The germ cell lineage in mice is established about a week after fertilization, in a group of cells that have left the epiblast and moved to an extraembryonic site. They migrate back into the embryo, along the hind gut and into the gonads. Germ cells in male and female embryos then pursue different pathways: in the testis the germ cells cease proliferating and enter mitotic arrest, while germ cells in the ovary, like those in male embryos that remain outside the gonads, enter meiotic prophase. Studies on explanted germ cells suggest that all germ cells may enter meiosis at a certain stage of their development, unless prevented from doing so by some inhibitory influence of the testis. Germ cells during the migratory stage can be cultured, but do not enter meiosis unless embedded in somatic tissue. Addition of certain growth factors and cytokines to the culture medium allows germ cells to proliferate indefinitely in vitro: Like embryonic stem cells, these immortalized EG (embryonic germ) cells will colonize all cell lineages if introduced into a blastocyst. After birth, germ cells undergo gametogenesis; oogenesis in the female, spermatogenesis in the male. Brinster and his colleagues have shown that spermatogonial stem cells injected into a germ-cell depleted testis will repopulate the seminiferous tubules and undergo spermatogenesis, giving rise to functional spermatozoa. Stem cells from frozen testicular tissue are still capable of giving rise to spermatogenesis in a host testis. Rat testicular tissue can undergo spermatogenesis in a mouse testis, to form morphologically normal rat spermatozoa, even though the Sertoli cells that support them are of endogenous mouse origin. These findings are of fundamental importance for our understanding of spermatogenesis and the interactions between germ cells and Sertoli cells; but they also have significant practical implications, in relation to both agricultural practice and clinical treatment of infertility.  相似文献

Cardiovascular (CV) calcification is known as sub-clinical atherosclerosis and is recognised as a predictor of CV events and mortality. As yet there is no treatment for CV calcification and conventional CV risk factors are not consistently correlated, leaving clinicians uncertain as to optimum management for these patients. For this reason, a review of studies investigating diet and serum levels of macro- and micronutrients was carried out. Although there were few human studies of macronutrients, nevertheless transfats and simple sugars should be avoided, while long chain ω-3 fats from oily fish may be protective. Among the micronutrients, an intake of 800 μg/day calcium was beneficial in those without renal disease or hyperparathyroidism, while inorganic phosphorus from food preservatives and colas may induce calcification. A high intake of magnesium (≥380 mg/day) and phylloquinone (500 μg/day) proved protective, as did a serum 25(OH)D concentration of ≥75 nmol/L. Although oxidative damage appears to be a cause of CV calcification, the antioxidant vitamins proved to be largely ineffective, while supplementation of α-tocopherol may induce calcification. Nevertheless other antioxidant compounds (epigallocatechin gallate from green tea and resveratrol from red wine) were protective. Finally, a homocysteine concentration >12 µmol/L was predictive of CV calcification, although a plasma folate concentration of >39.4 nmol/L could both lower homocysteine and protect against calcification. In terms of a dietary programme, these recommendations indicate avoiding sugar and the transfats and preservatives found in processed foods and drinks and adopting a diet high in oily fish and vegetables. The micronutrients magnesium and vitamin K may be worthy of further investigation as a treatment option for CV calcification.  相似文献

In a series of experiments using the serial reaction time paradigm, the authors compared the predictions of a powerful associative model of sequence learning (the simple recurrent network; J. L. Elman, 1990) with human performance on the problem devised by A. Maskara and W. Noetzel (1993). Even though the predictions made by the simple recurrent network for variants of this problem are often counterintuitive, they matched human performance closely, suggesting that performance was associatively based rather than rule based. Simple associative chaining models of sequence learning, however, have difficulty in accommodating these results. The authors' conclusion is that, under the conditions of the experiments, human sequence learning is associatively driven, as long as this is understood to mean that a sufficiently powerful means of extracting the statistical regularities in the sequences is in play. A double-blind placebo-controlled trial of intravenous prostaglandin PGE1 (40 micrograms/h) was conducted in adult orthotopic liver transplant recipients. Infusion was started intraoperatively and continued for up to 21 days. Patients were followed up for 180 days postoperatively. Among 172 patients eligible for treatment in the study, 160 could be evaluated (78 PGE1; 82 placebo). Patient and graft survival were similar (PGE1: 16 deaths, 9 retransplantations [7 survivors]; controls: 15 deaths, 6 retransplantations [3 survivors]). In patients with surviving grafts, however, PGE1 administration resulted in a 23% shorter mean duration of hospitalization following transplantation (PGE1: 24.4 days; controls: 31.8 days; P = .02) and a 40% shorter length of time postoperatively in the intensive care unit (PGE1: 8.2 days; controls 13.7 days; P = .05). Reduced needs for renal support (P = .03) or surgical intervention other than retransplantation (P = .02) were also noted with PGE1 use. Further, PGE1 administration resulted in a trend toward improved survival rates in patients with mild renal impairment (preoperative serum creatinine 1.5 mg percent or greater; P = .08). Neither the incidence of acute cellular rejection nor of primary nonfunction was significantly different in the two groups. Phlebitis was the only complication that was more common during PGE1 administration, (PGE1: 9; controls: 4). These results suggest that PGE1 use in hepatic allograft recipients reduces morbidity and may result in sizable cost reductions.  相似文献

