Cladribine (CLD) treats multiple sclerosis (MS) by selectively and transiently depleting B and T cells with a secondary long-term reconstruction of the immune system. This study provides evidence of CLD’s immunomodulatory role in peripheral blood mononuclear cells (PBMCs) harvested from 40 patients with untreated relapsing-remitting MS (RRMS) exposed to CLD. We quantified cytokine secretion from PBMCs isolated by density gradient centrifugation with Ficoll–Paque using xMAP technology on a FlexMap 3D analyzer with a highly sensitive multiplex immunoassay kit. The PBMC secretory profile was evaluated with and without CLD exposure. PBMCs isolated from patients with RRMS for ≤12 months had significantly higher IL-4 but significantly lower IFN-γ and TNF-α secretion after CLD exposure. PBMCs isolated from patients with RRMS for >12 months had altered inflammatory ratios toward an anti-inflammatory profile and increased IL-4 but decreased TNF-α secretion after CLD exposure. CLD induced nonsignificant changes in IL-17 secretion in both RRMS groups. Our findings reaffirm CLD’s immunomodulatory effect that induces an anti-inflammatory phenotype.  相似文献

Extracellular biological synthesis of gold nanoparticles was achieved by a simple biological procedure using coriander extract as the reducing agent. The aqueous gold ions when exposed to coriander leaf extract are reduced and resulted in the biosynthesis of gold nanoparticles in the size range from 6.75-57.91 nm. The gold nanoparticles were characterized by UV-Vis spectroscopy, X-ray diffraction (XRD), energy dispersive X-ray analysis (EDAX), fourier transform infra-red spectroscopy (FT-IR) and transmission electron microscopy (TEM). This eco-friendly approach for the synthesis of nanoparticles is simple, amenable for large scale commercial production and technical applications.  相似文献

This paper describes an experimental investigation carried out involving instrumentation, measurement and monitoring of stresses in a large diameter gas pipeline during its lowering operations. It also highlights the ingenious techniques developed and adopted to overcome the difficulties pertaining to long leadwires, measurement of existing strains on the curved surface of a pipeline and protection of strain gauge installations. These novel methods were used to ensure smooth and proper functioning of strain gauge instrumentation even under the severe field conditions at site for more than two weeks, till the lowering operations and pipeline monitoring was successfully completed.  相似文献

Subcontracting is noted for its adverse health and safety (H&S) influence in construction. Using interviews with five of the top 20 UK contractors and one medium-sized contractor, this study explored how main contractors manage the H&S influence of subcontracting with a focus on their in-house measures. Some in-house measures found are: restricting the layers of subcontractors on projects; working with a regular chain of subcontractors; implementing a H&S reward scheme for subcontractors; and insisting on non-working subcontractor foremen who have direct responsibility for the safety of workers in their trade. These measures appear to be influenced by clients, industry peer groups, and the moral justification for occupational H&S, and they offer inter-organisational learning opportunities for contractors in devising measures to mitigate the H&S influence of subcontracting. These findings should also allay concerns that removing some of the legislative hurdles in connection with on-going red tape debate will result in poorer H&S.  相似文献

The formation of phosphate coatings by cathodic electrochemical treatment using graphite and steel anodes and evaluation of their corrosion resistance is addressed in this paper. The type of anode used, graphite/steel, has an obvious influence on the composition of the coating, resulting in zinc–zinc phosphate composite coating with graphite anode and zinc–iron alloy–zinc phosphate–zinc–iron phosphate composite coating with steel anode. The corrosion resistance of the coating is found to be a function of the composition of the coating. The deposition of zinc/zinc–iron alloy along with the zinc phosphate/zinc and zinc–iron phosphate using graphite/steel anodes has caused a cathodic shift in the Ecorr compared to uncoated mild steel substrates. The icorr values of these coatings is very high. EIS studies reveal that zinc/zinc–iron alloy dissolution is the predominant reaction during the initial stages of immersion. Subsequently, the formation of zinc and iron corrosion products imparts resistance to the charge transfer process and increases the corrosion resistance with increase in immersion time. The corrosion products formed might consist of oxides and hydroxychlorides of zinc and iron. The study suggests that cathodic electrochemical treatment could be effectively utilized to impart the desirable characteristics of the coating by choosing appropriate anode materials, bath composition and operating conditions.  相似文献

