Characterization of allergoids from ovalbumin in vitro and in vivo

Several in vivo and in vitro methods for monitoring immunological properties of two allergoids obtained by formaldehyde treatment of ovalbumin (OA) were developed. The calculated molecular weight of allergoids was 80 kD (OA-F1) and 165 kD (OA-F2), respectively. The allergenic activity in vitro of allergoids in mast-cell histamine release assay was 1000 times lower than of OA. Both allergoids showed reduced ability to induce passive cutaneous anaphylaxis in the Sprague-Dawley rats or systemic anaphylaxis in Dunkin-Harley guinea-pigs. The ability of OA and allergoids to bind to the OA-specific IgE antibodies was measured in vivo by the inhibition of passive cutaneous anaphylaxis (PCA-inhibition). Allergoid binding to IgE was 51-66% lower than the native allergen. Moreover, the avidity of OA-specific IgG antibodies, measured by ELISA-inhibition, for allergoids and allergen was of the same order. Allergoids induced a different pattern of humoral immune response from that, induced by the native allergen. Thus, after immunization of BALB/c mouse, both allergoids induced a higher production of IgG and a lower production of IgE than OA, only OA-F2 induced a lower production of IgG1. The differences in the IgA response to the immunogens was not significant. Delayed hypersensitivity studies in the BALB/c mouse showed that allergoids were 5- to 12-times less effective in inducing a cell-mediated immune response than OA. The present study provides a battery of immunological methods for preclinical testing of modified allergens.

Enantiospecific synthesis of (R)-1,7-dioxaspiro [5.5]undecane [major component of olive fruit fly (Dacus Oleae) sex pheromone] from d-Fructose

The synthesis of the title compound13 has been carried out through the preparation of its precursor, (3R,4R,5S,6R)-3,4,5-trihydroxy-1,7-dioxaspiro[5.5]undecane (6), obtained fromd-fructose using Wittig's methodology, reduction, and spiroketalation. Compound6 was transformed into13 by a Barton deoxygenation at C-5 followed by a Corey dideoxygenation at C-3,4 of the appropriately protected derivatives.Enantiospecific synthesis of spiroacetals. Using dynamic demand information and zoning for the storage of non-uniform density stock keeping units

The warehouse order-picking operation is one of the most labour-intense activities that has an important impact on responsiveness and efficiency of the supply chain. An understanding of the impact of the simultaneous effects of customer demand patterns and order clustering, considering physical restrictions in product storage, is critical for improving operational performance. Storage restrictions may include storing non-uniform density stock keeping units (SKUs) whose dimensions and weight constrain the order-picking operation given that a priority must be followed. In this paper, a heuristic optimisation based on a quadratic integer programming is employed to generate a layout solution that considers customer demand patterns and order clustering. A simulation model is used to investigate the effects of creating and implementing these layout solutions in conjunction with density zones to account for restrictions in non-uniform density SKUs. Results from combining layout optimisation heuristics and density zoning indicate statistical significant differences between assignments that ignore the aforementioned factors and those that recognise it.     

Linear complete differential resultants and the implicitization of linear DPPEs

The linear complete differential resultant of a finite set of linear ordinary differential polynomials is defined. We study the computation by linear complete differential resultants of the implicit equation of a system of n$n$ linear differential polynomial parametric equations in n−1$n-1$ differential parameters. We give necessary conditions to ensure properness of the system of differential polynomial parametric equations.     

A heuristic approach for packing identical rectangles in convex regions

In this paper we propose a heuristic approach for the problem of packing equal rectangles within a convex region. The approach is based on an Iterated Local Search scheme, in which the key step is the perturbation move. Different perturbation moves, both combinatorial and continuous ones, are proposed and compared through extensive computational experiments on a set of test instances. The overall results are quite encouraging.
