The purpose of the present investigation was to test whether permanent skeletal muscle cells (rat L6 cells) could serve as an in vitro model for α-tocopherol (αTocH) biodiscrimination studies. L6 cells were incubated in the presence of high density lipoprotein (HDL), low density lipoprotein (LDL), and very low density lipoprotein (VLDL) labeled in the lipid moiety with either all-rac-or RRR-[14C]αTocH. These incubations were performed either in the absence or in the presence of exogenously added bovine lipoprotein lipase (LPL) since skeletal muscle is one of the major expression sites of LPL in vivo. Time-dependent uptake studies (up to 24 h) in the absence of LPL have shown that equipotent doses of all-rac- and RRR-[14C]αTocH (1.36∶1) led to almost identical accumulation of the tracer, independent of the lipoprotein class used as αTocH carrier. With regard to αTocH donor capacity, it appeared that HDL is the most potent αTocH donor, followed by LDL and VLDL. In the presence of LPL, all-rac- and RRR-[14C]αTocH uptake was significantly enhanced (between two- and tenfold). Biodiscrimination studies using chiral high-performance liquid chromatographic analysis with radiometric detection of the corresponding methyl ether derivatives on a Chiralcel OD column have demonstrated that the 2S-and 2R-isomers of αTocH were taken up in a 1∶1 ratio by L6 cells independent of the absence or presence of LPL. In addition, we have not observed biodiscrimination between the four 2R-isomers, i.e., there was no preferential accumulation of the RRR-isomer. These data suggest that L6 cells do not discriminate between different αTocH isomers and that the addition of endogenous LPL significantly enhances the uptake of RRR- and all-rac-αTocH.  相似文献

The exploratory study with homeless women presented in this paper aims to highlight two shortcomings in the current literature on homelessness, which accompany the predominant conceptualisation of ‘home’ as a place of safety and security, and homelessness as a primarily structural issue characterised by ‘residential instability’. The first is the paucity of research on homeless women in their own right and the second the lack of studies which examine underlying reasons for residential instability from homeless women's own perspectives. An intensive, small sample interview study was carried out with 12 regular women users of a day centre for homeless people (Brighton, south of England). They were asked about their patterns of residence, reasons for moving, definitions of ‘house’, ‘home’, and ‘ideal home’, and they gave their housing histories in some detail. In a quantitative analysis, their patterns of moving and definitions of ‘home’ were compared to those of a group of securely housed women. Number of moves was comparable in both groups, but for the homeless women the majority of moves had been made to avoid abuse and social services relocations. Whilst all of the securely housed women could confidently define a difference in meaning between a ‘house’ and a ‘home’, only 3 of the 12 homeless women did. They equated ‘home’ with safety and security, the same terms used in the literature to define what housing means, and the two most salient features largely absent in homeless women's experience of housing. The meaning of home is further explored in a qualitative analysis, where the themes of safety and security—or rather their absence in abusive relationships—are traced through the homeless women's childhood, adolescence and adulthood. These exploratory findings question an easy equation between ‘residential instability’ and homelessness, and highlight the need to investigate further the reasons why women leave housing, and the relationship this has to an understanding of what ‘home’ means. Whilst current formulations suggest that the homelessness of women is a problem, and housing the solution, this study suggests that housing is the problem—homelessness may well be a solution.  相似文献

The distinction between underlying and superficial linguistic structure is a staple of modern cognitive psychology. Despite increasingly diverse conceptions of syntactic relations in linguistic theory, the received view in psycholinguistics has remained one in which the entities assigned to underlying relations may assume different surface relations. This view is examined in the context of language production and evidence is reviewed that the disposition to bind animate entities to the surface subject relation is a basic feature of language use, suggesting that mappings from conceptual categories to syntactic relations form a main support of the bridge from conception to language. Proceeding on this assumption, an evaluation of competing accounts of the mapping process in production is given. The glycolytic modulator TP53-Inducible Glycolysis and Apoptosis Regulator (TIGAR) is overexpressed in several types of cancer and has a role in metabolic rewiring during tumor development. However, little is known about the role of this enzyme in proliferative tissues under physiological conditions. In the current work, we analysed the role of TIGAR in primary human lymphocytes stimulated with the mitotic agent Concanavalin A (ConA). We found that TIGAR expression was induced in stimulated lymphocytes through the PI3K/AKT pathway, since Akti-1/2 and LY294002 inhibitors prevented the upregulation of TIGAR in response to ConA. In addition, suppression of TIGAR expression by siRNA decreased the levels of the proliferative marker PCNA and increased cellular ROS levels. In this model, TIGAR was found to support the activity of glucose 6-phosphate dehydrogenase (G6PDH), the first enzyme of the pentose phosphate pathway (PPP), since the inhibition of TIGAR reduced G6PDH activity and increased autophagy. In conclusion, we demonstrate here that TIGAR is upregulated in stimulated human lymphocytes through the PI3K/AKT signaling pathway, which contributes to the redirection of the carbon flux to the PPP.  相似文献

