Ether, methanolic and aqueous extracts of lyophilized rambutan (Nephelium lappaceum L.) peels and seeds were evaluated for phenolic contents, antioxidant and antibacterial activities. High amounts of phenolic compounds were found in the peel extracts and the highest content was in the methanolic fraction (542.2 mg/g dry extract). Several potential antioxidant activities, including reducing power, β-carotene bleaching, linoleic peroxidation and free radical scavenging activity, were evaluated. The peel extracts exhibited higher antioxidant activity than the seed extracts in all methods determined (P < 0.05). The methanolic fraction was found to be the most active antioxidant as shown by their 50% DPPH inhibition concentration, 4.94 μg/mL. The results indicated this fraction exhibited greater DPPH radical scavenging activity than BHT and ascorbic acid (0.32 g dry extract/g BHT or ascorbic acid). Antibacterial activity against eight bacterial strains was assessed by disc diffusion and broth macrodilution methods. All peel extracts exhibited antibacterial activity against five pathogenic bacteria. The most sensitive strain, Staphylococcus epidermidis, was inhibited by the methanolic extract (MIC 2.0 mg/mL).  相似文献

The antioxidative properties and total phenolic contents of raw and dry heated seed coat of Tamarindus indica were examined. The raw and dry heated samples were extracted with methanol followed by 70% acetone, after treating with petroleum ether, solvents were removed using rotary vacuum evaporator and the extracts which contain residual moisture were freeze-dried, respectively. Methanol extracts for both raw and dry heated seed coat samples contained higher level of total phenolics and tannins than the aqueous acetone extracts. The extracts were screened for their potential antioxidant activities using such as O2, OH, α,α-diphenyl-β-picrylhydrazyl (DPPH), ABTS+, FRAP and linoleic acid emulsion model systems. At different concentration of respective solvent extracts, the maximum level of superoxide anion radical scavenging activity (79-85%) was observed at 200 μg of both the raw and dry heated seed coat extracts in the reaction mixture. The DPPH radical and ABTS cation radical scavenging activities were well proved with the ferric reducing antioxidant capacity of the extracts. Interestingly, among the extracts, methanol and aqueous acetone extracts of dry heated sample showed the highest hydroxyl radical scavenging activity of 56.6 and 45.7%, respectively. All extracts, exhibited good antioxidant activity (64.5-71.7%) against the linoleic acid emulsion system and the values were lower and higher than the synthetic antioxdiant, BHA and ascorbic acid, respectively.  相似文献

Black-bone silky fowl (Gallus gallus domesticus Brisson) (BSF) muscle was hydrolyzed by papain, and the hydrolysate was separated by preparative high performance liquid chromatography (HPLC). The amino acid composition of the BSF hydrolysate (BSFH) and its fractions was determined by HPLC precolumn derivation with 2,4-dinitrofluorobenzene. The molecular weight (MW) distribution of the BSFH and its fractions was measured by a peptide column on an HPLC system. Antioxidant activities of the BSFH and its fractions were studied by testing the reducing power and four radical scavenging systems: superoxide anion (O2), hydroxyl (·OH), 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS+) radicals. The results demonstrated that the BSFH had strong antioxidant capacity to scavenge O2, DPPH and ABTS+, and displayed strong reducing power, but revealed less powerful ability to scavenge ·OH. Fraction II of the BSFH exhibited the highest activity in scavenging O2 and DPPH, and reducing power, whereas fraction I displayed the strongest ·OH scavenging ability. Besides Glu, Asp and Gly, the rich amino acids of Ala and Leu played an important role in antioxidant activity. The small-size peptides with MW ranging from approximately 200-6000 Da probably contributed to higher antioxidant activity. Results from this study indicated that BSFH and its fractions could be used as food additives and diet nutrients.  相似文献

A simple flow injection (FI)-spectrophotometric system for the screening of antioxidant capacity in herbal extracts was developed. The analysis was based on the color disappearance due to the scavenging of 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical by antioxidant compounds. DPPH and ascorbic acid were used as reagent and antioxidant standard, respectively. Effects of the DPPH concentration, DPPH flow rate, and reaction coil length on sensitivity were studied. The optimized condition provided the linear range of 0.010-0.300 mM ascorbic acid with less than 5%RSD(n = 10). Detection limit and quantitation limit were 0.004 and 0.013 mM, respectively. Comparison of antioxidant capacity in some herbal extracts determined by the FI system and a standard method was carried out and no significant difference was obtained.  相似文献
