, cough, or fecal score within the study period, nor did it affect the probability of a calf receiving treatment for a disease a greater number of times. Prepartum iodine supplementation of cows at 15mg/kg of DM increased the iodine levels in their calves at birth and 24 h of age, but did not affect their ability to absorb IgG from colostrum. Supplementation with iodine above the minimum requirements established by the National Research Council was unnecessary to ensure appropriate iodine levels in calves at birth. 相似文献

The objective of this study was to evaluate hair cortisol concentration as an indicator for stress caused by chronic lameness in dairy cows. Sixty-eight cows were scored for lameness for 4 consecutive weeks. The hair of the tail switch was clipped at the beginning of the study and regrown hair was clipped after 4 wk. Hair samples were analyzed for cortisol concentration. Animals with 2 consecutive locomotion scores ≥3 or with an overall mean score >1.5 were classified as lame. After pair matching lame and nonlame cows, considering days in milk, lactation number, and milk yield, and excluding cows with less than 20 mg hair sample for analysis, 21 lame and 21 nonlame cows were included in the analysis. The mean hair cortisol concentration in this study was 2.32 ± 0.35 pg/mg (mean ± standard deviation). Cortisol concentration from hair regrown in the study period was 2.38 ± 0.95 and 2.26 ± 1.35 pg/mg for lame and nonlame cows (n = 21), respectively; we found no difference in mean cortisol level of primiparous and multiparous cows. Based on these data, hair cortisol concentration was not a useful indicator to differentiate cows with chronic lameness and healthy cows. 相似文献

The objective of this study was to investigate the effects of a milk byproduct–based starter pellet, high in lactose, on feed intakes, growth, and rumen development when calves were fed either a high or low level of milk. This was done by using a 2 × 2 factorial design, with 120 Holstein heifer calves randomly assigned to 1 of 2 milk treatments and 1 of 2 feed treatments upon enrollment at birth. Calves were either assigned to a low (LM) or high (HM) level of milk replacer by an automated milk-feeding system for individually-housed calves. The LM treatment calves were offered 6 L/d (150 g/L; 1.2 L, 5×/d), whereas HM treatment calves were offered 15 L/d (150 g/L; 3 L, 5×/d) of milk replacer. Calves were also assigned to a milk byproduct- (whey) based starter feed treatment (WF), or a grain-based starter ration (95% concentrate starter pellet, 5% chopped wheat straw) feed treatment (GF). All calves were offered their treatment feed starting on d 5, with water offered ad libitum from d 0. The WF treatment calves were offered 150 g/d of the milk byproduct–based pellet until they consumed the entirety for 2 out of 3 consecutive days. At this point, WF treatment calves received 150 g of the milk byproduct–based pellet top dressed onto the grain-based ration to allow for ad libitum feed consumption until weaned, when they only received the grain-based ration postweaning (d 64–77). Calves on the GF feed treatment received only the grain-based ration. All calves were gradually weaned from d 43 to 63. By design, calves on the HM treatment consumed greater levels of milk than LM treatment calves for the majority of the days in the preweaning phase (8.9 ± 0.11 vs. 5.5 ± 0.11 L/d; SE). Calves on the GF and WF treatments consumed similar amounts of milk in the preweaning phase (7.2 ± 0.11 vs. 7.2 ± 0.11 L/d); however, there was a milk and feed treatment interaction detected for milk consumption in the weaning phase, with HM-GF calves drinking more milk than the other treatments and HM-WF calves drinking more milk than both LM-GF and LM-WF calves. The WF treatment calves had greater solid feed intake during the preweaning phase than GF treatment calves, with no feed intake differences in the weaning or postweaning phases. The WF calves drank more water in the preweaning phase than GF calves. Average daily gain was similar throughout the study between the GF and WF treatment calves. Rumen physiological measures were all similar by the end of the trial (d 77) regardless of feed or milk treatment, suggesting similar rumen development postweaning. Overall, regardless of the level of milk fed, offering the milk byproduct–based starter pellet may increase preweaning feed intake, along with water consumption. However, both feed and milk treatments had similar performance and rumen development measures by the end of the trial. 相似文献

