Amyloid‐β peptides (Aβ) and the protein human serum albumin (HSA) interact in vivo. They are both localised in the blood plasma and in the cerebrospinal fluid. Among other functions, HSA is involved in the transport of the essential metal copper. Complexes between Aβ and copper ions have been proposed to be an aberrant interaction implicated in the development of Alzheimer's disease, where Cu is involved in Aβ aggregation and production of reactive oxygen species (ROS). In the present work, we studied copper‐exchange reaction between Aβ and HSA or the tetrapeptide DAHK (N‐terminal Cu‐binding domain of HSA) and the consequence of this exchange on Aβ‐induced ROS production and cell toxicity. The following results were obtained: 1) HSA and DAHK removed CuII from Aβ rapidly and stoichiometrically, 2) HSA and DAHK were able to decrease Cu‐induced aggregation of Aβ, 3) HSA and DAHK suppressed the catalytic HO. production in vitro and ROS production in neuroblastoma cells generated by Cu–Aβ and ascorbate, 4) HSA and DAHK were able to rescue these cells from the toxicity of Cu–Aβ with ascorbate, 5) DAHK was more potent in ROS suppression and restoration of neuroblastoma cell viability than HSA, in correlation with an easier reduction of CuII–HSA than Cu–DAHK by ascorbate, in vitro. Our data suggest that HSA is able to decrease aberrant CuII–Aβ interaction. The repercussion of the competition between HSA and Aβ to bind Cu in the blood and brain and its relation to Alzheimer's disease are discussed.  相似文献

There has accumulated a large amount of literature on confidence interval construction involving lognormal data owing to the fact that many data in scientific inquiries may be approximated by this distribution. Procedures have usually been developed in a piecemeal fashion for a single mean, a single mean with excessive zeros, a difference between two means, and a difference between two differences (net health benefit). As an alternative, we present a general approach for all these cases that requires only confidence limits available in introductory texts. Simulation results confirm the validity of this approach. Examples arising from health economics are used to exemplify the methodology.  相似文献

The direct laser interference patterning method was used to fabricate periodic structures on flat and cylindrical specimens of austenitic stainless steel (X5CrNi18-10) and martensitic bearing steel (100Cr6). In such laser processing the characterization of materials is a central issue. Therefore, this paper places special emphasis on the chemical, metallurgical and mechanical characterization of the laser processed metals. During the patterning process of 100Cr6, the carbides in the martensitic structure appear as elongated needles and become aligned to the direction of melt flow. The chemical composition analysis before and after laser interference patterning did not reveal any significant changes to either material. However, the surface hardness of 100Cr6 decreased when energy densities above 2.0 J/cm2 were applied. For cylindrical specimens, the grating period changes with position along the circumference. The observed difference, however, is less than ~6.4% for rotation angles smaller than 20°.  相似文献

There has been an explosion of interest in health sciences applications of case-based reasoning (CBR), not only in the traditional CBR in medicine domain, but also in bioinformatics, enabling home health-care technologies, CBR integration, and synergies between CBR and knowledge discovery. This special issue features the best papers from the third workshop on CBR in the health sciences, held at ICCBR-05 in Madrid. It is the third in a series of exciting workshops, the first two of which were held at ICCBR-03, in Trondheim, Norway, and at ECCBR-04, in Madrid, Spain. The nine high-quality papers introduced here represent the research and experience of twenty-two authors working in eight different countries on a wide range of problems and projects. These papers illustrate some of the major trends of current research in CBR in the health sciences, and represent overall an excellent sample of the most recent advances of CBR in the health sciences.  相似文献

This paper discusses the development of an enterprise domain model in an environment where part of the domain knowledge is vague and not yet formalised in company-wide business rules. The domain model was developed for a young company starting in the telecommunications sector. The company relied on a number of stand-alone business support systems and sought for a manner to integrate them. There was opted for the development of an enterprise-wide domain model that had to serve as an integration layer to coordinate the stand-alone applications. A specific feature of the company was that it could build up its information infrastructure form scratch, so that many aspects of its business were still in the process of being defined. The paper will highlight parts of the Enterprise Model where there was a need for co-designing business rules together with the domain model. A result of this whole effort was that the company got more insight into important domain knowledge and developed a common understanding across functional areas of the way of doing business.  相似文献

