Pomegranate peel rich in phenolics, and pomegranate seed which contain a conjugated fatty acid namely punicic acid in lipid fraction remain as by‐products after processing the fruit into juice. Ice cream is poor in polyunsaturated fatty acids and phenolics, therefore, this study was conducted to improve the functional properties of ice cream by incorporating pomegranate peel phenolics and pomegranate seed oil. Incorporation of the peel phenolics into ice cream at the levels of 0.1% and 0.4% (w/w) resulted in significant changes in the pH, total acidity, and color of the samples. The most prominent outcomes of phenolic incorporation were sharp improvements in antioxidant and antidiabetic activities as well as the phenolic content of ice creams. Replacement of pomegranate seed oil by milk fat at the levels of 2.0% and 4.0% (w/w) increased the conjugated fatty acid content. However, perception of oxidized flavor increased with the additional seed oil. When one considers the functional and nutritional improvements in the enrichment of the ice cream together with overall acceptability results of the sensory analysis, then it follows from this study that ice creams enriched with pomegranate peel phenolics up to 0.4% (w/w) and pomegranate seed oil up to 2.0% (w/w) could be introduced to markets as functional ice cream. Enrichment of ice creams with pomegranate by‐products might provide consumers health benefits with striking functional properties of punicalagins in pomegranate peel, and punicic acid in pomegranate seed oil.  相似文献

The inhibitory activities of banana peel extract against carbohydrate digestive enzymes were investigated. Cycloeucalenone and 31-norcyclolaudenone were obtained by bioassay-guided fractionation as effective inhibitors of α-glucosidase and α-amylase. The structure–activity relationships of four triterpenes isolated from banana peel, cycloeucalenone, 31-norcyclolaudenone and cycloeucalenol and its isomer were investigated. The IC50 values of cycloeucalenone and 31-norcyclolaudenone against α-glucosidase were 31.83 ± 2.46 μm and 38.85 ± 1.54 μm , respectively, and their IC50 values against α-amylase were 20.33 ± 0.59 μm and 27.63 ± 0.83 μm , respectively. In contrast, cycloeucalenol and its isomer had insufficient inhibitory activity against either enzyme. The primary active sites of cycloeucalenone and 31-norcyclolaudenone are the carbonyl group at C-3 and the double bond in the side chain. Cycloeucalenone induced a parabolic mixed-type inhibition with a Ki value of 73.86 μm in the α-glucosidase inhibitory assay. This study provides new evidence for the potential use of banana peel triterpenes as antidiabetic agents.  相似文献

Inhibitory mode and anti-diabetic activities of the Carols muscadine extracts on the α-glucosidase and pancreatic lipase were investigated. The IC50 values of the muscadine and its seed methanolic extracts against the α-glucosidase were 1.92 and 1.53 mg/mL, and those against the lipase were 34.41 and 8.63 mg/mL, respectively, which indicated the muscadine possessed the strong anti-diabetes activity. Particularly, the ethyl acetate (EtOAc) extract fractions exhibited the highest inhibitory activities against both enzymes. Since most of the phenolic compounds existed in the EtOAc fractions, it is suggested that phenolic compounds may be the main source against the α-glucosidase and pancreatic lipase. Twelve individual phenolics were identified by HPLC-MS. Ellagic acid and quercetin exhibited much stronger anti-diabetic activities among the selected phenolic standards. Besides, the extracts and the phenolic standards obeyed the competitive mode in their enzymatic inhibitions.  相似文献

Effects of white sorghum brewing process on free amino-acids, γ-aminobutyric acid (GABA), phenolics and bioactivity, including antioxidant (by ABTS+ and reducing power, RP, methods), antihypertensive (angiotensin converting enzyme-I, ACE-I inhibition assay), and hypoglycaemic activity (α-glucosidase inhibition assay) were evaluated. From the wort to the beer, free amino acids decreased, but GABA and phenolics increased significantly, positively modifying the bioactive potential. ABTS and α-glucosidase inhibition activity correlated positively with at least one of the phenolic acids evaluated. Ale white sorghum beer presented high content of GABA (7.8 mg L−1), phenolics (40.7 mg total phenolic acids L−1), antioxidant activity (9.14 mmol Trolox equivalent L−1, and 48.8 mmol ascorbic acid equivalent L−1, for ABTS+ and RP, respectively), and exhibit ACE-I inhibition (1.0 μg captopril equivalent L−1) and α-glucosidase inhibition (34.5 mg acarbose equivalent L−1) activities. The level of bioactive compounds and its low ethanol content (2.3%), make beer obtained from malted white sorghum a potential functional beverage.  相似文献

