The significance of fluorescence anisotropy in fluorescence intensity and lifetime measurements, and erroneous measurements and interpretations resulting from its disregard, are thoroughly discussed, formulated and quantified. In all fluorescence-related measurements--including excitation and emission spectra, relative fluorescence intensity (FI), fluorescenc life time (FLT), fluorescence resonance energy transfer (FRET), etc., with the exception of fluorescence polarization and anisotropy--it is generally true that the higher the fluorescence anisotropy, the greater the distortion of fluorescence measurements. Quantifiable distortions occur when fluorescence measurements are conducted without considering the influence of fluorescence anisotropy. Here, this influence is described by numerous newly developed mathematical expressions which are simulated and experimentally confirmed utilizing single and binary fluorescent solutions of fluorophores with different spectroscopic characteristics. A marked agreement is shown between the theory and experimental data, clearly indicating the legitimacy of the physical suppositions and the mathematical expressions presented in this paper. Practical and instructive implications are discussed. The following findings are of special applicative importance: 1) the existence of an infinite number of couples of Magic Angles; 2) the deviation between two equally fluorescing particles having different fluorescence anisotropies; 3) the relation between the detected fluorescence intensity and anisotropy when measured under various setups of emission and excitation polarizers; 4) the dependence of the artificial normalized steady-state weight of a single-exponentially decaying fluorophore on its fluorescence anisotropy.  相似文献

This paper presents a novel image-guided robot-based system to assist orthopedic surgeons in performing distal locking of long bone intramedullary nails. The system consists of a bone-mounted miniature robot fitted with a drill guide that provides rigid mechanical guidance for hand-held drilling of the distal screws' pilot holes. The robot is automatically positioned so that the drill guide and nail distal locking axes coincide, using a single fluoroscopic X-ray image. Since the robot is rigidly attached to the intramedullary nail or bone, no leg immobilization or real-time tracking is required. We describe the system and protocol and present a method for accurate and robust drill guide and nail hole localization and registration. The in vitro system accuracy experiments for fronto-parallel viewing show a mean angular error of 1.3 degrees (std = 0.4 degrees ) between the computed drill guide axes and the actual locking holes axes, and a mean 3.0 mm error (std = 1.1 mm) in the entry and exit drill point, which is adequate for successfully locking the nail.  相似文献

A new method for propulsion using a Ni2MnGa ferromagnetic shape memory flapper is introduced. We optically examine the magnetic field induced strain of pure shear by means of a state of the art generator that provides alternating magnetic fields of 7000 Oe at frequencies of up to 100 Hz. Preliminary measurements show local shear deformation of about 5%, which open new frontiers in propulsion mechanisms.  相似文献

Noncommensurate 2D interfaces hold great promise toward low friction and nanoelectromechanical applications. For identical constituents, the crystals interlock at specific rotational configurations leading to high barriers for slide. In contrast, nonidentical constituents comprising different lattice parameters should enable robust superlubricity for all rotational configurations. This is however not the case for gold–graphite interfaces, as both theory and experiments show scaling behavior of the sliding force as a function of the interface contact area. By simulating the sliding force for gold–graphite interfaces, this work shows that the origin for high force barriers at special angular configurations is a result of commensurability between the moiré structure and the contact geometry. Consequently, this paper suggests new geometries that can potentially overcome such commensurability effects to enable robust superlubricity.  相似文献

This paper investigates limitations and design tradeoffs of the closed-loop sensitivity/performance of linear-time-invariant nonminimum-phase uncertain multiple-input-multiple-output plants, with I inputs and m outputs, where m /spl les/ l. It is shown that if rows i/sub 1/...,i/sub k/ of the plant transfer function form a k /spl times/ l nonminimum phase transfer matrix, and if the design is such that the sensitivity gain of k - 1 rows among the rows i/sub 1/,...,i/sub k/ of the closed-loop transfer function is low, then by necessity the sensitivity gain of the remaining row is high. This sensitivity constraint is quantified with the help of the crossover frequency restriction of a specially constructed single-input-single-output transfer function that includes the right half plane zeros and poles of the k /spl times/ l transfer matrix.  相似文献

