A novel approach was applied in this study to directly evaluate the effect of muscle fiber type on postmortem protein degradation. Porcine muscle fibers were isolated from longissimus muscle at day 1, 3, and 6 postmortem. Fibers were sorted by immunochemical myosin heavy chain isoform typing. Western blot analysis of fibers pooled separately into type I or IIb showed that the relative amounts of 39- and 50-kDa desmin degradation fragments at day 6, and 28- to 31-kDa fragments of troponin T fast type isoform (fTnT) at day 1 and 6 postmortem were higher in type IIb than in type I fibers. At day 6 troponin T slow type isoform (sTnT) was less degraded than fTnT in type I fibers. These results indicated greater rate and extent of proteolysis in type IIb than in type I fibers and higher susceptibility of fTnT to proteolysis than that of sTnT isoform.  相似文献

NDF, ADF, ‘cellulose’ and IVOMD residues, from feeding value determinations, were prepared from some forages, including straw, alkali-treated straw, grass and lucerne samples. The crude protein, cellulose, hemicellulose, uronic acid, Klason lignin, lignin and ash contents of these residues and the corresponding original forage samples were determined together with the relative composition of the neutral sugar constituents after acid hydrolysis. Cellulose, hemicellulose and Klason lignin were the main components in the NDF fractions but substantial amounts of crude protein (1–6%) also remained. Cellulose and Klason lignin were the main components in the ADF fractions, but 7–14% of the fractions was hemicellulose and 1–4% crude protein. Cellulose was the main component in the ‘cellulose’ fractions, but they also contained 8–13% hemicellulose, 2–7% Klason lignin and 2–11% ash. The composition of the IVOMD residues showed that cellulose and hemicellulose had been dissolved to about the same amounts (70–80%) and that most of the Klason lignin remained.  相似文献

Flavour development and overall eating quality of pan-fried pork chops of longissimus dorsi from eight different raw meat qualities aged for 4 and 15 days were assessed by a trained sensory panel. The raw meat qualities were obtained through combinations of strategic feeding/fasting (control vs. low glycogen concentration), slaughter live-weight (84kg vs. 110kg), and gender (female vs. castrate). The flavour development was investigated for possible correlation with the concentrations of selected individual flavour precursors present in the raw meat: monosaccharides, IMP and degradation products, fatty acids, lactate and thiamine. Differences in precursor concentrations between the raw meat qualities were observed with feeding/fasting and ageing as the main factors with the largest influence of all experimental factors. However, the concentrations of the precursors could not explain the differences in sensory perception of the pan-fried pork chops. Overall, the differences were small.  相似文献

The atmospheric transport of polybrominated diphenyl ethers (PBDEs) and polychlorinated biphenyls (PCBs) was compared by measuring concentrations in air and deposition on an island located in the central basin of the Baltic Sea. Median sigmaPBDE and sigmaPCB concentrations (gaseous + particle) were 8.6 and 7.4 pg m(-3), respectively. Airborne PCBs were mainly found in the gaseous phase, while most of the PBDEs were detected on particles, which agrees with predicted particle/gas distributions. SigmaPBDE levels were dominated by the decabrominated BDE209 followed bythe tetrabrominated BDE47 and pentabrominated BDE99. BDE209 is a marker for the environmental distribution of the commercial deca-BDE formulation (>99.5% BDE209), whereas BDE47 and BDE99 are markers for the commercial penta-BDE mixture. General correlations between PBDEs and PCBs suggested similarities in sources and transport mechanism, while more detailed examination of the data identified notable behaviors and exceptions. Differences in regression slopes among tetra-, penta-, and decabrominated PBDEs may reflect different transport processes and the change in usage pattern. Tetra- and pentabrominated PBDEs may originate from secondary sources such as air surface exchange in a manner similar to that of the PCBs, while the deca-BDE209 formulation still has primary sources. The tribrominated BDE17 was also detected and is proposed to be a breakdown product due to atmospheric debromination processes. PBDEs had higher washout ratios than PCBs, explaining their higher concentrations compared to PCBs in precipitation (median of 6.0 and 0.5 ng L(-1) for sigmaBDE and sigmaPCB concentrations ("dissolved" + particle), respectively) than in air. The calculated yearly deposition of PBDEs and PCBs indicated that the atmospheric input of PBDEs to the Baltic Proper is currently exceeding that of the PCBs by a factor of 40, while that of the PCBs is decreasing.  相似文献