A grip-induced muscle tension maintenance task distinguished between schizophrenic patients, regardless of medication or hospitalization status, and both normals and controls with affective (unipolar and bipolar) disorders. Unaffected 1st-degree relatives of schizophrenic patients also showed a grip deficit. Coupling the grip task with a visual discrimination task that divided attention through instruction or reinforcement contingency increased grip error times for all groups. No group differences appeared on the discrimination task, regardless of difficulty level, and the tone used to provide corrective feedback was not implicated in the grip deficit. The results suggest that the grip task is tapping, in a systematic and reliable manner, a motor-control abnormality that may be useful as a behavioral marker of schizophrenia. The objective of this work was to test a molecular genetic method for in situ monitoring of aerobic benzene, toluene, and xylene (BTX) biodegrading microorganisms. Catechol 2,3-dioxygenase (C23DO) genes occur in bacteria that biodegrade benzene, toluene, xylenes, phenol, biphenyl, and naphthalene. A competitive quantitative polymerase chain reaction (QC-PCR) technique using a single set of primers specific for an entire subfamily of C23DO genes was recently developed. To determine whether bacteria containing these C23DO genes actually exist in environments contaminated by BTX, aerobic microcosms containing previously uncontaminated soil were amended with different aromatic hydrocarbons and DNA extracts were analyzed by QC-PCR for C23DO genes. Anaerobic microcosms were established to confirm that oxygen was also necessary for the enrichment of C23DO genes. Field testing was done at two sites undergoing monitored natural attenuation. In microcosm experiments naphthalene, m-xylene, and p-xylene strongly enriched for C23DO genes while benzene, toluene, and o-xylene produced only transient, weakly detectable genes. In the field study, C23DO genes were detected in groundwater samples contaminated with either xylenes or naphthalene. The results of this study demonstrated that molecular genetic techniques can provide an accurate and rapid method to detect microorganisms capable of aromatic hydrocarbon biodegradation. Such a technique would be useful for monitoring the effectiveness of aeration technologies and for documenting microbial processes for monitored natural attenuation.  相似文献

The higher point density and mobility of terrestrial laser scanning (light detection and ranging (lidar)) is desired when extremely detailed elevation data are needed for mapping vertically orientated complex features such as levees, dunes, and cliffs, or when highly accurate data are needed for monitoring geomorphic changes. Mobile terrestrial lidar scanners have the capability for rapid data collection on a larger spatial scale compared with tripod-based terrestrial lidar, but few studies have examined the accuracy of this relatively new mapping technology. For this reason, we conducted a field test at Padre Island National Seashore of a mobile lidar scanner mounted on a sport utility vehicle and integrated with a position and orientation system. The purpose of the study was to assess the vertical and horizontal accuracy of data collected by the mobile terrestrial lidar system, which is georeferenced to the Universal Transverse Mercator coordinate system and the North American Vertical Datum of 1988. To accomplish the study objectives, independent elevation data were collected by conducting a high-accuracy global positioning system survey to establish the coordinates and elevations of 12 targets spaced throughout the 12 km transect. These independent ground control data were compared to the lidar scanner-derived elevations to quantify the accuracy of the mobile lidar system. The performance of the mobile lidar system was also tested at various vehicle speeds and scan density settings (e.g. field of view and linear point spacing) to estimate the optimal parameters for desired point density. After adjustment of the lever arm parameters, the final point cloud accuracy was 0.060 m (east), 0.095 m (north), and 0.053 m (height). The very high density of the resulting point cloud was sufficient to map fine-scale topographic features, such as the complex shape of the sand dunes.  相似文献
