BACKGROUND: The apical potassium (K+) channels mediate K+ recycling in thick ascending limb (TAL) and K+ secretion in cortical collecting duct (CCD). Recently, the cDNAs for a family of renal K+ channels, ROMK1, -2 and -3, were identified. Based on the biophysical properties and mRNA distribution, it is believed that these ROMK cDNAs encode the apical K+ channels of TAL and CCD. However, the information for cellular and subcellular localization of the ROMK proteins in these tubules is still not available. METHODS: Paraffin or frozen kidney sections from adult Sprague-Dawley rats were stained by polyclonal antibodies against the N- and C-terminal domain of ROMK. Immunoreactive staining was visualized by color development from horseradish peroxidase reaction. Membrane homogenates from kidney were analyzed by Western blot analysis. RESULTS: The polyclonal antibodies against cytoplasmic epitope of ROMK recognized a approximately 42 kD protein in the membrane homogenates from kidney, but not from liver. Staining by immunocytochemistry revealed that ROMK channels were localized to the apical membranes of the distal nephron in cortex and outer medulla, including thick ascending limb and collecting tubule. ROMK staining was absent in glomerulus, proximal tubule and inner medulla. Double staining of the tissue section with both ROMK-specific and H+-ATPase-specific antibodies revealed labeling of ROMK in the principal cells of the collecting tubules. CONCLUSIONS: These results further strengthen the idea that ROMK channels play important roles in the recycling of K+ in TAL and the secretion of K+ in CCD. 相似文献

Recently, air-abrasion technology has been examined for potential applications within dentistry, including the field of orthodontics. The purpose of this study was to compare the traditional acid-etch technique with an air-abrasion surface preparation technique, with two different sizes of abrading particles. The following parameters were evaluated: (a) shear bond strength, (b) bond failure location, and (c) enamel surface preparation, as viewed through a scanning electron microscope. Sixty extracted human third molars were pumiced and divided into three groups of 20. The first group was etched with a 37% phosphoric acid gel for 30 seconds, rinsed for 30 seconds, and dried for 20 seconds. The second and third groups were air-abraded with (a) a 50 microm particle and (b) a 90 microm particle of aluminum oxide, with the Micro-etcher microabrasion machine (Danville Engineering Inc.). All three groups had molar stainless steel orthodontic brackets bonded to the buccal surface of each tooth with Transbond XT bonding system (3M Unitek). A Zwick Universal Testing Machine (Calitek Corp.) was used to determine shear bond strengths. The analysis of variance was used to compare the three groups. The Adhesive Remnant Index (ARI) was used to evaluate the residual adhesive on the enamel after bracket removal. The chi square test was used to evaluate differences in the ARI scores among the groups. The significance for all tests was predetermined at p < or = 0.05. The results indicated that there was a significant difference in shear bond strength among the three groups (p = 0.0001). The Duncan Multiple Range test showed a significant decrease in shear bond strength in the air-abraded groups. The chi square test revealed significant differences among the ARI scores of the acid-etched group and the air-abraded groups (chi(2) = 0.0001), indicating no adhesive remained on the enamel surface after debonding when air-abrasion was used. In conclusion, the current findings indicate that enamel surface preparation using air-abrasion results in a significant lower bond strength and should not be advocated for routine clinical use as an enamel conditioner at this time. 相似文献

Constructing communication protocols from component service specifications, each of which specifies a subfunction of the target protocol, enables efficient development of a large and complex communication protocol. Concerning this construction, related techniques have been already proposed: integration of component protocol specifications into a single protocol specification and transformation of service specifications to protocol specifications. However, the integration needs special knowledge of communication protocols, and the transformation requires that a large and complex service specification should be developed as input to produce the target protocol. In order to cope with these problems, this paper proposes a new method which at first integrates component service specifications into a single service specification, and then transforms the service specification into the target protocol by a protocol synthesis technique. The most important point of view is that component integration is performed at the service specification level rather than the protocol specification level. Additionally, we define a class of ‘well-formed’ service specification which ensures correctness of the target protocol. As a result, the integration and transformation can be efficiently executed in small state space without special knowledge of communication protocols. Finally, we have shown the effectiveness of the proposed method by constructing a part of the real-life OSI protocol FTAM. 相似文献

The electrochemical reduction of CO2 on a Cu electrode was investigated in aqueous NaHCO3 solution, at low temperature. A divided H-type cell was employed, the catholyte was 0.65 mol dm−3 NaHCO3 aqueous solution and the anolyte was 1.1 mol dm−3 KHCO3 aqueous solution. The temperature during the electrolysis of CO2 was decreased stepwise to 271 K. Methane and formic acid were obtained as the main products. The maximum Faradaic efficiency of methane was 46% at −2.0 V and 271 K. The efficiency of hydrogen formation, a competing reaction of CO2 reduction, was significantly depressed with decreasing temperature. Based on the results of this work, the proposed electrochemical method appears to be a viable means for removing CO2 from the atmosphere and converting it into more valuable chemicals. The synthesis of methane by the electrochemical method might be of practical interest for fuel production and the storage of solar energy. 相似文献

A toroidal simulator is used to study the magnetomechanical behavior of a superconducting D-shaped coil set at the center between sectored ferromagnetic plates. Since the magnetic field distribution between the two plates is expected to be almost the same as that created by toroidal field coils arrayed in a torus, the toroidal simulator may be expected to show almost the same magnetomechanical behavior as that of the full torus coils. To verify its validity experimentally, buckling and vibration tests were carried out in a liquid helium cryostat. In addition numerical analyses were performed for the magnetic field distribution to reveal its similarity and for the experimental results from the toroidal simulator to confirm its validity. The thermodynamic properties (solubility, vapour pressure, density, viscosity, heat capacity and heat of mixing) of the H2O + CH3OH + LiBr + ZnCl2 (9:1 H2O:CH3OH and 1:1 LiBr:ZnCl2 by mass) system using H2O + CH3OH as the working media and LiBr + ZnCl2 as the absorbents were measured. The solubility data were obtained in the temperature range from 270.35 to 389.55 K. The measurements of vapour pressure, density, viscosity and heat capacity were carried out at various temperatures and absorbent concentrations. The differential heat of dilution and differential heat of solution at 298.15 K were measured for solutionw with absorbent concentrations from 0 to 75.2 wt%. The integral heat of mixing data at 298.15 K were obtained from both sets of experimental data. The integral heats of mixing for this quaternary system showed exothermic behaviour. The vapour pressure data were correlated with an Antoine-type equation. An empirical formula for the heat capacity was obtained from experimental data. The experimental data for the basic thermodynamic properties of this quaternary system were compared with those of the basic H2O + LiBr system. 相似文献

The design and fabrication of a GaAs varactor diode for u.h.f. t.v. tuners were carried out using a GaAs double epitaxial layer. The capacitance ratio of diode. C3/C25, is 6.0 and the series resistance at 470 MHz is 0.24 ?. These characteristics are much better than those of an Si diode and well satisfy the requirements for t.v. tuners. 相似文献
