Thrombin/antithrombin III (AT-III) proteolytic system is well known for its function in blood coagulation. Thrombin is expressed in skeletal muscle but nothing is known about the presence of AT-III in the tissue. In postmortem muscle this system has been and is still totally ignored. We therefore successfully attempted to purify AT-III from bovine skeletal muscle and characterized the purified protein (identified as AT-III by N-ter sequencing and mass spectrometry finger print) for its physicochemical and inhibitory properties. As the human blood serpin, muscle AT-III is thermolabile and stable only at alkaline pH (pH 9–10). The muscle serpin inhibits strongly thrombin in a heparin dependent manner and trypsin. Phosphatidylserine (PS) externalization demonstrated in the present work suggested that prothrombin can be activated to thrombin through binding of the activator complex on the external PS groups. PS externalization is concomitant with shrinkage of muscle fibers indicating that muscle cells are engaged in the cell death program known as apoptosis few minutes after death. We then discussed the potential role of this proteolytic system in postmortem apoptosis development as well as in the control of low voltage electrical stimulation efficiency. 相似文献

Beef production under different local husbandry systems might have meat sensory quality implications for the marketing of these products abroad. In order to assess the effect of finishing diet systems on beef quality, a trained sensory taste panel assessed meat aged for 20 days from 80 Uruguayan Hereford steers that were finished on one of the following diets: T1 = Pasture [4% of animal live weight (LW)], T2 = Pasture [3% LW plus concentrate (0.6% LW)], T3 = Pasture [3% LW plus concentrate (1.2% LW)], or T4 = Concentrate plus hay ad libitum. Beef odour and flavour intensities decreased with an increase in the energy content of the diet. The meat from T2 had the lowest acid flavour and strange odours intensities. In general, steers fed only concentrate plus hay (T4) produced meat that had an inferior sensory quality because they had more pronounced off-flavours and was tougher. 相似文献

This multicenter, community-based, prospective, longitudinal study evaluated the safety of nicotine replacement therapy (NRT), bupropion, and combined therapy of NRT and bupropion for smokers seeking to quit, when these therapies were used under real-world conditions following a smoking cessation program. Participants were smokers aged 18 years or older who attended five smoking cessation clinics. Evaluations were made at 15, 30, 60, and 90 days. We investigated the possible existence of adverse effects as well as the severity of each adverse effect and its influence on the treatment course. The study included 904 smokers: 370 received NRT, 413 received bupropion, and 121 received combined therapy. At 15, 30, 60, and 90 days, adverse effects were reported by 43.8%, 33.1%, 22.3%, and 5.7% of subjects, respectively. Adverse effects were significantly more frequent in subjects receiving combined therapy or bupropion alone than in NRT-treated subjects at the 15-, 30-, and 60-day follow-ups. A total of 83 smokers (9.3%) withdrew from treatment and 116 (12.8%) stopped temporarily because of adverse effects. No differences were found in the percentages of discontinuation among the different treatment options. Adverse effects rarely were severe (n=10). Nevertheless, 41 subjects (4.5%) discontinued drug therapy indefinitely and 55 (6.1%) discontinued it temporarily because of mild adverse effects. Pharmacological therapies for smoking cessation are safe as long as they are appropriately prescribed and supervised by clinicians according to clinical practice guidelines. Adverse effects are primarily mild. Nonetheless, mild adverse effects may be perceived by patients as a serious enough problem to cause them to discontinue treatment. 相似文献

