Polyphosphorylated and perphosphorylated hexopyranose monosaccharides and disaccharides were synthesized from parent or partially protected carbohydrates as potential allosteric effectors of hemoglobin. A study toward the construction of seven- and eight-membered cyclic pyrophosphates was also performed on the sugars which had the proper orientation, protection, and number of phosphates. All final compounds were tested for their efficiency on oxygen release from human hemoglobin. Several compounds presented higher potency than myo-inositol hexakisphosphate, which is the most efficient of the known allosteric effectors of hemoglobin. Structure-activity relationships were analyzed. The affinity and efficiency depend on the number of phosphates attached to the carbohydrate skeleton and are related primarily to the number of negative charges present. Myo-inositol trispyrophosphate (ITPP), a synthetic allosteric effector of hemoglobin, increases the regulated oxygen-releasing capacity of red blood cells (RBCs), leading to suppression of hypoxia-inducible factor 1α (HIF-1α) and to down-regulation of hypoxia-inducible genes such as vascular endothelial growth factor (VEGF). As a consequence, tumor growth is markedly affected. The effect of weekly intravenous injection of ITPP on an orthotopic, syngenic rat hepatocellular carcinoma (HCC) model was compared to that for untreated animals and animals subjected to conventional Doxorubicin chemotherapy. The longitudinal examination of HCC was performed by microCT imaging, and the cellular and molecular changes were evaluated by histology and Western blotting analysis of HIF-1α, VEGF, and caspase-3 gene expression in the tumor and in the surrounding liver. Hematologic impact was evaluated by blood cell-count measurement and determination of P50 (oxygen partial pressure for a 50 % oxygen saturation of hemoglobin). The HCC evaluation by microCT revealed a high potency of ITPP for tumor growth inhibition, thus allowing long-term survival and even cure of almost all the treated animals. The P50 value of hemoglobin in RBCs underwent a shift of 30 % following ITPP injection. Under these conditions, HIF-1α activity was strongly decreased, VEGF expression was down-regulated, and apoptosis was induced in HCC and surrounding liver cells, as indicated by Caspase-3 expression. ITPP did not affect hematologic parameters during treatment. The observations of in vivo tumor eradication suggest a significant clinical potential for ITPP in cancer therapy. 相似文献

The aim of the present work is the study of some physicochemical properties of two current commercial dental self‐curing two component composites, i.e., of Concise? (3M?, Dental Products, St. Paul, USA), and Alfacomp (VOCO GmbH, Germany). Based on their filler type Concise is characterized as “conventional” or “macrofilled” composite and Alfacomp as “hybrid.” The resin matrix of Concise is a copolymer of 2‐bis[4‐2‐hydroxy‐3‐(methacryloxy)‐propyl]phenyl propane (Bis‐GMA)/triethyleneglycol dimethacrylate, while that of Alfacomp a homopolymer of Bis‐GMA. The composites were prepared in accordance with the manufacturers' instructions by mixing equal amounts of the two components at room temperature. The degree of conversion of double bonds of resin matrix during curing was determined in thin film of composites using FTIR transmission spectroscopy. The degree of conversion of Concise and Aflacomp was found to be correspondingly (73.63 ± 4.33)% and (47.75 ± 1.80)% after a day‐polymerization. Sorption, solubility, and volumetric change were determined after storage of composites in water or ethanol/water solution 75 vol % at 37°C (a good food/oral simulating fluid) for 30 days. These properties for both composites were higher in ethanol/water solution than in water. Also these properties for Concise were lower than those for Alfacomp in both liquids. Thermal stability of composites was studied by thermogravimetric analysis which performed in air atmosphere from 50 to 800°C. This analysis of composites showed the degradation of their resin matrix in three steps. The organic resin content of Concise was found to be 18.0% w/w and that of Alfacomp 20.1% w/w. Concise showed generally better properties than Alfacomp. Sea buckthorn is a promising source of bioactive compounds. However, there is limited information on the effect of post harvest drying and extraction of sea buckthorn on its antioxidant capacity. The effect of freeze, air, and solar dryingon the extraction yield of sea buckthorn berries and leaves was evaluated with respect to their bioactive content. Sequential extraction with solvents of different polarity and solvent-free microwave-assisted extraction were applied. Freeze-drying has better performance in the berries’ extracts, while air-drying has better performance in the leaves’ extracts. All data were analyzed by Multi Factor Analysis of Variance (ANOVA) and Duncan's post-hoc tests at a level of α = .05. Although the extraction yield in the sequential extraction of sea buckthorn berries is significantly better, the leaves’ extracts exhibit superior radical scavenging ability. Accelerated solar drying appears to preserve sea buckthorn leaves without degrading their antioxidant content. The components responsible for the high antioxidant activity of leaves’ extracts were found to be several flavonoids and polyphenols. The extract obtained by the microwave extraction of fresh berries exhibits moderate antioxidant activity compared to the polar extracts of freeze-dried berries. 相似文献

