Zeranol, an oestrogenic growth promoter in food animals, is banned within the European Union (EU). However, commercially available immunoassay kits for zeranol cross-react with toxins formed by naturally occurring Fusarium spp. fungi, leading to false-positive screening results. This paper describes the validation of a specificity enhanced, rapid dry reagent time-resolved fluoroimmunoassay (TR-FIA) for zeranol (recovery 99%, limit of detection 1.3 ng ml(-1)) demonstrating that up to 150 ng ml(-1) of Fusarium spp. toxins in urine do not lead to false-positive results. This assay will assist EU Member States to implement Council Directive 96/23/EC, which requires states to monitor for potential abuses of zeranol. A similar TR-FIA for the Fusarium spp. toxin alpha-zearalenol, using the same sample extract, is also described (recovery 68%, limit of detection 5.6 ng ml(-1)). Only the addition of diluted sample extract is required to perform these dry-reagent TRFIAs, the results being available within 1h of extract application. The EU-funded project 'Natural Zeranol' (FAIR5-CT97-3443) will use these fluoroimmunoassays to screen bovine urine in four Member States to gather data on the seasonality of Fusarium spp. toxin contamination of urine and the incidence of zeranol screening test positives.  相似文献

PURPOSE: Demonstration of a technique for three-dimensional (3-D) assessment of tracheal-stenoses, regarding site, length and degree, based on spiral computed tomography (S-CT). PATIENTS AND METHODS: S-CT scanning and automated segmentation of the laryngo-tracheal tract (LTT) was followed by the extraction of the LTT medial axis using a skeletonization algorithm. Orthogonal to the medial axis the LTT 3-D cross-sectional profile was computed and presented as line charts, where degree and length was obtained. Values for both parameters were compared between 36 patients and 18 normal controls separately. Accuracy and precision was derived from 17 phantom studies. RESULTS: Average degree and length of tracheal stenoses was found to be 60.5% and 4.32 cm in patients compared with minor caliber changes of 8.8% and 2.31 cm in normal controls (p < 0.0001). For the phantoms an excellent correlation between the true and computed 3-D cross-sectional profile was found (p < 0.005) and an accuracy for length and degree measurements of 2.14 mm and 2.53% respectively could be determined. The corresponding figures for the precision were found to be 0.92 mm and 2.56%. CONCLUSION: LTT 3-D cross-sectional profiles permit objective, accurate and precise assessment of LTT caliber changes. Minor LTT caliber changes can be observed even in normals and, in case of an otherwise normal S-CT study, can be regarded as artifacts.  相似文献

A discrete coil EIT system is investigated for the general case of an eccentric circular inhomogeneity. The solution methodology of the forward problem of this system is explained. An optimization procedure using this forward problem solution is developed to find optimum currents that maximize the distinguishability. For an eccentric inhomogeneity problem, it is shown that the coil currents can be optimized to focus the current density in a region of interest. Optimum coil currents under limited peak coil currents constraint and limited total power constraint are obtained. Representative examples that demonstrate the performance of the system are presented.  相似文献

The origin and history section provides a chronology of this work, emphasizing the milestones reached during the past three decades. We present a method for accurate image registration and motion compensation in multidimensional signals, such as two-dimensional (2-D) X-ray images and three-dimensional (3-D) computed tomography/magnetic resonance imaging volumes. The method is based on phase from quadrature filters, which makes it robust to noise and temporal intensity variations. The method is equally applicable to signals of two, three or higher number of dimensions. We use parametric models, e.g., affine models, finite elements or local affine models with global regularization. Experimental results show high accuracy for 2-D and 3-D motion compensation.  相似文献

A four-dimensional mapping (three spatial dimensions + time) of myocardial strain-rate would help to describe the mechanical properties of the myocardium, which affect important physiological factors such as the pumping performance of the ventricles. Strain-rate represents the local instantaneous deformation of the myocardium and can be calculated from the spatial gradients of the velocity field. Strain-rate has previously been calculated using one-dimensional (ultrasound) or two-dimensional (2-D) magnetic resonance imaging) techniques. However, this assumes that myocardial motion only occurs in one direction or in one plane, respectively. This paper presents a method for calculation of the time-resolved three-dimensional (3-D) strain-rate tensor using velocity vector information in a 3-D spatial grid during the whole cardiac cycle. The strain-rate tensor provides full information of both magnitude and direction of the instantaneous deformation of the myocardium. A method for visualization of the full 3-D tensor is also suggested. The tensors are visualized using ellipsoids, which display the principal directions of strain-rate and the ratio between strain-rate magnitude in each direction. The presented method reveals the principal strain-rate directions without a priori knowledge of myocardial motion directions.  相似文献

