Monitoring of nitrate (NO3-) and nitrite (NO2-) content in agricultural products in Slovenia has been carried out since 1996. The results of monitoring over the period 1996-2002 are presented. During this time 924 samples of 14 different agricultural products (potato, lettuce, apples, carrot, silage maize, cabbage, grapes, peaches, string beans, cereals, pears, cucumbers, strawberries and tomato) were analysed. The samples were taken at the time of maturity directly from growing sites and they were analysed using segmented flow analysis. The average nitrate contents were the highest in lettuce (1074 mg kg-1), cabbage (881 mg kg-1), string beans (298 mg kg-1) and carrot (264 mg kg-1), and they were moderately high in potato (158 mg kg-1), silage maize (122 mg kg-1), strawberries (94 mg kg-1), cucumbers (93 mg kg-1) and cereals (49 mg kg-1). Low nitrate contents (below 6 mg kg-1) were found in fruit (grapes, peaches, apples and pears) and tomato. With the exception of cereals (8.9 mg kg-1), apples (1.5 mg kg-1), potato (1.2 mg kg-1) and pears (1.0 mg kg-1) the content of nitrites did not exceed 0.5 mg kg-1. It may be concluded that the results of the monitoring were in most cases similar to the results of investigations obtained in other countries.  相似文献

Oxidative stress, oxidative DNA damage and resulting mutations play a role in colorectal carcinogenesis. Impaired equilibrium between DNA damage formation, antioxidant status, and DNA repair capacity is responsible for the accumulation of genetic mutations and genomic instability. The lesion-specific DNA glycosylases, e.g., hOGG1 and MUTYH, initiate the repair of oxidative DNA damage. Hereditary syndromes (MUTYH-associated polyposis, NTHL1-associated tumor syndrome) with germline mutations causing a loss-of-function in base excision repair glycosylases, serve as straight forward evidence on the role of oxidative DNA damage and its repair. Altered or inhibited function of above glycosylases result in an accumulation of oxidative DNA damage and contribute to the adenoma-adenocarcinoma transition. Oxidative DNA damage, unless repaired, often gives rise G:C > T:A mutations in tumor suppressor genes and proto-oncogenes with subsequent occurrence of chromosomal copy-neutral loss of heterozygosity. For instance, G>T transversions in position c.34 of a KRAS gene serves as a pre-screening tool for MUTYH-associated polyposis diagnosis. Since sporadic colorectal cancer represents more complex and heterogenous disease, the situation is more complicated. In the present study we focused on the roles of base excision repair glycosylases (hOGG1, MUTYH) in colorectal cancer patients by investigating tumor and adjacent mucosa tissues. Although we found downregulation of both glycosylases and significantly lower expression of hOGG1 in tumor tissues, accompanied with G>T mutations in KRAS gene, oxidative DNA damage and its repair cannot solely explain the onset of sporadic colorectal cancer. In this respect, other factors (especially microenvironment) per se or in combination with oxidative DNA damage warrant further attention. Base excision repair characteristics determined in colorectal cancer tissues and their association with disease prognosis have been discussed as well.  相似文献

Much of plant development depends on cell-to-cell redistribution of the plant hormone auxin, which is facilitated by the plasma membrane (PM) localized PIN FORMED (PIN) proteins. Auxin export activity, developmental roles, subcellular trafficking, and polarity of PINs have been well studied, but their structure remains elusive besides a rough outline that they contain two groups of 5 alpha-helices connected by a large hydrophilic loop (HL). Here, we focus on the PIN1 HL as we could produce it in sufficient quantities for biochemical investigations to provide insights into its secondary structure. Circular dichroism (CD) studies revealed its nature as an intrinsically disordered protein (IDP), manifested by the increase of structure content upon thermal melting. Consistent with IDPs serving as interaction platforms, PIN1 loops homodimerize. PIN1 HL cytoplasmic overexpression in Arabidopsis disrupts early endocytic trafficking of PIN1 and PIN2 and causes defects in the cotyledon vasculature formation. In summary, we demonstrate that PIN1 HL has an intrinsically disordered nature, which must be considered to gain further structural insights. Some secondary structures may form transiently during pairing with known and yet-to-be-discovered interactors.  相似文献

