Making use of insights gained through field research in design can be challenging. Some issues that design teams may face are making sense of fragmented data collected, sharing insight among the design team and presenting the data in ways that support the situated design work. This paper introduces layered scenario mapping, a technique aimed at meeting such issues when designing a ship’s bridge. The technique builds on and expands traditional techniques for representing user data in design and results in a map describing a typical scenario along several dimensions and at different levels of abstraction. It highlights the spatial and temporal aspects of the situation, and emphasises the use of visual presentations. This paper describes why and how the layered scenario mapping technique was created, it critically assesses the technique and discusses experiences with using it. The technique proved to be valuable in making sense of fragmented data, and supported the design team’s collaborative work when designing a ship’s bridge. It is expected that the technique can also prove valuable when designing for other contexts where the spatial and/or temporal dimensions are of importance.  相似文献

Bending oscillations of piezoelectric bimorph beams are effective sound sources in gases or fluids, and, therefore, of practical interest. On the basis of the piezoelectric constitutive relations and the elastodynamic equations, the differential equation of flexural vibrations of thin rectangular piezoelectric heterogeneous bimorph beams, consisting of a piezoelectric layer glued onto an elastic substrate, is derived. The piezoelectric layer is polarized in thickness direction and can be excited to thickness vibrations by an electric alternating current voltage applied to electrodes covering the upper and lower surface of the layer. This causes an oscillating transverse contraction in the piezoelectric layer but not in the substrate, and, thus, generates flexural vibrations of the beam. The differential equation is solved analytically for beams of finite length with both ends free, one clamped and one free end, as well as for both ends clamped. Their vibration behavior in viscous fluids is considered. For a piezo-ceramic composite layer joined to a steel plate vibrating in air and in water, the analytical results are evaluated numerically as function of frequency  相似文献

Peptidylarginine deiminases (PADs) and extracellular vesicles (EVs) may be indicative biomarkers of physiological and pathological status and adaptive responses, including to diseases and disorders of the central nervous system (CNS) and related to hypoxia. While these markers have been studied in hypoxia-intolerant mammals, in vivo investigations in hypoxia-tolerant species are lacking. Naked mole-rats (NMR) are among the most hypoxia-tolerant mammals and are thus a good model organism for understanding natural and beneficial adaptations to hypoxia. Thus, we aimed to reveal CNS related roles for PADs in hypoxia tolerance and identify whether circulating EV signatures may reveal a fingerprint for adaptive whole-body hypoxia responses in this species. We found that following in vivo acute hypoxia, NMR: (1) plasma-EVs were remodelled, (2) whole proteome EV cargo contained more protein hits (including citrullinated proteins) and a higher number of associated KEGG pathways relating to the total proteome of plasma-EVs Also, (3) brains had a trend for elevation in PAD1, PAD3 and PAD6 protein expression, while PAD2 and PAD4 were reduced, while (4) the brain citrullinome had a considerable increase in deiminated protein hits with hypoxia (1222 vs. 852 hits in normoxia). Our findings indicate that circulating EV signatures are modified and proteomic content is reduced in hypoxic conditions in naked mole-rats, including the circulating EV citrullinome, while the brain citrullinome is elevated and modulated in response to hypoxia. This was further reflected in elevation of some PADs in the brain tissue following acute hypoxia treatment. These findings indicate a possible selective role for PAD-isozymes in hypoxia response and tolerance.  相似文献

Although hot water pasteurisation of carcasses is accepted as a general intervention in USA, this is not the case in Europe. The aims of this study were (i) to evaluate the microbiological effects of hot water pasteurisation of lamb carcasses, both after slaughtering and dressing and following subsequent chilling and storage; (ii) to discuss hot water pasteurisation from a public health and cost-benefit perspective; (iii) to discuss the benefits of hot water pasteurisation compared with use of separate meat processing streams for high-risk carcasses; (iv) to evaluate the use of recycled hot water in a hygienic context and in relation to EU regulations; and (v) to consider the technological and sensory aspects of hot water pasteurisation of lamb carcasses. Samples were collected from 420 naturally contaminated lamb carcasses, with 50% of the carcasses (n = 210) subject to hot water pasteurisation at 82 °C for 8 s immediately after slaughter. Surface swab samples from 4500 cm2 areas on carcasses were collected at slaughter, after chilling for 24 h, and after chilling for five days. The microbial analyses included Escherichia coli, Enterobacteriaceae, Bacillus cereus, Clostridium perfringens and aerobic plate count (APC). A resuscitation step using Tryptone Soya Agar was included in the microbiological analyses. Hot water pasteurisation significantly reduced the levels of E. coli, Enterobacteriaceae, B. cereus and APC (all P < 0.001). E. coli colony forming unit (CFU) reduction was 99.5%, corresponding to a reduction of 1.85 log CFU per carcass. E. coli was isolated from 66% of control carcasses and from 26% of pasteurised carcasses. After 24 h storage, the reduction in E. coli was increased to 2.02 log, and after five days E. coli could not be isolated from the pasteurised carcasses. These results suggest that surface pasteurisation could be an important and efficient procedure (critical control point) for reducing generic E. coli and thereby shiga toxin-producing E. coli on carcasses, and thus the risk for disease among consumers. The recycled water had acceptable physical and chemical parameters and no spore-forming bacteria were detected. Although some carcass discolouration was observed, after 24 h the colour was acceptable. Our data provide relevant input for some of the data gaps regarding hot water pasteurisation and indicate that replacing the expensive system of separate processing of high-risk carcasses with hot water surface pasteurisation should be considered as a serious option.  相似文献