Comments on the article by K. T. Herbst-Damm and J. A. Kulik (see record 2005-02260-012) entitled Volunteer support, marital status, and the survival times of terminally ill patients. Can a simple and low-cost intervention, such as a visit by a volunteer, extend the life of terminally ill patients? The answer, according to a study by Herbst-Damm and Kulik (2005), is yes. When it comes to quality of life, however, the effects of volunteer visits remain murky. Thus, it is critical that we look beyond effects on longevity and assess how such a program would influence quality of life. Extending the life of terminally ill patients might not always a blessing. Prolonging their life might even cause harm, as it can extend unwanted physical and mental suffering. It may also go against some patients' desire to hasten their death. Despite Herbst-Damm and Kulik's important findings, the current author remains unconvinced as to the merits of implementing volunteer visits as an intervention policy. Salt damage to plants has been attributed to a combination of several factors including mainly osmotic stress and the accumulation of toxic ions. Recent findings in our laboratory showed that phospholipid hydroperoxide glutathione peroxidase (PHGPX), an enzyme active in the cellular antioxidant system, was induced by salt in citrus cells and mainly in roots of plants. Following this observation we studied the two most important enzymes active in elimination of reactive oxygen species, namely, superoxide dismutase (SOD) and ascorbate peroxidase (APX), to determine whether a general oxidative stress is induced by salt. While Cu/Zn-SOD activity and cytosolic APX protein level were similarly induced by salt and methyl viologen, the response of PHGPX and other APX isozymes was either specific to salt or methyl viologen, respectively. Unlike PHGPX, cytosolic APX and Cu/Zn-SOD were not induced by exogenously added abscisic acid. Salt induced a significant increase in SOD activity which was not matched by the subsequent enzyme APX. We suggest that the excess of H2O2 interacts with lipids to form hydroperoxides which in turn induce and are removed by PHGPX. Ascorbate peroxidase seems to be a key enzyme in determining salt tolerance in citrus as its constitutive activity in salt-sensitive callus is far below the activity observed in salt-tolerant callus, while the activities of other enzymes involved in the defence against oxidative stress, namely SOD, glutathione reductase and PHGPX, are essentially similar.  相似文献

The authors of this study investigated task switching following cerebellar damage. The study group consisted of 7 children and adolescents (M age = 13.8 years) who underwent surgical removal of a benign posterior fossa tumor. They were tested at a sufficient interval after surgery (M lag = 6.13 years) for restoration of normal cognitive skills and intelligence. Although all showed normal learning of the task compared with control participants, when rapid behavioral changes were required (short preparation time), they exhibited behavioral rigidity manifested by enhanced switching cost. These results are in line with another study on serial reaction time with the same patients (A. Berger et al., in press). They have important implications for our understanding of the cognitive sequelae of early cerebellar damage as well as the involvement of the cerebellum in task switching. Pseudoternary (Ge,Sn,Pb)Te compounds display favorable thermoelectric properties. Spinodal decomposition in the quasiternary (Ge,Sn,Pb)Te system is at the origin of a wide solubility gap at low Sn concentrations. The structural evolution of the spinodal decomposition was investigated as a function of aging time at 500°C, using x-ray diffraction, electron microscopy, and scanning electron microscopy. The evolution of the structure at 500°C consists initially of a short diffusion-controlled demixing stage into Pb- and Ge-rich coherent areas, with compositions corresponding to the inflection points of the free-energy curve. The Pb-rich areas adopt configurations associated with the directions of the soft elastic moduli of the cubic compound. Both the Pb- and Ge-rich areas are supersaturated and undergo in a second stage a nucleation and growth process and give rise to a biphased structure with equilibrium compositions corresponding to the boundaries of the miscibility gap. The resulting Pb-rich areas display a relatively stable microstructure suggesting the presence of long-range interactions between the Pb-rich precipitates in the Ge-rich matrix.  相似文献