The objective of our experiment was to evaluate the effects of prill size of a palmitic acid–enriched fatty acid supplement (PA; 85% C16:0) on feed intake, nutrient digestibility, and production responses of dairy cows. Twenty-four primiparous and multiparous Holstein cows were assigned based on parity and production level to replicated 4 × 4 Latin squares balanced for carryover effects with 21-d periods. Treatments were a control diet (no added PA), or 2.0% PA added as a small prill size (PA-SM; 284 ± 12.4 µm), a medium prill size (PA-MD; 325 ± 14.7 µm), or a large prill size (PA-LG; 600 ± 17.4 µm) supplement. Overall, PA treatments increased milk fat content (4.25 vs. 3.99%), milk fat yield (1.48 vs. 1.39 kg/d), 3.5% fat-corrected milk (39.2 vs. 37.7 kg/d), and improved feed efficiency (fat-corrected milk:dry matter intake; 1.51 vs. 1.42) compared with control. Compared with control, PA treatments did not affect dry matter intake, body weight, body condition score, or yields of milk, protein, and lactose. The PA treatments increased neutral detergent fiber digestibility (44.8 vs. 42.4%) and reduced the digestibility of 16-carbon fatty acids (72.3 vs. 79.1%) and total fatty acids (76.6 vs. 80.3%). Compared with control, PA treatments reduced the contents of de novo synthesized milk fatty acids (23.0 vs. 25.8 g/100 g of fatty acids) and preformed milk fatty acids (36.3 vs. 39.1 g/100 g of fatty acids), but did not affect their yields. In contrast, PA treatments increased the content (40.8 vs. 35.1 g/100 g of fatty acids) and yield (570 vs. 436 g/d) of 16-carbon milk fatty acids compared with control. The PA prill size had no effect on dry matter intake, yield of milk and milk components, or feed efficiency. However, PA-LG tended to increase milk fat content compared with PA-SM (4.28 vs. 4.22%), and it increased 16-carbon fatty acid digestibility compared with PA-MD (74.2 vs. 71.0%) and PA-SM (74.2 vs. 71.7%). Additionally, PA-LG increased total fatty acid digestibility compared with PA-MD (78.1 vs. 75.6%) and PA-SM (78.1 vs. 76.0%). Results demonstrate that PA increased milk fat content and yield, and feed efficiency. Reducing prill size decreased fatty acid digestibility, but it had no effect on animal performance under the dietary conditions and prill sizes evaluated. 相似文献

In vivo kinetics of oleic,linoleic, and α-linolenic acid biohydrogenation in the rumen of dairy cows

Ruminal biohydrogenation (BH) of unsaturated fatty acids (FA) reduces absorption of essential FA and can result in formation of bioactive FA that cause milk fat depression. Rates of biohydrogenation of unsaturated FA are commonly observed using in vitro systems and are not well described in vivo. Seven ruminally cannulated cows were enrolled in a 3 × 3 Latin square design study to quantify biohydrogenation of 18:1n-9, 18:2n-6, and 18:3n-3 using a recently developed in vivo BH assay. All cows were fed a common high corn silage basal diet. Biohydrogenation was quantified using a perturbation model that consisted of a bolus dose of 200 g of an oil enriched in each unsaturated FA (oleic acid, OA = 87% 18:1n-9 sunflower oil; linoleic acid, LA = 70% 18:2n-6 safflower oil; and α-linolenic acid, ALA = 54% 18:3n-3 flaxseed oil) and 12 g of 17:0 as a marker of rumen outflow. Rumen contents were sampled before and after the bolus and enrichment of the bolused FA modeled. Using first-order kinetics to model FA disappearance, the fractional rates of disappearance of 18:1n-9 was 0.597 per hour, 18:2n-6 was 0.618 per hour, and 18:3n-3 was 0.834 per hour, similar to rates previously reported with this approach. Rumen turnover of 17:0 was 0.123 per hour, 0.065 per hour, and 0.106 per hour during the OA, LA, and ALA treatments, respectively. The extents of BH were calculated to be 82.8, 90.4, and 88.6% for 18:1n-9, 18:2n-6, and 18:3n-3, respectively. Finally, compartmental modeling was used to quantify the amount of each unsaturated FA metabolized through trans-10 and trans-11 BH pathways. The recently developed in vivo BH assay was able to predict rates of BH and provide insight into rumen metabolism of individual FA and may be useful to future investigations. 相似文献

