Human alveolar macrophages (AM) can produce potent reactive oxygen intermediates (ROI) and arachidonic acid metabolites (eicosanoids), which have important roles in host defense and the pathogenesis of some diseases of the lung. Bacterial lipopolysaccharide (LPS) is believed to cause profound lung injury and can prime mouse peritoneal macrophages for the enhanced secretion of ROI and eicosanoids. Therefore, we investigated the effect of LPS pretreatment on the ability of AM to release superoxide anions (O2-) and leukotriene B4 (LTB4). LPS can prime AM for the enhanced secretion of O2- and LTB4, regardless of whether they are derived from nonsmokers or smokers. Moreover, judging from the time-response characteristics, this priming for LTB4 release could be inhibited in the later stages of pretreatment, when the O2(-)-releasing capacity was enhanced. The priming inhibition was prevented, at least in part, by cycloheximide, but not by SOD and/or catalase. In addition, cycloheximide also inhibited the priming for O2- release. Hence, protein synthesis might be necessary for the priming for O2- release and for inhibiting the priming for LTB4 release. This phenomenon of self-limiting the priming response with LPS seems to be very important when we consider the high oxygen tension in the lungs and the many bacterial substances inspired into alveoli.  相似文献

Abstract— A quantitative model has been proposed which predicts the extent of lifetime scatter in low-cycle fatigue due to the influence of bending caused by load misalignment. The main components of the model are the mechanism of bending, the type of extensometer used to control strain and the fatigue characteristics of the material being assessed. Three mechanisms of bending have been studied and it is argued that the most damaging one is a consequence of a lateral offset in the centre-lines of the load-train with respect to either a machine's frame or ram. Scatter in lifetime is a maximum when strain is controlled by a single extensometer (which is generally the case) and when fatigue behaviour is dictated by crack initiation at the largest surface defect. Two types of scatter have been examined, (i) repeatability scatter due to testing practice within a single laboratory, and (ii) reproducibility scatter between laboratories. An example of the magnitude of reproducibility scatter due to bending has been given by using an equation based on the universal slopes method due to Manson.  相似文献

Virtual instruments for an extracorporeal circulation (ECC) process were developed to simulate the reactions of a patient to different artificial perfusion conditions. The computer simulation of the patient takes into account the hydraulic, volume, thermal and biochemical phenomena and their interaction with the devices involved in ECC (cannulae dimensions, oxygenator and filter types, pulsatile or continuous pump and thermal exchangers). On the basis of the patient's initialisation data (height, weight, Ht) and perfusion variables (pump flow rate, water temperature, gas flow rate and composition) imposed by the operator, the virtual ECC monitors simulated arterial and venous pressure tracings in real time, along with arterial and venous flow rate tracings, urine production tracing and temperature levels. Oxyhemoglobin arterial and venous blood saturation together with other related variables (pO2, pCO2, pH, HCO3 are also monitored. A drug model which allows the simulation of the effect of vasodilator and diuretic drugs is also implemented. Alarms are provided in order to check which variables (pressure, saturation, pH, urine flow) are out of the expected ranges during the ECC simulation. Consequently the possibility of modifying the control parameters of the virtual devices of the ECC in run-time mode offers an interaction mode between the operator and the virtual environment.  相似文献

In this paper we report on the results of an exploratory study of knowledge exchange between disciplines and subfields of science, based on bibliometric methods. The goal of this analysis is twofold. Firstly, we consider knowledge exchange between disciplines at a global level, by analysing cross-disciplinary citations in journal articles, based on the world publication output in 1999. Among others a central position of the Basic Life Sciences within the Life Sciences and of Physics within the Exact Sciences is shown. Limitations of analyses of interdisciplinary impact at the journal level are discussed. A second topic is a discussion of measures which may be used to quantify the rate of knowledge transfer between fields and the importance of work in a given field or for other disciplines. Two measures are applied, which appear to be proper indicators of impact of research on other fields. These indicators of interdisciplinary impact may be applied at other institutional levels as well. Batch process industries are characterized by complex precedence relationships between operations, which renders the estimation of an acceptable workload very difficult. A detailed schedule based model can be used for this purpose, but for large problems this may require a prohibitive large amount of computation time. We propose a regression based model to estimate the makespan of a set of jobs. We extend earlier work based on deterministic processing times by considering Erlang-distributed processing times in our model. This regression-based model is used to support customer order acceptance. Three order acceptance policies are compared by means of simulation experiments: a scheduling policy, a workload policy and a regression policy. The results indicate that the performance of the regression policy can compete with the performance of the scheduling policy in situations with high variety in the job mix and high uncertainty in the processing times. A general concept is cited for the design of an underground transportation tunnel in the Lefortovo District. Attention is focused on problems of the tunnel's layout in plan and profile, the layout of sections, and selection of the driving technology. Results are presented for the numerical modeling of variations in the stress—strain state of the soil mass, which are associated with the tunnel's installation.
