Single crystals of hydroxyapatite were grown by a flux method using Ca3(PO4)2 and Ca(OH)2 under 100 MPa of Ar gas. The crystals obtained had stoichiometric composition of Ca10(PO4)6(OH)2 and some of them were twin-free single crystals. From X-ray diffraction analyses, the space group was confirmed to be monoclinic P21/b with cell parameters a=0.9419(3) nm, b=1.8848(6) nm, c=0.6884(2) nm, and =119.98(2)°. The detailed crystal structure was determined with a reliability factor Rw=0.033; the O atoms of OH were located just on the 21 axis while the H atoms of OH occupied the positions a little deviated from the 21 axis. The origin of this structure was ascribed to the formation of hydrogen bonds between the H atoms of the OH ions and the specific O atoms of the PO4 ions.  相似文献

Gamma irradiation treatment (GIT) is the most widely used sterilization method for biomaterials in spite of the potential to degrade polymers. The adverse effects of GIT on the mechanical properties and biological stabilities of porous hydroxyapatite/collagen (HAp/Col) composites were investigated in this study. Those properties of a porous HAp/Col composite stabilized by dehydrothermal treatment (DHT) drastically decreased by GIT in the same manner as conventional pure Col materials. DHT after GIT could partially eliminate the adverse effects, due to the introduction of cross-linkage among the degraded Col molecules.  相似文献

Threshold photoelectron spectra of N2+ were measured between 23.4 and 27.6 eV with high resolution and high intensity by using the penetrating field technique and synchrotron radiation. Five vibrational progressions were observed. The first of these progressions was the C2Sigmau+ state. The second progression was identified as the transition to the second state of 2Pig symmetry found by P. Baltzer, M. Larsson, L. Karlsson, B. Wannberg, and M. Carlsson (1992. Phys. Rev. A 46, 5545). The third progression, which was discovered by F. Merkt and P. M. Guyon (1993. J. Chem. Phys. 99, 3400), can be designated as the 2Sigmau- state by comparison with previous theoretical study (E. W. Thulstrup and A. Andersen, 1975. J. Phys. B 8, 965). The fourth and fifth progressions were designated as the 2Deltau and 2(2) Piu states by similar comparison with previous theories. DNA methylation of both viral and host DNA is one of the major mechanisms involved in the development of Epstein–Barr virus-associated gastric carcinoma (EBVaGC); thus, epigenetic treatment using demethylating agents would seem to be promising. We have verified the effect of MC180295, which was discovered by screening for demethylating agents. MC180295 inhibited cell growth of the EBVaGC cell lines YCCEL1 and SNU719 in a dose-dependent manner. In a cell cycle analysis, growth arrest and apoptosis were observed in both YCCEL1 and SNU719 cells treated with MC180295. MKN28 cells infected with EBV were sensitive to MC180295 and showed more significant inhibition of cell growth compared to controls without EBV infection. Serial analysis of gene expression analysis showed the expression of genes belonging to the role of BRCA1 in DNA damage response and cell cycle control chromosomal replication to be significantly reduced after MC180295 treatment. We confirmed with quantitative PCR that the expression levels of BRCA2, FANCM, RAD51, TOP2A, and CDC45 were significantly decreased by MC180295. LMP1 and BZLF1 are EBV genes with expression that is epigenetically regulated, and MC180295 could up-regulate their expression. In conclusion, MC180295 inhibited the growth of EBVaGC cells by suppressing DNA repair and the cell cycle.  相似文献

