InlA is a surface protein participating in the entry of Listeria monocytogenes into mammalian non-phagocytic cells. PrfA is a positive regulatory factor that regulates the expression of a set of virulence genes. Recent studies revealed that some L. monocytogenes strains have a truncated form of these proteins because of nonsense mutations in their sequences, and these truncations contribute to the significant reduction in virulence of this pathogen. In this study, sequence analyses of inlA and prfA among L. monocytogenes isolated from ready-to-eat seafood revealed that only one out of 59 isolates had a nonsense-mutated inlA and all had non-mutated prfA. This indicated that these strains could be fully virulent based on the sizes of these proteins. 相似文献

Sugarcane bagasse is a useful biomass resource. In the present study, we examined the efficacy of ammonia pretreatment for selective release of hemicellulose from bagasse. Pretreatment of bagasse with aqueous ammonia resulted in significant loss of xylan. In contrast, pretreatment of bagasse with anhydrous ammonia resulted in almost no xylan loss. Aqueous ammonia or anhydrous ammonia-pretreated bagasse was then subjected to enzymatic digestion with a xylanase from the glycoside hydrolase (GH) family 10 or a xylanase from the GH family 11. The hydrolysis rate of xylan in bagasse pretreated with aqueous ammonia was approximately 50 %. In contrast, in the anhydrous ammonia-treated bagasse, xylan hydrolysis was > 80 %. These results suggested that anhydrous ammonia pretreatment would be an effective method for preparation of sugarcane bagasse for enzymatic hydrolysis to recover xylooligosaccharides. 相似文献

A novel liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed for the trace residue determination of vedaprofen (VPF) in livestock products and seafoods. VPF was extracted from each sample with acidified acetone, and the crude extract was re-extracted with ethyl acetate and NaCl solution. Clean-up was performed using a weak anion exchange cartridge (Bond Elut DEA). The LC separation was performed on a C18 column using acetonitrile-0.0025 mol/L formic acid (3 : 2) as the mobile phase and MS was run in the negative ion electrospray ionization mode. The calibration curve was linear in the range of 0.001-0.1 μg/mL VPF. The mean recoveries from equine muscle, cattle muscle, cattle liver, cattle fat, salmon, eel, corbicula, milk, egg and buckwheat honey were 72-94%, and the relative standard deviations (RSDs) were 1.1-2.0%. The tests on fission product (FP) behavior in piping under severe accidents are being conducted in the wide range piping integrity demonstration (WIND) project at JAERI to investigate the piping integrity which may be threatened by decay heat from deposited FPs. In order to obtain the background information for future WIND experiment and to confirm analytical capabilities of the FP aerosol analysis codes, ART and VICTORIA, the FP behavior in safety relief valve (SRV) line of BWR during TQUX sequence was analyzed. The analyses showed that the mechanisms that control the FP deposition and transport agreed well between the two codes. However, the differences in models such as diffusiophoresis or turbulence, the treatment of chemical forms and aerosol mass distribution could affect the deposition in piping and, consequently, on the source terms. The WIND experimental analyses were also conducted with a three-dimensional fluiddynamic WINDFLOW, ART and an interface module to appropriately couple the fluiddynamics and FP behavior analyses. The analyses showed that the major deposition mechanism for cesium iodide (CsI) is thermophoresis which depends on the thermal gradient in gas. Accordingly, the coupling analyses were found to be essential to accurately predict the CsI deposition in piping, to which little attention has been paid in the previous studies. 相似文献

The energy resolved computed tomography (CT), which had advantage over conventional CT (twofold higher CT value for iodine contrast agent and being free from beam hardening effect), was shown practical by employing the transXend detector: it measured X-rays as electric current and gave energy distribution of incident X-rays after analysis. This article shows a new application of the transXend detector for estimating the thicknesses of acrylic, iodine, and aluminum in a phantom. For this purpose, the responses of the segment detectors in the transXend detector are changed intentionally with inserting filters. With previously obtained two-dimensional maps for acrylic–iodine and acrylic–aluminum thicknesses, which are shown by the ratios of electric currents measured by the segment detectors, the thickness of materials on the path of the X-rays are obtained by a transmission measurement. 相似文献

