Gut microbiota alterations are intimately linked to chronic constipation upon aging. We investigated the role of targeted changes in the gut microbiota composition in the relief of constipation symptoms after rhubarb extract (RE) supplementation in middle-aged volunteers. Subjects (95% women, average 58 years old) were randomized to three groups treated with RE at two different doses determined by its content of rhein (supplementation of 12.5 mg and 25 mg per day) vs. placebo (maltodextrin) for 30 days. We demonstrated that daily oral supplementation of RE for 30 days was safe even at the higher dose. Stool frequency and consistency, and perceived change in transit problem, transit speed and difficulty in evacuating, investigated by validated questionnaires, were improved in both groups of RE-treated volunteers compared to placebo. Higher abundance of Lachnospiraceae (mainly Roseburia and Agathobacter) only occurred after RE treatment when present at low levels at baseline, whereas an opposite shift in short-chain fatty acid (SCFA) levels was observed in both RE-treated groups (increase) and placebo (decrease). Fecal Lachnospiraceae and SCFA were positively correlated with stool consistency. This study demonstrates that RE supplementation promotes butyrate-producing bacteria and SCFA, an effect that could contribute to relieving chronic constipation in middle-aged persons. 相似文献

Prostate specific membrane antigen targeted radionuclide therapy (PSMA-TRT) is a promising novel treatment for prostate cancer (PCa) patients. However, PSMA-TRT cannot be used for curative intent yet, thus additional research on how to improve the therapeutic efficacy is warranted. A potential way of achieving this, is combining TRT with poly ADP-ribosylation inhibitors (PARPi), which has shown promising results for TRT of neuroendocrine tumor cells. Currently, several clinical trials have been initiated for this combination for PCa, however so far, no evidence of synergism is available for PCa. Therefore, we evaluated the combination of PSMA-TRT with three classes of PARPi in preclinical PCa models. In vitro viability and survival assays were performed using PSMA-expressing PCa cell lines PC3-PIP and LNCaP to assess the effect of increasing concentrations of PARPi veliparib, olaparib or talazoparib in combination with PSMA-TRT compared to single PARPi treatment. Next, DNA damage analyses were performed by quantifying the number of DNA breaks by immunofluorescent stainings. Lastly, the potential of the combination treatments was studied in vivo in mice bearing PC3-PIP xenografts. Our results show that combining PSMA-TRT with PARPi did not synergistically affect the in vitro clonogenic survival or cell viability. DNA-damage analysis revealed only a significant increase in DNA breaks when combining PSMA-TRT with veliparib and not in the other combination treatments. Moreover, PSMA-TRT with PARPi treatment did not improve tumor control compared to PSMA-TRT monotherapy. Overall, the data presented do not support the assumption that combining PSMA-TRT with PARPi leads to a synergistic antitumor effect in PCa. These results underline that extensive preclinical research using various PCa models is imperative to validate the applicability of the combination strategy for PCa, as it is for other cancer types. 相似文献

Microbial populations associated to poplar are well described in non-contaminated and metal-contaminated environments but more poorly in the context of polycyclic aromatic hydrocarbon (PAH) contamination. This study aimed to understand how a gradient of phenanthrene (PHE) contamination affects poplar growth and the fungal microbiome in both soil and plant endosphere (roots, stems and leaves). Plant growth and fitness parameters indicated that the growth of Populus canadensis was impaired when PHE concentration increased above 400 mg kg−1. Values of alpha-diversity indicators of fungal diversity and richness were not affected by the PHE gradient. The PHE contamination had a stronger impact on the fungal community composition in the soil and root compartments compared to that of the aboveground organs. Most of the indicator species whose relative abundance was correlated with PHE contamination decreased along the gradient indicating a toxic effect of PHE on these fungal OTUs (Operational Taxonomic Units). However, the relative abundance of some OTUs such as Cadophora, Alternaria and Aspergillus, potentially linked to PHE degradation or being plant-beneficial taxa, increased along the gradient. Finally, this study allowed a deeper understanding of the dual response of plant and fungal communities in the case of a soil PAH contamination gradient leading to new perspectives on fungal assisted phytoremediation. 相似文献

Seed size is often considered to be an important trait for seed quality, i.e., vigour and germination performance. It is believed that seed size reflects the quantity of reserve material and thus the C and N sources available for post-germinative processes. However, mechanisms linking seed size and quality are poorly documented. In particular, specific metabolic changes when seed size varies are not well-known. To gain insight into this aspect, we examined seed size and composition across different accessions of barrel medic (Medicago truncatula Gaertn.) from the genetic core collection. We conducted multi-elemental analyses and isotope measurements, as well as exact mass GC–MS metabolomics. There was a systematic increase in N content (+0.17% N mg−1) and a decrease in H content (–0.14% H mg−1) with seed size, reflecting lower lipid and higher S-poor protein quantity. There was also a decrease in 2H natural abundance (δ2H), due to the lower prevalence of 2H-enriched lipid hydrogen atoms that underwent isotopic exchange with water during seed development. Metabolomics showed that seed size correlates with free amino acid and hexoses content, and anticorrelates with amino acid degradation products, disaccharides, malic acid and free fatty acids. All accessions followed the same trend, with insignificant differences in metabolic properties between them. Our results show that there is no general, proportional increase in metabolite pools with seed size. Seed size appears to be determined by metabolic balance (between sugar and amino acid degradation vs. utilisation for storage), which is in turn likely determined by phloem source metabolite delivery during seed development. 相似文献

