In vivo Raman spectroscopy, using fiber-optic probes is hindered by the intense background signal, which is generated in the fused-silica fibers, in the fingerprint region of the Raman spectrum (approximately 0-2000 cm(-1)). Optical filtering is necessary to obtain tissue spectra of sufficient quality. The complexity of fiber-optic probes for fingerprint Raman spectroscopy, in combination with size constraints and flexibility requirements for in vivo use have been a major obstacle in the development of in vivo diagnostic tools based on Raman spectroscopy. A setup for remote Raman spectroscopic tissue characterization in the high-wavenumber region ( approximately 2400-3800 cm(-1)) is presented. It makes use of a single, unfiltered, optical fiber for guiding laser light to the sample and for collecting scattered light and guiding it back to a spectrometer. Such a simple configuration is possible because the fused-silica core and cladding of the fiber present almost no Raman background signal at these wavenumbers. Several commercially available optical fibers were tested with respect to Raman signal background, to determine their suitability for in vivo Raman spectroscopy measurements in the high-wavenumber region. Different fiber core, cladding, and coating materials were tested. Silica core-silica clad fibers, with an acrylate coating and a black nylon jacket, proved to be one of the best candidates. In vitro measurements on brain tissue of a 6-month-old pig were obtained with a remote high-wavenumber Raman setup. They illustrate the low background signal generated in the setup and the signal quality obtained with a collection time of 1 s.  相似文献

Detection of meningioma in dura mater by Raman spectroscopy   总被引:1,自引:0,他引:1  
Radical tumor resection is the treatment of choice for patients suffering from meningioma. However, recurrence of these tumors is a problem. Tumor recurrences are attributed to residual nests of meningioma within the regional dura. Therefore, complete removal of all tumor-infiltrated dura is important. Meningioma and normal dura were studied by Raman microspectroscopy to assess the possibility of developing an in vivo Raman method for guidance of meningioma resections. Pseudocolor Raman maps were constructed of cryosections of dura and meningioma, obtained from 20 patients. Comparison of these maps with histopathology enabled assignment of the spectra to either meningioma or dura. Large differences exist between the Raman spectra of dura and meningioma, because of the high collagen content of dura and the increased lipid content of tumors. A classification model for dura and tumor tissue based on linear discriminant analysis of Raman spectra yielded an accuracy of 100%. A first attempt was made to determine the minimum amount of meningioma in dura that is detectable by Raman spectroscopy. It is concluded that Raman spectra enable meningioma to be distinguished from dura, which makes Raman spectroscopy a viable candidate for guidance of surgical resection of meningioma.  相似文献

Comments on the article by Renner (see record 2007-09069-001), in which he seems to make the argument that the university is going to starve to death if levels of financing continue to decline, so universities should offer staff ways of getting away before the ship goes down completely. What universities must do as a first task, is to find a new base upon which to operate, one that will restore the undergraduate degree to a position of high value. If we do, this society will continue to support a cadre of people who bring students into touch with the lead-edge of intellectual currents within a relatively confined organic structure. CD34 is expressed on the surface of hematopoietic stem/progenitor cells, stromal cells, and on the surface of high-endothelial venules (HEV). CD34 binds L-selectin, an adhesion molecule important for leukocyte rolling on venules and lymphocyte homing to peripheral lymph nodes (PLN). We generated CD34-deficient mutant animals through the use of homologous recombination. Wild-type and mutant animals showed no differences in lymphocyte binding to PLN HEV, in leukocyte rolling on venules or homing to PLN, in neutrophil extravasation into peritoneum in response to inflammatory stimulus, nor in delayed type hypersensitivity. Anti-L-selectin monoclonal antibody (MEL-14) also inhibited these immune responses similarly in both CD34-deficient and wild-type mice. However, eosinophil accumulation in the lung after inhalation of a model allergen, ovalbumin, is several-fold lower in mutant mice. We found no abnormalities in hematopoiesis in adult mice and interactions between mutant progenitor cells and a stromal cell line in vitro were normal. No differences existed in the recovery of progenitor cells after 5-fluorouracil treatment, nor in the mobilization of progenitor cells after granulocyte colony-stimulating factor treatment compared with wild-type animals. Surprisingly, although CD34 was not expressed in these mice, a portion of its 90-kD band crossreactive with MECA79 remained after Western blot. Thus, we have identified an additional molecule(s) that might be involved in leukocyte trafficking. These results indicate that CD34 plays an important role in eosinophil trafficking into the lung.  相似文献

The concept of uncertainty is used to develop a theory of the innovation process for computer-aided manufacturing. The interrelated propositions trace the process by which computerized equipment is adopted, prepared for, and implemented. This is done in terms of problems arising from uncertainty and coping strategies used to respond to them. The theory explains why it is so difficult to introduce the technology into companies. Problems arise in deciding whether or not to purchase, in preparing the organization's supporting functions, and in deciding whether or not success has been achieved. The propositions are also used to suggest coping strategies that can alleviate these problems. They include infrastructure development, participation, and installation in stages. The use of the [S,S]-isomer of EDDS to enhance phytoextraction has been proposed for the remediation of heavy metal contaminated soils. Speciation of metals in soil solution in the presence of EDDS and dissolved organic matter (DOM) received, however, almost no attention, whereas metal speciation plays an important role in relation to uptake of metals by plants. We investigated the influence of EDDS on speciation of dissolved metals in batch extraction experiments using fourfield-contaminated soils with pH varying between 4.7 and 7.2. Free metal concentrations were determined with the Donnan membrane technique, and compared with results obtained with the chemical speciation program ECOSAT and the NICA-Donnan model using a multicomponent approach. Addition of EDDS increased total metal concentrations in our soil extracts by a factor between 1.1 and 32 (Al), 2.1-48 (Cu), 1.1-109 (Fe), 1.1-5.5 (Ni), and 1.3-17 (Zn). In general, Al, Cu, Fe, and Zn had the largest total concentrations in the EDDS-treated extracts, but the contribution of these metals to the sum of total metal concentrations varied significantly between our soils. Free metal concentrations varied between 7.0 and 8.9 (pCd2+), 3.9-9.9 (pCu2+), 6.3-10.2 (pNi2+), and 5.2-7.0 (pZn2+). Addition of EDDS decreased free metal concentrations by a factor between 1.4 and 1.9 (Cd), 3.4-216 (Cu), 1.3-186 (Ni), and 1.3-3.3 (Zn). Model predictions of free metal concentrations were very good, and predicted values were mostly within 1 order of magnitude difference from the measured concentrations. A multicomponent approach had to be used in our model calculations, because competition between Fe and other metals for binding with EDDS was important. This was done by including the solubility of metal oxides in our model calculations. Multicomponent models can be used in chelant-assisted phytoextraction experiments to predict the speciation of dissolved metals and to increase the understanding of metal uptake by plants.  相似文献
