Myocardial infarction is one of the major causes of mortality worldwide and is a main cause of heart failure. This disease appears as a final point of atherosclerotic plaque progression, destabilization, and rupture. As a consequence of cardiomyocytes death during the infarction, the heart undergoes unfavorable cardiac remodeling, which results in its failure. Therefore, therapies aimed to limit the processes of atherosclerotic plaque progression, cardiac damage during the infarction, and subsequent remodeling are urgently warranted. A hopeful therapeutic option for the future medicine is targeting and regulating non-coding RNA (ncRNA), like microRNA, circular RNA (circRNA), or long non-coding RNA (lncRNA). In this review, the approaches targeted at ncRNAs participating in the aforementioned pathophysiological processes involved in myocardial infarction and their outcomes in preclinical studies have been concisely presented.  相似文献

Atherosclerosis and nonalcoholic fatty liver disease are leading causes of morbidity and mortality in the Western countries. The renin–angiotensin system (RAS) with its two main opposing effectors, i.e., angiotensin II (Ang II) and Ang-(1–7), is widely recognized as a major regulator of cardiovascular function and body metabolic processes. Angiotensin-converting enzyme 2 (ACE2) by breaking-down Ang II forms Ang-(1–7) and thus favors Ang-(1–7) actions. Therefore, the aim of our study was to comprehensively evaluate the influence of prolonged treatment with ACE2 activator, diminazene aceturate (DIZE) on the development of atherosclerotic lesions and hepatic steatosis in apoE−/− mice fed a high-fat diet (HFD). We have shown that DIZE stabilized atherosclerotic lesions and attenuated hepatic steatosis in apoE−/− mice fed an HFD. Such effects were associated with decreased total macrophages content and increased α-smooth muscle actin levels in atherosclerotic plaques. Moreover, DIZE changed polarization of macrophages towards increased amount of anti-inflammatory M2 macrophages in the atherosclerotic lesions. Interestingly, the anti-steatotic action of DIZE in the liver was related to the elevated levels of HDL in the plasma, decreased levels of triglycerides, and increased biosynthesis and concentration of taurine in the liver of apoE−/− mice. However, exact molecular mechanisms of both anti-atherosclerotic and anti-steatotic actions of DIZE require further investigations.  相似文献

Summary A study on the copolymerization of glycolide with lactide and glycolide with ɛ-caprolactone was performed in the presence of zirconium (IV) acetylacetonate at moderate temperatures (100° and 150°C). Zirconium acetylacetonate appeared to be an efficient initiator of copolymerization. The obtained polymers were characterized by high molecular weights. The analytic structure of Rational Interpolants (R.I.) f (z) built from randomly perturbed data is explored; the interpolation nodes x j , j = 1,...,M, are real points where the function f reaches these prescribed data . It is assumed that the data are randomly perturbed values of a rational function (n) (m) (m / n is the degree of the numerator/denominator). Much attention is paid to the R.I. familyf (n+1) (m–1), in the small stochasticity régime. The main result is that the additional zero and pole are located nearby the root of the same random polynomial, called the Froissart Polynomial (F.P.). With gaussian hypothesis on the noise, the random real root of F.P. is distributed according to a Cauchy-Lorentz law, with parameters such that the integrated probability over the interpolation interval x 1, x M is always larger than 1/2; in two cases studied in detail, it reaches 2/3 in one case and almost 3/4 in the other. For the families f (n+k) (m+k), numerical explorations point to similar phenomena; inspection shows that, in the mean, the localization occurs in the complex and/or real vicinity of the interpolation interval.  相似文献

Understanding how forces propagate in granular assemblages is important for equipment design and process control in many technologies. Yet, it remains poorly understood. In this study, a cuboidal assembly comprising cylinders of various lengths (aspect ratios AR ranging from 0.9 to 3.6) were subjected to uniaxial confined compression tests. Samples were vertically compressed until the top platen exerted a pressure of 50 kPa on the uppermost particles. This maximum pressure corresponds to the hydrostatic pressure of an approximately 15 m high column of chopped wood that may be encountered in real storage structures. The nonlinear loading curves were obtained depended on the aspect ratios of the cylinders. The modulus of elasticity, calculated from the linear elastic part of the stress–strain curve, monotonically decreased from 10.2 to 8.6 MPa as the aspect ratio increased from 1.2 to 3.6. The elastic modulus and volume fraction exhibited similar trends as functions of the aspect ratio. The horizontal-to-vertical pressure ratio was calculated as the horizontal pressure exerted on the wider walls to the vertical pressure exerted on the top lid during loading–unloading cycles. For ARs up to 3.6, the pressure ratio was approximately 0.31; for the longest cylinders (AR = 3.6), it decreased to approximately 0.27, probably because the assumption of the representative chamber volume was invalidated at this AR.  相似文献

The paper provides comparison of three different approaches to on-line tuning of generalized adaptive notch filters (GANFs) — the algorithms used for identification/tracking of quasi-periodically varying dynamic systems. Tuning is needed to adjust adaptation gains, which control tracking performance of GANF algorithms, to the unknown and/or time time-varying rate of system nonstationarity. Two out of three compared approaches are classical solutions — the first one incorporates sequential optimization of adaptation gains while the second one is based on the concept of parallel estimation. The main contribution of the paper is that it suggests the third way — it shows that the best results can be achieved when both approaches mentioned above are combined in a judicious way. Such joint sequential/parallel optimization preserves advantages of both treatments: adaptiveness (sequential approach) and robustness to abrupt changes (parallel approach). Additionally the paper shows how, using the concept of surrogate outputs, one can extend the proposed single-frequency algorithm to the multiple frequencies case, without falling into the complexity trap known as the “curse of dimensionality”.  相似文献

The paper discusses the problem of stress and strain concentration in the entry and exit planes during the drawing process. Earlier investigations carried out by Pierlin, Pawelski and Armstroff as well as Chen and Kobayashi's finite element analysis are reviewed. Authors’ calculations based on the rigid-plastic finite element approach are compared with experimental and theoretical results published by Pawelski and Armstroff. Solution for rigid-plastic material behaviour confirmed the die pressure increase in the entry and exit zones questioning Pierlin's suggestion that the elastic strains are responsible for pressure concentrations. Analysis of the strain rate distribution led to the presumption that pressure increase is due to the strain rate concentrations in the areas close to the entry and exit zones.  相似文献

The development of methods to engineer and immobilize amine transaminases (ATAs) to improve their functionality and operational stability is gaining momentum. The quest for robust, fast, and easy-to-use methods to screen the activity of large collections of transaminases, is essential. This work presents a novel and multiplex fluorescence-based kinetic assay to assess ATA activity using 4-dimethylamino-1-naphthaldehyde as an amine acceptor. The developed assay allowed us to screen a battery of amine donors using free and immobilized ATAs from different microbial sources as biocatalysts. As a result, using chromatographic methods, 4-hydroxybenzylamine was identified as the best amine donor for the amination of 5-(hydroxymethyl)furfural. Finally, we adapted this method to determine the apparent Michaelis-Menten parameters of a model immobilized ATA at the microscopic (single-particle) level. Our studies promote the use of this multiplex, multidimensional assay to screen ATAs for further improvement.  相似文献
