Silver nanoparticles (AgNP) have been widely applied because of their broad spectrum of antimicrobial activities against bacteria, fungi, and viruses. However, little research has been done to evaluate their effects on Cronobacter sakazakii, an opportunistic pathogen usually infecting infants and having a high fatality rate. The aims of this work were to investigate the antibacterial property of novel, synthesized, positively charged silver nanoparticles against C. sakazakii and to discuss the potential antibacterial mechanisms involved. In this study, the spherical and face-centered cubic silver nanoparticles had a mean particle size of 31.2 nm and were synthesized by reducing Ag+ using citrate and dispersed by glycerol and polyvinylpyrrolidone (PVP) under alkaline conditions. Minimum inhibitory concentrations (MIC) and inhibition zone tests showed that the AgNP exhibited strong antibacterial activity against 4 tested C. sakazakii strains with mean MIC of 62.5 to 125 mg/L and average inhibition zone diameters of 13.8 to 16.3 mm. Silver nanoparticles caused cell membrane injury accompanied by adsorption of AgNP onto the cell surface, as shown by changes in cell morphology, cell membrane hyperpolarization, and accelerated leakage of intracellular reducing sugars and proteins outward from the cytoplasm. In addition, dysfunction of the respiratory chain was induced after treatment with AgNP, which was supported by a decrease in intracellular ATP and inhibition of related dehydrogenases. This research indicates that AgNP could be a novel and efficient antibacterial agent to control C. sakazakii contamination in environments producing powdered infant formulas from milk. 相似文献

Prevention of pathogen colonization of medical implants is a major medical and financial issue since infection by microorganisms constitutes one of the most serious complications after surgery or critical care. Immobilization of antimicrobial molecules on biomaterials surfaces is an efficient approach to prevent biofilm formation. Herein, the first self‐defensive coating against both bacteria and yeasts is reported, where the release of the antimicrobial peptide is triggered by enzymatic degradation of the film due to the pathogens themselves. Biocompatible and biodegradable polysaccharide multilayer films based on functionalized hyaluronic acid by cateslytin (CTL), an endogenous host‐defensive antimicrobial peptide, and chitosan (HA‐CTL‐C/CHI) are deposited on a planar surface with the aim of designing both antibacterial and antifungal coating. After 24 h of incubation, HA‐CTL‐C/CHI films fully inhibit the development of Gram‐positive Staphylococcus aureus bacteria and Candida albicans yeasts, which are common and virulent pathogens agents encountered in care‐associated diseases. Hyaluronidase, secreted by the pathogens, leads to the film degradation and the antimicrobial action of the peptide. Furthermore, the limited fibroblasts adhesion, without cytotoxicity, on HA‐CTL‐C/CHI films highlights a medically relevant application to prevent infections on catheters or tracheal tubes where fibrous tissue encapsulation is undesirable. 相似文献

Green-synthesized metal oxide nanoparticles have garnered considerable attention due to their simple, sustainable, and eco-friendly attributes, coupled with their diverse applications in biomedicine and environmental context. The current study shows a sustainable approach for synthesizing molybdenum oxide nanoparticles (MoONPs) utilizing an extract from Anabaena sp. A-1. This novel approach marks a significant milestone as various spectral approaches were employed for characterization of the green-synthesized MoONPs. Ultraviolet–visible (UV–Vis) spectroscopic analysis revealed a surface plasmon resonance (SPR) peak of MoONPs at 538 nm. Fourier transform infrared (FTIR) spectral analysis facilitated the identification of functional groups responsible for both the stability and production of MoONPs. Scanning electron microscopy (SEM) was utilized revealing a rod shape morphology of the MoONPs. X-ray diffraction (XRD) analysis yielded a calculated crystal size of 31 nm, indicating the crystalline nature of MoONPs. Subsequently, biological assays were employed to ascertain the potential of the bioengineered MoONPs. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay was used to quantify free radical scavenging activity, revealing an antioxidant capacity of 68.1% at 200 μg/mL. To evaluate antibacterial and antifungal efficacy, the disc diffusion method was employed across varying concentrations of MoONPs (6.25, 12.5, 25, 50, 100, 150, 200 μg/mL). Quantification of cytotoxicity was performed via a brine shrimp assay, yielding an IC50 value of 552.3 μg/mL, a metric of moderate cytotoxicity. To assess the biocompatibility of MoONPs, an antihemolytic assay was conducted, confirming their safety profile. Additionally, MoONPs exhibited non-toxic attributes in an insecticidal assay. Notably, in anti-inflammatory assay MoONPs showed an inactive nature towards the reactive oxygen species. In conclusion, these findings highlight the potential versatility of MoONPs in various biological applications, extending beyond their recognized anti-inflammatory and insecticidal properties. 相似文献

There are many practical situations where the underlying distribution of the quality characteristic either deviates from normality or it is unknown. In such cases, practitioners often make use of the nonparametric control charts. In this paper, a new nonparametric double exponentially weighted moving average control chart on the basis of the signed-rank statistic is proposed for monitoring the process location. Monte Carlo simulations are carried out to obtain the run length characteristics of the proposed chart. The performance comparison of the proposed chart with the existing parametric and nonparametric control charts is made by using various performance metrics of the run length distribution. The comparison showed the superiority of the suggested chart over its existing parametric and nonparametric counterparts. An illustrative example for the practical implementation of the proposed chart is also provided by using the industrial data set. 相似文献

A new control chart, namely, modified exponentially weighted moving average (EWMA) control chart, for monitoring the process variance is introduced in this work by following the recommendations of Khan et al.15 The proposed control chart deduces the existing charts to be its special cases. The necessary coefficients, which are required for the construction of modified EWMA chart, are determined for various choices of sample sizes and smoothing constants. The performance of the proposed modified EWMA is evaluated in terms of its run length (RL) characteristics such as average RL and standard deviation of RL. The efficiency of the modified EWMA chart is investigated and compared with some existing control charts. The comparison reveals the superiority of proposal as compared with other control charts in terms of early detection of shift in process variation. The application of the proposal is also demonstrated using a real-life dataset. 相似文献