Because dietary intake is supposed to be an important route of human exposure we quantified the dietary intake of perfluorooctane sulfonate (PFOS), perfluorooctanoate (PFOA), perfluorohexane sulfonate (PFHxS), perfluorohexanoate (PFHxA), and perfluorooctane sulfonamide (PFOSA) using 214 duplicate diet samples. The study was carried out with a study population of 15 female and 16 male healthy subjects aged 16-45 years. The participants collected daily duplicate diet samples over seven consecutive days in 2005. Duplicate samples were homogenized and their ultrasonic extracts were cleaned up by SPE and subjected to HPLC-ESI-MS/MS. In addition, individual intakes were estimated based on blood levels of PFOS and PFOA using a pharmacokinetic model. Blood samples were collected once during the sampling period. The median (90th percentile) daily dietary intake of PFOS and PFOA was 1.4 ng/kg b.w. (3.8 ng/kg b.w.) and 2.9 ng/kg b.w. (8.4 ng/kg b.w.), respectively. PFHxS and PFHxA could be detected only in some samples above detection limit with median (maximum) daily intakes of 2.0 ng/kg b.w. (4.0 ng/kg b.w.) and 4.3 ng/kg b.w. (9.2 ng/kg b.w.), respectively. Because PFOSA could not be detected above the limit of detection of 0.2 ng/g f.w. this indirect route of exposure seems to be of less significance. Overall, the results of this study demonstrate that the German population is exposed to PFOS and PFOA, but the median dietary intake did not reach the recommended tolerable daily intake by far. Biomonitoring data predict an exposure in a comparable range. We suppose that, normally, food intake is the main source of exposure of the general population to PFOS and PFOA.  相似文献

The evolving debate on "urban shrinkage" mirrors an increasing interest in demographic phenomena on the part of urban scholars. This paper discusses ambiguous evidence about recent population decline in the large cities of Poland and the Czech Republic, with a particular focus on ?ódz and Brno in general and their inner cities more specifically. By applying a mixed-method approach, the paper identifies indications of inner-city repopulation and socio-demographic diversification which are not yet apparent in register or census data. It is argued that there are indications of a silent transformation of traditional residential patterns and neighbourhoods in east central Europe. In the inner cities, this is reflected, amongst other things, by the presence of new households that may be called "transitory urbanites".  相似文献

Platelets and lipoproteins play a crucial role in atherogenesis, in part by their ability to modulate inflammation and oxidative stress. While oxidized low density lipoproteins (OxLDL) play a central role in the development of this disease, high density lipoproteins (HDL) represent an atheroprotective factor of utmost importance. As platelet function is remarkably sensitive to the influence of plasma lipoproteins, it was the aim of this study to clarify if HDL are able to counteract the stimulating effects of OxLDL with special emphasis on aspects of platelet function that are relevant to inflammation. Therefore, HDL were tested for their ability to interfere with pro-thrombotic and pro-inflammatory aspects of platelet function. We are able to show that HDL significantly impaired OxLDL-induced platelet aggregation and adhesion. In gel-filtered platelets, HDL decreased both the formation of reactive oxygen species and CD40L expression. Furthermore, HDL strongly interfered with OxLDL-induced formation of platelet-neutrophil aggregates in whole blood, suggesting that platelets represent a relevant and sensitive target for HDL. The finding that HDL effectively competed with the binding of OxLDL to the platelet surface might contribute to their atheroprotective and antithrombotic properties.  相似文献

The continuously operated laboratory scale Kaldnes moving bed biofilm reactor (MBBR) was used for thermophilic (55 degrees C) aerobic treatment of TMP whitewater. In the MBBR, the biomass is grown on carrier elements that move along with the water in the reactor. Inoculation with mesophilic activated sludge gave 60-65% SCOD removal from the first day onwards. During the 107 days of experiment, the 60-65% SCOD removals were achieved at organic loading rates of 2.5-3.5 kg SCODm(-3) d(-1), the highest loading rates applied during the run and HRT of 13-22h. Carbohydrates, which contributed to 50-60% of the influent SCOD. were removed by 90-95%, while less than 15% of the lignin-like material (30-35% of SCODin) was removed. The sludge yield was 0.23g VSSg SCOD(-1)removed. The results show that the aerobic biofilm process can be successfully operated under thermophilic conditions.  相似文献
