We cloned the murine full-length cDNA encoding Ahch, the mouse homologue of DAX1 (DSS-AHC Region on Human X Chromosome, Gene1) which is the gene responsible for human X-linked adrenal hypoplasia congenita (AHC) and hypogonadotropic hypogonadism (HH). Sequence analysis revealed that the murine and human cDNAs have 65% aa identity and 75% aa similarity overall. The cysteine residues in the putative DNA binding domain, which may interact with Zn2+ ions to form zinc fingers, are 100% conserved between the two species, indicating that the novel zinc-finger structures in DAX1 may be functional. In addition, mouse interspecific backcrosses show that the Ahch gene is closely linked to the glycerol kinase locus, GyK, on the mouse X chromosome, indicating that the order of the loci is conserved in this syntenic region between mouse and human. 相似文献

In recent years unmanned vehicles have grown in popularity, with an ever increasing number of applications in industry, the military and research within air, ground and marine domains. In particular, the challenges posed by unmanned marine vehicles in order to increase the level of autonomy include automatic obstacle avoidance and conformance with the Rules of the Road when navigating in the presence of other maritime traffic. The USV Master Plan which has been established for the US Navy outlines a list of objectives for improving autonomy in order to increase mission diversity and reduce the amount of supervisory intervention. This paper addresses the specific development needs based on notable research carried out to date, primarily with regard to navigation, guidance, control and motion planning. The integration of the International Regulations for Avoiding Collisions at Sea within the obstacle avoidance protocols seeks to prevent maritime accidents attributed to human error. The addition of these critical safety measures may be key to a future growth in demand for USVs, as they serve to pave the way for establishing legal policies for unmanned vessels. 相似文献

Methanol-induced conformational transitions of hen egg white lysozyme were investigated with a combined use of far- and near-UV CD and NMR spectroscopies, ANS binding and small-angle X-ray scattering. Addition of methanol induced no global change in the native conformation itself, but induced a transition from the native state to the denatured state which was highly cooperative, as shown by the coincidence of transition curves monitored by the far- and near-UV CD spectroscopy, by isodichroic points in the far- and near-UV CD spectra and by the concomitant disappearance of individual 1H NMR signals of the native state. The ANS binding experiments could detect no intermediate conformer similar to the molten globule state in the process of the methanol denaturation. However, at high concentration of methanol, e.g., 60% (v/v) methanol/water, a highly helical state (H) was realized. The H state had a helical content much higher than the native state, monitored by far-UV CD spectroscopy, and had no specific tertiary structure, monitored both by near-UV CD and NMR spectroscopy. The radius of gyration in the H state, 24.9 angstroms, was significantly larger than that in the native state (15.7 angstroms). The Kratky plot for the H state did not show a clear peak and was quite similar to that for the urea-denatured state, indicating a complete lack of globularity. Thus we conclude that the H state has a considerably expanded, flexible broken rod-like conformation which is clearly distinguishable from the "molten globule" state. Plant class I glucan endo-1,3-beta-glucosidases (beta-1,3-glucanase; 1,3-beta-D-glucan glucanohydrolase, EC 3.2.1.39) have been implicated in development and defense against pathogen attack. Nevertheless, beta-1,3-glucanase deficiencies generated by antisense transformation of Nicotiana sylvestris and tobacco have little biological effect. We report here that another beta-1,3-glucanase activity is induced in these deficient mutants after infection with necrotizing viruses. Induction of class I beta-1,3-glucanase was markedly inhibited in leaves of N. sylvestris and tobacco antisense transformants infected with tobacco necrosis virus and tobacco mosaic virus, respectively. A serologically distinct beta-1,3-glucanase activity was present in the infected antisense transformants but was absent in both healthy and infected control plants and in antisense transformants treated with the stress hormone ethylene. Immunoblot analyses, localization studies, and measurements of antibody specificity indicate that this compensatory beta-1,3-glucanase activity is an intracellular enzyme different from known tobacco beta-1,3-glucanases. Therefore, plants can compensate for a deficiency in enzyme activity by producing a functionally equivalent replacement--i.e., "ersatz"--protein or proteins. The fact that compensation for beta-1,3-glucanase activity occurs in response to infection argues strongly for an important role of these enzymes in pathogenesis. 相似文献