Zusammenfassung Die in den USA entwickelte sensorische Texturprofilanalyse stellt die aussagestärkste Methode zur quantitativen und qualitativen Bewertung der Texturunterschiede von Lebensmitteln dar. Vor der Anwendung in Regionen mit anderen Konsumgewohnheiten bedarf sie jedoch einer entsprechenden Adaptierung. Mitteilung 1 berichtet über modifizierte standardisierte Intensitätsskalen zur quantitativen Einstufung der mechanischen Textureigenschaften Festigkeit, Brüchigkeit, Kaubarkeit, weichelastisches Verhalten, Viskosität und Adhäsivität. Die Anpassung an das österreichische Lebensmittelspektrum erfolgte mittels rheologischer Vorprüfungen potentieller Standardpunkte und anschließender Fixierung der Normskalen durch ein geschultes Prüferpanel unter guter Korrelation zwischen instrumentellen und sensorischen Prüfungen.
Adaption of sensory texture profile analysis 1. Scaling of mechanical properties
Summary Texture profile analysis, originally developed in the United States, represents the most complete system of both quantitative and qualitative sensory texture measurements of foodstuffs. Due to regionally varying habits of consumption, an adequate adaptation is necessary prior to application. This paper deals with the evaluation of the mechanical properties firmness, brittleness, chewiness, gumminess, viscosity and adhesiveness. Modifications of standard rating scales for typical Austrian food materials are reported. Rheological examination of commodities was followed by final establishment of standard scales with a trained panel. Good correlation between instrumental and sensory results was obtained.

Several hemostatic strategies rely on the use of blood components such as fibrinogen and thrombin, which suffer from high cost and short shelf‐life. Here, a cost‐effective synthetic biomaterial is developed for rapid local hemostasis. Instead of using thrombin, thrombin‐receptor‐agonist‐peptide‐6 (TRAP6) is covalently engineered in polyvinyl alcohol (PVA) hydrogels. Soluble PVA‐TRAP6 is first prepared by covalent attachment of cysteine‐containing TRAP6 onto the backbone of PVA‐norbornenes (PVA‐NB) through photoconjugation. Cytotoxicity studies using C2C12 myoblasts indicate that PVA‐NB and PVA‐TRAP6 are nontoxic. Thromboelastography reveals that hemostatic activity of TRAP6 is retained in conjugated form, which is comparable to free TRAP6 solutions with equal concentrations. A 0.1% PVA‐TRAP6 solution can shorten the clotting time (CT) to ca. 45% of the physiological CT. High platelet‐activating efficiency is further confirmed by platelet aggregation assay and flow cytometry (FACS). For potential clinical applications, TRAP6‐presenting hydrogel particulates (PVA‐TRAP6‐P) are developed for local platelet activation and hemostasis. PVA‐TRAP6‐P is prepared by biofunctionalization of photopolymerized PVA‐NB hydrogel particulates (PVA‐NB‐P) with TRAP6. It is demonstrated that PVA‐TRAP6‐P can effectively shorten the CT to ca. 50%. FACS shows that PVA‐TRAP6‐P can activate platelets to a comparable extent as soluble TRAP6 control. Altogether, PVA‐TRAP6‐P represents a promising class of biomaterials for safe hemostasis and wound healing.  相似文献

Authentication based on the Merkle tree has been proposed as an energy efficient approach in a resource constrained sensor network environment. It replaces complicated certificate verification with more power efficient hash computations. While previous works assumed complete binary Merkle tree structures, which can be used efficiently only in sensor networks with a specific number of sensor nodes, we investigate incomplete Merkle trees to support any number of sensors. For the incomplete Merkle tree, we demonstrate that an optimal structure can be found through mathematical analysis and simulation. A novel tree indexing scheme is also proposed to reduce communication overhead and save sensors resources during authentication  相似文献