从21 种果皮中筛选出总酚、总黄酮含量较高的果皮,并选择1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基、2,2’-联氨-双-3-乙基苯并噻唑啉-6-磺酸(2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid),ABTS)阳离子自由基清除力评价其抗氧化活性。对筛选出酚类物质含量高且抗氧化活性强的果皮进行体外模拟胃肠液消化,研究不同消化时间对总酚、总黄酮释放量的影响,并通过透析袋模拟肠道的吸收作用,考察其生物利用率,同时利用超高效液相色谱-二极管阵列检测器/电喷雾飞行时间质谱(ultra performance liquid chromatography-diode array detector/electrospray ionization-time of fight-mass spectrometry,UPLC-DAD/ESI-TOF-MS)研究模拟胃肠液消化前后酚类物质变化。结果表明:21 种果皮中石榴皮(总酚含量为(58.09±0.46)mg/g、总黄酮含量为(47.50±0.39)mg/g)和山竹皮(总酚含量为(52.09±1.52)mg/g、总黄酮含量为(36.07±0.46)mg/g)中酚类物质含量较高;石榴皮的抗氧化能力强于山竹皮;模拟胃液消化阶段,石榴皮中总酚和总黄酮释放量均增加,模拟肠液消化阶段总酚和总黄酮的释放量先增加后减少;与胃液消化阶段相比,肠液消化阶段总酚和总黄酮含量明显降低;生物利用率测定结果表明总酚的生物利用率在每个时间点均强于总黄酮;通过UPLC-DAD/ESI-TOF-MS从石榴皮提取物中共检测出的5 种酚类化合物(分别为α-安石榴苷、β-安石榴苷、鞣花酸己糖苷、鞣花酸脱氧己糖苷、鞣花酸),其在胃液消化过程中损失率小于肠液消化,说明石榴皮中酚类物质在酸性环境中结构稳定。  相似文献

A new method for pomegranate peel application in food industries was developed based on the ultrasound-assisted extraction of phenolic compounds and their subsequent encapsulation by spray drying. The effects of various parameters on extraction yield, on encapsulation efficiency/yield, and on the main physical properties of the microcapsules (moisture content, bulk density, rehydration ability) were studied. Ultrasound was found to increase extraction yield, but mainly to shorten the treatment time by over 20 times. The maximum encapsulation efficiency was 99.80% and the optimum operating conditions were found to be: wall material, maltodextrin/whey protein isolate (50:50); inlet air temperature, 150 °C; drying air flow rate, 17.5 m3/h; ratio of wall to core material, 9/1; feed solid concentration, 30% (w/w). The encapsulated phenolic extract was found efficient in improving the shelf life of hazelnut paste, in spite of the limited solubility of the crude extract in such a high lipid content matrix.Industrial relevancePomegranate peels, a by-product of pomegranate juice and concentrate industries, present a wide range of pharmaceutical and nutraceutical properties. Therefore, the peels could have more beneficial applications in food industries instead of being used as animal feed or in commercial cosmetic products. Evaluation of phenolic contents and their in vitro bioactive functional properties of cowpea and horse gram flours in comparison to chickpea flour revealed that horse gram flour, which contain highest concentrations of total phenolics and total flavonoids was found to be the most active 2,2′-diphenyl-1-picrylhydrazyl (DPPH) radical scavenger. Hydrogen peroxide scavenging capacity, reducing power and metal chelating abilities were significantly higher in cowpea and horse gram than chickpea flour. Phenolic extracts from flours showed distinct variations in the inhibition of enzymes associated with hyperglycemia and hypertension. Although, α-glucosidase inhibitory activity was superior in cowpea (IC50 52.8 μg/ml), α-amylase (IC50 96.4 μg/ml) and angiotensin-I converting enzyme (IC50 32.8 μg/ml) inhibitory activities were predominant in horse gram. These results provided useful information for effective utilisation of legume flours as ingredients in composite flours and for the development of food products with health promoting functions.  相似文献

