This study demonstrates how the common pharmaceutical wetting agent sodium dodecyl sulfate (SDS) increases the absorption of drugs and peptides across the human intestinal epithelium. First, an assay that could follow the reversible and irreversible time-dependent effects of SDS on the permeability of Caco-2 cell monolayers with high reproducibility was developed. SDS (0.40 mM) exposure for 20 min resulted in reversible absorption enhancement of mannitol (M(r), 182 g/mol), 1-deamino-8-D-arginine-vasopressin (M(r), 1071 g/mol), and polyethylene glycol (M(r), 4000 g/mol). A longer (2 h) exposure to SDS resulted in irreversible absorption enhancement. Second, transepithelial electrical resistance measurements (TEER) together with fluorescence and transmission electron microscopy were used to study the effects of SDS on epithelial integrity, cell membranes, intracellular calcium concentration, cytoskeleton, and tight junctions. The effect of SDS (0.40 mM) on epithelial integrity was immediate. A significant decrease in transepithelial electrical resistance measurements was obtained with 1 min after exposure to SDS that was concomitant with increases in the permeability of the apical cell membranes and intracellular calcium concentration. SDS shortened the microvilli of the cells and produced apical (but not basolateral) membrane wounds, actin disbandment, disorganization of the terminal web, and structural separation of the tight junctions. The absorption enhancement was not reduced after repair of the apical cell membranes, indicating that SDS enhances drug and peptide absorption across the intestinal epithelium by the paracellular pathway.  相似文献

In this study a series of 3-arylisoquinoline derivatives were synthesized and cytotoxicity against human melanoma tumor cell evaluated, and a three dimensional quantitative structure-activity relationship was investigated using the comparative molecular field analysis (CoMFA). The results suggested that the electrostatic, steric and hydrophobic factors of 3-arylisoquinolines were strongly correlated with the antitumor activity. Considerable predictive ability (cross-validated r2 as high as 0.721) was obtained through CoMFA.  相似文献

Continuous improvements in insulating materials, manufacturing consistencies in sealed windings and extensive laboratory testing of various systems show that synchronous machines with NEMA II construction can replace totally enclosed machines. Tests on windings in corrosive atmospheres show that sealed insulation systems are impervious to the environment. Standard maintenance ensures that incendive arcing will not be generated in potentially hazardous environments. When compared to totally enclosed machines, the NEMA II-type construction is lighter, more efficient, has lower short-circuit current and associated torques and is easier to maintain  相似文献

Three methods are described for direct post-fracture, post-shadow labelling of individual classes of intramembrane particles (IMPs) in freeze-fracture replicas of biological membranes. The P-face IMPs corresponding to the acetylcholine receptor complexes (AChRs) of vertebrate neuroeffector junctions are identified by post-replication labelling with ferritin-antibody complexes and with neurotoxin-biotin-avidin-colloidal gold affinity ligands. (The freeze-etch nomenclature of Branton et al., 1975, is used in this report.) These post-shadow labelling techniques resemble conventional en bloc labelling techniques except that the labelling reagents must penetrate a thin but discontinuous layer of platinum superimposed on the molecules of interest. In the ‘sectioned labelled-replica technique’, the replicated and labelled tissues are stained, embedded in plastic and sectioned parallel to the replica-tissue interfaces. In the direct ‘labelled-replica techniques’, the replicated and labelled samples are freeze-dried or critical point dried, the labelled surfaces are stabilized by carbon coating, and the underlying tissues are dissolved, allowing the labelled-replicas to be examined as conventional freeze-fracture replicas. The unshadowed side of each AChR IMP is shown to retain sufficient biochemical information to permit both immunospecific and neurotoxin specific labelling despite formaldehyde fixation, freezing, fracturing, platinum shadowing, and thawing in aqueous media. A new mixed ferricyanide-osmium staining method reveals electron opaque structures spanning the membrane bilayer in the same size, number and distribution as the labelled IMPs. These experiments demonstrate the feasibility of identifying individual IMPs in freeze-fracture replicas and may allow the identification of specific membrane lesions in human disease.  相似文献

