The triglyceride (fatty acid) selectivity of an immobilized lipase from Thermomyces lanuginosa (Lipozyme TL IM) was investigated in lipase-catalyzed interesterification reactions between two nono-acid TG in n-hexane. Tristearin (tri-C18∶0) was used as a reference in a series of TG with saturated FA from tri-C4∶0 to tri-C20∶0, except for tri-C6∶0, and in a series of unsaturated FA from tri-C18∶1 to tri-C18∶3. The quantification was performed by HPLC, and different methods of selectivity evaluation were used. None of the methods used showed any significant differences between the performances of the lipase on the different TG, indicating that Lipozyme TL IM is nonselective toward FA or TG in the system used. A response surface design was used to investigate the influence of water activities (a w ) and reaction temperatures on the reactivity of Lipozyme TL IM with a system of tripalmitin (tri-C16∶0) and trilaurin (tri-C12∶0) in n-hexane. An increase in temperature (40 to 60°C) was found to affect the reactivity of the lipase significantly. The reactivity of Lipozyme TL IM was unaffected by the change in a w from 0.1130 to 0.5289. An increase in a w only led to an increase in FFA formation.  相似文献

A circumpolar survey of heavy metals in willow ptarmigan liver and kidney revealed considerable variations in Cd content in Canada and Scandinavia. The Cd content in central Canada was comparable with that in Scandinavia and Russia, at least for kidney. However, in both liver and kidney the median for Canada as a whole was much higher than in the other countries. Some Canadian locations had exceptionally high levels, several birds having >50 mg kg(-1) in liver and >400 mg kg(-1) in kidney. In Norway, the Cd content was highest in central mountain areas in south Norway and inland locations in the two northernmost counties. Five locations in central and north Norway showed mean Cd levels in kidney above 100 mg kg(-1). It is difficult to evaluate regional differences in Sweden, but most locations had the same Cd level as moderately contaminated locations in Norway. Cd levels in Russia were comparable to moderately contaminated locations in the other countries. Due to a high intake of willow, naturally rich in Cd, direct comparison of the Cd level in willow ptarmigan from different locations cannot reveal the effects of long-range pollution. The Pb concentration in willow ptarmigan kidney and liver varied significantly in Norway and Canada, but not in Sweden and Russia. Levels in Sweden and Russia were comparable to those in Canada and low levels in Norway. The highest median value from all locations within countries was found in Norway, both in liver and kidney. The highest Pb content was found in south Norway, indicating an effect of long-range pollution in willow ptarmigan. The level in western Canada was significantly higher than in central Canada. The Hg content in liver varied significantly from one location to another in all the countries and in kidney everywhere except Sweden. In Scandinavia, there is no distinct regional pattern. Canada had a significantly higher Hg level in central than western regions in both tissues the opposite of that found for Cd and Pb. Cu and Zn showed significant variations from one location to another in liver and kidney in Canada and Norway, but only in kidney in samples from Sweden. Comparison between western and central Canada revealed a significant difference for Cu in liver, samples from central Canada having more. There are no significant differences from one country to another, but some localities in Canada seem to have higher Cu concentrations in kidney than are found in Scandinavia and Russia.  相似文献

The properties of a new immobilized glucose isomerase, produced by a selected strain of Streptomyces murinus, are described. The major advantages of the new immobilized enzyme are a productivity of more than 10,000 kg syrup dry substance per kg enzyme under optimal industrial conditions, increased activity and a very low syrup by-product formation. The influence of process parameters (temperature, pH and feed syrup additives) on activity and stability is discussed based on laboratory and industrial plant data.  相似文献