ABSTRACTBackground: Food recommendations to improve cancer prevention are generally based on epidemiologic data and remain inconsistent. These epidemiologic studies, while controversial, have generally produced results that caution against the consumption of high-fat foods, including eggs, red meat, and full-fat dairy, such as butter and cheese. Yet, limited data exist assessing the quality of individual sources of these foods and the effect each has after its consumption. This study set out to assess the impact sources of food within the same groups from animals raised differently on variables associated with health in human studies. Methods and Materials: A search was conducted through MEDLINE, Embase, and PubMed. In total, twenty-nine studies met inclusion criteria, measuring physiologic changes in humans after consuming animal products following animal diet manipulation. A meta-analysis was attempted to assess the differences between the cohorts in these studies, but was aborted due to poor study quality, vast differences in study design, and a limited number of studies. Results: Studies varied by animal, animal diet manipulation, food product, and overall design. Significant differences were present between groups eating the same food (cheese, beef, eggs, and butter) from animals raised differently, including levels of: conjugated linoleic acid, omega-3 fatty acids (alpha linoleic acid [ALA], docosahexaenoic acid [DHA], and eicosapentaenoic acid [EPA]), and inflammatory factors (triacyl glycerol [TAG], interleukin-6 [IL-6], interleukin-8 [IL-8], tumor necrosis factor [TNF], and C-reactive protein [CRP]). Lipid levels were minimally affected. Conclusions: This work highlights differences in human health markers after consumption of the same foods from animals raised differently. Overall, lipid levels remained relatively neutral, but significant changes in inflammatory and other serum markers and phospholipids were present. Future studies and dietary recommendations should consider how animals are raised, as this can produce different effects on health markers. 相似文献

The primary aim of this study was to assess the influence of provision of information on lay citizens' opinions regarding 2 common management practices, zero-grazing and cow-calf separation. To aid in the interpretation of the findings, our secondary aim was to explore the awareness and opinions of Brazilian citizens about these practices. We surveyed a convenience sample of Brazilian citizens (192 men and 208 women), recruited in a public place, with the majority stating that they were largely unfamiliar with animal production and lived in urban environments. Participants were presented short scenarios with information on the primary production factors and welfare concerns for and against zero-grazing (n = 200) or cow-calf separation (n = 200). Participants were then asked to state their position (reject, indifferent, or support), and to provide the reason(s) justifying their position. Immediately following, participants were provided a short statement describing either zero-grazing or cow-calf separation, depending on what question they responded to in the first part. Two closed questions (Q) followed each of these statements: (Q1) “Are you aware of this practice?” with choices yes, somewhat, or no, and (Q2) “What is your position regarding this practice?” with choices reject, indifferent, or support. Only 31 and 33% of the respondents were aware of zero-grazing and cow-calf separation, respectively. Previous awareness of existence of practice did not influence levels of support. Provision of information resulted in more people rejecting the practices of zero-grazing and cow-calf separation. Participants' main justifications to reject zero-grazing and cow-calf separation focused on perceived negative effects of practices on farm animal welfare and product quality, and loss of naturalness. Survey participants, Brazilians living in urban environments, with little or no association with dairy production, were generally unaware that many cows do not have access to pasture and that cows are separated from their calf at birth. Independent of provision of additional information, most participants did not support these practices. Provision of brief explanatory information played a minor role in influencing people's views, but failed to result in general acceptance. 相似文献

Intersucking, i.e., cattle sucking the udder of heifers or cows, is a frequent problem in dairy herds and may lead to udder damage, mastitis, milk loss, and culling of breeding animals. Using epidemiological methods, we conducted an observational cross-sectional study to investigate risk factors for intersucking in Swiss dairy cows. We asked 114 randomly selected dairy farmers about a broad spectrum of environmental factors possibly associated with intersucking, such as housing conditions, management, and feeding of calves, heifers, and cows. Thirty of the 114 farms were confronted with intersucking in cows. The mean proportion of intersucking cows per farm was 1.6%. From a total of 3077 cows (Swiss Brown Cattle, Simmental, and Holstein Friesian) we recorded 49 cows that had performed or were currently intersucking. In 69% of these cows, intersucking had been observed as heifers. Using path analysis and multivariable stepwise backward logistic and linear regression analyses, we revealed that the most important risk factor for intersucking cows was the presence of intersucking heifers on a farm (odds ratio = 7.8). The results suggest that intersucking in cows is the continuation of a habit that was already established in a cow's subadult life. This emphasizes the importance of looking not only at the animal's current environmental situation but also considering its entire life history for the prevention of behavioral problems. 相似文献