The purpose of this study was to investigate the implications of selected chemopreventive parameters and metabolic conversion of resveratrol in vivo. In two 8-week long feeding experiments with rats, a low-resveratrol diet containing 50 mg resveratrol per kg body weight (bw) and day and a high-resveratrol diet with 300 mg per kg bw and day were administered. For chemopreventive evaluation selected phase I and phase II enzymes of the biotransformation system, the total antioxidant activity, and the vitamin E status of the animals were determined. The level of resveratrol and its metabolites in the feces, urine, plasma, liver, and kidneys was identified and quantitated by high-performance liquid chromatography-diode array detection (HPLC-DAD) using synthesized resveratrol conjugate standards. Feeding of different dosages of resveratrol revealed no effect on the different chemopreventive parameters, except for the total antioxidant activity, which was elevated in plasma by 19% after feeding 50 mg resveratrol per kg bw and day. The formation of trans-resveratrol-3-sulfate, trans-resveratrol-4'-sulfate, trans-resveratrol-3,5-disulfate, trans-resveratrol-3,4'-disulfate, trans-resveratrol-3,4',5-trisulfate, trans-resveratrol-3-O-beta-D-glucuronide, and resveratrol aglycone was detected by HPLC analysis, depending on the biological material. Total resveratrol recovery in urine and feces of rats fed on 50 mg resveratrol per kg bw and day was 15% and 13%, respectively. For rats fed the higher dosage of 300 mg resveratrol per kg bw and day recovery was 54% and 17%, respectively. This is the first study performed with synthesized standards of relevant resveratrol conjugates. The lack of effect on the chemopreventive parameters is probably due to the formation of various resveratrol conjugates reducing its bioavailability in the rat.  相似文献

The membrane fluidity characteristics of multilamellar (MLV) and extruded liposomes prepared with kaempferol (K), kaempferol-3-glucoside (KG), (−)-epigallocatechin (EGC) or (−)-epigallocatechin-3-gallate (EGCG) are presented. Kaempferol caused the highest increase in fluorescence polarisation of DPH in both liposomes (other compounds had not) indicating that K with nK/nLip below 0.2 or 0.1 decreased the membrane fluidity, while at higher molar ratios the membrane fluidity increased. EPR measurements with MLV and spin probes MeFASL(10,3) and MeFASL(2,11) showed a significant decrease in fluidity in the upper part of the membrane for all flavonoids measured, and in the core of the membrane an increase in fluidity for EGCG and EGC. Computer simulation of the EPR spectra showed that the membrane of the MLV used was composed of at least three coexisting domain types with different fluidity and that the order parameter of the most ordered domains is responsible for membrane fluidity alterations.  相似文献

Die ESL-Technologie stellt nach der H-Milch eine weitere Entwicklung mit hohem Zusatznutzen in Bezug auf Haltbarkeit und Vorratshaltung dar, wovon sowohl Handel als auch Verbraucher gleicherma?en profitieren k?nnen. Im Vergleich zum herk?mmlich pasteurisierten Produkt bringt das ESL-Konzept entscheidende Vorteile hinsichtlich der Haltbarkeit der Milch mit sich, wirkt sich dabei aber kaum oder nur geringfügig auf die für den Verbraucher entscheidenden bzw. wertrelevanten Produkteigenschaften aus. ESL-Produkte sind in diesem Sinne also nicht nur „l?nger haltbar”, sondern unserer Einsch?tzung nach auch „l?nger frisch”. Dass gerade die Produktcharakteristika der ESL-Milch den Wünschen der Verbraucher entgegenkommen, zeigt sich an der stetig steigenden Nachfrage nach diesen Produkten und dem damit einhergehenden Absatzrückgang von herk?mmlich pasteurisierter Frischmilch. Die Verbraucher haben also durch ihr Kaufverhalten in den letzten Jahren bereits deutlich gemacht, dass sie der ESL-Milch durchaus positiv gegenüberstehen und sich mit diesem Produktkonzept keineswegs „get?uscht” fühlen, was die hier zusammengefassten Fakten eindeutig untermauern.  相似文献

The aim of this study was to develop a combined method for measuring the total antioxidant activity, the reductive and the radical scavenging activity. Linoleic acid was used as the substrate for an iron-initiated lipid peroxidation to measure the total antioxidant activity. In addition, methyl esters of linoleic acid hydroperoxides were used as substrates to measure the reductive antioxidant activity. The radical scavenging antioxidant activity was calculated by subtracting the reductive antioxidative activity from the total antioxidative activity. As representative examples, the antioxidants alpha-tocopherol, ascorbic acid, trans-resveratrol and L-glutathione as well as commonly used food additives such as 2(3)-tert-butyl-4-hydroxyanisole (BHA) and 2,6-bis(1,1-dimethylethyl)-4-methylphenol (BHT) were analyzed. The results for the novel antioxidation test showed that alpha-tocopherol, BHA and BHT are primarily acting as radical scavengers, whereas ascorbic acid and L-glutathione show a strong reductive capacity. As linoleic acid as well as its hydroperoxides both are present in foods and in the organism, the test presented here can be considered representative of radical reactions occurring in food matrixes and in vivo. Further experiments are required to document the comprehensive applicability in foods and in vivo.  相似文献
