Cloning using nuclear transfer in bovine species: results and perspectives

A all in vitro cloning technique was developed in which the reconstituted embryos from the first cycle nuclear transfer (cloning) were used as blastomere donor for the second cycle nuclear transfer (recloning). Such method permitted to produce 14.5% of morulae and 14.9% of blastocysts after the first and second cycles of nuclear transfer, respectively. The rates of birth obtained after transfer of such embryos were 21.4% et 20.8% for first and second cycles respectively, corresponding to 6 et 5 calves for 28 et 24 transferred embryos. Unfortunately, gestation pathologies and an increase of birth weights were observed. It seems that the in vitro presence of gametes and/or embryos may be responsible of an alteration in the control of gene expression.     

Effect of polytetrafluoroethylene covering of Palmaz stents on the development of intimal hyperplasia in human iliac arteries

PURPOSE: The occurrence of neointimal hyperplasia within a stent may result in restenosis with recurrent symptoms of end-organ ischemia. This study evaluated the potential of a nonporous covering of a stent to function as a barrier to the formation of intrastent neointimal hyperplasia. MATERIALS AND METHODS: Twelve endovascular stent grafts were used to treat 12 high-risk patients with limb-threatening ischemia secondary to long-segment iliac artery occlusion. A 6-mm, thin-walled polytetrafluoroethylene graft was inserted and anchored to the common iliac artery with use of Palmaz stents. Each stent was covered by graft material over one-half of its length. Control angiograms obtained immediately after graft insertion were compared with follow-up angiograms obtained between 4 and 6 months after the initial procedure. On each angiogram, the region of the stent was magnified by 20x to permit computerized luminal diameter measurements. RESULTS: The mean luminal diameter within the stent was significantly greater on the covered (7.7 mm +/- 0.33 standard deviation) compared with the uncovered (6.7 mm +/- 0.85 standard deviation) portions (P < .01). CONCLUSIONS: Partially covered stents are a unique model for assessing the effects of an extrinsic stent covering on arterial healing and myointimal hyperplasia. These data suggest that a relatively nonporous covering of polytetrafluoroethylene may inhibit stent-related restenosis in iliac arteries.     

A new polymorphic restriction site in the human 11 beta-hydroxysteroid dehydrogenase type 2 gene

The 11 beta-hydroxysteroid dehydrogenase type II enzyme (11 beta HSD2) inactivates glucocorticoids in the kidney and thus prevents glucocorticoids from occupying the non-selective mineralocorticoid receptor in epithelial tissues. Mutations in the HSD11B2 gene have been found to cause the syndrome of apparent mineralocorticoid excess, a rare autosomal recessive disease characterized by severe hypertension. Thus, this locus could also be an ideal candidate involved in the etiology of primary hypertension. We identified a polymorphism in exon 3 characterized by a GAG to GAA transition at codon 178, with the loss of an Alu I restriction site and analysed it in an association study using end-stage renal disease patients, diabetic or essential hypertensive patients and control subjects. Two-hundred and eighty nine subjects and patients were analysed; the genotype was determined by amplification of genomic DNA and subsequent digestion with Alu I restriction enzyme. The prevalence of the Alu I allele was 8.6% in healthy control subjects (n = 116). This prevalence was lower (chi 2 P = 0.035 vs. controls) than the 18.0% in a group of renal transplant patients (n = 61). The corresponding values for patients with diabetes mellitus (n = 25), hypertension (n = 41) and patients on dialysis (n = 46) were 4.0%, 4.8% and 4.3%, respectively. There was no correlation between blood pressure and the marker in non-ESRD subjects. These data indicate the presence of a polymorphic marker in exon 3 of the HSD11B2 gene; this marker is associated with end-stage renal disease but not with essential hypertension in humans.     

A new oxisuran metabolite

1. An unidentified oxisuran metabolite which had been observed in animal urine was biosynthesized by incubating [14C]oxisuran with rat liver cytosol. 2. The metabolite, isolated by preparative t.l.c. and extraction, was identified as oxisuran alcohol sulphide by mass fragmentography. Confirmation of this identification was obtained by biosynthesis of the same compound from oxisuran sulphide. 3. The 9000 g supernatant liquid from rat liver was less effective than cytosol in reducing oxisuran to its alcohol sulphide. Neither rat liver fraction reduced oxisuran alcohol sulphoxides to sulphide. 4. The 9000 g fraction oxidized oxisuran and oxisuran alcohol sulphoxide to oxisuran alcohol sulphone.     

Congenital anomalies of the diaphragm. Personal experience with thirty-five consecutive cases

A personal experience of thirty-five consecutive patients with congenital anomalies of the diaphragm is reported. The anomalies included twenty-five Bochdalek hernias, seven diaphragmatic eventrations, and three esophageal hiatal hernias. Except for the patients with esophageal hiatal hernias, virtually all patients presented with respiratory embarrassment, twenty-seven of the thirty-five within 24 hours of birth. An especially high mortality is associated with this early onset of symptoms. The treatment was surgical with a thoracic approach used for right Bochdalek defects and eventrations, and an abdominal approach used for left Bochdalek hernias. The importance of proper preoperative and postoperative management in a well equipped neonatal intensive care unit is emphasized.
