Penile cancer (PeC) is a rare disease, and no prognostic biomarkers have been adopted in clinical practice yet. The objective of the present study was to identify differentially expressed miRNAs (DEmiRs) and genes (DEGs) as potential biomarkers for lymph node metastasis and other prognostic factors in PeC. Tumor samples were prospectively obtained from 24 patients with squamous cell carcinoma of the penis. miRNA microarray analysis was performed comparing tumors from patients with inguinal lymph node metastatic and localized disease, and the results were validated by qRT-PCR. Eighty-three gene expression levels were also compared between groups through qRT-PCR. Moreover, DEmiRs and DEGs expression levels were correlated with clinicopathological variables, cancer-specific (CSS), and overall survival (OS). TAC software, TM4 MeV 4.9 software, SPSS v.25.0, and R software v.4.0.2 were used for statistical analyses. We identified 21 DEmiRs in microarray analysis, and seven were selected for validation. miR-744-5p and miR-421 were overexpressed in tissue samples of metastatic patients, and high expression of miR-421 was also associated with lower OS. We found seven DEGs (CCND1, EGFR, ENTPD5, HOXA10, IGF1R, MYC, and SNAI2) related to metastatic disease. A significant association was found between increased MMP1 expression and tumor size, grade, pathological T stage, and perineural invasion. Other genes were also associated with clinicopathological variables, CSS and OS. Finally, we found changes in mRNA–miRNA regulation that contribute to understanding the mechanisms involved in tumor progression. Therefore, we identified miRNA and mRNA expression profiles as potential biomarkers associated with lymph node metastasis and prognosis in PeC, in addition to disruption in mRNA–miRNA regulation during disease progression.  相似文献

A measurement technique is developed to measure the characteristics of zinc oxide (ZnO) microvaristors directly. We used Cu wires, a micromanipulator, and an optical microscope to gain ohmic contact of the measurement apparatus with ZnO particles. With our system, the IV characteristics and C−2V characteristics were measured for 25 µm ZnO microvaristors. Our measurements show that these microparticles behave as varistors. Further measurement on the ZnO microvaristors and observation of their fine structure may help to understand the varistor behavior. It also can pave the way to the understanding of the electrical characteristics of grain boundaries as well as the double Schottky barrier formed at the grain boundaries. © 2011 Institute of Electrical Engineers of Japan. The structure and some physicochemical properties of endosperm starches from seven amylose-extender (ae) and two waxy (wx) alleles of maize (Zea mays L.) were studied. Starches prepared from mature kernels of six ae mutants, Oh43 inbred line ae (standard ae), ae-RWB-2 and ae-RWB-3, and W23 × L317 hybrid line ae-PP, ae-Bol 561 and ae-emll, were uniquely ae type, as was concluded from B type X-ray diffractograms; high gelatinization temperatures determined by differential scanning calorimetry (DSC); with poor starch-granule digestibility to amylase; high amylose (37—45 %) and high intermediate fraction (13—18 %) contents; and low ratios (1.0—1.2) of long α-1,4-chains to short α-1,4-chains of amylopectin determined by gel permeation chromatography (GPC) of isoamylase-debranched starches. The results also indicated that different ae alleles had different effects on the amylose content of endosperm starches. Mature kernels of Oh43 ae-RWB-1 mutant showed tarnished and translucent phenotype characteristics of the ae genotype but contained endosperm starch with 21—22 % of amylose which was lower than that of the Oh43 normal counterpart. Both wx-B and wx-C (standard wx) genes have similar effect on structure and physicochemical properties of waxy starches of A632, B37, C105, Oh43, and W64A inbred lines.  相似文献
