The aim of this study was to analyze the influence of nanoporous structure of polymeric biomaterials on the in vitro osteogenic induction of human stem cells. An electronic search in three databases (MEDLINE, SCOPUS, and Web of Science) was performed for articles that were published before May 2018. In vitro studies were included if they met the following criteria: (1) the use of polymeric scaffolds (natural or synthetic); (2) the co-culture of human stem cells with the scaffold; and (3) cell viability, proliferation, and osteogenic differentiation assays. The main characteristics of the published studies were summarized, and a quality assessment tool was used to analyze methodological features. Eighty-eight potential articles were firstly retrieved. Thirteen were eligible for qualitative analysis. Only three studies characterized cell stemness. Nanostructure of the scaffolds showed a significant influence on viability, proliferation, and osteogenic differentiation of human stem cells. Combination of porosity between 72 and 93% and a large range diameter between 50 and 224 μm resulted in more remarkable cellular proliferation and differentiation. Porous polymeric scaffolds can be functionalized by stem cells leading to osteogenic induction. High standards of laboratory practice and accurate methodological reporting are essential for the credibility of the results.  相似文献

Advances in research have boosted therapy development for congenital disorders of glycosylation (CDG), a group of rare genetic disorders affecting protein and lipid glycosylation and glycosylphosphatidylinositol anchor biosynthesis. The (re)use of known drugs for novel medical purposes, known as drug repositioning, is growing for both common and rare disorders. The latest innovation concerns the rational search for repositioned molecules which also benefits from artificial intelligence (AI). Compared to traditional methods, drug repositioning accelerates the overall drug discovery process while saving costs. This is particularly valuable for rare diseases. AI tools have proven their worth in diagnosis, in disease classification and characterization, and ultimately in therapy discovery in rare diseases. The availability of biomarkers and reliable disease models is critical for research and development of new drugs, especially for rare and heterogeneous diseases such as CDG. This work reviews the literature related to repositioned drugs for CDG, discovered by serendipity or through a systemic approach. Recent advances in biomarkers and disease models are also outlined as well as stakeholders’ views on AI for therapy discovery in CDG.  相似文献

Mass spectrometry (MS)-based techniques can be a powerful tool to identify neuropsychiatric disorder biomarkers, improving prediction and diagnosis ability. Here, we evaluate the efficacy of MS proteomics applied to human peripheral fluids of schizophrenia (SCZ) patients to identify disease biomarkers and relevant networks of biological pathways. Following PRISMA guidelines, a search was performed for studies that used MS proteomics approaches to identify proteomic differences between SCZ patients and healthy control groups (PROSPERO database: CRD42021274183). Nineteen articles fulfilled the inclusion criteria, allowing the identification of 217 differentially expressed proteins. Gene ontology analysis identified lipid metabolism, complement and coagulation cascades, and immune response as the main enriched biological pathways. Meta-analysis results suggest the upregulation of FCN3 and downregulation of APO1, APOA2, APOC1, and APOC3 in SCZ patients. Despite the proven ability of MS proteomics to characterize SCZ, several confounding factors contribute to the heterogeneity of the findings. In the future, we encourage the scientific community to perform studies with more extensive sampling and validation cohorts, integrating omics with bioinformatics tools to provide additional comprehension of differentially expressed proteins. The produced information could harbor potential proteomic biomarkers of SCZ, contributing to individualized prognosis and stratification strategies, besides aiding in the differential diagnosis.  相似文献

In this work the synthesis of cubic, FDU-1 type, ordered mesoporous silica (OMS) was developed from two types of silicon source, tetraethyl orthosilicate (TEOS) and a less expensive compound, sodium silicate (Na2Si3O7), in the presence of a new triblock copolymer template Vorasurf 504 (EO38BO46EO38). For both silicon precursors the synthesis temperature was evaluated. For TEOS the effect of polymer dissolution in methanol and the acid solution (HCl and HBr) on the material structure was analyzed. For Na2Si3O7 the influence of the polymer mass and the hydrothermal treatment time were the explored experimental parameters. The samples were examined by Small Angle X-ray Scattering (SAXS) and Nitrogen Sorption. For both precursors the decrease on the synthesis temperature from ambient, ?25 °C, to ?15 °C improved the ordered porous structure. For TEOS, the SAXS results showed that there is an optimum amount of hydrophobic methanol that contributed to dissolve the polymer but did not provoke structural disorder. The less electronegative Br? ions, when compared to Cl?, induced a more ordered porous structure, higher surface areas and larger lattice parameters. For Na2Si3O7 the increase on the hydrothermal treatment time as well as the use of an optimized amount of polymer promoted a better ordered porous structure.  相似文献

