Yacon (Smallanthus sonchifolius), a native Andean plant, has been cultivated as a crop and locally used as a traditional folk medicine for the people suffering from diabetes and digestive/renal disorders. However, the medicinal properties of this plant and its processed foods have not been completely established. This study investigates the potent antioxidative effects of herbal tea leaves from yacon in different free radical models and a ferric reducing model. A hot‐water extract exhibited the highest yield of total polyphenol and scavenging effect on 1,1‐diphenyl‐2‐picryl hydrazyl (DPPH) radical among four extracts prepared with hot water, methanol, ethanol, and ethylacetate. In addition, a higher reducing power of the hot‐water extract was similarly demonstrated among these extracts. Varying concentrations of the hot‐water extract resulted in different scavenging activities in four synthetic free radical models: DPPH radical (EC50 28.1 μg/mL), 2,2′‐azinobis(3‐ethylbenzothiazoline‐6‐sulfonic acid) cation radical (EC50 23.7 μg/mL), galvinoxyl radical (EC50 3.06 μg/mL), and chlorpromazine cation radical (EC50 475 μg/mL). The yacon tea‐leaf extract further demonstrated superoxide anion (O2?) radical scavenging effects in the phenazine methosulfate‐NADH‐nitroblue tetrazolium (EC50 64.5 μg/mL) and xanthine oxidase assay systems (EC50 20.7 μg/mL). Subsequently, incubating human neutrophilic cells in the presence of the tea‐leaf extract could suppress the cellular O2? radical generation (IC50 65.7 μg/mL) in a phorbol 12‐myristate 13‐acetate‐activated cell model. These results support yacon tea leaves may be a good source of natural antioxidants for preventing O2? radical‐mediated disorders.  相似文献

An overview is given of previously-proposed measurement-based connection admission control (CAC) methods. First we address requirements for CAC methods, and then provide a taxonomy for CAC methods. Measurement-based CAC methods are discussed in detail, classified according to the taxonomy, and compared against each other with respect to the requirements. We conclude that measurement-based CAC methods based on effective bandwidth and bufferless models are promising because they do not require complex hardware and are less dependent on assumptions regarding traffic than methods in which the effect of buffer is considered.  相似文献

Machine learning models have been developed to perform damage detection using images to improve bridge inspection efficiency. However, in damage detection using images alone, the 3D coordinates of the damage cannot be recorded. Furthermore, the accuracy of the detection depends on the quality of the images. This paper proposes a method to integrate and record the damage detected from multiple images into a 3D model using deep learning to detect the damage from bridge images and structure from motion to identify the shooting position. The proposed method reduces the variability of the detection results between images and can assess the scale of damage or, conversely, where there is no damage and the extent of inspection omissions. The proposed method has been applied to a real bridge, and it has been shown that the actual damage locations can be recorded as a 3D model.  相似文献

Direct reprogramming of cardiac fibroblasts to induced cardiomyocytes (iCMs) is a promising approach to cardiac regeneration. However, the low yield of reprogrammed cells and the underlying epigenetic barriers limit its potential. Epigenetic control of gene regulation is a primary factor in maintaining cellular identities. For instance, DNA methylation controls cell differentiation in adults, establishing that epigenetic factors are crucial for sustaining altered gene expression patterns with subsequent rounds of cell division. This study attempts to demonstrate that 5′AZA and miR-133a encapsulated in PLGA-PEI nanocarriers induce direct epigenetic reprogramming of cardiac fibroblasts to cardiomyocyte-like cells. The results present a cardiomyocyte-like phenotype following seven days of the co-delivery of 5′AZA and miR-133a nanoformulation into human cardiac fibroblasts. Further evaluation of the global DNA methylation showed a decreased global 5-methylcytosine (5-medCyd) levels in the 5′AZA and 5′AZA/miR-133a treatment group compared to the untreated group and cells with void nanocarriers. These results suggest that the co-delivery of 5′AZA and miR-133a nanoformulation can induce the direct reprogramming of cardiac fibroblasts to cardiomyocyte-like cells in-vitro, in addition to demonstrating the influence of miR-133a and 5′AZA as epigenetic regulators in dictating cell fate.  相似文献