In this study, effect of ellagic acid (EA), gallic acid (GA), grape seed extract (GSE), pomegranate peel extract (PPE), and peppermint essential oil (PEO) supplementation on sensorial and antioxidative properties of ice cream was determined. Additionally, survival of Lactobacillus casei Shirota in ice cream and its interaction with the supplements were also investigated. All the supplements increased total phenolic content of ice creams significantly (P?<?0.05). DPPH radical scavenging activities of ice creams supplemented with EA, GA, PPE, or GSE were higher than that of control group. Moreover, PPE made the best contribution to phenolic content and antiradical activity of ice cream. L. casei Shirota was highly adapted to ice cream media and survived with high numbers (7.21?log?cfu/mL) during the storage period for 60?days. Supplementation of ice creams provided lower decrease in bacterial number, and the best bacterial behaviors were observed in the ice creams supplemented with GA or PPE. All ice cream samples tested in this study had generally acceptable sensorial scores. In conclusion, EA, GA, PPE, GSE, and PEO could be used to enhance the phenolic content of ice cream and to gain antioxidant property, and there was a good interaction between L. casei and these phenolic compounds. Additionally, PPE supplementation provided the best result for all concerns.  相似文献

为获得超声波辅助纤维素酶提取石榴幼果多酚的最佳工艺,以多酚得率为考察指标,在单因素试验基础上,通过Plackett-Burman(PB)试验设计筛选提取工艺中影响多酚得率的显著性因素,Box-Behnken试验设计和响应面分析法优化得出超声波辅助纤维素酶提取石榴幼果多酚的最佳工艺条件,采用体外α-葡萄糖苷酶抑制模型研究石榴幼果多酚对α-葡萄糖苷酶的抑制作用及其动力学性质。结果表明:当超声功率318 W、加酶量23 U/m L、酶解温度46℃和酶解时间2.7 h时,石榴幼果多酚平均得率为11.65%,与预测值误差很小。石榴幼果多酚具有较强抑制α-葡萄糖苷酶的活性,质量浓度为1.20 mg/m L时,对α-葡萄糖苷酶抑制率达到70.3%,抑制作用的IC50为0.747 mg/m L。在质量浓度0.24~1.20 mg/m L范围内,石榴幼果多酚与对α-葡萄糖苷酶抑制效果之间呈现一定的正相关关系,其抑制机理属于可逆性抑制和非竞争性抑制。  相似文献

Grape juice residue was incorporated into ice cream at 2.5%, 5.0% and 10.0% to obtain a product with functional characteristics. The chemical composition, colour, total phenolic compounds, antioxidant capacity and sensory acceptance of the ice creams were analysed at day 0 and after 40 days of storage. Ice creams containing grape juice residue had a higher concentration of phenolic compounds and antioxidant activity compared to the control samples. Consumers’ acceptance was similar for all products. At the end of storage, the polyphenol content and antioxidant activity had decreased. Grape juice residue has potential as a natural source of antioxidants to develop new products.  相似文献

Cocoa brew showed a dose-dependent inhibitory activity against α-glucosidase (IC50 7.87 mg/mL). The cocoa brew was fractionated by ultrafiltration in a low molecular weight fraction (LMW < 10 kDa) and two melanoidin-rich fractions called high molecular weight (HMW > 30 kDa) and intermediate molecular weight (IMW 10–30 kDa) fractions. All fractions tested caused some inhibition with IMW that was the most active (IC50 2.37 mg/mL). LMW fraction was separated with Sephadex LH-20 in an unbound (containing monomeric and dimeric catechins) and a bound fraction. All the inhibitory activity was recovered in the unbound fraction. All the phenolic compounds identified with HPLC showed α-glucosidase inhibitory activity. IMW and HMW fractions were fractionated by ethanol precipitation. The fractions from IMW precipitated with 75 and 25% ethanol were found to contain power inhibitors of α-glucosidase activity (IC50 0.87 and 1.01 mg/mL, respectively). In the HMW sample, the fractions precipitated with 50 and 75% ethanol were found to be active against α-glucosidase activity. Most of the inhibitory activity against α-glucosidase of cocoa brew was due to the LMW fraction (56%) whereas IMW and HMW contributed for about 47% of the inhibitory activity. This study suggests that different components of cocoa may influence α-glucosidase activity.  相似文献

