Herein we report the first discovery of natural readthrough products that do not display antimicrobial activity. Two natural negamycins, 3‐epi‐deoxynegamycin and its leucine adduct, isolated 37 years ago, were found to be potent readthrough agents against nonsense mutations of eukaryotes, but not prokaryotes, without displaying antimicrobial activity. These results suggest that the compounds are valuable leads for the development of readthrough drugs against nonsense‐mediated genetic diseases without the potential for contributing to the emergence of drug‐resistant bacteria.  相似文献

Upgraded brown coal water mixture (UBCWM) preparation by using an Indonesian upgraded coal produced by upgraded brown coal (UBC) process, was carried out to study the effect of dispersing and stabilizing additives on rheological behavior of the UBCWM. Three kinds of anionic dispersing additives, naphthalene sulfonic formaldehyde condensate (NSF), poly (meth) acrylate (PMA) and poly styrene sulfonic acid (PSS) and three kinds of stabilizing additives, carboxyl methyl cellulose (CMC), rhansam gum (S-194) and gellan gum (S-60) were used in this study. Results indicate that the addition of NSF 0.3 wt.% together with S-194 0.01 wt.% is effective in preparing UBCWM with good slurryability and stability, based on its rheological characteristics with the apparent viscosity at shear rate of 100 s− 1 and yield stress at zero point of shear rate. The rheological behavior of all of the UBCWM that prepared, exhibits non-Newtonian Bingham plastic. From the economical point of view, the price of S-194 is expensive. On the other hand, CMC is cheap and abundant. Therefore, the addition of CMC 0.01 wt.% together with NSF 0.3 wt.% is also effective in preparing UBCWM with good fluidity and stability.  相似文献

Trace elements in teeth of northern fur seal Collorhinus ursinus were analyzed by PIXE and micro-PIXE (μ-PIXE). Trace elements such as Mn, Fe, Zn, Cu, As, Br, Sr, and Pb were detected in the teeth, which were composed of hydroxyapatite crystals. Among these elements, the concentrations of Fe and Zn were relatively higher in winter and lower in summer and also Zn varied along with growth of individuals. These elemental fluctuation seems to correspond to the life history of the northern fur seal. It suggests that the PIXE analysis will make a powerful tool to reconstruct the life history.  相似文献

Two methods were examined for the formation of poly-Si films. One is flux method and the other is Cat-CVD method. Flux method was used for forming poly-Si seed films on glass substrates covered with rear electrode. Poly-Si films of a few μm grain size and of mainly (1 1 1) crystalline orientation were obtained at less than 600°C. To make the seed films function as BSF layer for solar cell, boron doping was applied and carrier concentration of 2×1019/cm3 was obtained which is suitable for highly efficient solar cells. Cat (catalytic)-CVD method was examined for forming poly-Si photo-active layers on the seed films. The films showed deposition gas pressure-dependent crystalline orientations and there was no amorphous incubation layer in (1 1 1) oriented films by Cat-CVD method when deposited on (1 1 1) oriented seed films prepared by Flux method. The electrical properties of the film are insufficient at present, may be due to high defect density and the film structure which allows impurity contaminations of oxygen and carbon after film deposition. Although the film quality needs to be improved, poly-Si films whose crystal fraction is more than 85% were obtained at deposition rate of up to around 40 Å/s. This result indicates high potential of Cat-CVD method for high throughput photo-active formation process necessary for low production cost thin film silicon solar cells.  相似文献

The catalyst slurry used to form the catalyst layer of a polymer electrolyte fuel cell (PEFC) must be mixed for a sufficiently long period for good and stable cell performance. However, the optimum mixing duration must be determined from the viewpoint of process design. We prepared slurries with various amounts of Nafion and examined the influence of the slurry mixing time on the viscosity of the slurry, the size of the pores and distribution of elements in the catalyst layer, and the cell voltage of the PEFC. We found that when the Nafion content is optimum, these properties change gradually and stabilize after a sufficiently long mixing time as the progression of the adsorption of Nafion to catalyst particles. However, when the Nafion is comparatively low, although the properties stabilize after sufficient mixing, the Nafion molecules are first dispersed and then localized around aggregates. This reduces cell performance when mixing excessively long. These differences in the structural and electrochemical behaviors could be predicted by measuring the variation in viscosity during the mixing process.  相似文献

We have shown that stable lactic acid fermentation of model kitchen refuse occurs with intermittent pH adjustment under nonsterilized conditions. Nonetheless, the optical activity of the accumulated lactic acid was low, which is disadvantageous for the production of high-quality poly-l-lactic acid. Here, we attempt to increase optical purity by introducing l-lactic acid-producing strains under nonsterilized conditions and demonstrate that the inoculation of Lactobacillus rhamnosus or Lactococcus lactis, both of which are l-lactic acid producers, is partially effective in the early fermentation stage, but does not improve the final optical purity of the accumulated lactic acid. We confirmed by fluorescence in situ hybridization using group-specific and species-specific 16S rDNA probes that this is due to the selective proliferation of naturally existing L. plantarum. L. plantarum KY-1, which is isolated from model kitchen refuse, showing lactic acid racemase activity, as well as d-lactate dehydrogenase activity, in its membrane fraction. We conclude that racemase activity associated with L. plantarum is the main cause of decreased optical purity in the accumulated lactic acid.  相似文献

1. Alkaline phosphatase (EC 3.1.3.1) from human liver was solubilized from the homogenate using 0.2% Triton X-100 containing 0.2 M lithium 3,5-diiodosalicylate, and the pellet obtained was resolubilized with 20% n-butanol. The procedure resulting in 3842-fold purification included acetone fractionation, ammonium sulfate precipitation, DEAE-cellulose chromatography, Sephadex G-200 gel filtration, hydroxyapatite gel chromatography and further concanavalin A/Sepharose 4B affinity chromatography. 2. The highly purified enzyme showed one major protein band on acrylamide gel electrophoresis at pH 8.6, and exhibited one-seventh of the alkaline p-nitrophenylphosphatase activity in the hepatic enzyme preparation contains of the alkaline pyrophosphatase activity. 3. The highly purified enzyme was a sialic-acid containing glycoprotein. 4. Sialidase-treated hepatic enzyme clearly presented the phenomenon of delayed mobility, and the delayed enzyme fraction stained more strongly than that of non-treated hepatic alkaline phosphatase. 5. In order to investigate the role of the carbohydrate region(s) of the hepatic alkaline phosphatase molecule on substrate binding, the effect of sialidase treatment on the rate of substrate inhibition of alkaline phosphatase was studied. In the case of hepatic enzyme without sialidase, substrate inhibition of alkaline phosphatase activity was clearly shown, while in the case of the hepatic enzyme with sialidase, there was hardly any substrate inhibition in the range of 1-8 mM p-nitrophenylphosphate.  相似文献

To establish an environmentally friendly groundwater bioremediation process using a cellulose carrier combined with cellulose-utilizing, denitrifying microorganisms, a novel psychrophilic bacterium, designated CL-5, which can degrade a commercial-based cellulose carrier as the sole carbon source, was screened. Since the denitrification capability of CL-5 is low, complex microbial systems were constructed together with other denitrifying bacteria designated NR-1 and NR-2 that were also isolated from soil. The nitrate-reducing activities of mixed cultures were much higher than those of the pure cultures of CL-5, NR-1 and NR-2. The highest N(2)O and N(2) formation activities were observed in the mixed culture of CL-5+NR-2.  相似文献