Creep rupture data and microstructural degradation during aging of high Cr ferritic boiler steels with enhanced creep strength have been studied with special attention to prediction of long term creep rupture life. Tempered lath martensite structure in the high Cr ferritic steels remains unchanged during short term aging, whereas static recovery of the lath martensite structure proceeds when diffusion distance during aging becomes sufficiently long as is the case in long term creep. The static recovery brings about premature failure in long term creep and decreases in apparent activation energy for creep life. The decrease in activation energy is responsible for overestimation of rupture life reported in strength enhanced high Cr ferritic steels. The boundary from a short term region with high activation energy QH to a long term region with low activation energy QL moves towards longer time with decreasing Cr concentration. The difference in activation energy (QH − QL) primarily determines the extent of overestimation of rupture life predicted from short term data. In general, the extent of overestimation is less serious at 9%Cr as compared to 12%Cr. 相似文献

An international joint research has been conducted to investigate light degradation of low-resistivity Si CZ wafers and also to provide practical solutions to suppress the degradation. Ten kinds of CZ, MCZ and FZ Si wafers were evaluated under AM1.5 irradiation and processed to fabricate solar cells using low- and high-temperature processes. Lifetime degradation was suppressed using MCZ Si wafers with low oxygen content and Ga-doped CZ wafers with high oxygen content. In addition, high-temperature oxidation was also effective to reduce light degradation remarkably. No degradation of solar cells could be realized by using B-doped MCZ and Ga-doped CZ wafers combined with a high-temperature processing. 相似文献

An experimental study was conducted to investigate transient local heat transfer around a bubble at onset of boiling on a thin glass heating plate immersed in saturated n-hexane at low pressure. Eight rapid response Cu-Ni thermocouples consisting of a vacuum deposited thin film were used to measure the temperature change of the heating surface. Simultaneous high-speed video photographs were also obtained. The surface temperatures near a nucleation site decreased rapidly owing to the evaporation of a thin layer (microlayer) of liquid formed beneath the bubble in the early period and the rate of bubble growth increased with increasing incipient boiling superheat (ΔTIB). To determine a suitable initial water-soluble carbohydrate (WSC) content to make wheat straw natural fermentation successful and to study fermentation characteristics, glucose was used to adjust the initial WSC content to 1.4%, 4.0%, 5.0%, 6.0%, 7.0%, 8.0%, 9.0%, and 10.0% dry matter (DM) in the wheat straw. At 30 d of fermentation, there were three pHs: when the initial WSC content was 1.4%, the pH was 5.5; when the initial WSC contents were 4.0%, 5.0% and 6.0%, the pHs were near 5.1; and when the initial WSC contents were 7.0%, 8.0%, 9.0% and 10.0%, the pHs were near 4.0. The pattern of changes in WSC content during the fermentation was similar to that in pH. At 30 d of fermentation, there existed a dividing line in WSC remnants between the initial 6.0% WSC treatment and the initial 7.0% WSC treatment. When the initial WSC content was more than 7.0%, the remaining WSC content was more than 23.7 g/kg DM. When the initial WSC content was less than 6.0%, the remaining WSC content was less than 13.6 g/kg DM. Particularly for the 1.4% WSC treatment, the remaining WSC content was 2.1 g/kg DM. The results of the microbiological enumeration showed that with the increase in initial WSC content, the numbers of lactic acid bacteria (LAB) and other bacteria generally decreased. Denaturing gradient gel electrophoresis (DGGE) results showed that when the initial WSC content was beyond 7.0%, the LAB of the fermentation system were detected. 相似文献
