Decentralized Control System Design under Uncertainty Using Mixed‐Integer Optimization

Decentralized control system design comprises the selection of a suitable control structure and controller parameters. In this contribution, the optimal control structure and the optimal controller parameters are determined simultaneously using mixed‐integer dynamic optimization (MIDO) under uncertainty, to account for nonlinear process dynamics and various disturbance scenarios. Application of the sigma point method is proposed in order to approximate the expectation and the variance of a chosen performance index with a minimum number of points to solve the MIDO problem under uncertainty. The proposed methodology is demonstrated with a benchmark problem of an inferential control for a reactive distillation column. The results are compared with established heuristic design methods and with previous deterministic approaches.     

Lactic acid fermentation of anthocyanin-rich sweet potato (Ipomoea batatas L.) into lacto-juice

Sweet potato (SP) is an important root crop grown all over the world and consumed as a vegetable, boiled, baked or often fermented into food and beverages. The grated SP roots [non- boiled and fully boiled (boiled in water at 100 °C for 15 min) were treated with 0.05% of commercial pectinase enzyme (Pectinex, Novoenzyme) in order to extract the juice. The fresh juice was inoculated with Lactobacillus plantarum MTCC 1407 culture at 28 ± 2 °C for 48 h to produce lacto-juice (LJ). The anova analysis of analytical data revealed that there was significant effect of boiling conditions (fully boiled and non-boiled) on pH [ F (1, 4) = 220.5, P  < 0.001), TA [ F (1, 4) = 78.89, P  < 0.01], starch [ F (1, 4) = 26.63, P  < 0.01), total sugar [ F (1, 4) = 61.36, P  < 0.01) and anthocyanin [ F (1, 4) = 32.86, P  < 0.01) but not on reducing sugar [ F (1, 4) = 2.48, P  = 0.19). Sensory evaluation rated the SP LJ acceptable based on texture, taste, aroma, flavour and after taste. LJ prepared from fully boiled roots with 10% cane sugar was most preferred by a consumer's panelist based on Linear Discriminant Analysis. Principal component analyses (PCA) reduced the seven original analytical variables to three independent components (factors), which accounted for 99.9% of the total variations. Similarly, six original sensory variables were reduced to two independent components, which accounted for 65.7% of the total variations.     

Molecular characterization of Mycobacterium avium complex isolates giving discordant results in AccuProbe tests by PCR-restriction enzyme analysis, 16S rRNA gene sequencing, and DT1-DT6 PCR

Based on cultural and biochemical tests, a total of 84 strains (72 clinical and 12 environmental isolates from the Caribbean Isles, Europe, and the Indian subcontinent) were identified as members of the Mycobacterium avium complex (MAC). They were further characterized with MAC, M. avium, and M. intracellulare probes of the AccuProbe system, and this was followed by selective amplification of DT6 and DT1 sequences. Seventy isolates gave concordant results; 63 were identified as M. avium, 5 were identified as M. intracellulare, and 24 remained untypeable by both methods. Fourteen isolates gave discrepant results, as they were DT1 positive but gave negative results by the M. intracellulare AccuProbe test. Consequently, a detailed molecular analysis of all DT1-positive isolates (14 discrepant strains plus 5 M. intracellulare strains) was performed by PCR-restriction analysis (PRA) of the hsp65 gene and 16S rRNA gene sequencing. The results confirmed the reported heterogeneity of M. intracellulare, as only 6 of 19 isolates (32%) gave PRA results compatible with published M. intracellulare profiles while the rest of the isolates were grouped in four previously unpublished profiles. 16S rRNA gene sequencing showed that only 8 of 19 isolates (42%) were related to M. intracellulare IWGMT 90247 (EMBL accession no. X88917), the rest being related to MCRO19 (EMBL accession no. X93030) and MIWGTMR10 (EMBL accession no. X88915). In conclusion, we have characterized a significant number of MAC isolates which were not identified by the AccuProbe test, PRA, or 16S rRNA sequencing. However, all of them were identifiable by DT1-DT6 PCR (they were DT6 negative and DT1 positive) and could be tentatively identified as M. intracellulare based on previously published observations. It is noteworthy that the majority of such isolates (14 of 19) were from the Indian subcontinent, with 12 of 14 being environmental isolates. Our study confirms the marked heterogeneity of M. intracellulare isolates and shows the utility of in-house DT1 PCR to detect this group of isolates, which would otherwise have been missed by the AccuProbe system in a routine clinical microbiology laboratory.