A combined process of lipase (E.C. 3.1.1.3) catalysis and extraction of product with supercritical carbon dioxide was studied. The effect of different flow rates of the extraction fluid on the selective removal of the ethyl esters (EE) synthesized in a lipase-catalyzed alcoholysis of cod liver oil with ethanol was investigated. The faster the flow rate, the faster the extraction rate and the higher the recovery of EE. For example, after a 270-min extraction, the total recovery of EE was 1520 mg for a flow rate of 0.3 liter carbon dioxide at atmospheric pressure and room temperature/min (NL/min) as compared to 250 mg when 0.015 NL/min was used. The concentration of EE in the carbon dioxide was found to decrease with increasing flow rate, which indicates that the rate of diffusion of EE limits their extraction at fast flow rates. A high flow rate was found to result in a more selective extraction of EE, i.e., less amounts of other lipid components present in the reaction mixture were coextracted with the EE. Further, by increasing the flow rate, the equilibrium of the reaction was shifted slightly toward ester synthesis. An increase in the flow rate from 0.015 to 0.075 NL/min resulted in an approximately 10% increase in total conversion (from 73 to 82%), whereas only a negligible increase was obtained when the flow rate was increased further to 0.15 NL/min.  相似文献

Porous carbon materials represent prospective materials for absorbers, filters, and electronic applications. Carbon fibers with high surface areas can be produced from polyacrylonitrile and spun as thin fibers from solution. The resulting polymer fibers are first stabilized to obtain conjugated ribbons and then carbonized to graphitic structures in a second high‐temperature step in an inert atmosphere. In this study, we investigated a previously described fast laser‐heating process that delivered fibers with a higher crystallinity and surface area compared to the thermally carbonized fibers. In a subsequent KOH‐activation step, the crystalline domains were exfoliated, and the surface of the fibers became macroporous. This led to a reduced specific surface area but a higher capacitance compared to thermally carbonized nanofibers. 1‐Alkynylcyclopropyl tosylates react with alkylmagnesium halides in the presence of catalytic ferric acetylacetonate [Fe(acac)3] under net propargylic substitution; allene formation, which is the prevalent reactivity mode of propargylic substrates otherwise, was noticed as a side reaction only when branched alkyl‐ or aryl‐Grignard reagents were used. These transformations represent the first successful iron‐catalyzed cross‐coupling reactions of tert‐alkyl electrophiles.

Non-targeted effects (NTE) of ionizing radiation may initiate myeloid neoplasms (MN). Here, protein mediators (I) in irradiated human mesenchymal stromal cells (MSC) as the NTE source, (II) in MSC conditioned supernatant and (III) in human bone marrow CD34+ cells undergoing genotoxic NTE were investigated. Healthy sublethal irradiated MSC showed significantly increased levels of reactive oxygen species. These cells responded by increasing intracellular abundance of proteins involved in proteasomal degradation, protein translation, cytoskeleton dynamics, nucleocytoplasmic shuttling, and those with antioxidant activity. Among the increased proteins were THY1 and GNA11/14, which are signaling proteins with hitherto unknown functions in the radiation response and NTE. In the corresponding MSC conditioned medium, the three chaperones GRP78, CALR, and PDIA3 were increased. Together with GPI, these were the only four altered proteins, which were associated with the observed genotoxic NTE. Healthy CD34+ cells cultured in MSC conditioned medium suffered from more than a six-fold increase in γH2AX focal staining, indicative for DNA double-strand breaks, as well as numerical and structural chromosomal aberrations within three days. At this stage, five proteins were altered, among them IQGAP1, HMGB1, and PA2G4, which are involved in malign development. In summary, our data provide novel insights into three sequential steps of genotoxic signaling from irradiated MSC to CD34+ cells, implicating that induced NTE might initiate the development of MN.  相似文献