This paper presents an alternative method to estimate somatic cell count (SCC) in cows' milk. SCC is an important indicator in the detection of inflammatory reactions within the udder in cows and Direct Optical Microscopy (DOM) is the present reference method for SCC but, owing to its dependence on human operators, it is extremely costly, time-consuming and potentially subjective. The industrial method of choice is Epifluorescence (EF), which has the potential for impressive throughput and acceptable precision, but requires huge inversions and handling of highly toxic reactives and waste. In this paper, an advantageous method that involves application of a low-cost Video Microscopy (VM) system is analysed and discussed, including a comparison between DOM and VM, and an example of application of both methods to evaluate EF counts. Separation and detection of native anhydrous carbohydrates derived from the combustion of biomass using an electrophoretic microchip with pulsed amperometric detection (PAD) is described. Levoglucosan represents the largest single component of the water extractable organics in smoke particles and can be used to trace forest fires or discriminate urban air pollution sources. Detection of levoglucosan and other sugar anhydrides in both source and ambient aerosol samples is typically performed by gas chromatographic (GC) separation with mass spectrometric (MS) detection. This method is cost, time, and labor intensive, typically involving a multistep solvent extraction, chemical derivatization, and finally analysis by GC/MS. However, it provides a rich wealth of chemical information as the result of the combination of a separation method and MS and exhibits good sensitivity. In contrast, microchip capillary electrophoresis offers the possibility of performing simpler, less expensive, and faster analysis. In addition, integrated devices can be fabricated and incorporated with an aerosol collection system to perform semicontinuous, onsite analysis. In the present report, the effect of the separation potential, buffer pH and composition, injection time, and pulsed amperometric detection parameters were studied in an effort to optimize both the separation and detection of anhydrous sugars. Using the optimized conditions, the analysis can be performed in less than a minute, with detection limits ranging from 22 fmol (16.7 microM) for levoglucosan to 336 fmol (258.7 microM) for galactosan. To demonstrate the capabilities of the device, a comparison was made between GC/MS and microchip electrophoresis using an aerosol source sample generated in a wood-burning chamber. Maize drying is an important process, especially for storage and conservation. For this study, the experimental stage was carried out using a forced convection dryer with air heated at different temperature conditions (306.05–441.85 K) and flow (0.13–0.256 m3/hr), totalizing 15 drying curves. Then the performances of the classic drying kinetics methodology and the approach proposed in this paper, in which the increase in moisture content of the product with time was represented combining exponential models and neural networks based on wavelets, were compared. Good performance was obtained in predictions using the proposed approach. One of the main differentials of the methodology adopted was the obtainment of a model that has a global predictive capacity, within the range of tested operating conditions, which can be used in predicting drying curves for different operating conditions.
Practical applications
The drying process is also one of the most widely used methods for preserving food, and has the advantage of reducing the costs of storage and transport because of the low volume and weight of the end product. During the last years, this topic has attracted a broad industrial interest, resulting in many research studies investigating the drying process. Usually, with regard to the classic approach for modeling of the drying process, the kinetics of drying curves obtained in different operating conditions is affected separately, that is, the parameters are estimated independently, resulting in different regression problems. With the classical approach, in general, it is not possible to obtain a comprehensive prediction model with regards to operating conditions. We have proposed an alternative modeling method. Aiming to obtain a modeling tool with an overall predictive ability, an approach for drying kinetics prediction that combines exponential models and neural networks was proposed. The presence of pathogens such as Escherichia coli O157:H7 (EC) represents risks to public health and to economy of Brazilian beef industry. In this context, the application of lactic acid (LA) is an efficient practice employed to reduce bacterial count without compromising consumer safety. Our aim was to verify the inhibitory effect of LA application combined with aging and different packaging on EC and its effects on beef color. The LA effect on EC counts was concentration dependent during aging and storage, with T2 (10% of LA added) demonstrating greater (p < .05) reductions than T1 (5% of LA added). Aging did not affect (p > .05) EC counts, however reduced (p < .05) the total aerobic mesophilic bacteria. LA application promoted a decrease on beef redness (p > .05) after application and during storage. LA treatment (T2) promoted a reduction in E. coli O157:H7, despite the effects on beef color.
Practical applications
The present data evidence a breakthrough in lactic acid (LA) researches once evaluate the inhibitory effect of aging, LA concentration and package on Escherichia coli O157:H7 and the influence of these technologies in beef color.