Electrical impedance tomography (EIT) is an imaging technique that attempts to reconstruct the impedance distribution inside an object from the impedance between electrodes placed on the object surface. The EIT reconstruction problem can be approached as a nonlinear nonconvex optimization problem in which one tries to maximize the matching between a simulated impedance problem and the observed data. This nonlinear optimization problem is often ill-posed, and not very suited to methods that evaluate derivatives of the objective function. It may be approached by simulated annealing (SA), but at a large computational cost due to the expensive evaluation process of the objective function, which involves a full simulation of the impedance problem at each iteration. A variation of SA is proposed in which the objective function is evaluated only partially, while ensuring boundaries on the behavior of the modified algorithm.  相似文献

In the present investigation, a scanning electron microscopy analysis was performed to evaluate the effects of the topical application of ethylenediaminetetraacetic acid (EDTA) gel associated with Cetavlon (EDTAC) in removing the smear layer and exposing collagen fibers following root surface instrumentation. Twenty-eight teeth from adult humans, single rooted and scheduled for extraction due to periodontal reasons, were selected. Each tooth was submitted to manual (scaling and root planing) instrumentation alone or combined with ultrasonic instruments, with or without etching using a 24% EDTAC gel. Following extraction, specimens were processed and examined under a scanning electron microscope. A comparative morphological semi-quantitative analysis was performed; the intensity of the smear layer and the decalcification of cementum and dentinal surfaces were graded in 12 sets using an arbitrary scale ranging from 1 (area covered by a smear layer) to 4 (no smear layer). Root debridement with hand instruments alone or combined with ultrasonic instruments resulted in a similar smear layer covering the root surfaces. The smear layer was successfully removed from the surfaces treated with EDTAC, which exhibited numerous exposed dentinal tubules and collagen fibers. This study supports the hypothesis that manual instrumentation alone or instrumentation combined with ultrasonic instrumentation is unable to remove the smear layer, whereas the subsequent topical application of EDTAC gel effectively removes the smear layer, uncovers dentinal openings and exposes collagen fibers.  相似文献

Robotic soccer is nowadays a popular research domain in the area of multi-robot systems. In the context of RoboCup, the Middle Size League is one of the most challenging. This paper presents an efficient omnidirectional vision system for real-time object detection, developed for the robotic soccer team of the University of Aveiro, CAMBADA. The vision system is used to find the ball and white lines, which are used for self-localization, as well as to find the presence of obstacles. Algorithms for detecting these objects and also for calibrating most of the parameters of the vision system are presented in this paper. We also propose an efficient approach for detecting arbitrary FIFA balls, which is an important topic of research in the Middle Size League. The experimental results that we present show the effectiveness of our algorithms, both in terms of accuracy and processing time, as well as the results that the team has been achieving: 1st place in RoboCup 2008, 3rd place in 2009 and 1st place in the mandatory technical challenge in RoboCup 2009, where the robots have to play with an arbitrary standard FIFA ball.  相似文献

A content analysis was conducted to examine the portrayal of social aggression in the 50 most popular television programs among 2‐ to 11‐year‐old children. Results revealed that 92% of the programs in the sample contained some social aggression. On average, there were 14 different incidents of social aggression per hour in these shows, or one every 4 minutes. Compared to the portrayals of physical aggression, social aggression was more likely to be enacted by an attractive perpetrator, to be featured in a humorous context, and neither rewarded or punished. In these ways, social aggression on television poses more of a risk for imitation and learning than do portrayals of physical aggression. Findings are discussed in terms of social cognitive theory.  相似文献

A dynamic latched comparator can suffer from three non-idealities: offset voltage, random noise and kickback noise. Specifically in an analog-to-digital converter (ADC) the kickback noise of a comparator can noticeably affect the settling time and accuracy of the decision. This work offers an analytical treatment of kickback noise generation, and proposes a synchronized kickback noise cancellation technique, which is achieved via placing clocked NMOS-PMOS capacitors at the proper nodes to cancel out effectively those unwanted charges (electrons or holes) under different operating regions of the MOS devices. The technique is clock-rate insensitive and particularly suitable for the SAR-type ADC as it will not alter the charge stored in the capacitor array. Optimized in 65-nm CMOS the kickback noise is <±0.3 mV in 30× Monte–Carlo simulations at both 50- and 500-MHz clock rates. For 10-bit resolution in a full scale of 1 Vpp, the kickback noise of the proposed comparator comparing with the conventional one is improved by 48×, from 6.27 to 0.13 LSB.  相似文献