Tropical root and tuber crops [cassava, sweet potato, yams, colocasia (taro), etc] are important staples for food security for about a fifth of the world population. Bulk of cassava in Africa and Latin America are processed into fermented foods and food additives such as organic (acetic, citric and lactic) acids, mono-sodium glutamate, etc. The fermented foods from cassava are gari , fufu , lafun , chickwanghe , agbelima , attieke and kivunde in Africa, tape in Asia and ' cheese ' bread , and ' coated peanut ' in Latin America. Lactic acid bacteria and yeasts are the major group of micro-organisms associated with cassava fermentation. Similarly, sweet potatoes can be fermented into soy sauce, vinegar, lacto-juices, lacto-pickles and sochu (an alcoholic drink produced in Japan), and yams into fermented flour. Most of these fermented food products are functional foods rich in phytochemicals, dietary fibres, anti-oxidant compounds (β-carotene, anthocyanin, etc) and probiotic components (lactic acid bacteria and yeasts).

Bactericidal action of ofloxacin, sulbactam-ampicillin, rifampin, and isoniazid on logarithmic- and stationary-phase cultures of Mycobacterium tuberculosis

The bactericidal actions of ofloxacin and sulbactam-ampicillin, alone and in combination with rifampin and isoniazid, on exponential-phase and stationary-phase cultures of a drug-susceptible isolate of Mycobacterium tuberculosis were studied in vitro. In exponential-phase cultures, all drugs were bactericidal, with the higher concentrations of ofloxacin (5 micrograms/ml) and sulbactam-ampicillin (15 micrograms of ampicillin per ml) being as bactericidal as 1 microgram of isoniazid per ml or 1 microgram of rifampin per ml. In two-drug combinations, both drugs increased the levels of activity of isoniazid and rifampin and were almost as bactericidal as isoniazid-rifampin; they also appeared to increase the level of activity of isoniazid-rifampin in three-drug combinations. In contrast, ofloxacin and sulbactam-ampicillin had little bactericidal activity against stationary-phase cultures and were less active than isoniazid or rifampin alone. Furthermore, in two-drug or three-drug combinations, they did not increase the level of activity of isoniazid, rifampin, or isoniazid-rifampin. These findings suggest that ofloxacin and sulbactam-ampicillin are likely to be most useful in the early stages of treatment and in preventing the emergence of resistance to other drugs but are unlikely to be effective as sterilizing drugs helping to kill persisting lesional bacilli.     

Solar photocatalytic detoxification of cyanide by different forms of TiO<Subscript>2</Subscript>

The photocatalytic efficiencies of TiO₂ nanocrystals of different modifications (anatase, rutile, P25 Degussa, Hombikat), to oxidize cyanide ion and subsequently the cyanate also, under natural sunlight at 950±25W m⁻² in alkaline solution have been compared. The oxides have been characterized by powder XRD, UV-visible diffuse reflectance and impedance spectroscopies. Under identical solar irradiance, the reaction follows Langmuir-Hinshelwood kinetics on cyanide, and depends on the apparent area of the catalyst bed and dissolved oxygen. However, the adsorption of cyanide on TiO₂ in dark is too small to be measured analytically. The photocatalytic activity of TiO₂ is not solely governed by the band gap or charge-transfer resistance or capacitance or phase composition but is in accordance with the specific surface area or the average crystallite size; rutile is an exception.

Inferential Control of Reactive Destillation Columns – An Algorithmic Approach

Here, mixed integer optimization is used to determine the optimal control structure and the optimal controller parameters simultaneously. The process dynamics is included explicitly into the constraints using a rigorous nonlinear dynamic process model. Depending on the objective function, which is used for the evaluation of competing control systems, two different formulations are proposed which lead to mixed‐integer dynamic optimization (MIDO) problems. A MIDO solution strategy based on the sequential approach is adopted in the present paper. Here, the MIDO problem is decomposed into a series of nonlinear programming (NLP) subproblems (dynamic optimization) where the binary variables are fixed, and mixed‐integer linear programming (MILP) master problems which determine a new binary configuration for the next NLP subproblem. The proposed methodology is applied to inferential control of reactive distillation columns as a challenging benchmark problem for chemical process control.
