Enamel matrix derivative (EMD) is widely used in periodontal tissue regeneration therapy. However, because the bioactivity of EMD varies from batch to batch, and the use of a synthetic peptide could avoid use from an animal source, a completely synthetic peptide (SP) containing the active component of EMD would be useful. In this study an oligopeptide synthesized derived from EMD was evaluated for whether it contributes to periodontal tissue regeneration. We investigated the effects of the SP on cell proliferation and osteoblast differentiation of human mesenchymal stem cells (MSCs), which are involved in tissue regeneration. MSCs were treated with SP (0 to 1000 ng/mL), to determine the optimal concentration. We examined the effects of SP on cell proliferation and osteoblastic differentiation indicators such as alkaline phosphatase activity, the production of procollagen type 1 C-peptide and osteocalcin, and on mineralization. Additionally, we investigated the role of extracellular signal-related kinases (ERK) in cell proliferation and osteoblastic differentiation induced by SP. Our results suggest that SP promotes these processes in human MSCs, and that ERK inhibitors suppress these effects. In conclusion, SP promotes cell proliferation and osteoblastic differentiation of human MSCs, probably through the ERK pathway.  相似文献

We have recently developed a new model and measurement technique for the dynamics of conversion and shrinkage during the photopolymerization of multi-acrylates. The model development and the application to different UV intensities were investigated in this paper. In this new model, instead of assigning the excess volume entirely as excess free volume, it is partitioned into both free and occupied volume components. The new measurement technique allowed us to collect real-time conversion and shrinkage data simultaneously and synchronously to verify our new model. When utilized on a broad range of incident UV intensities (3, 16 and 130 mW/cm2), there was considerable success for predicting conversion for all the cases, but the result is less satisfactory for shrinkage prediction at the highest UV intensity. Possible explanations of this discrepancy are discussed.  相似文献

Ductile regime machining is an alternative method for polishing of brittle materials to obtain a high quality surface finish by a ductile or plastic material removal process. Hence, there is a growing interest to study ductile regime machining over several decades. This paper reviews current state of research and development in ductile regime machining. In this study, experiments were carried out to establish the effect of tool rake angle on the performance of single crystal diamond tools in micro-grooving of electroless nickel plated molding dies. Diamond tools with rake angles of 00,+50 and ?50 were used in the study, and it was found that the diamond tool with 00 rake angle has superior performance in terms of tool wear, cutting forces, and machined surface roughness. For a cutting distance of up to 11.69 km, the 00 rake tool machined satisfactorily without any sign of tool wear while the tools with +50 and ?50 rake angles suffered from progressive wear with a corresponding increase in cutting forces. However, it was noted that despite the increase in wear on tools with +50 and ?50 rake angles, and cutting forces on the diamond tools with all three different rake angles did not significantly affect the surface roughness. The effects of various cutting parameters such as spindle speed and infeed rate on the cutting forces and surface roughness are also presented in the study. It was observed that cutting forces increase proportionately with spindle speed and infeed rate without any significant variation of surface roughness. Surface quality of up to 3 nm Ra was achieved during micro-grooving of electroless nickel.  相似文献

Neuropsin (kallikrein-related peptidase 8) is concentrated in the hippocampus, amygdala, olfactory bulb, and prefrontal cortex. Earlier studies showed that protease deficiency causes a significant impairment of early-phase long-term potentiation in the Schaffer collateral pathway and hippocampus-dependent memory in the Y maze and Morris water maze (Z. Chen et al., 1995; A. Hirata et al., 2001; H. Tamura et al., 2006). In addition to neuropsin's participation in the hippocampal memory, amygdalar and cortical localization of the gene suggests extrahippocampal behavioral function, and the authors therefore examined neuropsin-deficient mice, including tests of sensory motor reflex, open field, light-dark transition, Rota-Rod, elevated plus-maze, hot plate, startle response-prepulse inhibition, Porsolt forced swim, Barnes maze, eight-arm radial maze, and contextual and cued fear conditioning tests. Here, the authors found increased anxiety in neuropsin-deficient mice, suggesting the involvement of this protease in emotional responses. Though the k-nearest neighbor (k-NN) pattern classifier is an effective learning algorithm, it can result in large model sizes. To compensate, a number of variant algorithms have been developed that condense the model size of the k-NN classifier at the expense of accuracy. To increase the accuracy of these condensed models, we present a direct boosting algorithm for the k-NN classifier that creates an ensemble of models with locally modified distance weighting. An empirical study conducted on 10 standard databases from the UCI repository shows that this new Boosted k-NN algorithm has increased generalization accuracy in the majority of the datasets and never performs worse than standard k-NN.  相似文献

In this paper, we focus on the microscopic structure of the liquid-vapor interface and the effect of impurities such as surfactants or electrolytes in the aqueous solution on interfacial properties. We perform molecular dynamics (MD) simulations of a simple planar interface system and a bubble system in which a nanometer-size void region is maintained. In these MD systems, some static and dynamic properties of molecules near the interface are investigated and we elucidate some microscopic features of the interface. Especially, we show the difference of the dynamics of water molecules between the surfactant and electrolyte solutions. We also consider the definition of the interface at the molecular level which can treat the local and instantaneous structure, and discuss the physical properties which we calculate including the effect of molecular fluctuations of the interface.  相似文献