A fully integrated transceiver suitable for low-data-rate wireless telemetry and sensor networks operating in the license-free ISM frequency bands at 433, 868, or 915 MHz implemented in 0.25-/spl mu/m CMOS is presented. G/FSK, ASK, and OOK modulation formats are supported at data rates from 0.3 to 200 kb/s. The transceiver's analog building blocks include a low-noise amplifier, mixer, channel filter, received signal-strength indication, frequency synthesizer, voltage-controlled oscillator, and power amplifier. FSK demodulation is implemented using a novel digital complex-frequency correlator that operates over a wide modulation-index range and approximates matched filter detection performance. Automatic gain control, automatic frequency control, and symbol timing recovery loops are included on chip. Operating in the 915-MHz band in FSK mode at 9.6 kb/s, the receiver consumes 19.7 mA from a 3-V supply and achieves a sensitivity of -112.8dBm at 0.1% BER. The transmitter consumes 28.5 mA for an output power of 10 dBm and delivers up to 14 dBm.  相似文献

Comparisons were made of the effect on cooling rate, weight loss, texture, bacterial numbers, drip and appearance of pork sides (average dead weight 75 kg) in refrigeration systems using high humidity (ice bank) or conventional chilling, both with and without a rapid pre-chill or delayed chilling, with and without a water spray. All treatments took between 15·7 and 19h post mortem to cool the deep leg of sides to 7°C. Weight loss varied between 0·95% for sides in the delay and spray treatment to 2·17% for conventional chilling. The texture of the M. longissimus dorsi of sides from the rapid pre-chill and conventional chilling treatment was significantly tougher than from the other methods, including those from the rapid pre-chill and high humidity system. Variation in texture between animals within treatments was far greater than between treatments, and could not be explained by variations in cooling and glycolytic rate. There were no significant differences (P > 0·05) in bacterial numbers, drip and appearance between treatments. The choice of chilling system can be made on the basis of weight loss and capital and running costs. The delay and spray treatment would save £37 800 on an annual throughout of 3 080 tonnes of pork compared with a conventional system.  相似文献

Microstrip antennas aperture coupled to a grounded coplanar waveguide (GCPW) have been fabricated on a single layer substrate and studied experimentally and theoretically. Through-plated via holes are used to suppress spurious parallel plate modes. Measurements demonstrate that this antenna element has good performance for both bandwidth and radiation patterns. In this paper, we derive the maximum-likelihood (ML) location estimator for wideband sources in the near field of the sensor array. The ML estimator is optimized in a single step, as opposed to other estimators that are optimized separately in relative time-delay and source location estimations. For the multisource case, we propose and demonstrate an efficient alternating projection procedure based on sequential iterative search on single-source parameters. The proposed algorithm is shown to yield superior performance over other suboptimal techniques, including the wideband MUSIC and the two-step least-squares methods, and is efficient with respect to the derived Cramer-Rao bound (CRB). From the CRB analysis, we find that better source location estimates can be obtained for high-frequency signals than low-frequency signals. In addition, large range estimation error results when the source signal is unknown, but such unknown parameter does not have much impact on angle estimation. In some applications, the locations of some sensors may be unknown and must be estimated. The proposed method is extended to estimate the range from a source to an unknown sensor location. A double-bind communication was broken into its 2 components, punishment and contradictory material, and administered to 4 groups of 39 female college sophomores. There were 2 punishment conditions, 76% variable-interval white noise and no punishment, and 2 levels of stimulus material, contradictory and noncontradictory. These 4 factors were manipulated in a 2 * 2 factorial design over 3 levels of trait anxiety. The criterion measure was change in state anxiety as measured by 5 tests. It was found that the interaction of the 2 components, that is, the full double-bind condition, was significant at the .01 level. Six cases of posterior cervical fusion with rib grafts in children are reported. Four of the children had sustained cervical spine injuries in accidents, and two had congenital absence of the odontoid. Three-level fusions (C1-3) were done in four children, and four-level fusions (C1-4) in two. One child died of unrelated causes 3 months after the operation. The other five children have been followed for 5 to 13 years. All are doing well and each has a remarkably supple, stable neck and no neurological deficit.  相似文献
