A new continuous processing method is presented for the production of continuous-fiber-reinforced metal matrix composite (MMC) wires. The process currently yields MMC wires with the diameter of 0.5–1.6 mm at a maximum speed of 0.3 m/s for (fiber)–(metal) combinations of (Al2O3, Si–Ti Carbide, SiC, Carbon)–(Aluminum, 2024A1, 6061A1) with a fiber volume fraction of 0.50–0.65. Oral administration of lactobacilli as probiotics is gaining importance in the treatment of intestinal inflammations. We investigated the effect of non-starter lactobacilli Lactobacillus casei subsp casei 2756, Lactobacillus curvatus 2775, and Lactobacillus plantarum 2142 as well as their spent culture supernatants (SCS) on Salmonella enteritidis 857 growth, interleukin (IL)-8 and heat shock protein 70 (Hsp70) synthesis in undifferentiated crypt-like and differentiated villus-like Caco-2 cells. The cells were infected with graded numbers of non-starter lactobacilli or S. enteritidis 857 for 1 h and allowed to recover for 24 h or exposed to 200 bacteria/cell for 1 h and allowed to recover for different periods of time. In another experiment S. enteritidis 857 was first pre-treated with SCS-lactobacilli for 1 h before infecting the cells. The levels of IL-8 and Hsp70 were assessed using sandwich ELISA and immunostaining of Western blots, respectively. The effect of SCS-lactobacilli on S. enteritidis 857 growth was evaluated by agar plate diffusion test. The non-starter lactobacilli induced a significant increase in the levels of both IL-8 and Hsp70. However, compared with the S. enteritidis 857 induced IL-8 synthesis, the levels of IL-8 induced by the lactobacilli at any equivalent bacterial number were far lower. After exposure of Caco-2 cells to S. enteritidis 857 pre-treated with SCS-lactobacilli, it appeared that their SCS inhibited the S. enteritidis 857 growth and IL-8 synthesis and in addition induced the expression of Hsp70. The differences in response of crypt- and villus-like Caco-2 cells are merely a reflection of their differentiation status. Our data suggest that the beneficial effect of non-starter lactobacilli to the intestinal inflammations might be associated with a decrease of the IL-8 levels. This effect could be mediated, at least in part, by the bacteria themselves or via a secreted antimicrobial product(s) either directly against the pathogens or indirectly through the synthesis of Hsp70. 相似文献

Physicochemical and functional properties of arabinoxylans (AXs) can be significantly influenced by their isolation method. Finding balanced process conditions that allow optimal extraction yields while preserving AXs functionality is a challenge. The aim of this study was to determine the effect of different chemical solvents with neutral and alkaline pH on the intrinsic properties and extraction yield of AXs isolated from rye bran. Additionally, the application of xylanases and other cell wall degrading enzymes (Pentopan Mono BG, Deltazym XL‐VR, Viscoflow BG) to solubilize bound AXs was investigated. Results show that the use of Ca(OH)2 for isolation was superior to water and Na2CO3, as it selectively solubilized AXs and delivered isolates with a purity of up to 43.92% AX and a moderate ferulic acid (FA) content (209.35 ± 16.79 mg FA/100 g AX). Application of xylanases was further able to duplicate these achieved AX yields (7.50 to 9.85g AX/100 g bran). Additionally, isolates displayed highest ferulic acid contents (445.18 to 616.71 mg FA/100 g AX) and lowest impurities in comparison to chemical extracted AXs. Rheological characterization of the isolates showed a pronounced shear thinning behavior which fitted well to the power‐law model (R2 > 0.989). Differences in pseudoplasticity of the isolates suggested that structural and chemical properties might have been responsible for this behavior. 相似文献

A human prostate tumour cell line, LNCaP C4-2, when injected into athymic male nude mice, produced tumours containing: (1) only human cancer cells similar to those injected; (2) only murine stromal cells containing abnormal chromosome constitutions; or (3) both human prostate cancer cells similar to those injected and the transformed murine stromal cells with altered chromosome constitutions. Karyotypic analysis of murine metaphases from all the host-derived tumours showed mostly pseudodiploid chromosome constitutions, with multiple copies (amplification) of mouse chromosome 15 and the absence of a typical Y chromosome. Fluorescence in situ hybridization analysis of these murine cells, using a biotin-labelled total human DNA painting probe, further demonstrated the absence of human DNA and the presence of only mouse metaphase and interphase cells in these transformed stromal cells. These results suggest that cancer cells are capable of inducing neoplastic transformation in stromal cells of the host organ by some, as yet unknown, epigenetic mechanism(s). 相似文献

Micropolar beam models are developed for the static, free vibration and buckling analysis of repetitive spatial beamlike lattices with rigid joints. The micropolar beam models have independent microrotation and displacement fields and are characterized by their strain energy, potential energy due to initial stresses and kinetic energy from which the governing differential equations and boundary conditions can be derived. The procedure for developing the expression for the strain energy of the micropolar beam involves introducing basic assumptions regarding the variation of the displacement and microrotation components in the plane of the cross-section, and obtaining effective elastic coefficients of the continuum in terms of the material properties and geometry of the original lattice structure. The high accuracy of the solutions obtained by the micropolar beam models is demonstrated by means of numerical examples for vierendeel and double-laced lattice girders with triangular cross-sections. 相似文献

Process manufacturing is increasingly being driven by market forces, customer needs, and perceptions, resulting in more and more complex multiproduct manufacturing technologies. The increasing automation and tighter quality constraints related to these processes make the operator's job more and more difficult. This makes decision support systems (DSSs) for the operator more important than ever before. A traditional operator support system (OSS) focuses only on specific tasks that are performed. In the case of complex processes, the design of an integrated information system is extremely important. The proposed data-warehouse-based OSS makes possible linking complex and isolated production units based on the integration of the heterogenous information collected from the production units of a complex production process. The developed OSS is based on a data warehouse designed by following the proposed focus-on-process data-warehouse-design approach, which means stronger focus on the material and information flow through the entire enterprise. The resulting OSS follows the process through the organization instead of focusing separate tasks of the isolated process units. For human-computer interaction, front-end tools have been worked out, where exploratory data analysis and advanced multivariate statistical models are applied to extract the most informative features of the operation of the technology. The concept is illustrated by an industrial case study, where the OSS is designed for the monitoring and control of a high-density polyethylene (HDPE) plant. 相似文献

The question as to how far the development of chronic diseases in humans depends on diet still remains open. Simultaneously, epidemiological studies suggest the consumption of a flavonoids rich diet might decrease the risk of degenerative changes and certain diseases. The intake of this group of compounds as to quality and quantity depends on dietary habits and a widespread presence of quercetin in the diet makes this compound one of the key factors. Onion, one of the richest and most common quercetin sources, was particularly often studied in different aspects. Quercetin is present in onion mainly as glycosides, of which the distribution within the onion bulb changes in onion processing, and biological activities attracted a lot of attention. Especially antioxidative activity demonstrated in vitro was initially associated with most of the beneficial effects of quercetin on the human body. However, after ingestion quercetin undergoes extensive metabolism and microbial action resulting in its altered or degraded structure; therefore, most of the effects shown in in vitro experiments with the pure compound cannot be directly extrapolated to in vivo systems. Yet, this does not mean that quercetin simultaneously loses its positive impact on consumer health. Even after being metabolized it may still affect the redox balance by inducing antioxidative and detoxifying enzymes or compounds which may be involved in sustaining homeostasis. 相似文献
