An extruding wire knife was used to give adult male CFHB rats a minimally traumatic unilateral mechanical lesion of the medial forebrain bundle. In addition, some rats received bilateral intrastriatal injections of one of three fluorescent retrograde tracers either eight days before or eight days after the lesion. Injections made after the lesion revealed that about half of the animals had complete lesions of the nigrostriatal tract, while the other half were incompletely lesioned, the mean proportion of non-axotomized neurons being 23%. Over the 10 weeks following the lesions, the number of tyrosine hydroxylase-immunoreactive cells in the lesioned substantia nigra fell linearly, reaching a mean of 29% of that of the control substantia nigra. In the animals which were completely lesioned, neuronal survival at 10 weeks varied between 6 and 12%. That the disappearance of tyrosine hydroxylase-immunoreactive neurons was due to cell death rather than the loss of tyrosine hydroxylase itself was confirmed by labelling the cells with Fluoro Gold before axotomy; the tracer was seen in survival neurons, microglia and in a few involuted neurons which continued to be tyrosine hydroxylase-immunoreactive. This percentage of neurons surviving axotomy corresponds to the proportion of substantia nigra neurons which project to the contralateral striatum, and these neurons were in the region of the substantia nigra from which the contralateral projection originated. It is concluded that following mechanical transection of the nigrostriatal tract, all truly axotomized substantia nigra neurons die over a period of about 10 weeks.  相似文献

Abstract

We have studied the decoherence of motional-state superpositions of single trapped ions caused by their coupling to fluctuating fields. We illustrate these studies by showing how Schrödinger-cat state superpositions decohere in the presence of purposely-applied uniform stochastic electric fields and we compare these results to the decoherence caused by ambient fields in the experiments.  相似文献

This paper presents a dynamic load flow technique for power system simulators. The approach presented can be applied to a system when there is a net accelerating or decelerating power in the system. It possesses all the attractive features and basic structure of the normal fast decoupled method, and it does not depend on the selected reference bus. The modifications to the standard fast decoupled power flow algorithm are derived, and power system simulator implementation considerations are discussed. The algorithm has been tested on standard IEEE test cases and on a metropolitan utility 559 bus model. A numerical example of the above algorithm on the 559 test case is presented which demonstrates convergence characteristics and computation requirements of the proposed algorithm. These results are compared with those of normal fast decoupled load flow.  相似文献

The symptoms of poor product data management (PDM), data visibility, accessibility and reliability, are evident in many engineering environments. Data discipline has been lost in the proliferation of computer-based tools in a fragmented product development lifecycle. In this paper the authors try to isolate the golden rules of PDM. Process management and product structure management procedures have been identified. By embodying these, one can bring discipline to the management of product data. In application, implementation of `success' is limited by the ability of the technology. A balance is necessary between enablers and business requirements that may demand functionality beyond the cost effective operational parameters of the technology. Technology solutions must be driven by business needs, where the functionality and features of any particular solution are in line with the objectives of the implementation  相似文献

Low concentrations of guanidine hydrochloride (GuHCl) increase the rate (and to a lesser degree, the extent) of tubulin polymerization as assessed by light scattering. Maximum enhancement occurs at 120-160 mM GuHCl followed by decreases at higher GuHCl. The latent period is decreased, and there is a 3-4 fold reduction in the critical concentration of polymerization. Electronmicrographs reveal microtubules in the controls and an increasing fraction of total polymers present as aberrant microtubules as the GuHCl concentration is increased from 20 to 100 mM. The GuHCl effect is markedly reduced, but not abolished, in tubulin S (in which the anionic C termini of both monomers have been removed). The GuHCl-induced polymerization has an absolute requirement for GTP and taxol or DMSO, is very sensitive to podophyllotoxin inhibition, and can overcome urea-mediated inhibition of polymerization. Guanidinium analogues mimic the GuHCl effect roughly as a function of the number of potential hydrogen bonds. The anions of the guanidine salts superimpose their inhibitory action on the guanidinium cation effect according to the lyotropic series. At higher GuHCl concentrations (peak effect 500-700 mM), a different polymer (type II) is formed that is GTP and taxol independent, but whose polymerization is retarded but not prevented by podophyllotoxin. Its structure resembles the fibrillar network seen in unfolding intermediates of other proteins. We conclude that both charge and hydrogen-bonding ability are major contributors to the GuHCl-induced promotion of tubulin polymerization, and that charge-shielding is likely to be the basis for this effect.  相似文献