Although Governance, Risk and Compliance (GRC) is an emerging field of study within the information systems (IS) academic community, the concept behind the acronym has to still be demystified and further investigated. The study investigates GRC systems in depth by (a) reviewing the literature on existing GRC studies, and (b) presenting a field study on views about GRC application by professional experts. The aim of this exploratory study is to understand the aspects and the nature of the GRC system following an enterprise systems approach. The result of this study is a framework of particular GRC characteristics that need to be taken into consideration when these systems are put in place. This framework includes specific areas such as: goals and objectives, purpose of the system, key stakeholders, methodology and requirements prior to implementation, critical success factors and problems/barriers. Further discussion about the issues, the concerns and the diverse views on GRC would assist in developing an agenda for the future research on the GRC field. 相似文献

This paper constitutes a literature review on student modeling for the last decade. The review aims at answering three basic questions on student modeling: what to model, how and why. The prevailing student modeling approaches that have been used in the past 10 years are described, the aspects of students’ characteristics that were taken into consideration are presented and how a student model can be used in order to provide adaptivity and personalisation in computer-based educational software is highlighted. This paper aims to provide important information to researchers, educators and software developers of computer-based educational software ranging from e-learning and mobile learning systems to educational games including stand alone educational applications and intelligent tutoring systems. In addition, this paper can be used as a guide for making decisions about the techniques that should be adopted when designing a student model for an adaptive tutoring system. One significant conclusion is that the most preferred technique for representing the student’s mastery of knowledge is the overlay approach. Also, stereotyping seems to be ideal for modeling students’ learning styles and preferences. Furthermore, affective student modeling has had a rapid growth over the past years, while it has been noticed an increase in the adoption of fuzzy techniques and Bayesian networks in order to deal the uncertainty of student modeling. 相似文献

In the present study, complexes of vanillin with β-cyclodextrin (β-CD) were prepared by the freeze-drying method. The formation of the inclusion complex was confirmed by differential scanning calorimetry (DSC). DSC thermograms indicated that the endothermic peak of vanillin and the physical mixture of vanillin with β-CD, due to the melting of vanillin crystals, were absent in DSC thermograms obtained for the freeze-dried inclusion complex. Moreover, the DSC studies under oxidation conditions indicate that the complex of vanillin with β-CD is protected towards oxidation since it remains intact at temperatures where the free vanillin is oxidising. The structure of the complex in aqueous solutions was established by nuclear magnetic resonance (NMR) studies and specifically by two-dimensional rotational frame NOE spectra. NMR studies showed inclusion of the entire vanillin molecule in the β-CD in a tilted manner and with the aldehyde group in the primary side. The stoichiometry of the complex was 1:1. A phase solubility study was performed by mixing an excess amount of vanillin with aqueous solutions containing increasing amounts of β-CD. The results indicated that the complex of a vanillin/β-CD inclusion is more soluble in water than vanillin alone. 相似文献

The objectives of the present study were (a) to produce gluten-free bread, fortified with iron (GFB-Fe), using selected iron compounds (ferric pyrophosphate, ferric pyrophosphate with emulsifiers, NaFeEDTA, electrolytic iron, ferrous gluconate, ferrous lactate and ferrous sulphate) (b) to test sensory characteristics of the GFB-Fe (feel-mouth texture, crumb colour, aroma and taste) (c) to compare iron dialyzability of various iron compounds in GFB-Fe. The most acceptable products were those fortified with ferric pyrophosphate with emulsifiers and ferric pyrophosphate. Ferrous dialyzable iron (ferrous iron with molecular weight lower than 8000 Da, an index for prediction of iron bioavailability) was measured under simulated gastrointestinal conditions. Ferrous dialyzable iron in GFB-Fe fortified with ferric pyrophosphate with emulsifiers, NaFeEDTA, ferrous bis-glycinate, ferrous gluconate or ferrous sulphate was higher than that in GFB-Fe fortified with electrolytic iron, ferrous lactate or ferric pyrophosphate (P < 0.05). These results are promising for the development of GFB-Fe products in the future. 相似文献