BACKGROUND: Experimental studies have shown that endothelial dysfunction is an early event preceding restenosis. Monocytes and neutrophils have been shown to bind to damaged endothelium via the cell adhesion molecules (CAMs). The selectins are involved in capturing the leukocytes and tethering them to the endothelium. E-selectin is a CAM that is only expressed on activated endothelial cells. Its ligands are expressed on monocytes and neutrophils and it has been found to exist in a soluble form. This soluble form may represent a marker for endothelial damage and may be a precursor of smooth muscle proliferation. METHODS AND RESULTS: Fifty-four patients who were undergoing peripheral arterial balloon angioplasty had blood sampled before angioplasty. E-selectin was measured in plasma with the use of an ELISA. At follow-up angiogram, 30% (n=14) of the patients had restenosed at 1 year. There was a significant difference in baseline E-selectin levels in patients who restenosed compared with those who did not (65.3 ng/mL [58.25 to 78.05] versus 52.3 [34.2 to 62.1], Mann-Whitney U, P<.007). Endothelial activation with subsequent adherence of white blood cells is an important step in restenosis. CONCLUSIONS: We have shown an increased level of shed E-selectin in patients destined for restenosis and suggest that this work further supports a role for white blood cell/endothelial interaction in restenosis after angioplasty.  相似文献

We have measured the xenon near infrared (NIR) scintillation yield in the wavelength range from about 700 to 850 nm for gas pressures from 0.3 to 1.5 atm. We have found the NIR yield to be nearly constant above 0.9 atm, increasing by about a factor of 4.5 as pressure is lowered to 0.3 atm. Measured in number of photons emitted we estimate that at 0.3 atm the NIR yield is 45% of the ultraviolet yield near 170 nm. The NIR yield may be sufficient to form the basis of detector systems utilizing low-pressure xenon gas.  相似文献

An epornitic of conjunctivitis in free-flying house finches (Carpodacus mexicanus) occurred in several mid-Atlantic and eastern states of the USA in 1994. Clinical signs and gross lesions ranged from mild to severe unilateral or bilateral conjunctival swelling with serous to mucopurulent drainage and nasal exudate. Microscopic lesions consisted of chronic lymphoplasmacytic conjunctivitis, rhinitis, and sinusitis. Notably slow-growing mycoplasmas were isolated from conjunctival and/or infraorbital sinus swabs from clinically affected birds. Isolates were identified as Mycoplasma gallisepticum (MG) by direct immunofluorescence and DNA probe-based polymerase chain reactions. These findings suggest that MG is the likely etiology for this epornitic of conjunctivitis in house finches.  相似文献

Tested whether referential communication training of young speakers can be generalized to novel tasks. 19 1st graders were taught to use 2 general communication rules through a verbal self-regulation procedure. Three groups were tested: a practice control and 2 training groups. In the 1st training group, Ss were trained on 2 tasks. In the 2nd group, Ss received the same training but in only 1 context. Results show that both training groups performed better than the controls on both direct training posttests and novel generalization tasks, immediately as well as 2 wks later. However, there were no significant differences between the 2 training groups. It is concluded that verbal self-regulation training appears to be a promising means of obtaining generalizaion of speaking skills in young children, in support of findings by D. Meichenbaum and J.
