Reconstitution of tissue architecture in vitro is important because it enables researchers to investigate the interactions and mutual relationships between cells and cellular signals involved in the three-dimensional (3D) construction of tissues. To date, in vitro methods for producing tissues with highly ordered structure and high levels of function have met with limited success although a variety of 3D culture systems have been investigated. In this study, we reconstituted functional hepatic tissue including mature hepatocyte and blood vessel-like structures accompanied with bile duct-like structures from E15.5 fetal liver cells, which contained more hepatic stem/progenitor cells comparing with neonatal liver cells. The culture was performed in a simulated microgravity environment produced by a rotating wall vessel (RWV) bioreactor. The hepatocytes in the reconstituted 3D tissue were found to be capable of producing albumin and storing glycogen. Additionally, bile canaliculi between hepatocytes, characteristics of adult hepatocyte in vivo were also formed. Apart from this, bile duct structure secreting mucin was shown to form complicated tubular branches. Furthermore, gene expression analysis by semi-quantitative RT-PCR revealed the elevated levels of mature hepatocyte markers as well as genes with the hepatic function. With RWV culture system, we could produce functionally reconstituted liver tissue and this might be useful in pharmaceutical industry including drug screening and testing and other applications such as an alternative approach to experimental animals.  相似文献

ABSTRACT

Heat treatment, such as quenching and carburising, often involves volume change caused by phenomena such as thermal expansion, phase transformation, and carbide precipitation during tempering. During the tempering process, an external force induces additional plastic deformation. The authors termed this phenomenon ‘tempering plasticity’. In this study, we performed crystal plasticity analysis using fast Fourier transform considering the volume change in carbides to assess the mechanism of tempering plasticity. As a result, tempering plastic strain occurred as the volume fraction of carbide increased, and the tempering plastic phenomenon could be reproduced based on the transformation model proposed by Greenwood–Johnson. The result supports the idea that the volume change accompanied by carbide precipitation is an important mechanism that invokes tempering plasticity.  相似文献

A new method is proposed in this paper to determine the permittivity of materials. The general consideration is to use only the amplitudes of the transmission and reflection coefficients to do the permittivity determination. According to the analysis, the permittivity can be uniquely determined by measuring these two amplitudes when a sample is prepared with large enough attenuation that the multiple reflections between the two surfaces of the sample can be neglected. The validity of the method was proven by experiments. Using the method, the instantaneous and noncontacting measurements of the amplitudes can be realized. Thus, the dynamic measurement of the permittivity becomes possible  相似文献

In order to develop life assessment techniques for aged components made of modified 9Cr–1Mo steel, specimens were artificially deteriorated by aging, creep and fatigue tests at elevated temperatures, and associated changes in the microstructure and mechanical properties were examined. It was observed that aging resulted in formation of Laves phase causing a decrease in toughness. The creep damage in base metal could be correlated with decrease in hardness, while creep damage in weldments could be correlated with the area fraction and density of creep voids. Creep rupture in weldments occurred in the fine-grained heat affected zone by the formation and growth of creep voids. The fatigue damage in base metal correlated to the maximum length of a crack among micro-cracks initiated during fatigue cycles.  相似文献

A BiCMOS logic circuit with very small input capacitance has been developed, which operates at low supply voltages. A High-beta BiCMOS (Hβ-BiCMOS) gate circuit which fully utilizes the bipolar transistor features achieves 10 times the speed of a CMOS gate circuit with the same input capacitance and operating at 3.3 V supply voltage. In order to lower the minimum supply voltage of Hβ-BiCMOS, a BiCMOS circuit configuration using a charge pump to pull up the output high level of the BiCMOS gate circuit is proposed. By introducing a BiCMOS charge pump, Hβ-BiCMOS achieves very high speed operation at sub-2.0 V supply voltage. It has also been demonstrated that only a very small number of charge pump circuits are required to drive a large number of Hβ-BiCMOS gate circuits  相似文献

A method of measurement of solids holdup in a three-phase reactor by analyzing the shape and the phase lag or lead of an ultrasonic wave has been proposed. The solids holdup in a solids-dilute three-phase fluidized bed could be measured without being affected by the presence of gas bubbles. The experimental values of solids axial dispersivity measured by the ultrasonic technique are in reasonable agreement with an empirical correlation of previous data obtained by other methods. The possibility of the application of the ultrasonic technique to the measurement of solids holdup in a three-phase fluidized bed has been suggested.  相似文献

A tangential Fast Ion Charge eXchange Spectroscopy is newly applied on a Large Helical Device (LHD) for co/countercirculating fast ions, which are produced by high energy tangential negative-ion based neutral beam injection. With this new observation geometry, both the tangential-neutral beam (NB) and a low-energy radial-NB based on positive ions can be utilized as probe beams of the measurement. We have successfully observed Doppler-shifted H-alpha lights due to the charge exchange process between the probing NB and circulating hydrogen ions of around 100 keV in LHD plasmas.  相似文献

Ground water contaminated with trichloroethylene (TCE) used in electronic, electric, dry cleaning and the like industries is often treated by air-stripping. In this treatment process, TCE in its vapor form is stripped from ground water by air stream and sometimes emitted into the atmosphere without any additional treatments. Activated carbon adsorption is one of the practical and useful processes for recovering the TCE vapor from the exhaust air stream. However, adsorption of the TCE vapor from the stripping air stream onto activated carbons is not so simple as that from dry air, since in the exhaust air stream the TCE vapor coexists with water vapor with relatively high concentrations. The understanding of the adsorption characteristics of the TCE vapor to be adsorbed on activated carbon in the water vapor-coexisting system is essential for successfully designing and operating the TCE recovery process. In this work, the adsorption equilibrium relations of the TCE vapor adsorption on activated carbons were elucidated as a function of various relative humidity. Activated carbon fibers (ACFs) were used as model activated carbon. The adsorption equilibrium relations were studied by the column adsorption method. The adsorption isotherms of TCE vapor adsorbed on sample ACFs were successfully correlated by the Dubinin-Radushkevich equation for both cases with and without coexistent water vapor. No effects of coexistent water vapor were found on the limiting adsorption volume. However, the adsorption characteristic energy was significantly reduced by the coexistence of water vapor and its reduction was successfully correlated with the equilibrium amount of water vapor adsorbed under the dynamic condition.  相似文献

Tomakomai Large Scale Crude Oil Fire Experiments   总被引:6,自引:0,他引:6  
This paper summarizes the results of large-scale crude oil fire experiments conducted in Tomakomai, Japan, in 1998 to obtain information that could be applied to the development of firefighting strategies for, and the design of, huge petroleum storages. Arabian light-equivalent crude oil was burned in pans 5-, 10-, and 20-m in diameter. Most of the experiments were performed under favorable conditions. Measured data include external radiation, infrared image of the flame, flame temperature, gas concentration inside the flame, and other burning characteristics. The height of the strongest radiant emittance was H/D=0.1 to 0.2, where D=pan diameter and H=height from the initial fuel surface, and a kind of fireball appeared occasionally at the intermittant flame zone. Emitted smoke particles were sampled on the ground and observed with a scanning electron micrograph, and the distribution of the diameters of primary smoke particles was examined. The average diameter of primary smoke particles is 53.0 (±10.5)nm. The dependence of burning characteristics and flame structure on pan diameter is discussed. The flame height of the 20-m diameter pan fire is 1.9 (±0.3) D. The burning rate increases as the pan diameters increase, but the radiative fraction decreases as pan diameter increases.  相似文献
