Type 2 transglutaminase (TG2) is the main autoantigen in coeliac disease (CD), a widespread inflammatory enteropathy caused by the ingestion of gluten-containing cereals in genetically predisposed individuals. As a consequence, serum antibodies to TG2 represent a very useful marker in CD diagnosis. However, TG2 is also an important player in CD pathogenesis, for its ability to deamidate some Gln residues of gluten peptides, which become more immunogenic in CD intestinal mucosa. Given the importance of TG2 enzymatic activities in CD, several studies have sought to discover specific and potent inhibitors that could be employed in new therapeutical approaches for CD, as alternatives to a lifelong gluten-free diet. In this review, we summarise all the aspects regarding TG2 involvement in CD, including its enzymatic reactions in pathogenesis, the role of anti-TG2 antibodies in disease management, and the exploration of recent strategies to reduce deamidation or to use transamidation to detoxify gluten.  相似文献

Plants evolved an impressive arsenal of multifunctional specialized metabolites to cope with the novel environmental pressures imposed by the terrestrial habitat when moving from water. Here we examine the multifarious roles of flavonoids in plant terrestrialization. We reason on the environmental drivers, other than the increase in UV-B radiation, that were mostly responsible for the rise of flavonoid metabolism and how flavonoids helped plants in land conquest. We are reasonably based on a nutrient-deficiency hypothesis for the replacement of mycosporine-like amino acids, typical of streptophytic algae, with the flavonoid metabolism during the water-to-land transition. We suggest that flavonoids modulated auxin transport and signaling and promoted the symbiosis between plants and fungi (e.g., arbuscular mycorrhizal, AM), a central event for the conquest of land by plants. AM improved the ability of early plants to take up nutrients and water from highly impoverished soils. We offer evidence that flavonoids equipped early land plants with highly versatile “defense compounds”, essential for the new set of abiotic and biotic stressors imposed by the terrestrial environment. We conclude that flavonoids have been multifunctional since the appearance of plants on land, not only acting as UV filters but especially improving both nutrient acquisition and biotic stress defense.  相似文献

The forthcoming 5G technology aims to provide massive device connectivity and ultra-high capacity with reduced latency and costs. These features will be enabled by increasing the density of the base stations, using millimeter-wave (mmWave) bands, massive multiple-input multiple-output systems, and non-orthogonal multiple access techniques. The ability to support a large number of terminals in a small area is in fact a great challenge to guarantee massive access. In this context, this paper proposes a new receiver model for the uplink of 5G mmWave cellular networks. The receiver, called Iterative Decoding and Interference Cancellation (IDIC), is based on the Slotted Aloha (SA) protocol and exploits the capture effect alongside the successive IC process to resolve packet collisions. A 5G propagation scenario, modeled according to recent mmWave channel measurements, is used to compare IDIC with the widely adopted Contention Resolution Diversity SA (CRDSA) scheme to show the performance gain of IDIC, when elements of practical relevance, like imperfect cancellation and receive power diversity, are considered. The impact of packet and power diversity is also investigated to derive the preferable uplink random access strategy that maximizes the system throughput according to the offered channel load.

Artificial intelligent actuators are extensively explored for emerging applications such as soft robots, human-machine interfaces, and biomedical devices. However, intelligent actuating systems based on synthesized polymers suffer from challenges in renewability, sustainability, and safety, while natural polymer-based actuators show limited capabilities and performances due to the presence of abundant hydrogen-bond lockers. Here this study reports a new hydrogen bond-mediated strategy to develop mimosa-inspired starch actuators (SA). By harnessing the unique features of gelatinization and abundant hydrogen bonds, these SA enable high-sensitivity and multi-responsive actuation in various scenarios. The non-gelatinized SA can be irreversibly programmed into diverse shapes, such as artificial flowers, bowl shapes, and helix structures, using near-infrared light. Furthermore, the gelatinized SA exhibit reversibly multi-responsive actuation when exposed to low humidity (10.2%), low temperature (37 °C), or low-energy light (0.42 W cm−2). More importantly, the SA demonstrate robust applications in smart living, including artificial mimosa, intelligent lampshade, and morphing food. By overcoming the hydrogen-bond lockers inherent in natural polymers, SA open new avenues for next-generation recyclable materials and actuators, bringing them closer to practical applications.  相似文献

Testosterone is the most important hormone in male health. Aging is characterized by testosterone deficiency due to decreasing testosterone levels associated with low testicular production, genetic factors, adiposity, and illness. Low testosterone levels in men are associated with sexual dysfunction (low sexual desire, erectile dysfunction), reduced skeletal muscle mass and strength, decreased bone mineral density, increased cardiovascular risk and alterations of the glycometabolic profile. Testosterone replacement therapy (TRT) shows several therapeutic effects while maintaining a good safety profile in hypogonadal men. TRT restores normal levels of serum testosterone in men, increasing libido and energy level and producing beneficial effects on bone density, strength and muscle as well as yielding cardioprotective effects. Nevertheless, TRT could be contraindicated in men with untreated prostate cancer, although poor findings are reported in the literature. In addition, different potential side effects, such as polycythemia, cardiac events and obstructive sleep apnea, should be monitored. The aim of our review is to provide an updated background regarding the pros and cons of TRT, evaluating its role and its clinical applicability in different domains.  相似文献

The human oral pathobionts Aggregatibacter actinomycetemcomitans, Streptococcus mitis and Streptococcus mutans, in dysbiosis-promoting conditions, lead to oral infections, which also represent a threat to human systemic health. This scenario may be worsened by antibiotic misuse, which favours multi-drug resistance, making the research on pathogen containment strategies more than crucial. Therefore, we aimed to in vitro select the most promising probiotic strains against oral pathogen growth, viability, biofilm formation, and co-aggregation capacity, employing both the viable probiotics and their cell-free supernatants (CFSs). Interestingly, we also assessed probiotic efficacy against the three-pathogen co-culture, mimicking an environment similar to that in vivo. Overall, the results showed that Lactobacillus CFSs performed better than the Bifidobacterium, highlighting Limosilactobacillus reuteri LRE11, Lacticaseibacillus rhamnosus LR04, Lacticaseibacillus casei LC04, and Limosilactobacillus fermentum LF26 as the most effective strains, opening the chance to deeper investigation of their action and CFS composition. Altogether, the methodologies presented in this study can be used for probiotic efficacy screenings, in order to better focus the research on a viable probiotic, or on its postbiotics, suitable in case of infections.  相似文献

The fundamental dynamic behaviour of a middle-vessel continuous distillation column is studied in this paper. The interaction between the middle vessel (MV) design and control parameters is identified by means of a linear analysis. Transfer functions relating the dynamics of the product composition responses to changes in the disturbance inputs and manipulated inputs are derived. The role of the MV holdup and MV level controller gain in altering the product composition responses is identified. It is shown that the MV level controller gain can significantly affect the control performance when feed flow disturbances must be compensated for. However, it has a much lower impact in the control performance when feed composition disturbances need to be rejected; in this case, exploiting the “buffering” effect of the MV is more important for control. The analysis is carried out in detail for a LV control configuration, and is subsequently extended to an “on-demand” DB configuration. Finally, simple guidelines for the choice of the MV level controller gain and MV holdup are provided.  相似文献