The distribution of different iron (Fe) species in soils, sediments, and surface waters has a large influence on the mobility and availability of Fe, other nutrients, and potentially toxic trace elements. However, the knowledge about the specific forms of Fe that occurs in these systems is limited, especially regarding associations of Fe with natural organic matter (NOM). In this study, extended X-ray absorption fine structure (EXAFS) spectroscopy was used to characterize Fe(III) in organic soils (pH 4.6-6.0) with varying natural Fe content. The EXAFS data were subjected to wavelet transform analysis, to facilitate the identification of the nature of backscattering atoms, and to conventional EXAFS data fitting. The collective results showed the existence of two pools of iron: mononuclear Fe(III)-NOM complexes and precipitated Fe(III) (hydr)oxides. In the soil with lowest pH (4.6) and Fe content mononuclear organic complexes were the completely dominating fraction whereas in soils with higher pH and Fe content increasing amounts of Fe (hydr)oxides were detected. These results are of environmental importance, as the different iron pools most likely have markedly different reactivities.  相似文献

A comparative study of linear methods for prediction of sensory profiles from gas chromatography (GC) measurements was performed. The data used came from an experiment on the effect of storing apples at various oxygen concentrations. Partial least-squares regression and continuum regression showed the best performance, measured by a two-step cross-validation principle. The traditional prediction error sum of squares (PRESS) overestimated the predictive ability of a multiple linear regression approach. The quality of the predictions of sensory properties from GC analyses was measured in terms of a ‘panel size equivalent’. Thus, the predictions obtained in the present study were as accurate as predictions from an assessor panel consisting of 4–9 assessors, depending on the sensory property in question.  相似文献

The common assumption that chloride (Cl-) is conservative in soils and can be used as a groundwater tracer is currently being questioned, and an increasing number of studies indicate that Cl- can be retained in soils. We performed lysimeter experiments with soil from a coniferous forest in southeast Sweden to determine whether pore water residence time and nitrogen and Cl- loads affected Cl- retention. Over the first 42 days there was a net retention of Cl- with retention rates averaging 3.1 mg CI- m(-2) d(-1) (68% of the added Cl- retained over 42 days). Thereafter, a net release of Cl- at similar rates was observed for the remaining experimental period (85 d). Longer soil water residence time and higher Cl- load gave higher initial retention and subsequent release rates than shorter residence time and lower Cl- load did. Nitrogen load did not affect Cl transformation rates. This study indicates that simultaneous retention and release of Cl- can occur in soils, and that rates may be considerable relative to the load. The retention of Cl- observed was probably due to chlorination of soil organic matter or ion exchange. The cause of the shift between net retention and net release is unclear, but we hypothesize that the presence of O2 or the presence of microbially available organic matter regulates Cl- retention and release rates.  相似文献

Bacillus cereus is an endospore-forming bacterium able to cause food-associated illness. Different treatment processes are used in the food industry to reduce the number of spores and thereby the potential of foodborne disease. Chitosan is a polysaccharide with well-documented antibacterial activity towards vegetative cells. The activity against bacterial spores, spore germination and subsequent outgrowth and growth (the latter two events hereafter denoted (out)growth), however, is poorly documented. By using six different chitosans with defined macromolecular properties, we evaluated the effect of chitosan on Bacillus cereus spore germination and (out)growth using optical density assays and a dipicolinic acid release assay. (Out)growth was inhibited by chitosan, but germination was not. The action of chitosan was found to be concentration-dependent and also closely related to weight average molecular weight (Mw) and fraction of acetylation (FA) of the biopolymer. Chitosans of low acetylation (FA = 0.01 or 0.16) inhibited (out)growth more effectively than higher acetylated chitosans (FA = 0.48). For the FA = 0.16 chitosans with medium (56.8 kDa) and higher Mw (98.3 kDa), a better (out)growth inhibition was observed compared to low Mw (10.6 kDa) chitosan. The same trend was not evident with chitosans of 0.48 acetylation, where the difference in activity between the low (19.6 kDa) and high Mw (163.0 kDa) chitosans was only minor. In a spore test concentration corresponding to 102-103 CFU/ml (spore numbers relevant to food), less chitosan was needed to suppress (out)growth compared to higher spore numbers (equivalent to 108 CFU/ml), as expected. No major differences in chitosan susceptibility between three different strains of B. cereus were detected. Our results contribute to a better understanding of chitosan activity towards bacterial spore germination and (out)growth.  相似文献

Variations in texture were determined for 10 day aged raw and cooked Longissimus thoracis (LT) muscle from 436 bulls of 15 European cattle breeds slaughtered at an age of 13–16 months. Variations in texture were related to differences in pH 24 h post-mortem, sarcomere length, collagen characteristics and lipid content. The shear force of cooked meat samples varied from 43.8 to 67.4 N/cm2. Simmental, Highland and Marchigiana cattle had the highest shear force values and Avileña-Negra Ibérica, Charolais, Casina and Pirenaica cattle had the lowest values. Cooked meat toughness showed a weak negative correlation to lipid content (P < 0.001) but no correlation to collagen characteristics. Raw meat texture measured by compression correlated positively (P < 0.001) with total and insoluble collagen. In conclusion, collagen characteristics showed correlation to raw meat texture but not to cooked meat toughness of LT muscle in European young bulls.  相似文献