Docosahexaenoic acid (DHA; 22∶6n−3) is found in extremely high levels in human ejaculate with the majority occurring in the spermatozoa. However, the relative concentration of DHA and other fatty acids, in blood serum, seminal plasma, and spermatozoa of asthenozoospermic vs. normozoospermic individuals is not known. We analyzed the phospholipid fatty acid composition of blood serum, seminal plasma, and spermatozoa of normozoospermic men and asthenozoospermic men in order to determine if DHA levels, as well as the levels of other fatty acids, differed. The serum phospholipid DHA levels were similar in the two groups, suggesting similar intakes of dietary DHA. On the other hand, seminal plasma levels of DHA (3.0 vs. 3.7%) and total polyunsaturated fatty acids (PUFA) (11.8 vs. 13.5%) were significantly lower in asthenozoospermic vs. normozoospermic men, respectively, while 18∶1 (19.0 vs. 16.8%) and monounsaturated fatty acids (MUFA) (24.2 vs. 21.7%) were significantly higher in the asthenozoospermic vs. the normozoospermic men. Spermatozoa from asthenozoospermic men had higher levels of 18∶1, 20∶0, 22∶0, 22∶1, and 24∶0 than sperm from normozoospermic men, and lower levels of 18∶0 and DHA (8.2 vs. 13.8%). Furthermore, total MUFA (19.3 vs. 16.5%) was higher and total PUFA (19.0 vs. 24.0%), n−3 fatty acids (9.3 vs. 14.6%), and the ratio of n−3 to n−6 fatty acids (1.0 vs. 1.6) were lower in the asthenozoospermic men. Therefore, in asthenozoospermic individuals, lower levels of DHA in the seminal plasma, but not in the blood serum, mimic the decreased concentrations of DHA in the spermatozoa. This suggests that the lower concentrations of spermatozoon DHA in these individuals are due not to dietary differences but to some type of metabolic difference in the asthenozoopermic men. 相似文献

Using an environmental transmission electron microscope (ETEM), we show that a significant amount of water, far exceeding the multilayers caused by surface adsorption, is reversibly associated prior to deliquescence with substrate-supported NaCl particles (dry diameters of ~ 40 nm to 1.5 μ m; ~ 18°C). We hypothesize that the water is present as an aqueous solution containing dissolved Na and Cl ions. Water uptake occurs at relative humidities (RH) as low as 70%, and the resulting liquid layer coating the particles is stable over extended times if the RH is held constant. We exposed CaSO 4 and CaSO 4 · 2H 2 O particles to elevated RH values in the ETEM to show that chemically nonspecific condensation of gas-phase water on the TEM substrate does not explain our observations. Furthermore, damage to the NaCl surface induced by the electron beam and small fluctuations in RH do not seem to contribute to or otherwise affect water uptake. We have similar observations of water association for other alkali halide particles, including NaBr and CsCl, prior to deliquescence. To explain the observations, we derive the phase rule for this geometry and show that it allows for the coexistence of liquid, solid, and vapor for the binary NaCl/H 2 O system across a range of RH values. The derivation includes the effects of heterogeneous pressure because of the Laplace-Young relations for the subsystems. Furthermore, in view of the lever rule and the absence of similar observations for free-floating pure NaCl aerosol particles, we hypothesize that the surface energy necessary to support these effects is provided by sample-substrate interactions. Thus, the results of this study may be relevant to atmospheric systems in which soluble compounds are associated with insoluble materials. 相似文献

The potential use of plasma polymer coatings as substrates for serum-free expansion of limbal epithelial cells was investigated.
Preliminary studies using a human corneal epithelial cell line showed that acrylic acid-coated surfaces performed better than
allyl amine and allyl alcohol coated surfaces in terms of cell metabolic activity and confluence as assessed using the MTT
assay. Subsequently, the proliferation and maturity of primary human limbal epithelial cells in co-culture with growth arrested
3T3 fibroblasts on a range of acrylic acid plasma coated surfaces, octadiene plasma coated surfaces and tissue culture plastic
was investigated using MTT and cytokeratin 3 immunostaining. The cells performed better in the presence of serum on all surfaces.
However, the acrylic acid coated surfaces successfully sustained a serum-free fibroblast/epithelial cell co-culture. The metabolic
activity of the epithelial cells was superior on the acrylic acid coated surfaces than on tissue culture plastic in serum-free
conditions and their levels of differentiation were not significantly higher than in the presence of serum. These results
suggest that these surfaces can be used successfully for the serum-free expansion of human limbal epithelial cells. 相似文献

Bioanalytical automation expanded at Glaxo Inc. from 1987 to
1991 by cycling through periods of justification, planning,
implementation, obstacle-jumping and success, which justified
continued cycling. In 1990 it became evident that the technology and
its growth needed to be planned and the resources had to be
managed. A Strategic Plan was researched and prepared. The plan
describes the mission, values, goals and structure of the Bioanalytical
Automation Group and the most important requirements for
achieving those planned goals, including: (1) Long-term management
commitment; (2) Trained, dedicated personnel; (3) Quality
facilities; (4) Teamwork; and (5) Investment in automationcompatible
equipment. The strategic plan has been in effect for over
