在羟基磷灰石色谱上施加电场 ,发展了一种新的电色谱分离技术 .对蛋白质电泳和溶液电渗对分离特性的影响进行了实验研究 ,测量了羟基磷灰石电色谱装置中膜、膜与填充介质在不同浓度磷酸缓冲溶液和在不同电流下产生的电渗流 ,分别计算出由膜和羟基磷灰石电渗而产生的推动力 .实验结果表明电渗流量随缓冲溶液离子强度的降低和电流的增加而增加 .在施加电场的情况下 ,羟基磷灰石对牛血清白蛋白的吸附速度加快 ,动态吸附量增加 .分别考察了不同洗脱条件下电场对洗脱效果的影响 .在高浓度的洗脱溶液下 ,电渗的作用很小 ,电场的作用主要是电迁移的作用 ,可以通过选择适宜的电场方向来提高分离精度 ;在低浓度的洗脱溶液下 ,电渗的作用有效地促进了洗脱液与介质颗粒的传质 ,提高了洗脱效果 .在羟基磷灰石色谱上施加电场提高了该色谱分离过程的速率和效率 . 相似文献

本文介绍了一种新型制备色谱分离技术──连续环状离子交换色谱CAC,建立了二维稳定连续环状离子交换色谱的数学模型。在一定条件下经过适当转化,二维稳态CAC过程可转化为一维非稳定色谱过程。分别改变CAC进料浓度、进料流速及冲洗流速等操作条件,测定了CAC对果葡糖的分离性能。同时,用固定床线性色谱理论对CAC进行预测,其预测值与实验结果较好地相吻合。实验得出,连续环状离子交换色谱对果葡糖具有较好的分离性能。 相似文献

A counteracting chromatographic electrophoresis(CACE)separation device has been success-fully built and tested.Experiments are performed with protein mixtures of BSA,Hg,Mg and Cyto-cto study the separation performances,concentration profiles,start-up methodology as well as influ-ences of pH and ionic strength on the separation process.Mathematical models based on the fluiddynamics of the porous medium are proposed.Experiments indicate that the CACE process has agood commercial potential in protein engineering and fine chemicals purification. 相似文献

Dynamic field gradient focusing (DFGF) uses an electric field gradient opposed by a counter‐flow of buffer to separate milligrams of proteins according to their electrophoretic mobilities. A nonlinear model of protein separation in a preparative‐scale DFGF device was developed to aid in refining the instrument's design and finding optimal run conditions prior to performing experiments. The model predicted the focal points of bovine serum albumin (BSA), and bovine hemoglobin (Hb) to within the 95% confidence intervals about the means of the experimental values. The resolution between the proteins in the model was 2.08, which was 3% less than the lower limit of the 95% confidence interval about the experimental value. The model predicted 67% more dispersion than was present in the experimental device, which made the simulated BSA peak 22% wider than the experimentally measured width. © 2008 American Institute of Chemical Engineers AIChE J, 2009 相似文献

Uniformly sized molecularly imprinted polymers (MIPs) for nimodipine have been prepared in an aqueous system by multi‐step swelling and polymerization method, utilizing 4‐vinylpyridine (4‐VPY) or methacrylic acid (MAA) as a functional monomer, ethylene glycol dimethacrylate (EDMA) as a cross‐linking agent. Scanning electron microscopy was used to identify the structure features of the obtained polymers. Further, the influences of some chromatographic conditions were examined to explore the possible recognition mechanism. The results reveal that stable molecularly imprinted polymeric microspheres with good size monodispersity were obtained, and the polymer beads showed specific recognition for the template molecule and some other dihydropyridine calcium antagonists (DHPs). Besides hydrophobic interaction, the molecular shape complementation of DHPs and the MIPs seems to play an important role in the retention and recognition of DHPs. The Scatchard analysis showed that two kinds of binding sites existed in the MIPs. The MIPs was then used as a high‐performance liquid chromatography (HPLC) separation medium to simultaneously concentrate and purify nimodipine in plasma. The results reveal that the obtained MIPs could be used for on‐line concentration, purification, and measurement of nimodipine in biological samples. © 2008 Wiley Periodicals, Inc. 用浸渍法制备了负载型MxOy-V2O5/Al2O3催化剂,同时考察了不同的制备方法对MoO3-V2O5/Al2O3催化剂的性能影响。用BET、H2-TPR、TPD平衡法等考察了催化剂的比表面积、氧化还原性能、表面氧脱附性能,结果表明,添加变价助剂Mo、Fe和Cr氧化物会增加供氧数目,提高α-蒎烯转化率。MoO3-V2O5/Al2O3催化剂表面O 2-和O-物种较多,活性也较好,桃金娘烯醛收率较高。MoO3-V2O5/Al2O3催化剂的制备方法影响其催化性能,表面化学改性法制备的催化剂表面具有较多O2-和O-物种,有利于催化剂的活性提高,α-蒎烯的转化率达88.9%,桃金娘烯醛的收率为79.1%。 相似文献
