Using maltohexaose G6 as a substrate, a convenient analytical method for amylase-catalyzed hydrolytic reaction was developed by application of an enzymatic assay-kit (glucose oxidase-peroxidase) to measurement of glucose, produced on the reaction: G6→G5 + G. The initial velocity was confirmed to be proportional to the initial concentration of amylase, examined, ∼ 100 μm. The new method was found in a good correlation with the Nelson and Somogyi method. Rate assay was also examined: The initial velocity is proportional to the enzyme concentration, ∼ 30 μm. Hence, this enzymatic assay method is recognized to be convenient and useful for the measurement of α-amylase.  相似文献

Using the pH-jump (neutral to above 11) for Tyr-OH ionization as a probe, two glucoamylases from Rhizopus niveus and Rhizopus delemar were found to be caused the change in conformation, which is in a kinetically single step. The pH titration at pHs below 12 was carried out on a multidimensional correlation spectrophotometer. Correlations among the spectrophotometric properties: fluorescence, absorption and circular dichroism are almost identical for the two enzymes. These results suggest that conformational characteristics of the niveus enzyme is almost the same as that of the delemar one.  相似文献

Unpolarized optical spectra were measured in the wavelength range 322–1666 nm by the diffuse reflection technique from spinel powders synthesized in the system MgAl2O4–MgCr2O4. The spectra were interpreted by the crystal-field theory on the basis of trigonally distorted spinel octahedra with D3d symmetry. For chromium-rich solid solutions, including the MgCr2O4 end-member, results after peak fittings showed octahedral D3d local symmetry around Cr3+ ions, identical to the crystallographic site symmetry. For chromium-poor solid solutions, however, octahedral C3v local symmetry was suggested around Cr3+ ions, different from the D3d crystallographically expected.  相似文献

The positional distribution of fatty acids (FA) of triacylglycerols (TAG) and major phospholipids (PL) prepared from four cultivars of peas (Pisum sativum L.) were investigated as well as their tocopherol contents. The lipids extracted from these peas were separated by thin-layer chromatography (TLC) into seven fractions. The major lipid components were PL (52.2–61.3%) and TAG (31.2–40.3%), while the other components were also present in minor proportions (5.6–9.2%). γ-Tocopherol was present in the highest concentration, and α- and δ-tocopherols were very small amounts. The main PL components isolated from the four cultivars were phosphatidylcholine (42.3–49.2%), followed by phosphatidylinositol (23.3–25.2%) and then phosphatidylethanolamine (17.7–20.5%). Small but significant differences (P < 0.05) in FA distribution existed when different pea cultivars were determined. However, the principal characteristics of the FA distribution in the TAG and the three PL were evident among the four cultivars; unsaturated FA were predominantly located in the sn-2 position, and saturated FA primary occupied the sn-1 or sn-3 position in the oils of the peas. These results suggest that the regional distribution of tocopherols and fatty acids in peas is not dependent on the climatic conditions and the soil characteristics of the cultivation areas during the growing season.  相似文献

Endogenous tocochromanols in extracted lipids from rice brans of the five cultivars were determined by high-performance liquid chromatography, and were investigated in relation to the fatty acid (FA) distribution of triacylglycerols (TAG) and phospholipids (PL). The dominant tocols were α-tocopherol and γ-tocotrienol, followed by α-tocotrienol and with much smaller amounts of γ-tocopherol and δ-tocotrienol. The lipids of these rice brans comprised mainly TAG (80.6–86.0 wt.%), free FA (4.2–9.0 wt.%), and phospholipids (5.5–6.7 wt.%), whilst other components were also detected in minor proportions (0.2–2.1 wt.%). The PL components included phosphatidyl choline (31.8–46.8 wt.%), phosphatidyl ethanolamine (25.0–38.9 wt.%) and phosphatidyl inositol (20.2–23.2 wt.%). Comparison of these different cultivars showed, with a few exceptions, no significant differences (P > 0.05) in FA distribution. FA distribution of TAG among the five cultivars was evident in the rice brans: unsaturated FA were predominantly concentrated at the sn-2 position and saturated FA primarily occupying the sn-1 or sn-3 position. These results suggest that the tocopherol content, lipid component, and FA distribution in rice brans are not dependent on the cultivation areas during the growing season.  相似文献

In this paper, a novel control method for a matrix converter is proposed. The proposed method is a PAM (Pulse Amplitude Modulation) control method based on a virtual AC/DC/AC conversion method. Output voltage amplitude is controlled by controlling a virtual DC link voltage with a virtual rectifier. Output frequency is controlled by a virtual inverter. First, the proposed method is explained. Next, the validity of the proposed method is confirmed through simulation and experiment using a 750‐W prototype matrix converter. Moreover, various characteristics of the proposed method and conventional virtual AC/DC/AC method are compared through experiment. As a result of the experiment, it has been understood that total harmonic distortion of output current and common‐mode voltage can be reduced compared with the case of using a conventional virtual AC/DC/AC conversion method. © 2008 Wiley Periodicals, Inc. In a sample of 263 male GO players at 48 levels of expertise and ranging from 18 to 78 years of age, it was found that factors of expertise deductive reasoning (EDR) and expertise working memory (EWM) were independent of factors of fluid reasoning (Gf) and short-term working memory (STWM) that, along with cognitive speed (Gs), have been found to characterize decline of intelligence in adulthood. The main effects of analyses of cross-sectional age differences indicated age-related decline in EDR and EWM as well as in Gf, STWM, and Gs. However, interaction and partialing analyses indicated that decline in EDR and EWM decreased to no decline with increase in level of expertise. The results thus suggest that with increase in factors known to raise the level of expertise—particularly, intensive, well-designed practice there may be no age-related decline in the intelligence that is measured in the abilities of expertise.