This work discusses the extent to which differences in technological or environmental conditions affect the sensory properties of cheeses made in mountain or valley farms. The work is focused on Idiazabal cheese, frequently made with lamb rennet paste and with milk from sheep flocks managed under extensive grazing systems. Fourteen sensory attributes were assessed, and free fatty acid content and rennet lipase activity were analyzed in cheeses from mountain and valley farms. Sensory differences between cheeses were mainly dependent on the type and amount of rennet used for cheesemaking, particularly on the level of rennet lipase activity. The sensory attributes did not appear to be influenced by pasture type, grazing practices, or environmental conditions associated with farm location. Rennet lipase activity was responsible for the high scores of strong sensory attributes such as pungent mouthfeel, and these attributes masked the potential sensory differences that could be found due to farm location. 相似文献

Free fatty acid receptors (FFAR) play significant roles in various physiological processes, including energy metabolism, through interaction with their ligands, fatty acids. To determine whether the receptors FFAR1 and FFAR2 are involved in the regulation of liver metabolism during the peripartal period, we selected 13 German Holstein multiparous dairy cows and grouped them as high β-hydroxybutyrate (H-BHB; n = 8) or low β-hydroxybutyrate (L-BHB; n = 5) according to their individual maximum plasma BHB concentration observed within wk 2 or 3 postpartum (H-BHB: >1 mmol/L and L-BHB: <0.77 mmol/L). The selected cows had a milk yield of more than 10,000 kg/305 d during a previous lactation. The cows were fed a total mixed ration according to their requirements during the far-off dry period [5.9 MJ of net energy for lactation (NEL)/kg of dry matter (DM), crude protein (CP) 126 g/kg of DM], close-up dry period (6.5 MJ of NEL/kg of DM, CP 137 g/kg of DM), and lactation (7 MJ of NEL/kg of DM, CP 163 g/kg of DM). Blood samples were taken weekly, from d ?34 to d 40 relative to parturition. Liver biopsies were taken on d ?34, ?17, 3, 18, and 30 relative to parturition and at slaughter (d 40). The protein abundance of FFAR1 was lower during the whole peripartal period in the H-BHB group. The abundance of FFAR2 increased over time and tended to be higher in H-BHB cows. The abundance of FFAR1 might be associated with imbalances of liver metabolism in peripartal dairy cows. 相似文献

Since many years, the role of the colonic microbiota in maintaining the host's overall health and well-being has been recognized. Dietary modulation of the microbiota composition and activity has been achieved by the use of pre-, pro- and synbiotics. In this review, we will summarize the available evidence on the modification of bacterial metabolism by dietary intervention with pre-, pro- and synbiotics. Enhanced production of SCFA as a marker of increased saccharolytic fermentation is well documented in animal and in vitro studies. Decreased production of potentially toxic protein fermentation metabolites, such as sulfides, phenolic and indolic compounds, has been less frequently demonstrated. Besides, pre-, pro- and synbiotics also affect other metabolic pathways such as the deconjugation of secondary bile acids, bacterial enzyme activities and mineral absorption. Data from human studies are less conclusive. The emergence of new analytical techniques such as metabolite profiling has revealed new pathways affected by dietary intervention. However, an important challenge for current and future research is to relate changes in bacterial metabolism to concrete health benefits. Potential targets and expected benefits have been identified: reduced risk for the metabolic syndrome and prevention of colorectal cancer. 相似文献

Food labelling can play a key role in increasing consumers’ food-related consciousness, and improving the healthiness of their food choices. However consumers’ use of food labels is affected by a number of variables. The aim of this paper is to explore the role of health-orientation on consumers’ use of different labelled information, making a distinction between mandatory and voluntary information (namely, nutrition facts panel vs nutrition and health claims). Data were collected in Italy through face-to-face interviews on a sample of 300 consumers in charge of their grocery shopping. To evaluate the relationship between health-orientation and label usage we developed an index strictly based on the definition of health-orientation itself and included such measure in our analysis, which is based on a set of OLS regressions. The main results overall stress the idea that different degrees of health-orientation are, indeed, reflected in a diverse use of labelled information. Highly health-oriented consumers are more likely to refer to the extensive information reported on nutrition facts panel, whereas claims are of main interest for consumers with low orientation to health. The results also strengthen the key role played by nutrition knowledge and education in favouring a frequent usage of the nutrition facts panel and confirm previous findings on the relationship between socio-economic status and consumers use of labelled information. 相似文献