Mycoplasma bovis is an important pathogen causing disease and substantial economic losses in cattle. However, knowledge of the dynamics of antibody responses in individual cows in the face of an outbreak is currently extremely limited. The use of commercial antibody tests to support clinical decision-making and for surveillance purposes is therefore challenging. Our objective was to describe the dynamics of M. bovis antibody responses in 4 Danish dairy herds experiencing an acute outbreak of M. bovis-associated disease, and to compare the antibody dynamics between dairy cows with different disease manifestations. A total of 120 cows were examined using a standardized clinical protocol and categorized into 4 disease groups: “mastitis,” “systemic,” “nonspecific,” and “none.” Paired blood and milk samples were collected and tested using a commercial M. bovis antibody–detecting ELISA. Plots of raw data and generalized additive mixed models with cow and herd as random effects were used to describe serum and milk antibody dynamics relative to the estimated time of onset of clinical disease. Cows with mastitis had high optical density measurement (ODC%) of antibodies in both milk and serum at disease onset. The estimated mean ODC% in milk was below the manufacturer's cut-off for the other groups for the entire study period. The estimated mean serum ODC% in the “systemic” group was high at onset of disease and stayed above the cut-off until 65 d after disease onset. However, the lower 95% confidence interval (CI) for the mean ODC% was only above the manufacturer's cut-off between 7 and 17 d after onset of disease. The CI of the “systemic” and “none” groups did not overlap at any time between the day of disease onset and 65 d after disease onset, and the estimated mean ODC% for both the “nonspecific” and “none” groups were generally below the cut-off for the majority of the study period. In conclusion, the serum antibody responses were highly dynamic and showed a high level of variation between individual cows. This strongly suggests that serology is unlikely to be useful for individual diagnosis of M. bovis-associated disease in dairy cows. However, it might still be useful for herd- or group-level diagnosis. Antibodies in milk were only increased in cows with M. bovis mastitis, indicating that milk antibody measurements only have diagnostic utility for cows with mastitis.  相似文献

On the background of positive survival data from farms in Mississippi, treating calves with antiserum injection in addition to normal colostrum administration, the objective of the present study was to evaluate the influence of a single subcutaneously administered bovine antiserum injection (0.031 g of IgG/kg of body weight) and pooled colostrum administration on efficiency of Ig absorption and on 24-h plasma IgG concentration in neonatal bull calves. Twenty-nine male dairy calves (21 Holsteins and 8 Jerseys) were assigned randomly at parturition to receive one of four treatments: 1) colostrum (n = 9), 2) colostrum and bovine antiserum injection (n = 7), 3) milk replacer (n = 5), or 4) milk replacer and bovine antiserum injection (n = 8). At birth, calves either did or did not receive an injection of bovine antiserum and were fed pooled colostrum or milk replacer (Holsteins, 3.8 L; Jerseys, 1.9 L) via an esophageal feeder. Blood was collected immediately before administration of the colostrum or milk replacer, then again at 24 and 48 h postpartum. Immunoglobulin G concentrations of colostrum, milk replacer, antiserum, and plasma were monitored by single radial immunodiffusion. Colostrum administration and injection of bovine antiserum each increased plasma Ig concentration at 24 h posttreatment. In addition, antiserum injection increased the apparent efficiency of absorption of colostral Ig by 42% over that for calves fed colostrum alone. The increase in plasma IgG for antiserum-treated calves exceeded the total amount of IgG administered in the antiserum injection; hence, this increase appeared to be the result of an increase in total absorption of colostral IgG, or possibly antiserum injection somehow triggered active synthesis of IgG. Injection of antiserum might possibly serve as a beneficial adjunct to a colostrum management program by enhancing the acquisition of passive immunity from colostral sources.  相似文献

Adsorption of three sulfonamide antimicrobials to clay minerals was investigated as a function of pH, ionic strength, and type of exchangeable cation. Sulfonamide antimicrobial adsorption exhibited pronounced pH dependence consistent with sorbate speciation and clay properties. Sulfonamide antimicrobials did not intercalate into montmorillonite, and surface charge density influenced sorption by determining adsorption domain size. Adsorption edge data were best fit to a model including terms for the cationic and uncharged species. Adsorption of uncharged sulfamethazine to montmorillonite was relatively insensitive to pH, ionic strength, and type of exchangeable cation, while that to kaolinite was highly sensitive to ionic strength. Adsorption of cationic sulfamethazine to montmorillonite exceeded that of the neutral species by 1-2 orders of magnitude, but was unimportant for kaolinite atthe pH values examined. Cation exchange appeared to contribute to sorption of cationic sulfonamide species to montmorillonite. Anionic sulfamethazine adsorption was negligible. The nature of the sulfonamide R group influenced the degree of adsorption of cationic and neutral species. Our results highlight the importance of considering sulfonamide speciation and clay surface charge density in predicting the transport of these antimicrobials.  相似文献

