Understanding the regulation of the testicular endocrine function leading to testosterone production is a major objective as the alteration of endocrine function is associated with the development of many diseases such as infertility. In the last decades, it has been demonstrated that several endogenous molecules regulate the steroidogenic pathway. Among them, bile acids have recently emerged as local regulators of testicular physiology and particularly endocrine function. Bile acids act through the nuclear receptor FXRα (Farnesoid-X-receptor alpha; NR1H4) and the G-protein-coupled bile acid receptor (GPBAR-1; TGR5). While FXRα has been demonstrated to regulate testosterone synthesis within Leydig cells, no data are available regarding TGR5. Here, we investigated the potential role of TGR5 within Leydig cells using cell culture approaches combined with pharmacological exposure to the TGR5 agonist INT-777. The data show that activation of TGR5 results in a decrease in testosterone levels. TGR5 acts through the PKA pathway to regulate steroidogenesis. In addition, our data show that TGR5 activation leads to an increase in cholesterol ester levels. This suggests that altered lipid homeostasis may be a mechanism explaining the TGR5-induced decrease in testosterone levels. In conclusion, the present work highlights the impact of the TGR5 signaling pathway on testosterone production and reinforces the links between bile acid signaling pathways and the testicular endocrine function. The testicular bile acid pathways need to be further explored to increase our knowledge of pathologies associated with impaired testicular endocrine function, such as fertility disorders. 相似文献

The low-frequency noise observed in thermal lens spectrometry (TLS) can be modeled by assuming that the heated region, constituted by the thermal lens gradient and associated convective stream, behaves as a weakly damped harmonic oscillator with a natural frequency, vo, which is forced to move at an externally imposed pump frequency, vp. Out-of-phase lower-frequency oscillations of the TLS signal can be produced both by transient events, such as the beginning of the TLS experiment and small changes in the pump beam stability, and by drift of boundary conditions, such as the temperature of the surroundings. A model is developed and checked using 1-(2-pyridylazo)-2-napthol (PAN) solutions in silicone oil. Consequences of analytical interest are drawn; e.g., the signal-to-noise ratio of the TLS experiments is improved by pumping at the resonance frequency, where vp = vo. 相似文献

Following the development of a high-throughput (HT) methodology for the evaluation of diesel soot oxidation catalysts in a 16 parallel channels reactor, a library of over 60 catalysts was tested under optimized conditions. The catalyst compositions were chosen to include solids which specific properties like oxygen storage capacity, oxygen mobility and ionic conductivity. The key parameters for high activity appear related to the presence of active and mobile surface oxygen species, and to an appropriate catalyst particle size in order to favour the number of contacts with the soot. In contrast, high oxygen storage capacity and bulk oxygen ion mobility do not appear as relevant properties for high catalytic activity. The biphasic catalytic reduction of the C–C double bond of dimethylitaconate with a water soluble rhodium/triphenylphosphinetrisulphonated sodium salt (TPPTS) complex is investigated. Kinetic studies in a well-mixed batch reactor provide kinetics parameters and an activation energy of 71 kJ mol−1 but cannot discriminate between a first order or a complex kinetic model within the range of substrate concentration where the approximation of linear liquid/liquid partition is respected. Previous studies have shown the worldwide presence of six congeners of polybrominated diphenyl ethers (PBDEs) in marine biota (BDE-28, -47, -99, -100, -153 and -154). The objective of the present study was to document their presence, their level and their transfer in the food web of two major estuaries in France, the Loire and the Seine. PBDEs were quantified in eight principal species from the Loire, representing primary consumers (the bivalve Scrobicularia plana), omnivores (the worm Nereis diversicolor, the shrimps Crangon crangon, Palaemon elegans and Palaemon serratus, the flatfish Platichthys flesus and Solea solea) and supercarnivores (the eel Anguilla anguilla). In the Seine, only worms, bivalves, sole and eels have been studied. Parameters, which can interfere with the interpretation of contamination data (organ distribution, influence of weight or size of specimens, lipid richness, intrinsic variability), have been examined. BDE-47 was the predominant congener in all biota. Higher contamination was observed in most of the species collected from the Seine, in agreement with the higher human presence and economic activity in the Seine than in the Loire basin. PBDEs have been shown to biomagnify in both of the studied estuarine food webs. However, assessment of PBDE transfer from seafood products exposed to contaminants in the Seine estuary showed that human daily intake is far below the no observed adverse effect levels. 相似文献

A field campaign was conducted in Ny-Alesund (78 degrees 54'N, 11 degrees 53'E), Svalbard (Norway) during April and May 2005. An Atmospheric Mercury (Hg) Depletion Event (AMDE) was observed from the morning of April 24 until the evening of April 27. Transport of already Hg and ozone (O3) depleted air masses could explain this observed depletion. Due to a snowfall event during the AMDE, surface snow Hg concentrations increased two fold. Hg deposition took place over a short period of time corresponding to 3-4 days. More than 80% of the deposited Hg was estimated to be reemitted back to the atmosphere in the days following the event. During the campaign, we observed night and day variations in surface snow Hg concentrations, which may be the result of gaseous elemental mercury (GEM) oxidation to divalent Hg at the snow/air interface by daylight surface snow chemistry. Finally, a decrease in the reactive Hg (HgR) fraction of total Hg (HgT) in the surface snow was observed during spring. Construction and demolition waste (CDW) are the largest waste products in the world today and competes as a viable recycled additive material in place of natural aggregates. Due to the increase in compressive strength of different mix proportions of CDW, it is also considered for reuse in concrete and subbase construction. This study shows the effect of CDW in expansive soil stabilization. The chemical and mechanical properties of these materials have shown that they are capable of developing compressive strength properties for replacement of cement with significant reduction in carbon emission. The inherent compositional properties of recycled CDW compared in this review suggests that CDW have good filler properties in highly expansive soils. Mixtures of crushed brick and recycled aggregates characterised based on chemical properties of different replacement ratios suggests that CDW of good-quality aggregates reduces swell potential of expansive soils and increased mechanical strength in pavement construction. 相似文献

The effect of metal exposure on the accumulation and cytosolic speciation of metals in livers of wild populations of European eel with special emphasis on metallothioneins (MT) was studied. Four sampling sites in Flanders showing different degrees of heavy metal contamination were selected for this purpose. An on-line isotope dilution method in combination with size exclusion (SE) high pressure liquid chromatography (HPLC) coupled to Inductively Coupled Plasma time-of-flight Mass Spectrometry (ICP-TOFMS) was used to study the cytosolic speciation of the metals. The distribution of the metals Cd, Cu, Ni, Pb and Zn among cytosolic fractions displayed strong differences. The cytosolic concentration of Cd, Ni and Pb increased proportionally with the total liver levels. However, the cytosolic concentrations of Cu and Zn only increased above a certain liver tissue threshold level. Cd, Cu and Zn, but not Pb and Ni, were largely associated with the MT pool in correspondence with the environmental exposure and liver tissue concentrations. Most of the Pb and Ni and a considerable fraction of Cu and Zn, but not Cd, were associated to High Molecular Weight (HMW) fractions. The relative importance of the Cu and Zn in the HMW fraction decreased with increasing contamination levels while the MT pool became progressively more important. The close relationship between the cytosolic metal load and the total MT levels or the metals bound on the MT pool indicates that the metals, rather than other stress factors, are the major factor determining MT induction. 相似文献