Corchorus olitorius leaf is consumed in various parts of the world as leafy vegetable and folk remedy for the management of some degenerative diseases with dearth of information on its biochemical rationale. Therefore, this study sought to characterize the inhibitory action of polyphenol-rich extracts (free and bound) of C. olitorius on α-amylase, α-glucosidase and angiotensin I converting enzyme (ACE), as well as to identify the phenolic compound responsible for these activities. Our findings revealed that the extracts inhibited α-amylase and α-glucosidase (12.5–50.0 μg/mL), and ACE (10.0–50.0 μg/mL) in dose-dependently with free extracts having significantly (P < 0.05) higher α-amylase (17.5 μg/mL), α-glucosidase (11.4 μg/mL) and ACE (15.7 μg/mL) inhibitory activities as revealed by the IC50. Reversed-phase HPLC analysis of the extracts revealed chlorogenic acid (7.5 mg/100 g) and isorhamnetin (51.1 mg/100 g) as the main phenolics in the free extract and caffeic acid (58.1 mg/100 g) in the bound extract. Therefore, the enzyme inhibitory activity of C. olitorius extracts may be attributed to the presence of caffeic acid, chlorogenic acid and isorhamnetin, thus justifying its use in folklore for the management of diabetes and hypertension.  相似文献

ABSTRACT: Ascophyllum nodosum is a brown seaweed that grows abundantly in the Northeast coastal region. In this study, the potential of A. nodosum for type 2 diabetes management through antioxidant-mediated α-glucosidase and α-amylase inhibition was investigated. After the initial screening of 4 locally harvested seaweeds, A. nodosum was chosen for its highest phenolic content and was subjected to water extraction. Among extraction ratios of 50 g to 100 to 1000 mL at room temperature, 50 g/400 mL yielded the highest phenolic content of 4.5 mg/g wet weight. For evaluation of extraction temperature ranging from 20 to 80 °C, 50 g/400 mL was chosen as a minimum amount of extractant. Among temperatures studied, extraction at 80 °C resulted in the highest total phenolic contents (4.2 mg/g wet weight). All extracts had similar levels of antioxidant activity in the range of 60% to 70% in terms of 1, 1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity. The 80 °C extract had the highest α-glucosidase and α-amylase inhibitory activity with IC50 of 0.24 and 1.34 μg phenolics, respectively, compared to the IC50 of acarbose, reference inhibitor, being 0.37 and 0.68 μg. The results show that fresh A. nodosum has strong α-glucosidase and mild α-amylase inhibitory activities that correlated with phenolic contents. This study suggests a nutraceutical potential of A. nodosum based on phytochemical antioxidant and antihyperglycemia activities.  相似文献

Immature faba bean seeds of five varieties were investigated for changes in phenolic composition, antioxidant capacity and inhibitory effects on α-glucosidase and lipase at three edible stages (S1, S2 and S3). Phenolics and antioxidant capacity in seed coats were significantly higher than those in cotyledons, and the highest was found of FD18 at stage S3. While phenolic content in cotyledons decreased steadily with the harvest time delayed. Eight phenolic compounds were confirmed with epicatechin and catechin prominent and their levels showed a great variability depending on varieties, maturation stages and plant parts of faba bean seeds. Unexpectedly, the inhibitory effects of purified phenolics on α-glucosidase were even stronger than that of the positive control acarbose, and the inhibitory effects on lipase in the cotyledons declined gradually from stage S1 to S3. Significant positive correlations between phenolics from seed coats and antioxidant capacity, and phenolics from cotyledons and the enzyme inhibitory effects were observed. Overall, the immature faba bean seed, especially its seed coat, has great potential as a functional food.  相似文献

