1. To see whether presynaptic activation paired with postsynaptic depolarization is necessary for the induction of long-term potentiation (LTP) in visual cortex or whether an activation of postsynaptic receptors in conjunction with depolarization is sufficient, we carried out perforated patch-clamp recordings with nystatin from cultured cortical neurons of rats. 2. Recorded neurons were monosynaptically activated either by electrical stimulation of an adjacent neuron or by direct activation of glutamate on "hot spots" of dendrites through iontophoresis or pressure ejection. In experiments in which cultured neurons were stained immunocytochemically with antibody against synaptophysin after electrophysiological recordings, hot spots were found to correspond to probable synaptic sites. 3. Excitatory postsynaptic currents (EPSCs) evoked by test stimulation applied to the adjacent neuron at 0.1 Hz were recorded at a holding potential of -60 or -70 mV for 5-10 min after an establishment of the whole cell recording configuration. Then, stimulation was paired with postsynaptic depolarization (0 mV for 200 ms) at 1 Hz for 30 or 60 s. LTP of EPSCs was induced in 7 of the 15 cells from which stable recordings were obtained for 18-30 min after pairing. 4. When postsynaptic depolarization was paired with direct glutamate application in the absence of presynaptic stimulation in 12 cells, only 1 showed LTP. Postsynaptic depolarization alone did not induce LTP in any of the six cells tested. Also, presynaptic stimulation alone did not induce LTP in any of the five cells tested. 5. These results suggest that the concurrent activation of presynaptic elements with postsynaptic depolarization is necessary for the induction of LTP in visual cortex.  相似文献

This study focused ice slurry formation in an ice storage system using W/O emulsions with 70 and 80% water contents. Emulsions consisted of a silicone oil–water mixture with a small amount of amino-group-modified silicone oil additive. Ice slurry was formed by cooling the emulsion without ice adhesion to the cooling wall, as water in the emulsion did not directly contact the cooling wall. As the structure of W/O emulsion slowed the propagation rate of supercooling dissolution, voltage and ultrasonic wave were applied to the W/O emulsion to propagate dissolution more quickly and decrease maximum supercooling degree, respectively. Thus, the effects of voltage and ultrasonic wave applications on propagation rate were clarified.  相似文献

In the partial oxidation of tar derived from the pyrolysis of cedar wood, the effect of H2S addition was investigated over non-catalyst, steam reforming Ni catalyst, and Rh/CeO2/SiO2 using a fluidized bed reactor. In the non-catalytic gasification, the product distribution was not influenced by the presence of H2S. Steam reforming Ni catalyst was effective for the tar removal without H2S addition, however, the addition of H2S deactivated drastically. In contrast, Rh/CeO2/SiO2 exhibited higher and more stable activity than the Ni catalyst even under the presence of high concentration of H2S (280 ppm). On the Ni catalyst, the adsorption of sulfur was observed by XPS and Ni species was oxidized during the partial oxidation of tar. In the case of Rh/CeO2/SiO2, the adsorption of sulfur was below the detection limit of XPS. This can be related to the self-cleaning of catalyst surface during the circulation in the fluidized bed reactor for the partial oxidation of tar derived from cedar pyrolysis.  相似文献

A compact sodium-cooled reactor is an important candidate as a fast breeder reactor (FBR) and has been investigated in the feasibility study of FBR cycle. Due to the compact sizing of the reactor vessel, gas entrainment at the free surface of sodium coolant becomes one of the significant issues for reactor design, and it is required to clarify the criterion of gas entrainment at free surface and the tolerance. In the present study, some visualization experiments were performed in a water-air system focusing on the gas entrainment due to surface vortex and its transient phenomena. Influences of horizontal velocity were clarified by the visualization. The gas entrainment due to the surface vortex occurs intermittently. Time trends of circulation and length of gas core for the intermittent surface vortices were measured by the particle image velocimetry and visualization. It was found that the gas core length extends with time delay to the increase of circulation around the vortex.  相似文献