Measurements of the growth rate of roughness on a cathode surface can be used to study the physics of cathodic processes. But these growth rates are cell wide variables and they ordinarily depend on what is taking place at the anode as well as at the cathode. Our aim is to derive a simple condition under which the anode makes no contribution to the interpretation of growth rate measurements at the cathode.Our condition stems from the contribution of ion diffusion to the growth rate and it is satisfied for all non-small values of the wave number of a growing disturbance. This, together with the fact that surface tension is the only stabilizing factor and the fact that the surface tension coefficient multiplying the wave number is small, means that the anode ought not to be important for the fastest growing wave numbers.Once the effect of the anode is eliminated, the growth rate at the cathode can be expressed in a formula so simple that pencil and paper calculations are possible and this makes important questions in cell design easy to answer, e.g., is the growth rate diffusion controlled?  相似文献

The study reported in this article deals with understanding the physical, chemical and microstructural differences in sulfate attack from seawater and groundwater. Portland cement mortars were completely immersed in solutions of seawater and groundwater. Physical properties such as length, mass, and compressive strength were monitored periodically. Thermal analysis was used to study the relative amounts of phases such as ettringite, gypsum, and calcium hydroxide, and microstructural studies were conducted by scanning electron microscopy. Portland cement mortars performed better in seawater solution compared to groundwater solution. The difference in performance could be attributed to the reduction in the quantity of the expansive attack products (gypsum and ettringite). The high Cl concentration of seawater could have played an important role by binding the C3A to form chloroaluminate compounds, such as Friedel's salt (detected in the microstructural studies), and also by lowering the expansive potential of ettringite. Furthermore, the thicker layer of brucite forming on the specimens in seawater could have afforded better protection against ingress of the solution than in groundwater.  相似文献

Poly(ethylene‐co‐vinyl alcohol)/poly(vinyl pyrrolidone) (EVAL/PVP) blend membranes with antifouling properties were prepared by nonsolvent induced phase separation. Residual PVP in the sample was calculated by infrared spectroscopic data and confirmed by thermogravimetric analysis. The effect of residual PVP on hydrophilicity and permeation characteristics of the membranes was evaluated. Porosity and equilibrium water content of the membranes were influenced by the addition of PVP. The effect of protein fouling on flux using bovine serum albumin as a model system was studied in detail. The residual PVP content could enhance the antifouling property of the membrane. All membranes proved to have sufficient mechanical strength to withstand pressure‐driven filtrations.  相似文献

The susceptibility of tin-plated contacts to fretting corrosion is a major limitation for its use in electrical connectors. The present paper evaluates the influence of a variety of factors, such as, fretting amplitude (track length), frequency, temperature, humidity, normal load and current load on the fretting corrosion behaviour of tin-plated contacts. This paper also addresses the development of fretting corrosion maps and lubrication as a preventive strategy to increase the life-time of tin-plated contacts. The fretting corrosion tests were carried out using a fretting apparatus in which a hemispherical rider and flat contacts (tin-plated copper alloy) were mated in sphere plane geometry and subjected to fretting under gross-slip conditions. The variation in contact resistance as a function of fretting cycles and the time to reach a threshold value (100 mΩ) of contact resistance enables a better understanding of the influence of various factors on the fretting corrosion behaviour of tin-plated contacts. Based on the change in surface profile and nature of changes in the contact zone assessed by laser scanning microscope (LSM) and surface analytical techniques, the mechanism of fretting corrosion of tin-plated contacts and fretting corrosion maps are proposed. Lubrication increases the life-time of tin-plated contacts by several folds and proved to be a useful preventive strategy.  相似文献

This paper presents an attempt to model the abrasive waterjet (AWJ) turning process considering material removal from the circumference of a rotating cylindrical specimen. The methodology involves the use of Finnie's theory of erosion to estimate the volume of material removed by the impacting abrasive particles. The proposed model considers the impact of jet at an angle to the workpiece surface to account for the curvature of the workpiece. Unlike earlier works, this model considers the continuous change in local impact angle caused by the change in workpiece diameter. The flow stress of the workpiece material is determined using a novel experiment involving the same abrasive and workpiece materials. The adequacy of the proposed model is examined through AWJ turning tests under various process parameter combinations. The final diameters predicted by the model are found to be in good agreement with the experimental results.  相似文献