Summary A new group of low-molecular weight channel-forming oligo(hydroxybutyric acids) (cPHBs, 1 with n = 8–30; main component MW ≈ 1300 dalton) was isolated from microorganisms of different origin. Inclusion bodies were electron-microscopically visible in cells in the state of autolysis, not in cells in the exponential phase of growth. cPHB and high-molecular poly(l3-hydroxybutyric acid) (sPHB) is cleaved by phenylethylamine and forms the corresponding monomeric hydroxybutyramide and – under drastic conditions, the crotylamide. One of these compounds, the 3-hydroxy-N-phenethyl-butyramide (5), was isolated as a new natural product now. A study was conducted at Lake J?ms?nvesi in Central Finland, to identify the potential ecotoxicological risks of the remediation operation of a creosote-/PAH-contaminated lake sediment, made by capping during the years 1998-1999. Mussels (Anodonta anatina) were deployed to the lake at the same time as the remediation operation was started in November 1998. The contaminated area (0.5 ha) was covered by a filter geotextile (polypropylene), gravel and sand (1-1.5m) which were spread out on the ice and let to sink onto the bottom of the lake when the ice melted in May 1999. The possible impacts of capping to the adjacent environment were assessed from mussels exposed and particulate material settled (SPM) to collectors placed on the lake bottom. Mussel tissue, SPM, the water inside the collector were analyzed for polycyclic aromatic compounds (PAH)-compounds. Biological endpoints included body condition, glycogen and protein contents of adductor muscle. Mussels and SPM exposed downstream to the contaminated site (Site 3) contained the highest total PAH concentrations. Biota-sediment accumulation factors of acenaphthene, phenanthrene, anthracene, fluoranthene, pyrene and benzo(a)anthracene of mussels varied from 0.79 to 1.45. The glycogen and protein concentrations were lowest in adductor muscle from mussels exposed to conditions at Site 3. Concentrations of some PAH-compounds were found distinctly increased adjacent to the remediated area, possibly due to the agitation of contaminated sediment due to the capping. It is also possible that resuspension of sediment around remediated area (containing some PAHs) spread the deposited PAH-compounds.  相似文献

The development of non-petrochemical sources for the plastics industry continues to progress as large multinationals focus on renewable resources to replace fossil carbon. Many bacteria are known to accumulate polyoxoesters as water-insoluble granules in the cytoplasm. The thermoplastic and/or elastomeric behaviour of these biodegradable polymers holds promise for the development of various technological applications. Here, we report the synthesis and characterization of microbial polythioesters (PTEs), a novel class of biopolymers of general technological relevance. Biosynthesis of PTE homopolymers was achieved using a recombinant strain of Escherichia coli that expressed a non-natural pathway consisting of a butyrate kinase, a phosphotransbutyrylase, and a PHA synthase. Different homopolymers were produced, consisting of either 3-mercaptopropionate, 3-mercaptobutyrate, or 3-mercaptovalerate repeating units, if the respective mercaptoalkanoic acids were provided as precursor substrates to the fermentative process. The PTEs contributed up to 30% (w/w) of the cellular dry weight and were identified as hydrophobic inclusions in the cytoplasm. The chemical and stereochemical homogeneity of the purified PTEs were identified by different methods, and the estimated physical properties were compared to the oxypolyester equivalents, revealing low crystalline order and, for the poly(3-mercaptopropionate) improved thermal stability. The ability to produce PTEs through a biosynthetic route opens up new avenues in the field of biomaterials.  相似文献

Using the planar waveguide concept in surface acoustic wave (SAW) technology is often advantageous when the modeling of transversely distributed phenomena is indispensable for an accurate design of SAW devices. This is especially true when complex multi-track structures such as transversely coupled resonator filters (TCRFs) are under consideration where, e.g., transverse velocity and stiffness profiles have to be incorporated in the device simulation. The interdigital transducers (IDTs) and the reflector gratings composing those devices behave as planar waveguides, supporting, in principle, all kinds of modes such as bound, semi-bound, and radiation modes. Therefore, to model these SAW propagation effects, we subdivide the SAW structures in transverse direction into several parallel waveguiding channels (N regions), and take, as the wave-describing quantity, a two-dimensional scalar potential function. By doing so, we obtain a complete set of orthonormal modes into which an arbitrary transverse excitation function can be expanded to study its propagation. The general mode spectrum includes a discrete spectrum of bound modes and continuous spectra of semi-bound and radiation modes. We calculate all types of modes by making use of the stack matrix technique. The present work, which arose from the requirement of creating an efficient mathematical tool for the simulation of TCRFs, provides the complete analysis of general SAW multi-channel structures.  相似文献