The primary objective of this study was to evaluate the repeatability of measurements of β-hydroxybutyrate (BHB) concentrations in capillary blood obtained from minimal-invasive puncturing of different sites at the external vulva skin of dairy cows. A total of 240 multiparous cows sampled once within 21 d in milk were enrolled in the study. Capillary blood samples were obtained from the right and left labium of the external vulva skin and additionally from a site close to the first puncture. Samples were analyzed by using an electronic hand-held device [FreeStyle Precison Neo (FSP-Neo), Abbott GmbH & Co., Wiesbaden, Germany)]. Additionally, samples from a coccygeal vessel were analyzed as reference in a laboratory. Mean deviations (± standard deviation) between the 3 capillary sampling sites ranged from 0.01 ± 0.14 (left vs. right labium) to 0.02 ± 0.13 mmol/L (left or right labium vs. additional site). The BHB concentrations in capillary blood compared with the reference showed a mean deviation of 0.14 ± 0.18 mmol/L. The Spearman correlation coefficient between BHB concentrations in capillary blood and coccygeal blood was 85.4%. Mixed model analyses showed no significant effect of sampling site or squeezing the skin during sampling procedures on the differences toward the reference. Based on laboratory BHB concentrations of ≥1.2 and ≥1.4 mmol/L, respectively, the prevalence of hyperketonemia was 10.4 (n = 25) and 5.0% (n = 12). Thresholds to detect hyperketonemia with the FSP-Neo were optimized by receiver operating characteristic analyses. Based on a laboratory BHB concentration of ≥1.2 mmol/L thresholds were determined as 1.1 (left labium, 2nd puncture) and 1.0 mmol/L (right labium), with sensitivities between 94 and 99% and specificities between 89 and 97%. Based on a serum BHB concentration of 1.4 mmol/L, receiver operating characteristic analyses resulted in optimized cutoffs of 1.4 mmol/L for all sampling sites at the vulva, with sensitivities of 100% and specificities between 96 and 99%. In conclusion, vulvar capillary blood is suitable for accurate measuring of BHB concentrations by using the FSP-Neo. After adjusting the thresholds, the FSP-Neo provides very good test characteristics independent of the capillary sampling site or squeezing the skin and allows a practical and reliable screening tool for dairy cows. 相似文献

Several temperature-humidity indexes (THI) have been used to estimate the degree of thermal stress experienced by dairy cows. The present objectives were to develop equations using meteorological variables that predicted rectal temperature of lactating cows in a subtropical environment and compare the goodness of fit of these equations to those using 8 different THI. Rectal temperature was measured between 1500 and 1700 h in 1,280 lactating Holstein cows in north central Florida between August and December. Meteorological data recorded in the barn where cows were located included dry bulb temperature (Tdb), relative humidity (RH), dew point temperature, and wind speed. Wet bulb temperature was calculated. In the first series of analyses, regression analysis was used to model rectal temperature using the meteorological variables as well as THI. The r2 using Tdb (0.41) was slightly less than for models using all but one THI (r2 between 0.42 and 0.43). The r2 for equations using Tdb could be improved by adding RH (r2 = 0.43) or RH and RH2 (r2 = 0.44) to the model. In the second analysis, regression analysis was performed using forward selection, backward elimination, and stepwise selection procedures with the meteorological variables. All models gave a similar goodness of fit (r2 = 0.44). An analysis of variance with rectal temperature as a class variable was performed to determine the least squares means of meteorological measurements associated with hyperthermia. A Tdb of 29.7°C was associated with rectal temperature of 39°C, and a Tdb of 31.4°C was associated with rectal temperature of 39.5°C. In conclusion, Tdb is nearly as good a predictor of rectal temperatures of lactating Holsteins in a subtropical environment as THI. Estimates of values of meteorological variables associated with specific rectal temperatures should prove valuable in relating environmental conditions to the magnitude of hyperthermia experienced by heat-stressed cows. 相似文献