Phenolic extracts of 4 Vigna species of legumes (mung bean, moth bean, and black and red varieties of adzuki beans) were evaluated for phenolic contents, antioxidant activities, and inhibitory properties against α-glucosidase and pancreatic lipase. Results showed that adzuki bean varieties contain higher phenolic indexes than mung bean and moth beans. Adzuki bean (black) variety was found to be the most active 2,2′-diphenyl-1-picrylhydrazyl and superoxide anion scavenger. However, the hydrogen peroxide scavenging and metal chelating abilities were significantly higher in adzuki bean (red) variety. Mung bean exhibited least antioxidant activities in all the methods tested. Phenolic extracts from these legumes also showed distinct variations in the inhibition of enzymes associated with hyperglycemia and hyperlipidemia. Inhibitory activities of all the extracts against lipase were found to be more potent than α-glucosidase. Although, α-glucosidase inhibitory activity was superior in the black variety of adzuki bean (IC(50,) 26.28 mg/mL), both adzuki bean varieties (black and red) along with moth bean showed strong inhibitory activities on lipase with no significant difference in their IC(50) values (7.32 to 9.85 mg/mL). These results suggest that Vigna species of legumes are potential source of antioxidant phenolics and also great sources of strong natural inhibitors for α-glucosidase and lipase activities. This information may help for effective utilization of these legumes as functional food ingredients for promoting health. Practical Application: Vigna species of legumes are good sources of phenolic antioxidants and strong natural inhibitors of enzymes associated with diabetes and obesity. Therefore, utilization of these legumes in the development of functional foods with increased therapeutic value would be a significant step toward health promotion and wellness.  相似文献

The optimum extraction conditions of phenolic compounds from ginger were evaluated with respect to antioxidant activity and angiotensin converting enzyme and α-glucosidase inhibitory activities. Free phenolics were extracted under conditions that varied according to extraction time, temperature, and solvent type (water, acetone, and methanol). Acid and base hydrolysis reactions were used to obtain bound phenolic compounds from ginger. The results showed that the type of solvent used and the temperature and time of extraction needed for maximal total phenolic content, antioxidant activity, and angiotensin converting enzyme inhibitory activity differed greatly from solvent conditions and showed the greatest α-glucosidase inhibitory activity. The predominant free phenolics in the methanol extracts included diosmin, thymol, and carvacrol, which varied greatly according to solvent extraction conditions (i.e., time and temperature). Diosmin was the predominant bound phenolic compound of the methanol extracts. The present study findings indicate that differing solvent extraction protocols involving extraction time and temperature for ginger need to be explored to generate specific optimal bioactivities of the extracts, which are related to the pattern of predominant phenolics in those extracts.  相似文献

Pomegranate is an important source of bioactive compounds and has been used for folk medicine for many centuries. Pomegranate juice has been demonstrated to be high in antioxidant activity and is effective in the prevention of atherosclerosis. In a previous study, we found that pomegranate peel had the highest antioxidant activity among the peel, pulp and seed fractions of 28 kinds of fruits commonly consumed in China as determined by FRAP (ferric reducing antioxidant power) assay. In this study, we extracted antioxidants from pomegranate peel, using a mixture of ethanol, methanol and acetone, and the antioxidant properties of the extract were further investigated as compared with the pulp extract. The contents of total phenolics, flavonoids, proathocyanidins and ascorbic acid were also measured. The results showed that pomegranate peel extract had markedly higher antioxidant capacity than the pulp extract in scavenging or preventive capacity against superoxide anion, hydroxyl and peroxyl radicals as well as inhibiting CuSO4-induced LDL oxidation. The contents of total phenolics, flavonoids and proathocyanidins were also higher in peel extract than in pulp extract. The large amount of phenolics contained in peel extract may cause its strong antioxidant ability. We concluded that pomegranate peel extract appeared to have more potential as a health supplement rich in natural antioxidants than the pulp extract and merits further intensive study.  相似文献

In the current study, we evaluated total phenolics, phenolic profile, and oxygen radical absorbance capacity (ORAC) of onion (Allium cepa L.). The inhibitory activity of onion extracts against porcine pancreatic α-amylase and rat intestinal α-glucosidase was also investigated. Ethyl alcohol extract of onion skin had the highest α-glucosidase inhibitory activity, ORAC value and total phenolic content, followed by water extract of skin, ethyl alcohol extract of pulp, and water extract of pulp. The α-glucosidase inhibitory activity of the extracts was compared to selected specific phenolics detected in the extracts using high performance liquid chromatography (HPLC). Quercetin, a major phenolic compound in onion extract had high α-glucosidase inhibitiory activity. The α-glucosidase inhibitory activity of the onion extracts correlated to the phenolic content and antioxidant activity of the extracts. These results suggest that onion which has high quercetin content has the potential to contribute as a dietary supplement for controlling hyperglycemia and oxidative stress-linked diabetes complications.  相似文献