Information Systems Security (ISS) has constantly been ranked as a key concern for Information Systems (IS) managers. Research in the field has largely assumed rational choice (functional) approaches to managing ISS. Such approaches do not give due recognition to the role of improvisation in ISS work. Empirical evidence in organisations suggests that in the context of dynamic, volatile and uncertain environments practitioners are both rational and adaptive (a manifestation of improvisation). In this paper, we conceptualise and demonstrate the manifestation of improvisation in ISS. In order to develop a better understanding of improvisation in ISS activities, hermeneutical and exegetical techniques were employed. Empirical data were collected through in-depth interviews in a single case study. The data obtained were analysed and interpreted hermeneutically. Generally it was found that improvisation is manifested in ISS activities. Implications of these and other findings for the scholarly community and for practical use are discussed. 相似文献

Multiplexers based on the modulation of superconducting quantum interference devices are now regularly used in multi-kilopixel arrays of superconducting detectors for astrophysics, cosmology, and materials analysis. Over the next decade, much larger arrays will be needed. These larger arrays require new modulation techniques and compact multiplexer elements that fit within each pixel. We present a new in-focal-plane code-division multiplexer that provides multiplexing elements with the required scalability. This code-division multiplexer uses compact lithographic modulation elements that simultaneously multiplex both signal outputs and superconducting transition-edge sensor (TES) detector bias voltages. It eliminates the shunt resistor used to voltage bias TES detectors, greatly reduces power dissipation, allows different dc bias voltages for each TES, and makes all elements sufficiently compact to fit inside the detector pixel area. These in-focal plane code-division multiplexers can be combined with multi-GHz readout based on superconducting microresonators to scale to even larger arrays. 相似文献

The clock distribution and generation circuitry forms a critical component of current synchronous digital systems and is known to consume at least a quarter of the power budget of existing microprocessors. We propose and validate a high level model for evaluating the energy dissipation of the clock generation and distribution circuitry, including both the dynamic and leakage power components. The validation results show that the model is reasonably accurate, with the average deviation being within 10% of SPICE simulations. Access to this model can enable further research at high-level design stages in optimizing the system clock power. To illustrate this, a few architectural modifications are considered and their effect on the clock subsystem and the total system power budget is assessed. 相似文献

The square root covariance Kalman filter (SRCF) and the square root covariance Kalman predictor (SRCP) are derived from a least squares viewpoint. A new systolic array architecture is presented which is suitable for implementing both forms of the filter. The systolic SRCF is found to be comparable with other architectures in the literature in terms of size, speed and processor utilization. The recent development of open-source 3-D printers makes scaling of distributed additive-based manufacturing of high-value objects technically feasible and offers the potential for widespread proliferation of mechatronics education and participation. These self-replicating rapid prototypers (RepRaps) can manufacture approximately half of their own parts from sequential fused deposition of polymer feedstocks. RepRaps have been demonstrated for conventional prototyping and engineering, customizing scientific equipment, and appropriate technology-related manufacturing for sustainable development. However, in order for this technology to proliferate like 2-D electronic printers have, it must be economically viable for a typical household. This study reports on the life-cycle economic analysis (LCEA) of RepRap technology for an average US household. A new low-cost RepRap is described and the costs of materials and time to construct it are quantified. The economic costs of a selection of 20 open-source printable designs (representing less than 0.02% of those available), are typical of products that a household might purchase, are quantified for print time, energy, and filament consumption and compared to low and high Internet market prices for similar products without shipping costs. The results show that even making the extremely conservative assumption that the household would only use the printer to make the selected 20 products a year the avoided purchase cost savings would range from about $300 to $2000/year. Assuming the 25 h of necessary printing for the selected products is evenly distributed throughout the year these savings provide a simple payback time for the RepRap in 4 months to 2 years and provide an ROI between >200% and >40%. As both upgrades and the components that are most likely to wear out in the RepRap can be printed and thus the lifetime of the distributing manufacturing can be substantially increased the unavoidable conclusion from this study is that the RepRap is an economically attractive investment for the average US household already. It appears clear that as RepRaps improve in reliability, continue to decline in cost and both the number and assumed utility of open-source designs continues growing exponentially, open-source 3-D printers will become a mass-market mechatronic device. 相似文献