Genetic parameters for the major milk proteins were estimated in the 3 main French dairy cattle breeds (i.e. Montbéliarde, Normande, and Holstein) as part of the PhénoFinlait program. The 6 major milk protein contents as well as the total protein content (PC) were estimated from mid-infrared spectrometry on 133,592 test-day milk samples from 20,434 cows in first lactation. Lactation means, expressed as a percentage of milk (protein contents) or of protein (protein fractions), were analyzed with an animal mixed model including fixed environmental effects (herd, year × month of calving, and spectrometer) and a random genetic effect. Genetic parameter estimates were very consistent across breeds. Heritability estimates (h2) were generally higher for protein fractions than for protein contents. They were moderate to high for αS1-casein, αS2-casein, β-casein, κ-casein, and α-lactalbumin (0.25 < h2 < 0.72). In each breed, β-lactoglobulin was the most heritable trait (0.61 < h2 < 0.86). Genetic correlations (rg) varied depending on how the percentage was expressed. The PC was strongly positively correlated with protein contents but almost genetically independent from protein fractions. Protein fractions were generally in opposition, except between κ-casein and α-lactalbumin (0.39 < rg < 0.46) and κ-casein and αS2-casein (0.36 < rg < 0.49). Between protein contents, rg estimates were positive, with highest values found between caseins (0.83 < rg < 0.98). In the 3 breeds, β-lactoglobulin was negatively correlated with caseins (?0.75 < rg < ?0.08), in particular with κ-casein (?0.75 < rg < ?0.55). These results, obtained from a large panel of cows of the 3 main French dairy cattle breeds, show that routinely collected mid-infrared spectra could be used to modify milk protein composition by selection. 相似文献

Metabolism in most organisms can show variations between the day and night. These variations may also affect the composition of products derived from livestock. The aim of the present study was to investigate the difference in composition between the day milk and night milk of dairy cows. Ten multiparous Holstein cows (milk yield = 25.2 ± 5.00 kg/d) were randomly selected during mid lactation. Milk samples were collected at 0500 h (“night milk”) and 1500 h (“day milk”) and analyzed to determine their composition. Mid-infrared spectroscopy was used to analyze macronutrient content of milk. Metabolomics and lipidomics were used to detect and analyze small molecules and fatty acids, respectively. An automatic biochemical analyzer and ELISA kits were used to determine biochemical indicators, as well as antioxidant and immune parameters in the milk. Though milk fat, protein, lactose, and total milk solids were not different between day milk and night milk, small molecules, metabolites and lipids, and hormones and cytokines differed between day milk and night milk. Regarding biochemical and immune-related indicators, the concentrations of malondialdehyde, HSP70, and HSP90 in night milk were lower than that in day milk. However, interferon-γ levels were higher in night milk. Additionally, night milk was naturally rich in melatonin. Lipidomics analyses showed that the levels of some lipids in night milk were higher than those in day milk. Metabolomics analyses identified 36 different metabolites between day milk and night milk. Higher concentrations of N-acetyl-d-glucosamine, cis-aconitate, and d-sorbitol were observed in day milk. However, the other 33 metabolites analyzed, including carbohydrates, lipids, AA, and aromatic compounds, showed lower concentrations in day milk than in night milk. The present findings show that the composition of night milk differs considerably from that of day milk. Notable changes in the circadian rhythm also altered milk composition. These results provide evidence to support the strategic use and classification of day milk and night milk. 相似文献

The construction and use of haploblocks [adjacent single nucleotide polymorphisms (SNP) in strong linkage disequilibrium] for genomic evaluation is advantageous, because the number of effects to be estimated can be reduced without discarding relevant genomic information. Furthermore, haplotypes (the combination of 2 or more SNP) can increase the probability of capturing the quantitative trait loci effect compared with individual SNP markers. With regards to haplotypes, the allele frequency parameter is also of interest, because as a selection criterion, it allows the number of rare alleles to be reduced, and the effects of those alleles are usually difficult to estimate. We have proposed a simple pipeline that simultaneously incorporates linkage disequilibrium and allele frequency information in genomic evaluation, and here we present the first results obtained with this procedure. We used a population of 2,235 progeny-tested bulls from the Montbéliarde breed for the tests. Phenotype data were available in the form of daughter yield deviations on 5 production traits, and genotype data were available from the 50K SNP chip. We conducted a classical validation study by splitting the population into training (80% oldest animals) and validation (20% youngest animals) sets to emulate a real-life scenario in which the selection candidates had no available phenotype data. We measured all reported parameters for the validation set. Our results proved that the proposed method was indeed advantageous, and that the accuracy of genomic evaluation could be improved. Compared with results from a genomic BLUP analysis, correlations between daughter yield deviations (a proxy for true) and genomic estimated breeding values increased by an average of 2.7 percentage points for the 5 traits. Inflation of the genomic evaluation of the selection candidates was also significantly reduced. The proposed method outperformed the other SNP and haplotype-based tests we had evaluated in a previous study. The combination of linkage disequilibrium–based haploblocks and allele frequency–based haplotype selection methods is a promising way to improve the efficiency of genomic evaluation. Further work is needed to optimize each step in the proposed analysis pipeline. 相似文献