Polylactide (PLA)-grafted polysaccharides with various lengths and numbers of graft chains were synthesized using a trimethylsilyl protection method. The properties of the cast films prepared from graft-copolymers were investigated through thermal and dynamic mechanical analyses. The graft-copolymer films exhibited a lower glass transition temperature (Tg), melting temperature, and crystallinity, and higher viscosity properties compared to PLA films. Moreover, the usefulness of graft-copolymer as a plasticizer was investigated with 1:4 blend films prepared from the graft-copolymers and PLA. The blend films showed lower Tg and crystallinity, and higher viscosity properties compared to PLA films.  相似文献

The effect of polyethyleneglycol on the radical polymerization of methyl methacrylate initiated with an aqueous solution of sodium polystyrenesulfonate was studied. Under the definite condition, the conversion of methyl methacrylate raised from 6.6 to 100% by the addition of polyethyleneglycol. It was concluded that polyethyleneglycol acted only as a host of Na+, but not as a phase transfer catalyst.  相似文献

Leukocytes comprise less than 1% of all blood cells. Enrichment of their number, starting from a sample of whole blood, is the required first step of many clinical and basic research assays. We created a microfluidic device that takes advantage of the intrinsic features of blood flow in the microcirculation, such as plasma skimming and leukocyte margination, to separate leukocytes directly from whole blood. It consists of a simple network of rectangular microchannels designed to enhance lateral migration of leukocytes and their subsequent extraction from the erythrocyte-depleted region near the sidewalls. A single pass through the device produces a 34-fold enrichment of the leukocyte-to-erythrocyte ratio. It operates on microliter samples of whole blood, provides positive, continuous flow selection of leukocytes, and requires neither preliminary labeling of cells nor input of energy (except for a small pressure gradient to support the flow of blood). This effortless, efficient, and inexpensive technology can be used as a lab-on-a-chip component for initial whole blood sample preparation. Its integration into microanalytical devices that require leukocyte enrichment will enable accelerated transition of these devices into the field for point-of-care clinical testing.  相似文献

BACKGROUND: Relationships between buckwheat (Fagopyrum esculentum Moench) flour lipase, lipoxygenase and peroxidase activity, along with levels of individual free fatty acids (FFAs) and levels of headspace volatile compounds of boiled buckwheat noodles, were investigated for 12 different buckwheat varieties. Enzyme activities and FFA levels in flour were correlated with their respective varietal arrays of boiled noodle headspace volatile compounds, measured by gas chromatography–mass spectrometry. RESULTS: The volatiles hexanal, tentative butanal, tentative 3‐methylbutanal and tentative 2‐methylbutanal showed significant positive correlation with one another, indicating that they may be generated through similar mechanisms. These important volatile components of buckwheat flavor were also positively correlated with lipase and/or peroxidase activity, indicating that enzymatic reactions are important in flavor generation in boiled buckwheat noodles. On the other hand, pentanal, which showed no significant correlation with any enzyme activity, showed a significant positive correlation to the levels of C18:2 and C18:3 FFAs, suggesting the existence of a ‘non‐enzymatic’ and/or ‘uncertain enzymatic pathway’ for flavor generation in boiled buckwheat noodles. CONCLUSION: Lipase and peroxidase in buckwheat flour are important for flavor generation of boiled buckwheat noodles. Pitting corrosion is typical corrosion observed on coated hold frames in way of cargo holds of bulk carriers, which exclusively carry coal and iron ore. Extensive survey on the effect of pitting corrosion on structural strength under a wide variety of loading conditions is necessary to clarify the relationship between pitting intensity and residual strength in detail. In the present study, a series of tests has been conducted on structural models which consist of web, shell and face plates to investigate the effect of pitting corrosion on strength of web plates subjected to patch loading. In these tests, artificial pitting was made on the web plates and two equal concentrated loads have been applied vertically at the one third points of simply supported models. It was found that web crippling behavior is strongly affected by the pit distribution on the web plates. According to the FE-analyses following the experiment, ultimate strength of the web plates with pitting under patch loading is a little smaller than or almost the same as that of the web plates with uniform corrosion in terms of average thickness loss.  相似文献
