Proteomics has been widely used to study muscle biology and meat quality traits from different species including beef. Beef proteomics studies allow a better understanding of the biological processes related to meat quality trait determination. This study aimed to decipher by means of two-dimensional electrophoresis (2D-PAGE), mass spectrometry and bioinformatics the changes in post-mortem muscle with a focus on proteins differentially expressed in the Longissimus thoracis (LT) muscle of immunocastrated young heifers and steers. Carcass traits, chemical composition, pH, instrumental color (L*, a*, b*), cooking loss and Warner-Bratzler shear force (WBSF) of meat from F1 Montana-Nellore cattle were also evaluated. Backfat thickness (BFT) and intramuscular fat content (IMF) were 46.8% and 63.6% higher in heifers (p < 0.05), respectively, while evaporation losses (EL) were 10.22% lower compared to steers. No differences (p > 0.05) were observed for tenderness evaluated by WBSF (3, 10, and 17 days post-mortem), pH, and color traits (L*, a* and b*) between the experimental groups. The study revealed several proteins to be differentially expressed proteins in heifers compared steers (p < 0.05). In heifers, proteins involved in nutrient transport (TF, ALB, and MB), energy metabolism (ALDOA, GAPDH, and PKM), and oxidative stress and response to stress (HSPA8 and CA3) were associated with a greater BFT and IMF deposition. The higher expression of these proteins indicated greater oxidative capacity and lower glycolytic activity in the LT muscle of heifers. In steers, there was greater abundance of protein expression related to muscle contraction and proteins of structure (ACTA1, TPM2 and TNNT3), energy metabolism (ENO1, ENO3, PYGM, PGM1 and TPI1) and ATP metabolism (ATP5F1B, PEBP1 and AK1), indicating greater glycogenolysis in LT muscle, suggesting a shift in the glycolytic/oxidative fibers of steers. 相似文献

Despite the major advances in health psychology over the past decade and the changes that have taken place in the Canadian health sector, there remains a paucity of information available about the structure of psychological services in Canadian health-care settings. The most current information about psychology in Canada's hospitals was gathered in 1982 - almost 20 years ago (Arnett, Martin, Steiner, & Goodman, 1987). This article updates the previous information, as it presents the results of a survey that was sent to 975 acute, psychiatric and continuing care health facilities with a minimum patient bed count of 100. Detailed information about the number of psychologists and administrative organizations of psychological services in Canada are reported. The range of services provided by psychologists in health-care settings has expanded, and professional autonomy, as shown by the existence of independent departments of psychology, has changed. The implications of these results for the organization and delivery of psychological services in Canadian healthcare, and for advocacy on the part of psychology, are provided. Cathodic corrosion protection and corrosion protection coatings in sea-water It is a common method to protect ship hulls and underwater constructions against corrosion by multi-layer organic coatings with additional cathodic protection (impressed current, sacrificial anodes). In practice, however, the organic coatings are susceptible to blistering under the conditions of cathodic protection, and as a consequence the effectivity of the passive corrosion protection becomes questionable. In a parameter study with chlorinated rubber an attempt was made at evaluating the influence of the formulation–pigments, extenders, plasticizers, solvents, pigment volume concentration, drying time, film thickness, shop-primer (with/without) – on the tendency to blistering. For the laboratory tests scheduled for up to 18 months' duration an automatic testing equipment was installed in which the coated panels–mild steel, 100 × 150 × 3 mm3–were held in areated synthetic sea water (according ASTM D 1141-75) with simultaneous cathodic protection potentials of UH = ?850 and ?950 mV. On the basis of the results of this study it is possible to formulate chlorinated rubber coatings resistant to blistering even at overprotection potentials down to UH = ?950 mV. The results of this study, however, can not or only with much reservation be transferred to other vehicles such as epoxy, PUR and PVC-copolymers. Factors affecting blistering are discussed. 相似文献

This paper presents the results obtained from selenium fractionation in plasma, muscle and liver samples of Nile tilapia’s (Oreochromis niloticus) after protein separation. The plasma, muscle and liver proteome was obtained by 2D-PAGE, and selenium in protein spots was qualitatively and quantitatively determined by synchrotron radiation X-ray fluorescence and graphite furnace atomic absorption spectrometry (GFAAS). The fluorescence spectra indicated the presence of selenium in three protein spots of plasma, two of muscle and one of liver. Selenium was found to be distributed mainly in proteins with a molar mass smaller than 57.0 kDa and with pI in the range of 5.9–9.6, with one exception in the plasma sample, which presented protein with a molar mass of 60.0 kDa. After acid mineralization of the protein spots, a GFAAS determination of the concentration of selenium bound to these proteins indicated a range of 1.35–6.82 mg per g of protein. 相似文献

The Canadian legal system has made a number of significant reforms in the last few decades regarding the rights of child witnesses and, in particular, the rights of those who have disclosed sexual abuse. This paper provides an overview of the law in Canada as it pertains to child sexual abuse victims and witnesses, and reflects on the role and responsibilities of psychologists who work with child witnesses. It reviews the effects of protection reforms and preparation programs on children who must testify, examines some of the current major issues in the research literature in the area of interviewing and assessing sexually abused children, and considers the role of the psychologists as expert witness in court. In addition, it discusses potential ethical dilemmas for psychologists who work with child witnesses, and proposes recommendations for research and clinical practice.