Total mixed rations containing conventional forage sorghum, brown midrib (bmr)-6 forage sorghum, bmr-18 forage sorghum, or corn silage were fed to Holstein dairy cows to determine the effect on lactation, ruminal fermentation, and total tract nutrient digestion. Sixteen multiparous cows (4 ruminally fistulated; 124 d in milk) were assigned to 1 of 4 diets in a replicated Latin square design with 4-wk periods (21-d adaptation and 7 d of collection). Diets consisted of 40% test silage, 10% alfalfa silage, and 50% concentrate mix (dry basis). Acid detergent lignin concentration was reduced by 21 and 13%, respectively, for the bmr-6 and bmr-18 sorghum silages when compared with the conventional sorghum. Dry matter intake was not affected by diet. Production of 4% fat-corrected milk was greatest for cows fed bmr-6 (33.7 kg/d) and corn silage (33.3 kg/d), was least for cows fed the conventional sorghum (29.1 kg/d), and was intermediate for cows fed the bmr-18 sorghum (31.2 kg/d), which did not differ from any other diet. Total tract neutral detergent fiber (NDF) digestibility was greatest for the bmr-6 sorghum (54.4%) and corn silage (54.1%) diets and was lower for the conventional (40.8%) and bmr-18 sorghum (47.9%) diets. In situ extent of NDF digestion was greatest for the bmr-6 sorghum (76.4%) and corn silage (79.0%) diets, least for the conventional sorghum diet (70.4%), and intermediate for the bmr-18 sorghum silage diet (73.1%), which was not different from the other diets. Results of this study indicate that the bmr-6 sorghum hybrid outperformed the conventional sorghum hybrid; the bmr-18 sorghum was intermediate between conventional and bmr-6 in most cases. Additionally, the bmr-6 hybrid resulted in lactational performance equivalent to the corn hybrid used in this study. There are important compositional differences among bmr forage sorghum hybrids that need to be characterized to predict animal response accurately.  相似文献

Appital is a herbal medicine. The aim was to examine its effect in patients with irritable bowel syndrome (IBS). Other diseases were excluded by physical examination, rectoscopy, blood tests and in patients older than 35 years X-ray of the colon or colonoscopy. The study was designed as a double-blind placebo-controlled trial. The patients were randomized to either Appital or placebo. Following two weeks without medicine, the patient had Appital or placebo for eight weeks. Fifty-nine patients were randomized, 47 completed the study. The results were based on symptom scores registered by the patients. The symptom score was significantly reduced in patients treated with Appital (p = 0.002), but when compared to placebo, the difference was insignificant (p = 0.081). We concluded that Appital has no effect in relieving symptoms of IBS compared to placebo, although due to the possibility of type two error we cannot exclude a small, but hardly clinically relevant effect.  相似文献

Proximal separation (PS) refers to isolating pups in small cages so dams can hear, smell, and see pups but have very limited physical contact with them. Six days of PS diminished the number of discernible oxytocin (OT)- immunostaining perikarya in forebrain areas of rat dams compared with 6 days of total separation (TS) or no separation (NS) from pups. Dams exhibited a more rapid resurgence of maternal behavior after 4–6 days of PS than after 4–6 days of TS. Bilateral infusion of the OT antagonist (OTA; 1 μg/μl/side) into the ventral tegmental area blocked the resurgence of maternal behavior after 3–6 days of PS but not after 2 days of PS or 4–6 days of NS. The conclusion was that PS for 3 or more days reinstates OT as necessary and sufficient for the activation of maternal behavior in experienced rat mothers. These findings suggest that some aspects of somatosensory stimulation from pups regulate the role of OT in the control of maternal behavior.