Trichothecene mycotoxins such as deoxynivalenol, 4,15-diacetoxyscirpenol, and T-2 toxin, are potent protein synthesis inhibitors for eukaryotic organisms. The 3-O-acetyl derivatives of these toxins were shown to reduce their in vitro activity significantly as assessed by assays using a rabbit reticulocyte translation system. The results suggested that the introduction of an O-acetyl group at the C-3 position in the biosynthetic pathway works as a resistance mechanism for Fusarium species that produce t-type trichothecenes (trichothecenes synthesized via the precursor trichotriol). A gene responsible for the 3-O-acetylation reaction, Tri101, has been successfully cloned from a Fusarium graminearum cDNA library that was designed to be expressed in Schizosaccharomyces pombe. Fission yeast transformants were selected for their ability to grow in the presence of T-2 toxin, and this strategy allowed isolation of 25 resistant clones, all of which contained a cDNA for Tri101. This is the first drug-inactivating O-acetyltransferase gene derived from antibiotic-producing organisms. The open reading frame of Tri101 codes for a polypeptide of 451 amino acid residues, which shows no similarity to any other proteins reported so far. TRI101 from recombinant Escherichia coli catalyzes O-acetylation of the trichothecene ring specifically at the C-3 position in an acetyl-CoA-dependent manner. By using the Tri101 cDNA as a probe, two least overlapping cosmid clones that cover a region of 70 kilobase pairs have been isolated from the genome of F. graminearum. Other trichothecene biosynthetic genes, Tri4, Tri5, and Tri6, were not clustered in the region covered by these cosmid clones. These new cosmid clones are considered to be located in other parts of the large biosynthetic gene cluster and might be useful for the study of trichothecene biosynthesis.  相似文献

Thermotropic liquid-crystalline compounds were applied as membrane materials (membrane solvent and neutral carrier) for neutral carrier-type ion sensors to investigate how the ordered arrangement of neutral carriers affects the property of the resulting ion sensors. Nematic, smectic, and cholesteric liquid-crystalline compounds were used as the membrane solvents and crown ether derivatives with a molecular structure similar to the liquid-crystalline solvent as the K+ neutral carriers. Polarized IR spectroscopy and X-ray diffraction experiments confirmed that the highly ordered arrangement of membrane components was retained in the liquid-crystal-based ion-sensing membranes containing a neutral carrier and a lipophilic salt. The ordered arrangement of neutral carriers in the liquid-crystalline membranes enhanced the ion selectivity significantly, probably due to the efficient cooperation of two adjacent crown ether moieties in the highly ordered and aggregated state.  相似文献

A mullite/SiC whisker/SiC particle multi-composite, having excellent crack-healing ability and mechanical properties, was hot pressed in order to investigate the crack-healing behavior under stress and the resultant fatigue strength at the temperature of healing. A semi-elliptical surface crack 100 μm in surface length was introduced on each specimen. The pre-cracked specimens were crack healed under cyclic or constant stress by using a three-point bending stress at 1473 K, and the resultant bending strength and cyclic fatigue strength were measured at 1473 K. The pre-crack on the surface of the specimens could be healed even under stress. The threshold stresses for crack healing, as determined by evaluating the strengths of crack-healed specimens at a healing temperature of 1473 K, were 170 MPa for both constant and cyclic stresses, corresponding to 77% of the bending strength of the pre-cracked specimens. The static and cyclic fatigue behaviors of crack-healed specimens were also investigated at a healing temperature of 1473 K.  相似文献

We have investigated the hot carrier (HC) reliability of nMOSFETs with an ultrashallow source/drain (S/D) extension, and found that lightly doped drain (LDD)-type HC degradation is accelerated. The lifetime strongly depends on the extension implantation dose or the implantation angle. A reduced overlap region between the gate electrode and drain diffusion seemed to exaggerate the LDD-type HC degradation.