Defects in Bruton's tyrosine kinase (Btk) result in B cell immunodeficiencies in humans and mice. Recent studies showed that Btk is required for maximal activation of JNK, a family of stress-activated protein kinases, induced by several extracellular stimuli including interleukin (IL)-3. On the other hand, IL-3-induced JNK activation is dependent on Ras. In the present study we have investigated whether Ras is involved in Btk-mediated JNK activation in BaF3 mouse pro-B cells. Overexpression of wild-type Btk protein in these cells enhanced JNK activation upon IL-3 stimulation, whereas expression of kinase-dead Btk partially suppressed JNK activation. Induced expression of the dominant negative Ras(N17) in the cells overexpressing wild-type Btk suppressed JNK activation. Importantly, overexpression of Btk enhanced the level of the GTP-bound, active form of Ras in response to IL-3 stimulation. Btk overexpression also increased the Shc-Grb2 association induced by IL-3 stimulation. Expression of either N17Ras or V12Ras did not impose any effects on Btk kinase activity. These data collectively indicate that Ras plays a role of an intermediary signaling protein in Btk-mediated JNK activation induced by the IL-3 signaling pathway.  相似文献

BACKGROUND: The authors analyzed the radiation dose to the periphery of the cervix and area of the cervix in relation to local failure of radiotherapy for carcinoma of the cervix using computed tomography (CT) images. METHODS: Between 1981-1990, 127 consecutive patients were treated with definitive radiotherapy. Ninety-nine of these patients had CT images taken at the time of intracavitary therapy. Of these 99 patients, 80 were eligible for this analysis. After CT scanning, isodose curves relative to the point A dose were superimposed on the CT images. The minimum percent dose and minimum dose at the periphery of the cervix were estimated. The area of the cervix also was measured. These factors were examined in relation to the local tumor control rate. RESULTS: Histograms of both the minimum percent dose and the cervical area showed significant differences between the local control and local failure groups (P <0.001). The local control rates were related to both the minimum percent dose and the cervical area, and differed significantly over and below the values of 60% and 18 cm2 (P <0.001 each), respectively. The local control patients, over and below the line: Y = -0.220X + 21.2, in which X (gray [Gy]) and Y (Gy) are the whole pelvis dose and the minimum dose, respectively, could be well differentiated with significance (91.7% vs. 25.0%; P <0.001). CONCLUSIONS: Computed tomography analysis indicated that the local tumor control rate was related strongly to the minimum percent dose, the cervical area, and the pair of whole pelvis and minimum dose values. These factors were found to be more useful than the point A dose in predicting local tumor control.  相似文献

Vascular endothelial growth factor (VEGF) is one of the angiogenic factors. We examined both thyroid volume and intrathyroidal vascular area by color flow Doppler ultrasonography in patients with Graves' disease (GD), Hashimoto's thyroiditis (HT), and subacute thyroiditis. The serum concentrations of thyroid hormones, TSH, TSH receptor antibodies, and VEGF were also examined. There was a significant increase in serum VEGF levels in patients with untreated GD and goitrous HT compared with those in healthy subjects. The serum VEGF levels in untreated patients with subacute thyroiditis were significantly higher than those in patients with untreated GD or HT. There was a significant correlation between serum VEGF levels and the ratio of intrathyroidal vascular area and thyroid area in untreated patients with GD who had a goiter larger than or equal to 40 cm3. There was also a significant correlation between serum VEGF and TSH levels in patients with HT who were hypothyroid and had a goiter. Serum VEGF levels decreased significantly in these patients after treatment; this was accompanied by a significant decrease in intrathyroidal vascular area and thyroid volume. Our study demonstrates that VEGF appears to play an important role in intrathyroidal angiogenesis in patients with GD and goitrous HT.  相似文献

For use as electrical and electronics parts, or automobile and mechanical parts, toughened poly(phenylene sulfide) (PPS) is desired. For these applications, our investigation centered on improving the toughness of PPS and developing elastomer-toughened PPS and elastomer-toughened compounds of PPS. Using chemically treated PPS and an olefinic elastomer with a functional group, we developed elastomer-toughened PPS using a reactive processing method. In the PPS matrix, the elastomer is finely dispersed. While the notched Izod impact strength of the original PPS is about 1 kg · cm/cm. clastomer-toughened PPS has a notched impact strength around 50 kg · cm/cm. The notched fracture surface of elastomer-toughened PPS is observed using a scanning electron microscope. We concluded that the mechanism for the toughening is attributed to energy dissipation by matrix yield.  相似文献
